PARP and NAD+ metabolism

To coincide with PARP 2023 https://parp2023.febsevents.org/ Expert Reviews in Molecular Medicine will publish a special collection on PARP and NAD+ metabolism. There has been substantial progress in our understanding of PARP biology in health and disease. The role of PARP1 and 2 in DNA repair has been well characterised. Inhibitors of these enzymes have been approved for the treatment of various cancers and there is potential to re-purpose these drugs for other indications. Our knowledge of the role of other members of this superfamily of ADP-ribosylation proteins is expanding and inhibitors specific to these other enzymes are in development some of which are entering early phase clinical trial. Since these enzymes utilize NAD+ as their substrate the implications for NAD+ homeostasis are key to health and disease.
