Evidence for genes controlling resistance to Heligmosomoides bakeri on mouse chromosome 1

Resistance to infections with Heligmosomoides bakeri is associated with a significant quantitative trait locus (QTL–Hbnr1) on mouse chromosome 1 (MMU1). We exploited recombinant mice, with a segment of MMU1 from susceptible C57Bl/10 mice introgressed onto MMU1 in intermediate responder NOD mice (strains 1094 and 6109). BALB/c (intermediate responder) and C57Bl/6 mice (poor responder) were included as control strains and strain 1098 (B10 alleles on MMU3) as NOD controls. BALB/c mice resisted infection rapidly and C57Bl/6 accumulated heavy worm burdens. Fecal egg counts dropped by weeks 10–11 in strain 1098, but strains 1094 and 6109 continued to produce eggs, harbouring more worms when autopsied (day 77). PubMed search identified 3 genes (Ctla4, Cd28, Icos) as associated with ‘Heligmosomoides’ in the B10 insert. Single nucleotide polymorphism (SNP) differences in Ctla4 could be responsible for regulatory changes in gene function, and a SNP within a splice site in Cd28 could have an impact on function, but no polymorphisms with predicted effects on function were found in Icos. Therefore, one or more genes encoded in the B10 insert into NOD mice contribute to the response phenotype, narrowing down the search for genes underlying the H. bakeri resistance QTL, and suggest Cd28 and Ctla4 as candidate genes.

Objective
To examine the use of vitamin D supplements during infancy among the participants in an international infant feeding trial.
DesignLongitudinal study.
SettingInformation about vitamin D supplementation was collected through a validated FFQ at the age of 2 weeks and monthly between the ages of 1 month and 6 months.
SubjectsInfants (n 2159) with a biological family member affected by type 1 diabetes and with increased human leucocyte antigen-conferred susceptibility to type 1 diabetes from twelve European countries, the USA, Canada and Australia.
ResultsDaily use of vitamin D supplements was common during the first 6 months of life in Northern and Central Europe (>80 % of the infants), with somewhat lower rates observed in Southern Europe (>60 %). In Canada, vitamin D supplementation was more common among exclusively breast-fed than other infants (e.g. 71 % v. 44 % at 6 months of age). Less than 2 % of infants in the USA and Australia received any vitamin D supplementation. Higher gestational age, older maternal age and longer maternal education were study-wide associated with greater use of vitamin D supplements.
ConclusionsMost of the infants received vitamin D supplements during the first 6 months of life in the European countries, whereas in Canada only half and in the USA and Australia very few were given supplementation.

A silicon multi-cathode detector (SMCD) has been developed for microanalysis and x-ray mapping applications. The SMCD has a large active area (∼0.5 cm2), excellent energy resolution, and high count rate capability. The detector utilizes novel structures that have produced very low dark current, high electric field, uniform charge collection, low noise and high sensitivity to low energy x-rays. The detector's spectral response was evaluated using a 55Fe radioisotope source, as well as by fluorescing materials with an x-ray generator. Figure 1 shows a 55Fe spectrum with an energy resolution of 125 eV FWHM at 5.9 keV collected at 12 μs peaking time. This energy resolution has been repeatably measured on many different detectors. To evaluate the high count rate x-ray performance, which is very important for fast x-ray mapping, a Cu sample was fluoresced using a Rh-anode x-ray tube.

A new class of silicon multi-cathode detector (SMCD) has been developed for microanaiysis spectrometry applications. The detector has excellent energy resolution (< 150 eV FWHM) and high count rate capability (>1 Mcps). An energy resolution of 143 eV FWHM at 5.9 keV was measured with the SMCD at 6 μs peaking time.

The concept utilized in charge coupled devices (CCD’s) for detection and imaging of light signals involving lateral movement of charges and extremely low capacitance of the detector and readout electronics has spawned a variety of new ideas in the design of nuclear detectors. Initially, silicon drift detectors (SDD’s) were developed for high energy physics applications. More recently, a vigorous effort to develop new structures for x-ray spectroscopy and light detection has started. Drift structures have been designed in a variety of topologies and materials (such as Si, CdZnTe, and HgI2) to satisfy the requirements of many different applications. The most interesting features that can be achieved with drift structures include: a) Large active area devices with low capacitance and low electronic noise, b) Very high signal throughput, c) Operation at or near room temperature, d) High sensitivity over the large entrance electrode to low energy xrays and short wavelength light, f) Single carrier charge collection allowing for elimination of hole contribution to the spectral broadening in compound semiconductor detectors such as HgI2, CdTe, and CdZnTe, f) 2D resolution of few tens of micrometer in both directions over few cm2 active areas, and g) Possibility of using more sophisticated schemes of charge collection by switching between integration and drift mode.

A nineteen element mercuric iodide (HgI2) detector array has been developed in order to investigate the potential of using this technology for in-vivo x-ray and gamma-ray imaging. A prototype cross-grid detector array was constructed with hexagonal pixels of 1.9 mm diameter (active area = 3.28 mm2) and 0.2 mm thick septa. The overall detector active area is roughly 65 mm2. A detector thickness of 1.2 mm was used to achieve about 100% efficiency at 60 keV and 67% efficiency at 140 keV The detector fabrication, geometry and structure were optimized for charge collection and to minimize crosstalk between elements. A section of a standard high resolution cast-lead gamma-camera collimator was incorporated into the detector to provide collimation matching the discrete pixel geometry. Measurements of spectral and spatial performance of the array were made using 241-Am and 99m-Tc sources. These measurements were compared with similar measurements made using an optimized single HgI2 x-ray detector with active area of about 3 mm2 and thickness of 500 μm.
