Hyperglycemia is reported to predict worse outcome in patients with stroke, including intracerebral hemorrhage (ICH). In 83 consecutive cases of ICH at a tertiary stroke center, hyperglycemia (serum glucose >7 mmol/L) compared to normoglycemia at presentation was associated with higher rates of in-hospital mortality (51.2% vs. 26.2%, OR 2.3, CI 1.2–7.6, p = 0.02). The association with in-hospital mortality withstood adjustment for age, ICH volume, intraventricular hemorrhage, and infratentorial ICH location, but not baseline Glasgow Coma Scale. Acute hyperglycemia is associated with in-hospital mortality in spontaneous ICH patients, though this may be an indirect, rather than a causal relationship.
Bi-dimensional acculturation and depressive symptom trajectories from pregnancy to 1 year postpartum in marriage-based immigrant women in Taiwan

Background
Childbirth may pose many challenges to the psychological well-being of marriage-based immigrant mothers in interracial marriages, who must negotiate bi-dimensional acculturation – adaptation to the host culture and maintenance of her own heritage culture. We examined the temporal relationships between bi-dimensional acculturation and depressive symptoms from pregnancy to 1 year postpartum among marriage-based immigrant mothers in Taiwan using the cross-lagged structural equation modeling.
MethodsThis study recruited 310 immigrant mothers, who were examined in the second and third trimesters, and again at 1 month, 3 months, 6 months, and 1 year postpartum from March 2013 to December 2015. Depressive symptoms and bi-dimensional acculturation were measured using the Edinburgh Postnatal Depression Scale and Bidimensional Acculturation Scale for Marriage-Based Immigrant Women, respectively.
ResultsThe study found that adaptation to the host culture followed a downward linear trajectory, while maintenance of the mother's own heritage culture followed an upward linear trajectory from pregnancy to 1 year postpartum. All but one cross-lagged path between bi-dimensional acculturation and depressive symptoms was statistically insignificant, though almost all cross-sectional associations were significant. Adaptation to host culture was negatively associated with depressive symptoms at all time points. The association between maintenance of heritage culture and depressive symptoms reversed from positive to negative after 6 months postpartum.
ConclusionsAdaptation to the host culture and maintenance of the mother's heritage culture differed in their associations with maternal depressive symptoms. Health professionals should assist immigrant mothers in adapting to the host culture while supporting their heritage culture in the childbearing period.
2207: A Phase I dose escalation trial of nab-paclitaxel and fixed dose radiation in patients with unresectable or borderline resectable pancreatic cancer

OBJECTIVES/SPECIFIC AIMS: Patients with locally advanced pancreatic cancer typically have poor outcomes, with a median survival of ~16 months. Novel methods to improve local control are needed. Nab-paclitaxel (abraxane) has shown efficacy in pancreatic cancer and is FDA approved for metastatic disease in combination with gemcitabine. Nab-paclitaxel is also a promising radiosensitizer based on laboratory studies, but it has never been clinically tested with definitive radiotherapy for locally advanced disease. METHODS/STUDY POPULATION: We performed a phase 1 study using a 3+3 dose-escalation strategy to determine the safety and tolerability of dose escalated nab-paclitaxel with fractionated radiotherapy for patients with unresectable or borderline resectable pancreatic cancer. Following induction chemotherapy with 2 cycles of nab-paclitaxel and gemcitabine, patients were treated with weekly nab-paclitaxel and daily radiotherapy to a dose of 52.5 Gy in 25 fractions. Final dose-limiting toxicity (DLT) determination was performed at day 65 after the start of radiotherapy. RESULTS/ANTICIPATED RESULTS: Nine patients received nab-paclitaxel at a dose level of either 100 mg/m2 (n=3) or 125 mg/m2 (n=6). One DLT (grade 3 neuropathy) was observed in a patient who received 125 mg/m2 of nab-paclitaxel. Other grade 3 toxicities included fatigue (11%), anemia (11%), and neutropenia (11%). No grade 4 toxicities were observed. With a median follow-up of 8 months (range 5–28 months), median survival was 19 months and median progression-free survival was 10 months. Following chemoradiation, 3 patients underwent surgical resection, all with negative margins and limited tumor viability. Of the 3 patients, 2 initially had borderline resectable tumors and 1 had an unresectable tumor. Tumor (SMAD-4, Caveolin-1) and peripheral (circulating tumor cells and microvesicles) biomarkers were collected and are being analyzed. DISCUSSION/SIGNIFICANCE OF IMPACT: The combination of fractionated radiation and weekly nab-paclitaxel was safe and well tolerated. This regimen represents a potentially promising therapy for patients with unresectable and borderline resectable pancreatic cancer and warrants further investigation.

The three tropical species Gomphandra luzoniensis, Nothapodytes nimmoniana and Goniothalamus amuyon contain important cancer-fighting drugs; however, little is known about how to propagate these species from seeds. Thus, the aim of this study was to determine the germination requirements of seeds of each of these three species in order to provide an effective protocol to produce plants. Fresh seeds of G. luzoniensis germinated up to 73% at high temperatures in light in 4 weeks, and embryos were underdeveloped. Most seeds had morphological dormancy (MD), but a proportion of them had morphophysiological dormancy (MPD). Fresh seeds of N. nimmoniana germinated up to 50% in light in 4 weeks, embryo length increased by 17% before radicle emergence and ≥ 89% of the seeds had germinated after incubation for 6 weeks in light at high temperatures. Thus, about 50% of the seeds have MD and about 50% MPD. Fresh seeds of G. amuyon incubated at 30°C in light for 4 weeks germinated to 69%, whereas at the other incubation temperatures germination took longer than 4 weeks. Embryo length increased 213% before radicle emergence, and after 8 weeks of incubation at high temperatures ≥ 80% of the seeds had germinated. As in the other two species, the seed population consisted of a mixture of MD and MPD. Incubation of seeds of these three species at high temperatures (e.g. 25, 30 and 30/20°C) for up to 2 months is recommended for germination and thus seedling production.

We introduce a new multigrid method to study the lattice statics model arising from nanoindentation. A constrained Cauchy-Born elasticity model is used as the coarse-grid operator. This method accelerates the relaxation process and considerably reduces the computational cost. In particular, it saves quite a bit when dislocations nucleate and move, as demonstrated by the simulation results.
Underdeveloped embryos and kinds of dormancy in seeds of two gymnosperms: Podocarpus costalis and Nageia nagi (Podocarpaceae)

Although it has been speculated that seeds of the gymnosperm family Podocarpaceae have an underdeveloped embryo, no detailed studies have been done to definitively answer this question. Our purpose was to determine if embryos in seeds of two species of Podocarpaceae, Podocarpus costalis and Nageia nagi, from Taiwan are underdeveloped and to examine the kind of dormancy the seeds have. Embryos in fresh seeds of P. costalis were 4.6 ± 0.5 mm long, and they increased in length by about 54% before radicle emergence (germination), demonstrating that the embryo is underdeveloped at seed maturity. Seeds germinated to >90% at 30/20, 25/15 and 25°C in light in ≤ 4 weeks, without any cold stratification pretreatment. Thus, seeds of P. costalis have morphological dormancy (MD). Embryos in fresh seeds of N. nagi were 7.4 ± 0.8 mm long and they increased in length by about 39% before radicle emergence (germination) occurred, indicating that the embryo is underdeveloped at seed maturity. Seeds germinated to < 25% at 30/20 and 25°C in light in 4 weeks but to >90% at the same temperatures in 12 weeks. Thus, most seeds of N. nagi have morphophysiological dormancy (MPD). Although underdeveloped embryos are considered to be a primitive condition in seed plants, they also occur in the most advanced orders. The occurrence of underdeveloped embryos in Podocarpaceae documents that they are not restricted to a basal clade in gymnosperms.

We use samples of local main-sequence stars to show that the radial gradient of [Fe/H] in the thin disk of the Milky Way decreases with mean effective stellar temperature. We use the angular momentum of each star about the Galactic center to eliminate the effects of epicyclic motion, which would otherwise blur the estimated gradients. We use the effective temperatures as a proxy for mean age, and conclude that the decreasing gradient is consistent with the predictions of radial mixing due to transient spiral patterns. We find some evidence that the trend of decreasing gradient with increasing mean age breaks to a constant gradient for samples of stars whose main-sequence life-times exceed the likely age of the thin disk.
Deep simple morphophysiological dormancy in seeds of the basal taxad Cephalotaxus

Although mature seeds of the monogeneric conifer family Cephalotaxaceae sensu stricto have underdeveloped embryos, no definitive studies have been done to classify dormancy in this family. Our primary purpose was to determine the kind of dormancy in seeds of Cephalotaxus wilsoniana and to put the results into a broad phylogenetic context for gymnosperms. The species is of horticultural and medicinal value, and information is needed on how to propagate it efficiently from seeds. Embryo growth and germination were monitored for seeds at warm, cold and warm plus cold temperatures, and germination was monitored for seeds subjected to: (1) cold → warm → cold → warm; and (2) warm → cold → warm → cold → warm temperature sequences. The effects of gibberellic acids GA3 and GA4 were tested on radicle emergence in ungerminated seeds and on shoot emergence in root-emerged seeds. Germination was promoted by ≥ 36 weeks of warm stratification followed by ≥ 8 weeks of cold stratification, but only if seeds were returned to high temperatures. The underdeveloped embryo must increase in length by >120% before the radicle emerges. Neither GA3 nor GA4 was effective in promoting radicle emergence; however, both plant growth regulators increased rate (but not percentage) of shoot emergence in root-emerged seeds. We conclude that seeds of C. wilsoniana have the deep simple level of morphophysiological dormancy (MPD), C1b-C3-B1b; thus, warm stratification followed by cold stratification and then warm-temperature incubation are required for germination. In gymnosperms, MPD is known in cycads, Ginkgo and now in three families of conifers.

Germination of Viburnum odoratissimum seeds: a new level of morphophysiological dormancy

Previous studies indicated that seeds of Viburnum odoratissimum had only physiological dormancy (PD), but no measurements of embryos were made during the dormancy-break treatments. Thus, we investigated embryo growth and radicle and cotyledon emergence over a range of temperatures. Seeds have underdeveloped embryos, and their length increased about 300% before radicle emergence. Embryos also had PD, as evidenced by delays in beginning of embryo growth (2–3 weeks) and of germination after embryos were elongated (4 weeks). After radicle emergence, epicotyl emergence was delayed 1–8 weeks, depending on incubation temperature, but cold stratification was not required to break PD of the epicotyl. Unlike seeds of many previously studied Viburnum spp., epicotyls of V. odoratissimum have non-deep, rather than deep, PD. Hence, a new level of MPD called non-deep, simple, epicotyl MPD has been identified.

The embryo length/seed length (E/S) ratio of the early diverging eudicot Trochodendron aralioides is 0.34. Embryos in fresh seeds were 0.36±0.01 mm long, and they increased in length by about 250% (in 20 d) before radicle emergence (germination) occurred, demonstrating that the embryo is underdeveloped at seed maturity. Seeds germinated to 95–100% at 20/10, 25/15 and 30/15°C in light in ≤4 weeks, without any pretreatment, but no seeds germinated in darkness. Thus, seeds of T. aralioides have morphological dormancy (MD), which is considered to be the primitive condition in seed plants, and MD probably has existed in the genus Trochodendron since its origin in the early Tertiary.

The evolution equation governing wavemaker-generated cross-waves near a cutoff frequency in an infinitely deep, infinitely long channel is shown to be the nonlinear Schrödinger equation with a homogeneous boundary condition at the wavemaker. With the inclusion of an empirically determined damping coefficient, numerical results for growth rate, slow modulation period, and wave amplitude show good agreement with previous experiments. The results also describe observations of trapped and propagating solutions.

The assembly of plastic microfluidic devices, MOEMS and microarrays requiring high positioning and welding accuracy in the micrometer range, has been successfully achieved using a new technology based on laser transmission welding combined with a photolithographic mask technique. This paper reviews a laser assembly platform for the joining of microfluidic plastic parts with its main related process characteristics and its potential for low-cost and high volume manufacturing. The system consists of a of diode laser with a mask and an automated alignment function to generate micro welding seams with freely definable geometries. A fully automated mask alignment system with a resolution of < 2 μm and a precise, non-contact energy input allows a fast welding of micro structured plastic parts with high reproducibility and excellent welding quality.

A fast way to monitor the quality of high-k dielectric layers is wet etching, either monitored by Open Circuit Potential analysis or by Scanning Electron Microscopy. Defect densities in the order of 1.109 defects/cm2 are observed for as-deposited HfO2 layers. It is assumed that the mechanism for wet chemical defect observation is either due to crystallization and/or due to an oxygen deficient HfO2 layer resulting in Si/SiO up-diffusion upon thermal treatment. However, after appropriate post deposition annealing wet etch defect free layers can be prepared.

ZrO2 and HfO2 and their alloys with SiO2 are currently among the leading high-k materials for replacing SiOxNy as the gate dielectric for the sub-100 nm technology nodes. International SEMATECH (ISMT) is currently investigating integration issues associated with this required change in materials. Our work has focused on the integration of ALCVD deposited ZrO2 and HfO2 with an industry standard conventional MOSFET process flow with poly-Si electrode. Since the impact of contamination by these new high-k materials introduced in a production fab has not yet been established, it becomes very critical to prevent cross- contamination through the process tools in the fab. A baseline study was completed within ISMT's fab and appropriate protocols for handling high-k materials have been established. The integrated high-k gate stack in a conventional transistor flow should not only meet all the performance requirements of scaled transistors, but the gate dielectric film should be able withstand high-temperature anneal steps. Reactions between ZrO2 and Si have been observed at temperatures as low as 560°C (during the amorphous Si deposition process). Various wet chemistries were also evaluated for removing the high-k film inadvertently deposited on wafer backside, and it was found that ZrO2 etches at extremely slow rates in the majority of the common wet etch chemistries available in a fab. A new hot HF based process was found to be successful in lowering Zr contamination on the wafer backside to as low as 1.8 E10 atoms/cm2. The patterning of a high-k gate stack with poly-Si electrode is another area that required considerable focus. Various dry (plasma) etch and wet etch chemistries were evaluated for etching ZrO2 using both blanket films as well as wafers with patterned poly-Si gate over the high-k films. On the full CMOS flow device wafers, most of these wet chemistries resulted in severe pitting in the ZrO2 film remaining over the source/drain (S/D) areas, as well as in the Si substrate and the field oxide. A poly-Si gate over ZrO2 gate dielectric film was successfully patterned using the standard poly-Si gate etch (Cl2/HBr) for the main etch, followed by a combination of HF and H2SO4 clean for removing all of the ZrO2 remaining over the S/D area. This allowed the fabrication of low-resistance contacts to transistor S/D areas, which ultimately resulted in demonstration of functional transistors with high-k gate dielectric films.

Formation of aggregates has been observed as a general phenomenon for a wide variety of organic molecules, especially aromatic compounds and dyes. Aggregation is most commonly encountered in crystals or in thin films. However, it has been increasingly observed in microheterogenous media or in other situations where high local concentrations occur or where specific orientation is favored. Two limiting types of aggregation have been defined based on the orientation of transition dipoles and their absorption spectral characteristics. These are the “J” aggregate, in which head-to-tail arrangements of transitiondipole moments are characterized by a sharp, intense, red-shifted transition compared to the isolated (solvated) monomer, and the “H” aggregate, where head-to-head (card-pack) orientations are characterized by a blue shift of the prominent transition compared with the monomer. Several treatments have been proposed to correlate the observed spectral shifts with the aggregate structure. For a number of compounds, the association of known x-ray-determined structures with spectral features has supported the theoretical predictions developed by Kasha and Hochstrasser or by Czikkely, Försterling, and Kuhn. The focus of the studies described here has been on aggregation occurring in Langmuir-Blodgett (LB) films and related media, such as bilayer vesicles, which are characterized by an assembly of molecules in an interfacial situation where a polar-nonpolar or water-hydrocarbon boundary should provide a strong organizing influence. In early cases where aggregates were encountered as prominent components of mixed LB films (even when relatively dilute mixtures were used), the phenomenon was usually dismissed as “microcrystallization,” which was considered an unavoidable nuisance and not really due to fundamental intermolecular interactions. More recent studies have shown that aggregation in these media is really a significant molecular phenomenon that shows dependence both on the specific molecules and the topology of the film-forming surfactant. Although some previous investigations have been carried out with different results for various substrates, we have embarked on a study to correlate aggregation behavior for a number of different chromophores incorporated into amphiphilic structures to obtain a general picture of the relative importance of different factors that can control aggregation phenomena.
