OBJECTIVES/GOALS: Supported by the State of Alabama, the Alabama Genomic Health Initiative (AGHI) is aimed at preventing and treating common conditions with a genetic basis. This joint UAB Medicine-HudsonAlpha Institute for Biotechnology effort provides genomic testing, interpretation, and counseling free of charge to residents in each of Alabama’s 67 counties. METHODS/STUDY POPULATION: Launched in 2017, as a state-wide population cohort, AGHI (1.0) enrolled 6,331 Alabamians and returned individual risk of disease(s) related to the ACMG SF v2.0 medically actionable genes. In 2021, the cohort was expanded to include a primary care cohort. AGHI (2.0) has enrolled 750 primary care patients, returning individual risk of disease(s) related to the ACMG SF v3.1 gene list and pre-emptive pharmacogenetics (PGx) to guide medication therapy. Genotyping is done on the Illumina Global Diversity Array with Sanger sequencing to confirm likely pathogenic / pathogenic variants in medically actionable genes and CYP2D6 copy number variants using Taqman assays, resulting in a CLIA-grade report. Disease risk results are returned by genetic counselors and Pharmacogenetics results are returned by Pharmacists. RESULTS/ANTICIPATED RESULTS: We have engaged a statewide community (>7000 participants), returning 94 disease risk genetic reports and 500 PGx reports. Disease risk reports include increased predisposition to cancers (n=38), cardiac diseases (n=33), metabolic (n=12), other (n=11). 100% of participants harbor an actionable PGx variant, 70% are on medication with PGx guidance, 48% harbor PGx variants and are taking medications affected. In 10% of participants, pharmacists sent an active alert to the provider to consider/ recommend alternative medication. Most commonly impacted medications included antidepressants, NSAIDS, proton-pump inhibitors and tramadol. To enable the EMR integration of genomic information, we have developed an automated transfer of reports into the EMR with Genetics Reports and PGx reports viewable in Cerner. DISCUSSION/SIGNIFICANCE: We share our experience on pre-emptive implementation of genetic risk and pharmacogenetic actionability at a population and clinic level. Both patients and providers are actively engaged, providing feedback to refine the return of results. Real time alerts with guidance at the time of prescription are needed to ensure future actionability and value.
Using 3D Models to Understand the Changing Role of Fluting in Paleoindian Point Technology from Clovis to Dalton

Fluting is a technological and morphological hallmark of some of the most iconic North American Paleoindian stone points. Through decades of detailed artifact analyses and replication experiments, archaeologists have spent considerable effort reconstructing how flute removals were achieved, and they have explored possible explanations of why fluting was such an important aspect of early point technologies. However, the end of fluting has been less thoroughly researched. In southern North America, fluting is recognized as a diagnostic characteristic of Clovis points dating to approximately 13,000 cal yr BP, the earliest widespread use of fluting. One thousand years later, fluting occurs more variably in Dalton and is no longer useful as a diagnostic indicator. How did fluting change, and why did point makers eventually abandon fluting? In this article, we use traditional 2D measurements, geometric morphometric (GM) analysis of 3D models, and 2D GM of flute cross sections to compare Clovis and Dalton point flute and basal morphologies. The significant differences observed show that fluting in Clovis was highly standardized, suggesting that fluting may have functioned to improve projectile durability. Because Dalton points were used increasingly as knives and other types of tools, maximizing projectile functionality became less important. We propose that fluting in Dalton is a vestigial technological trait retained beyond its original functional usefulness.
Recovery of Transmembrane Potentials in Plants Resistant to Aryloxyphenoxypropanoate Herbicides: A Phenomenon Awaiting Explanation

Aryloxyphenoxypropanoate (APP) herbicides, such as diclofop, depolarize membranes in parenchyma cells of coleoptiles and root tips, and isolated tonoplast or plasma membrane vesicles from a variety of plant species. Some APP-resistant biotypes of rigid ryegrass and wild oat repolarize membranes after removal of herbicide from a bathing medium. The repolarization ability does not require presence of either APP-insensitive acetyl coenzyme A carboxylase or an increased capacity for herbicide detoxification. The kinetics of depolarization and repolarization depend upon the herbicide, the herbicide concentration, the biotype, and the pH of the bathing solution. For rigid ryegrass, depolarization in the presence of diclofop acid is more rapid than in the presence of diclofop-methyl, and 50% depolarization required about 4 μM diclofop acid. Both the nonherbicidal S(–) and the herbicidal R(+) enantiomers of diclofop acid depolarized membranes in susceptible and resistant ryegrass. Susceptible biotypes regenerated transmembrane potentials following removal of the S(–) but not the R(+) enantiomer, whereas resistant biotypes repolarized following exposure to either enantiomer or a mixture of the two. The herbicide 2,4-D affected, in a complex manner, the ability of both susceptible and resistant ryegrass biotypes to depolarize and repolarize. It is postulated that the intracellular concentration of diclofop acid in susceptible and resistant plants is not the same due to differences in the partitioning of diclofop acid between the extracellular spaces and the cytoplasm. The mechanism producing the postulated difference is unknown, but observations on the proton extrusion capacity of both ryegrass and wild oats, the responses of ryegrass to [K+] and PCMBS, and the single-gene inheritance pattern of resistance in wild oats indicate that changes in the diclofop sensitivity of a plasma membrane protein involved in the generation of proton or ion gradients may be involved.

Prevalence of overweight, obesity and underweight in Western Australian school-aged children; 2008 compared with 2003

Objective
Due to rising rates of obesity globally, the present study aimed to examine differences in overweight and underweight prevalence in Western Australian schoolchildren in 2008 compared with 2003.
DesignCross-sectional study at two time points; using two-stage stratified sampling, primary and secondary schools in both metropolitan and non-metropolitan Western Australia; sample selected was representative of the State's population figures.
SettingsSeventeen primary and thirteen secondary (2008) and nineteen primary and seventeen secondary (2003) schools. Government and non-government funded schools in metropolitan and non-metropolitan (regional/rural) Western Australia were recruited.
SubjectsHeight and weight were measured for 1708 (961 primary and 747 secondary) students in 2008 and 1694 (876 primary and 817 secondary) students in 2003.
ResultsOverweight and obesity prevalence in primary students was similar in 2008 (22·9 %) to 2003 (23·2 %; P > 0·05). In secondary girls overweight and obesity prevalence dropped from 23·1 % (2003) to 15·9 % (2008; P = 0·002). Secondary boys showed a slight decrease in overweight and obesity prevalence; however, this was not statistically significant (P = 0·102). Higher proportions of underweight in primary girls were observed in 2008 (9·9 %) compared with 2003 (4·2 %; P < 0·001) and in secondary girls in 2008 (9·4 %) compared with 2003 (5·5 %; P < 0·001).
ConclusionsPrevalence of overweight and obesity in Western Australian primary students was stable; however, it declined in secondary students. Both primary and secondary girls showed an increase in underweight prevalence. Public health interventions are needed for the high percentage of youth still overweight, whereas the observed increase in underweight girls warrants attention and further investigation.
Laboratory experiments simulating a coastal river inflow

The dynamics of buoyant water entering a rotating basin are studied using a series of laboratory experiments designed to elucidate the alongshore transport mechanisms in river plumes. Inflowing water, which is discharged perpendicular to the tank wall, is observed to form a growing anticyclonic bulge and a coastal current downstream of the bulge. Detailed simultaneous measurements of the velocity and buoyancy fields in the plume confirm that the bulge momentum is in a gradient–wind balance and the coastal current is geostrophic. The growth of the bulge and accumulation of fluid within it coincides with a reduction in coastal current transport to approximately 50% of the inflow discharge. The bulge is characterized by a depth scale, $h$, which is proportional to the geostrophic depth, $h_{g}$, and two time-dependent horizontal length scales, $y_{c}$, the displacement of the bulge centre from the wall, and $r_{b}$, the effective radius of the bulge. These two length scales are proportional to the inertial radius, $L_{i}$, and the local Rossby radius, $L_{b}$, respectively. When $r_{b}\gg y_{c}$, the bulge is held tightly to the wall, and a relatively large fraction of the inflow discharge is forced into the coastal current. For plumes with $y_{c}$ approaching $r_{b}$, the bulge is further from the wall, and the coastal current flux is reduced. Once ${y_{c}}/{r_{b}}\,{>}\,0.7$, the bulge separates from the wall causing flow into the coastal current to cease and the bulge to become unstable. In this state, the bulge periodically detaches from and re-attaches to the wall, resulting in pulsing transport in the coastal current. Scaling of the bulge growth based on $h_{g}$, $L_{i}$ and $L_{b}$ predicts that it will increase as $\hbox{\it Ro}^{1/4}$, where $\hbox{\it Ro}$ is the inflow Rossby number. The bulge growth, inferred from direct measurements of the coastal current transport, is proportional to $\hbox{\it Ro}^{0.32}$ and agrees with the predicted dependence within the experimental error.

We report the synthesis of photoreactive and light-emitting liquid crystals which can be patterned photolithographically to form polymer networks for use in organic light-emitting diodes. The resolution capability of the photopatterning is investigated using a phase mask of period one micron to spatially modulate the irradiance of ultraviolet light incident to the monomer thin film. A surface relief grating of the same period and depth 85 nm is formed on exposure and washing. An OLED incorporating a novel green light-emitting liquid-crystal has a luminous efficiency of 4.3 lm W-1 at 100 cd m-2 and gives a luminance of 990 cd m-2 at an operating voltage of 10 V.
Immediate and delayed stimulus repetitions evoke different ERPs in a serial-probe recognition task

In this study we examined whether event-related potentials (ERPs) associated with stimulus repetition and recognition in a serial-probe recognition task were comparable to ERPs in other tasks that are more typically used to investigate old/new ERP effects. The experiment consisted of 320 trials in which a recognition probe followed a four-item memory set; 160 trials consisted of images depicting common objects that were easy to label (EL task), and 160 trials consisted of images depicting abstract patterns that were difficult to label (DL task). Nineteen participants indicated whether a probe that followed each memory set was or was not presented in the memory set. Half of the probes matched, and half did not match, an item in the preceding memory set. ERPs appeared to reflect two processes—one that differentiated between recently presented stimuli and other stimuli and another that distinguished between repeated stimuli and new stimuli. ERPs to recent probes were more positive than ERPs to other probes in the EL and DL tasks. ERPs to match (old) probes were more positive than ERPs to nonmatch (new) probes only in the EL task.

In two experiments, we explored the utility of using event-related brain potentials (ERPs) evoked during picture recognition to examine the cognitive and neural processes underlying primacy and recency effects. Each experiment consisted of 210 trials in which a recognition probe followed a 12-picture sequence (105 match and 105 nonmatch trials). The 105 match-probe trials consisted of 35 trials in which the probe matched a prime memory set item (Positions 1–3), 35 in which the probe matched a middle memory set item (Positions 6–8), and 35 in which the probe matched a recent memory set item (Positions 10–12). Behavioral results revealed recency but not primacy effects in both experiments. Recent probes, compared with prime and middle probes, evoked ERPs that were more positive from approximately 300 to 400 ms; this enhanced positivity occurred in a positive component peaking around 315 ms and a negative component peaking around 365 ms. These findings fit more closely with the notion of short-term memory as an activation of elements in long-term memory than as a distinct memory store (or stores) separate from long-term memory.
