The self-organization of functional proteins directly onto solid materials is attractive to a wide range of biomaterials and systems that need to accommodate a biological recognition element. In such systems, inorganic binding peptides may be an essential component due to their high affinity and selective binding features onto different types of solid surfaces. This study demonstrates a peptide-enabled self-assembly technique for designing well-defined protein arrays over a metal surface. To illustrate this concept, we designed a fusion protein that simultaneously displays a red fluorescence protein (DsRed-monomer), which is highly selective for copper ions, and a gold binding peptide AuBP. The peptide tag, AuBP, self-directs the organization of DsRed-monomer protein onto a gold surface and forms arrays built upon an efficient control of the organic/inorganic interface at the molecular level. The peptide-assisted design offers a modular approach for fabrication of fluorescent-based protein arrays with copper ion sensing ability.

Bone is a natural protein (collagen)-mineral (hydroxyapatite) nanocomposite with hierarchically organized structure. Our previous work has demonstrated orientational differences in stoichiometry of hydroxyapatite resulting from orientationally dependent collagen-mineral interactions in bone. The nature of these interactions has been investigated both through molecular dynamics simulations as well as nanomechanical and infrared spectroscopic experiments. In this study, we report experimental studies on human cortical bone with osteogenesis imperfecta (OI), a disease characterized by fragility of bones and other tissues rich in type I collagen. About 90% of OI cases result from causative variant in one of the two structural genes (COL1A1 or COL1A2) for type I procollagens. OI provides an interesting platform for investigating how alterations of collagen at the molecular level cause changes in structure and mechanics of bone. Fourier transform spectroscopy, electron microscopy (SEM), and nanomechanical experiments describe the structural and molecular differences in bone ultrastructure due to presence of diseases. Photoacoustic-Fourier transform infrared spectroscopy (PA-FTIR) experiments have been conducted to investigate the orientational differences in molecular structure of OI bone, which is also compared with that of healthy human cortical bone. Further, in situ SEM static nanomechanical testing is conducted in the transverse and longitudinal directions in the OI bone. Microstructural defects and abnormities of OI bone were ascertained using scanning electron microscopies. These results provide an insight into molecular basis of deformation and mechanical behavior of healthy human bone and OI bone.

This paper presents the first attempts to study the large conductance mechano-sensitive channel (MscL) activity in an artificial droplet interface bilayer (DIB) system. A novel and simple technique is developed to characterize the behavior of an artificial lipid bilayer interface containing mechano-sensitive (MS) channels. The experimental setup is assembled on an inverted microscope and consists of two micropipettes filled with PEG-DMA hydrogel and containing Ag/AgCl wires, a cylindrical oil reservoir glued on top of a thin acrylic sheet, and a piezoelectric oscillator actuator. By using this technique, dynamic tension can be applied by oscillating axial motion of one droplet, producing deformation of both droplets and area changes of the DIB interface. The tension in the artificial membrane will cause the MS channels to gate, resulting in an increase in the conductance levels of the membrane. The results show that the MS channels are able to gate under an applied dynamic tension. Moreover, it can be concluded that the response of channel activity to mechanical stimuli is voltage-dependent and highly related to the frequency and amplitude of oscillations.

In this study, biodegradable foams were produced using cellulose nanofibrils (CNFs) and starch (S). The availability of high volumes of CNFs at lower costs is rapidly progressing with advances in pilot-scale and commercial facilities. The foams were produced using a freeze-drying process with CNF/S water suspensions ranging from 1 to 7.5 wt. % solids content. Microscopic evaluation showed that the foams have a microcellular structure and that the foam walls are covered with CNF`s. The CNF's had diameters ranging from 30 nm to 100 nm. Pore sizes within the foam walls ranged from 20 nm to 100 nm. The materials` densities ranging from 0.012 to 0.082 g/cm3 with corresponding porosities between 93.46% and 99.10%. Thermal conductivity ranged from 0.041 to 0.054 W/m-K. The mechanical performance of the foams produced from the starch control was extremely low and the material was very friable. The addition of CNF's to starch was required to produce foams, which exhibited structural integrity. The mechanical properties of materials were positively correlated with solids content and CNF/S ratios. The mechanical and thermal properties for the foams produced in this study appear promising for applications such as insulation and packaging.

Study on oxidizing cellulose scaffold to dialdehyde cellulose by sodium periodate (NaIO4) was carried out. Concentration of sodium periodate and the reaction time were effected for aldehyde introduction to cellulose scaffolds. Cellulose powder was dissolved in 1-butyl-3-methylimidazolium chloride, an ionic liquid, at 100°C and maintained at room temperature for 7 days, providing flexible cellulose scaffold. The cellulose scaffold was oxidized using periodate oxidation (Malaprade oxidation), which oxidizes carbohydrate by glycol cleavage to provide dialdehyde. Aldehyde groups introduced into cellulose were quantified by simple iodometry. Oxidized cellulose scaffold was degraded in the amino acid solution triggered by the reaction between aldehyde groups and amino groups. During immersion of the cellulose scaffolds in the amino acid solution, the mass loss of the scaffolds was evaluated by measuring of weight of oxidized cellulose scaffold before and after degradation.

The iridescent colors of Morpho butterflies have captured scientific intrigue for over a century. However, only recently photonic structures of the wing scales of Morpho butterflies have inspired new ideas in the diverse areas of technology including sensing. In this study, we performed theoretical and experimental evaluation of vapor-induced reflectance changes of the Morpho scales. These experiments provided additional details of the origin and the magnitude of vapor response selectivity in these natural photonic nanostructures and facilitated our design and fabrication of highly selective biomimetic photonic nanostructures.

Black liquor is a by-product of the paper mill Kraft process that deserves more valorization than its present use as low-grade fuel. In this work, SiC/C composite foams were prepared for the first time from concentrated emulsions by carbothermal reduction of bio-sourced precursors combining sodium silicate by lignin at 1400°C. The composition of the materials was determined by XRD, FTIR and Raman analyses. Their porous structure was characterized by SEM, mercury intrusion porosimetry, and nitrogen sorption, while their thermal properties were measured by TGA and dynamic DSC. Concerning their heat transport properties, we found out that when the starting lignin content was increased, the final C/Si ratio, the specific surface area and the heat diffusivity increased as well. Its high values were attributed to a cooperative effect between radiative heat transfer and the presence of partially graphitized carbon.

B-type carbonate apatite (B-CAp) powders were prepared by a wet method using Ca(OH)2 suspension and H3PO4 solution including NaHCO3 as a carbonate source, and porous B-CAp ceramics with two different amounts of carbonate contents were fabricated by sintering freeze-dried mixtures of the powders and gelatin composite. The porous B-CAp ceramics prepared had three-dimensionally interconnected pores. The sinterability of B-CAp ceramics was dependent on the chemical composition, especially sodium content and vacancy of OH site, and the carbonate contents did not directly influence the dissolution rate of porous B-CAp ceramics.

Swimming is dynamically a part of the hydrodynamic field and can be considered as a field of the optimal control motion. Animals move by instinct according to the situation which they are confronting with. Therefore, their instinctive motion is optimal most of the time. The movement of animals can be classified roughly into two kinds: the fast motion with the maximum speed and the motion with the minimum energy consumption. Considering the foreleg of the soft shelled turtle as a flat plane, several sets of movement of the foreleg were observed and calculated theoretically. The theoretical results agreed the observation results in the both cases with the maximum speed and the minimum energy consumption. Applying the theoretical movement of the soft shelled turtle foreleg to human movement in swimming, the general S-shaped pull stroke is the minimum energy consumption motion in free-style. It became clear that there was a different stroke for generating the maximum speed in free-style. That was the soft shelled turtle style of fast swimming, the I-shaped pull strokes. In 2002 when the author announced this theory, there was only one fast swimmer whose free-style swimming strokes coincidentally accorded with the I-shaped pull with fewer numbers of strokes at that time. He was the Olympic gold medalist Ian Thorp. Now the I-shaped stoke has become main stream in free style.

Several strategies have been explored from viewpoint of biomimetics to accomplish artificial photosynthesis by using macromolecules as a medium such as liposomes, supramolecules, and hydrogels.1 Differing from disordered solution systems in which multiple components such as photosensitizer and catalytic nanoparticle are diffusively mixed, the photochemical reactions occur efficiently in medium due to maintenance of the dipersibility of the components and specific molecular arrangement. Here we attempt to clarify the effect of medium hierarchy for photoinduced electronic transmission among multiple components. By conjugating each component on tubulin and integrating them via self-assembly to microtubules, ideal component arrangements with optimum distance for the electronic transmission will be possible.

Silicon has been used as one of the primary substrates for micro-machined intra-cortical neural implants (INI). The presence of various ions in the extracellular environment combined with cellular biological activity establishes a harsh, corrosive environment in the brain for INI, and as such, a long-term implant’s construction materials must be able to resist these environments. We have examined if environmental components could contribute to changes in the material, which in turn may be a contributing factor to the decreased long-term reliability in INI optimal neural recordings, which have prevented clinical use these devices for the last 4 decades. We tested silicon in artificial cerebrospinal fluid (ACSF), Dulbecco's modified eagle medium (DMEM), and H4 cells cultured within DMEM for 96 hours at 37°C as three various physiological environments to investigate the material degradation. We have observed that Si samples immersed in only DMEM and ACSF showed very minor surface alterations. However, Si samples cultured with H4 cells exhibited a large change in surface roughness from 0.24±0.04 nm to 4.85 nm. The scanning electron microscope (SEM) micrographs showed the presence of pyramid shaped pits. Further characterization with atomic force microscope (AFM) verified this result and quantified the severe changes in the surface roughness of these samples. At this initial stage of the investigation, we are endeavoring to identify the cause of these changes to the Si surface, but based on our observations, we believe that the increased corrosion could be result of chemical products released into the surrounding environment by the cells.

Topographical features are known to influence the axonal outgrowth of neurons. Understanding what kinds of topographical features are most effective at growth cone guidance and how outgrowth responds to these structures is of great importance to the study of nerve regeneration. To this end we analyze axonal outgrowth on tilted nanorod substrates which have been shown to impart directional bias to neuron growth. We utilize the Atomic Force Microscope to characterize the surface features present on these substrates and how such features are influencing the axonal outgrowth. Additionally, using a model which considers the neuronal growth cone as an object influenced by an effective potential we determine an effective force imparted on the growth cone by the surface topography.

Neuromodulation devices such as deep brain stimulators (DBS), spinal cord stimulators (SCS) and cochlear implants (CIs) use electrodes in contact with tissue to deliver electrical pulses to targeted cells. In general, the neuromodulation industry has been evolving towards smaller, less invasive devices. Improving power efficiency of these devices can reduce battery storage requirements. Neuromodulation devices can realize significant power savings if the impedance to charge transfer at the electrode-tissue interface can be reduced. High electrochemical impedance at the surface of stimulation microelectrodes results in larger polarization voltages. Decreasing this polarization voltage response can reduce power required to deliver the current pulse. One approach to doing this is to reduce the electrochemical impedance at the electrode surface. Previously we have reported on a novel electrochemically deposited 60:40% platinum-iridium (Pt-Ir) electrode material that lowered the electrode impedance by two orders of magnitude or more.

This study compares power consumption of an electrochemically deposited Pt-Ir stimulating microelectrode to that of standard Pt-Ir probe microelectrode produced using conventional techniques. Both electrodes were tested using in-vitro in phosphate buffered saline (PBS) solution and in-vivo (live rat) models.

Long-term functionality and stability of neural interfaces with complex geometries is one of the major challenges for chronic clinic applications due to lack of effective encapsulation. We present an encapsulation method that combines atomic layer deposited Al2O3 and Parylene C for encapsulation of biomedical implantable devices, focusing on its application on Utah electrode array based neural interfaces. The alumina and Parylene C bi-layer encapsulated wired Utah electrode array showed relatively stable impedance during the 960 equivalent soaking days at 37 °C in phosphate buffered solution. For the bi-layer coated wireless neural interfaces, the power-up frequency was constantly ∼ 910 MHz and the RF signal strength was stably around -73 dBm during equivalent soaking time of 1044 days at 37 °C (still under soak testing).
