Combining traditional dietary assessment methods with novel metabolomics techniques: present efforts by the Food Biomarker Alliance

FFQ, food diaries and 24 h recall methods represent the most commonly used dietary assessment tools in human studies on nutrition and health, but food intake biomarkers are assumed to provide a more objective reflection of intake. Unfortunately, very few of these biomarkers are sufficiently validated. This review provides an overview of food intake biomarker research and highlights present research efforts of the Joint Programming Initiative ‘A Healthy Diet for a Healthy Life’ (JPI-HDHL) Food Biomarkers Alliance (FoodBAll). In order to identify novel food intake biomarkers, the focus is on new food metabolomics techniques that allow the quantification of up to thousands of metabolites simultaneously, which may be applied in intervention and observational studies. As biomarkers are often influenced by various other factors than the food under investigation, FoodBAll developed a food intake biomarker quality and validity score aiming to assist the systematic evaluation of novel biomarkers. Moreover, to evaluate the applicability of nutritional biomarkers, studies are presently also focusing on associations between food intake biomarkers and diet-related disease risk. In order to be successful in these metabolomics studies, knowledge about available electronic metabolomics resources is necessary and further developments of these resources are essential. Ultimately, present efforts in this research area aim to advance quality control of traditional dietary assessment methods, advance compliance evaluation in nutritional intervention studies, and increase the significance of observational studies by investigating associations between nutrition and health.

Multi-Walled Carbon Nanotubes (MWNTs) mounted on commercial Scanning Force Microscopy (SFM) cantilevers have proven to be excellent probes for high resolution Tapping Mode-Scanning Force Microscopy (TM-SFM). Because of their robust nature and high electrical conductance, MWNTs are also attractive for use in Conductive Probe-Scanning Force Microscopy (CP-SFM). To be used in this application, however, the MWNT must be mounted via a high conductance contact to a conductive cantilever. A technique has been developed that produces such a contact. First, a MWNT is attached using acrylic adhesive to a commercial SFM cantilever that has been vacuum coated with gold. Then, a cap of gold is deposited over the junction between the MWNT and the SFM cantilever via spatially selective electro-deposition.

A new strategy for stabilizing inorganic nanoparticles in nonpolar solutions is described. Resorcinarenes 1-3 were synthesized and evaluated as surfactants because of their large concave headgroups with multiple contact sites. Au nanoparticles ranging from 3-20 nm in diameter were generated in the vapor phase and dispersed into dilute hydrocarbon solutions of 1-3, where they were stabilized for up to several months. Chemisorption is most likely mediated by multiple Au-O interactions, as indicated by several control experiments and by surface-enhanced Raman spectroscopy. The resorcinarenes were readily displaced by dodecanethiol, which resulted in the precipitation of particles >5 nm as determined by absorption spectroscopy and transmission electron microscopy. This suggests that the mobility of the resorcinarene tailgroups are important for maintaining the larger nanoparticles in a dispersed state. Resorcinarene surfactants with stronger chemisorptive properties are currently being explored.

“Fish-eye” particles consisting of metal clusters (Ag, Cu) a few nanometers in diameter encapsulated within a thin layer (-1 nm) of silica are produced using aerosol synthesis procedures. We present a method for predicting stable “fish-eye” nanostructures and describe synthesis techniques for producing significant quantities of silica-encapsulated metal nanoparticles.
For many metal/oxide pairs, gas phase formation of oxide encapsulated metal particles is thermodynamically favorable. Using known surface free energies and binary phase diagrams, it is possible to predict whether SiO2-encapsulated metal clusters will form in the gas phase. Two conditions which must be satisfied are: 1) that the surface free energy of the metal is higher than that of Si; and 2) that the metal composition in the particle is greater than the eutectic composition in the metal/Si phase diagram. Ag-SiO2 and Cu-SiO2 are two examples of systems which readily form “fish-eye” structures.
Two types of gas phase cluster sources are used at Purdue for producing encapsulated metal nanoparticles. The Multiple Expansion Cluster Source (MECS) is a well established apparatus which produces small quantities (- 50 mg/hr) of very uniform materials using resistive heating for evaporation. The new Arc Cluster Evaporation Source (ACES) offers much higher production rates (>1 g/hr) using DC arc evaporation. These two cluster sources make possible the study of a unique class of materials.

Indentation using the AFM is a powerful method for determining elastic properties of small supported clusters. However, a theoretical framework has yet to be developed to interpret such measurements. The elastic deformation of nanometer sized gold clusters are modeled using the Embedded Atom Method (EAM) potential of Foiles et al. [1]. Force versus deformation curves are obtained for a series of truncated octahedral clusters having FCC symmetry (N=38, 201, 586, 1289, 2406). It is found that the MD results both for static compression and for harmonic vibration can be analytically estimated by using an elastic constant for the clusters analogous to the elastic modulus of a bulk material. However MD predictions for static compression are not in agreement with the deformation results of Schaefer et al. [2].

Cluster-assembled thin metal films exhibit properties which are different from those of films obtained by conventional atomic-deposition. We present TEM data on the evolution of 2-D Microstructure and SFM data on the evolution of 3-D Microstructure in thin films grown by vacuum deposition of preformed silver clusters and of preformed acetylene-silver clusters on flat SiO2, and Mica. Electrical resistivity measurements of cluster-based Ag and Ag/C2H2 films deposited on glass substrates with nominal film thicknesses of 5 nm - 50 nm are also presented and discussed.
