P.011 Efficacy and safety of ravulizumab in adults with AQP4+ NMOSD: interim analysis from the ongoing phase 3 CHAMPION-NMOSD trial

Background: CHAMPION-NMOSD (NCT04201262) is an ongoing global, open-label, phase 3 study evaluating ravulizumab in AQP4+ NMOSD. Methods: Adult patients received an intravenous, weight-based loading dose of ravulizumab on day 1 and a maintenance dose on day 15 and every 8 weeks thereafter. Following a primary treatment period (PTP; up to 2.5 years), patients could enter a long-term extension (LTE). Results: 58 patients completed the PTP; 56/2 entered/completed the LTE. As of June 16, 2023, median (range) follow-up was 138.4 (11.0-183.1) weeks for ravulizumab (n=58), with 153.9 patient-years. Across the PTP and LTE, no patients had an adjudicated on-trial relapse during ravulizumab treatment. 91.4% (53/58 patients) had stable or improved Hauser Ambulation Index score. 91.4% (53/58 patients) had no clinically important worsening in Expanded Disability Status Scale score. The incidence of treatment-emergent adverse events (TEAEs) and serious adverse events was 94.8% and 25.9%, respectively. Most TEAEs were mild to moderate in severity and unrelated to ravulizumab. TEAEs leading to withdrawal from ravulizumab occurred in 1 patient. Conclusions: Ravulizumab demonstrated long-term clinical benefit in the prevention of relapses in AQP4+ NMOSD with a safety profile consistent with prior analyses.

Water wave overwash of a step by small steepness, regular incident waves is analysed using a computational fluid dynamics (CFD) model and a mathematical model, in two spatial dimensions. The CFD model is based on the two-phase, incompressible Navier–Stokes equations, and the mathematical model is based on the coupled potential-flow and nonlinear shallow-water theories. The CFD model is shown to predict vortices, breaking and overturning in the region where overwash is generated, and that the overwash develops into fast-travelling bores. The mathematical model is shown to predict bore heights and velocities that agree with the CFD model, despite neglecting the complicated dynamics where the overwash is generated. Evidence is provided to explain the agreement in terms of the underlying agreement of mass and energy fluxes.

A theoretical model and an experimental model of surge motions of an ice floe due to regular waves are presented. The theoretical model is a modified version of Morrison’s equation, valid for small floating bodies. The experimental model is implemented in a wave basin at a scale 1:100, using a thin plastic disc to model the floe. The processed experimental data display a regime change in surge amplitude when the incident wavelength is approximately twice the floe diameter. It is shown that the theoretical model is accurate in the high-wavelength regime, but highly inaccurate in the low-wavelength regime.

Background
Major depressive disorder (MDD) is moderately heritable, however genome-wide association studies (GWAS) for MDD, as well as for related continuous outcomes, have not shown consistent results. Attempts to elucidate the genetic basis of MDD may be hindered by heterogeneity in diagnosis. The Center for Epidemiological Studies Depression (CES-D) scale provides a widely used tool for measuring depressive symptoms clustered in four different domains which can be combined together into a total score but also can be analysed as separate symptom domains.
MethodWe performed a meta-analysis of GWAS of the CES-D symptom clusters. We recruited 12 cohorts with the 20- or 10-item CES-D scale (32 528 persons).
ResultsOne single nucleotide polymorphism (SNP), rs713224, located near the brain-expressed melatonin receptor (MTNR1A) gene, was associated with the somatic complaints domain of depression symptoms, with borderline genome-wide significance (pdiscovery = 3.82 × 10−8). The SNP was analysed in an additional five cohorts comprising the replication sample (6813 persons). However, the association was not consistent among the replication sample (pdiscovery+replication = 1.10 × 10−6) with evidence of heterogeneity.
ConclusionsDespite the effort to harmonize the phenotypes across cohorts and participants, our study is still underpowered to detect consistent association for depression, even by means of symptom classification. On the contrary, the SNP-based heritability and co-heritability estimation results suggest that a very minor part of the variation could be captured by GWAS, explaining the reason of sparse findings.

A theoretical model of water wave overwash of a thin floating plate is proposed. The nonlinear shallow-water equations are used to model the overwash, and the linear potential-flow/thin-plate model to force it. Model predictions are compared with overwash depths measured during a series of laboratory wave basin experiments. The model is shown to be accurate for incident waves of low steepness or short length.

In laboratory experiments adults and nymphs of the western conifer seed bug, Leptoglossus occidentalis Heidemann, were allowed to feed on mature seeds of Douglas-fir, Pseudotsuga menziesii (Mirbel) Franco. Weight-loss measurements and scanning electron microscopy provided strong supporting evidence for the use of simple radiographic diagnosis as a method of classifying feeding damage to seeds into four categories: light (greater than two thirds of seed contents remaining), moderate (one third to two thirds of seed contents remaining), severe (less than one third of seed contents remaining), and extreme (seed empty). Scanning electron micrographs showed the apparent depletion of lipid and protein storage reserves which was verified by quantitative analyses that showed significant loss of lipid and buffer-insoluble (crystalloid) storage protein from seeds in all damage categories. The amount of buffer-soluble (matrix) protein was reduced in seeds from the severe and extreme damage categories. The increase in buffer-soluble protein observed in lightly damaged seeds was likely due to the solubilization of crystalloid storage protein, as a result of its breakdown into smaller peptides. Our results suggest that through the action of both lipases and proteases, L. occidentalis can have a serious impact on the major storage reserves of conifer seeds. Moreover, our data suggest that L. occidentalis feeds in a different manner than the laceration and flushing method found in other seed-feeding Hemiptera.

The relative concentration of elements in adults of the red turnip beetle, Entomoscelis americana Brown, as determined by X-ray energy-dispersive spectroscopy (XES), was used with discriminant analysis to identify source populations of beetles from different fields in the same district. Classification of dispersed beetles from the same district, using discriminant functions, suggests that XES is a promising technique for dispersal studies. However, multivariate analysis of variance and canonical analysis of XES results of groups of beetles given different foods over varying times in the laboratory indicate that highly significant changes occur in the chemoprint. Methods of minimizing the effect of such changes are suggested for future studies of dispersal.

Many agricultural systems around the world are challenged by declining soil resources, a dry climate and increases in input costs. The cultivation of plants that are better adapted than current crop species to nutrient poor soils, a dry climate and low-input agricultural systems would aid the continued profitability and environmental sustainability of agricultural systems. This paper examines herbaceous native Australian legumes for their capacity to be developed as grain crops adapted to dry environments. The 14 genera that contain herbaceous species are Canavalia, Crotalaria, Cullen, Desmodium, Glycine, Glycyrrhiza, Hardenbergia, Indigofera, Kennedia, Lotus, Rhynchosia, Swainsona, Trigonella and Vigna. A number of these genera (e.g., Glycine, Crotalaria, Trigonella and Vigna) include already cultivated exotic grain legumes. Species were evaluated based on the extent to which their natural distribution corresponded to arid and semi-arid climatic regions, as well as the existing information on traits related to harvestability (uniformity of ripening, propensity to retain pod, pod shattering and growth habit), grain qualities (seed size, chemistry, color and the absence of toxins) and fecundity. Published data on seed yield were rare, and for many other traits information was limited. The Australian species of Vigna, Canavalia and Desmodium mainly have tropical distributions and were considered poorly suited for semi-arid temperate cropping systems. Of the remaining genera Glycyrrhiza and Crotalaria species showed many suitable traits, including an erect growth habit, a low propensity to shatter, flowers and fruits borne at the end of branches and moderate to large seeds (5 and 38 mg, respectively). The species for which sufficient information was available that were considered highest priority for further investigation were Glycine canescens, Cullen tenax, Swainsona canescens, Swainsona colutoides, Trigonella suavissima, Kennedia prorepens, Glycyrrhiza acanthocarpa, Crotalaria cunninghamii and Rhynchosia minima.

In a community hepatitis A outbreak in the Rochester, New York area, 64 of 79 (81 %) people with anti-hepatitis A 1gM-antibodies and onset of symptoms from 9 April–31 May 1994, recalled eating food obtained from a retail buyer's club. Eleven (65%) of 17 households with cases contained club members compared with 7 (21%) of 34 neighbourhood-matched control-households (matched odds ratio 8·5; 95% CI 1·7–41·6). Club employees who ate sugar-glazed baked goods were at fourfold increased risk for hepatitis. The source of infection was an 1gM- positive baker who contaminated baked goods while applying sugar glaze. Computer-generated purchase lists implicated 11–12 March and 21–24 March as the most likely dates when contamination occurred. This investigation demonstrates the importance of food workers adhering to established hygiene practices. Computer-generated commercial datasets can be useful in epidemiologic investigations.

1. It has been confirmed that Trichomonas foetus fails to survive freezing and thawing in the presence of 10 % glycerol in egg-yolk-citrate diluent. Under the same conditions adequate sperm survival was demonstrated.
2. Trichomonads were particularly sensitive to the toxic effects of glycerol when suspended in egg-yolk citrate.
3. Trichomonads will survive freezing and thawing when suspended in other diluents such as egg-yolk phosphate or milk.

Background: As people live longer, there is increasing potential for mental disorders to interfere with testamentary distribution and render older people more vulnerable to “undue influence” when they are making a will. Accordingly, clinicians dealing with the mental disorders of older people will be called upon increasingly to advise the courts about a person's vulnerability to undue influence.
Method: A Subcommittee of the IPA Task Force on Testamentary Capacity and Undue Influence undertook to establish consensus on the definition of undue influence and the provision of guidelines for expert assessment of risk factors for undue influence.
Results: International jurisdictions differ in their approach to the notion of undue influence. Despite differences in legal systems, from a clinical perspective, the subcommittee identified some common “red flags” which might alert the expert to risk of undue influence. These include: (i) social or environmental risk factors such as dependency, isolation, family conflict and recent bereavement; (ii) psychological and physical risk factors such as physical disability, deathbed wills, sexual bargaining, personality disorders, substance abuse and mental disorders including dementia, delirium, mood and paranoid disorders; and (iii) legal risk factors such as unnatural provisions in a will, or provisions not in keeping with previous wishes of the person making the will, and the instigation or procurement of a will by a beneficiary.
Conclusion: This review provides some guidance for experts who are requested by the courts to provide an opinion on the risk of undue influence. Whilst international jurisdictions require different thresholds of proof for a finding of undue influence, there is good international consensus on the clinical indicators for the concept.
Sire breed comparisons for meat and eating quality traits in Australian pig populations

Sire breed comparisons for carcase (n = 1169), meat and eating quality (n = 686) traits were obtained using data recorded on progeny of known pedigree sired by Duroc (DU), Large White (LW), Landrace (LR) and Duroc Synthetic (DS) boars from crossbred (LW/LR) sows. Animals were reared in eco-shelters in large single-sex contemporary groups and slaughtered on an age constant basis at 22 weeks of age. Compared to progeny from other sire groups, animals sired by purebred Duroc boars tended to have improved eating quality traits (higher intramuscular fat and lower shear force) at the expense of poorer carcase characteristics (higher subcutaneous and belly fat). Animals sired by DS boars tended to be heavier and leaner than those sired by LW, LR or DU boars; intramuscular fat but not belly fat was correspondingly lower, while tenderness was generally consistent with that of the DU progeny. Significant variability of sire progeny groups within sire breed suggests that sire breed selection, potentially used for improving traits such as meat quality in commercial progeny, will be less accurate in the absence of sire-specific information, which is typically poorly recorded in this class of traits.

PLANET, the Probing Lensing Anomaly NETwork, is an international team conducting observations of on-going gravitational microlensing events from five sites in the southern hemisphere. Our primary goal is to detect or to put constraints on sub-stellar companions of M dwarfs from the galactic disk. We report the current status and discuss the future prospects. A 2 m robotic telescope at Dome C which would benefit from continuous coverage and dream like seeing (median of 0.27 arcsec) is currently the best option for a ground based aggressive search for Earth-mass planets in the habitable zone.

In this study, we applied a stepped-combustion approach to dating post-bomb lake sediment from north-central Mississippi. Samples were combusted at a low temperature (400 °) and then at 900 °. The CO2 was collected separately for both combustions and analyzed. The goal of this work was to develop a methodology to improve the accuracy of 14C dating of sediment by combusting at a lower temperature and reducing the amount of reworked carbon bound to clay minerals in the sample material. The 14C fraction modern results for the low and high temperature fractions of these sediments were compared with well-defined 137Cs determinations made on sediment taken from the same cores. Comparison of “bomb curves” for 14C and 137Cs indicate that low temperature combustion of sediment improved the accuracy of 14C dating of the sediment.
