Recent experiments and simulations have sparked growing interest in the study of Rayleigh–Bénard convection in very slender cells. One pivotal inquiry arising from this interest is the elucidation of the flow structure within these very slender cells. Here we employ tomographic particle image velocimetry, for the first time, to capture experimentally the full-field three-dimensional and three-component velocity field in a very slender cylindrical cell with aspect ratio $\Gamma =1/10$. The experiments cover a Rayleigh number range $5.0 \times 10^8 \leqslant Ra \leqslant 5.0 \times 10^9$ and Prandtl number 5.7. Our experiments reveal that the flow structure in the $\Gamma =1/10$ cell is neither in the multiple-roll form nor in the simple helical form; instead, the ascending and descending flows can intersect and cross each other, resulting in the crossing events. These crossing events separate the flow into segments; within each segment, the ascending and descending flows ascend or descend side by side vertically or in the twisting manner, and the twisting is not unidirectional, while the segments near the boundary can also be in the form of a donut like structure. By applying the mode decomposition analyses to the measured three-dimensional velocity fields, we identified the crossing events as well as the twisting events for each instantaneous flow field. Statistical analysis of the modes reveals that as $Ra$ increases, the average length of the segments becomes smaller, and the average number of segments increases from 2.5 to 3.9 in the $Ra$ range of our experiments.

We present an experimental study on the drag reduction by polymers in Taylor–Couette turbulence at Reynolds numbers ($Re$) ranging from $4\times 10^3$ to $2.5\times 10^4$. In this $Re$ regime, the Taylor vortex is present and accounts for more than 50 % of the total angular velocity flux. Polyacrylamide polymers with two different average molecular weights are used. It is found that the drag reduction rate increases with polymer concentration and approaches the maximum drag reduction (MDR) limit. At MDR, the friction factor follows the $-0.58$ scaling, i.e. $C_f \sim Re^{-0.58}$, similar to channel/pipe flows. However, the drag reduction rate is about $20\,\%$ at MDR, which is much lower than that in channel/pipe flows at comparable $Re$. We also find that the Reynolds shear stress does not vanish and the slope of the mean azimuthal velocity profile in the logarithmic layer remains unchanged at MDR. These behaviours are reminiscent of the low drag reduction regime reported in channel flow (Warholic et al., Exp. Fluids, vol. 27, no. 5, 1999, pp. 461–472). We reveal that the lower drag reduction rate originates from the fact that polymers strongly suppress the turbulent flow while only slightly weaken the mean Taylor vortex. We further show that polymers steady the velocity boundary layer and suppress the small-scale Görtler vortices in the near-wall region. The former effect reduces the emission rate of both intense fast and slow plumes detached from the boundary layer, resulting in less flux transport from the inner cylinder to the outer one and reduces energy input into the bulk turbulent flow. Our results suggest that in turbulent flows, where secondary flow structures are statistically persistent and dominate the global transport properties of the system, the drag reduction efficiency of polymer additives is significantly diminished.

Montmorillonite (Mnt)-based solid acids have a wide range of applications in catalysis and adsorption of pollutants. For such solid acids, the acidic characteristic often plays a significant role in these applications. The objective of the current study was to examine the effects of H3PO4-activation and supporting WO3 on the textural structure and surface acidic properties of Mnt. The Mnt-based solid acid materials were prepared by H3PO4 treatment and an impregnation method with a solution of ammonium metatungstate (AMT) and were examined as catalysts in the dehydration of glycerol to acrolein. The catalysts were characterized by nitrogen adsorption-desorption, powder X-ray diffraction (XRD), Fourier-transform infrared (FTIR) spectroscopy, scanning electronic microscopy (SEM), X-ray photoelectron spectroscopy (XPS), diffuse reflectance ultraviolet-visible (DR UV-Vis) spectroscopy, temperature programmed desorption of NH3 (NH3-TPD), diffuse reflectance Fourier-transform infrared (DR FTIR) spectroscopy of adsorbed pyridine, and thermogravimetric (TG) analyses. The phosphoric acid treatment of Mnt created Brönsted and Lewis acid sites and led to increases in specific surface areas, porosity, and acidity. WO3 species influenced total acidity, acid strength, the numbers of Brönsted and Lewis acid sites, and catalytic performances. A high turnover frequency (TOF) value (31.2 h−1) based on a maximal 60.7% yield of acrolein was reached. The correlation of acrolein yield with acidic properties indicated that the cooperative role of Brönsted and Lewis acid sites was beneficial to the formation of acrolein and a little coke deposition (<3.3 wt.%). This work provides a new idea for the design of solid acid catalysts with cooperative Brönsted and Lewis acidity for the dehydration of glycerol.

Cysticercosis caused by the metacestode larval stage of Taenia hydatigena formerly referred to as Cysticercus tenuicollis is a disease of veterinary importance that constitutes a significant threat to livestock production worldwide, especially in endemic regions due to condemnation of visceral organs and mortality rate of infected young animals. While the genetic diversity among parasites is found to be potentially useful in many areas of research including molecular diagnostics, epidemiology and control, that of T. hydatigena across the globe remains poorly understood. In this study, analysis of the mitochondrial DNA (mtDNA) of adult worms and larval stages of T. hydatigena isolated from dogs, sheep and a wild boar in China showed that the population structure consists of two major haplogroups with very high nucleotide substitutions involving synonymous and non-synonymous changes. Compared with other cestodes such as Echinococcus spp., the genetic variation observed between the haplogroups is sufficient for the assignment of major haplotype or genotype division as both groups showed a total of 166 point-mutation differences between the 12 mitochondrial protein-coding gene sequences. Preliminary analysis of a nuclear protein-coding gene (pepck) did not reveal any peculiar changes between both groups which suggests that these variants may only differ in their mitochondrial makeup.

We present an experimental study on controlling the number of vortices and the torque in a Taylor–Couette flow of water for Reynolds numbers from 660 to 1320. Different flow states are achieved in the annulus of width $d$ between the inner rotating and outer stationary cylinders through manipulating the initial height of the water annulus. We show that the torque exerted on the inner cylinder of the Taylor–Couette system can be reduced by up to 20 % by controlling the flow at a state where fewer than the nominal number of vortices develop between the cylinders. This flow state is achieved by starting the system with an initial water annulus height $h_0$ (which nominally corresponds to $h_0/d$ vortices), then gradually adding water into the annulus while the inner cylinder keeps rotating. During this filling process the flow topology is so persistent that the number of vortices does not increase; instead, the vortices are greatly stretched in the axial (vertical) direction. We show that this state with stretched vortices is sustainable until the vortices are stretched to around 2.05 times their nominal size. Our experiments reveal that by manipulating the initial height of the liquid annulus we are able to generate different flow states and demonstrate how the different flow states manifest themselves in global momentum transport.

This paper presents new LA-ICP-MS zircon U–Pb chronology, whole-rock geochemical and zircon Hf isotopic data for the felsic lavas of the Huili Group from the southwestern Yangtze Block. LA-ICP-MS zircon U–Pb dating shows that these rocks were emplaced in Late Mesoproterozoic time (∼1028 to 1019 Ma). Relative to typical I-type and S-type granitoids, all the samples are characterized by low Sr and Eu, and high high-field-strength element contents, high TFeO/MgO, enriched rare earth element compositions and negative Eu anomalies, indicating that they share the geochemical signatures of A-type granitoid. They can be further divided into two groups: Group I and Group II. Group I are A1-type felsic rocks and were produced by fractional crystallization of alkaline basaltic magmas. The Group II felsic lavas belong to the A2-type and were derived by partial melting of a crustal source with mixing of mantle-derived magmas. Both Group I and Group II felsic lavas may erupt in a continental back-arc setting. The coexistence of A1- and A2-type rocks in the southwestern Yangtze Block suggests that they can occur in the same tectonic setting.

We present experimental studies of higher-order modes of the flow in turbulent thermal convection in cells of aspect ratio ($\unicode[STIX]{x1D6E4}$) 1 and 0.5. The working fluid is water with the Prandtl number ($Pr$) kept at around 5.0. The Rayleigh number ($Ra$) ranges from $9\times 10^{8}$ to $6\times 10^{9}$ for $\unicode[STIX]{x1D6E4}=1$ and from $1.6\times 10^{10}$ to $7.2\times 10^{10}$ for $\unicode[STIX]{x1D6E4}=0.5$. We found that in $\unicode[STIX]{x1D6E4}=1$ cells, the first mode, which corresponds to the large-scale circulation (LSC), dominates the flow. The second mode (quadrupole mode), the third mode (sextupole mode) and the fourth mode (octupole mode) are very weak, on average these higher-order modes each contains less than 4 % of the total flow energy. In $\unicode[STIX]{x1D6E4}=0.5$ cells, the first mode is still the strongest but less dominant, the second mode becomes stronger which contains 13.7 % of the total flow energy and the third and the fourth modes are also stronger (containing 6.5 % and 1.1 % of the total flow energy respectively). It is found that during a reversal/cessation, the amplitude of the second mode and the remaining modes experiences a rapid increase followed by a decrease, which is opposite to the behaviour of the amplitude of the first mode – it decreases to almost zero then rebounds. In addition, it is found that during the cessation (reversal) of the LSC, the second mode dominates, containing 51.3 % (50.1 %) of the total flow energy, which reveals that the commonly called cessation event is not the cessation of the entire flow but only the cessation of the first mode (LSC). The experiment reveals that the second mode and the remaining higher-order modes play important roles in the dynamical process of the reversal/cessation of the LSC. We also show direct evidence that the first mode is more efficient for heat transfer. Furthermore, our study reveals that, during the cessation/reversal of the LSC, $Nu$ drops to its local minimum and the minimum of $Nu$ is ahead of the minimum of the amplitude of the LSC; and reversals can be distinguished from cessations in terms of global heat transport. A direct velocity measurement reveals the flow structure of the first- and higher-order modes.

The present study was performed to identify the genotype of a hypertrophic cardiomyopathy family and investigate the clinicopathogenic characteristics and prognostic features of relevant genetic abnormalities. Target sequence capture sequencing was performed to screen for pathogenic alleles in a 32-year-old female patient (proband). Sanger sequencing was carried out to verify the results. Sanger sequencing was also performed on other family members to identify allele carriers. A survival analysis was carried out using published literature and our findings. We found that the proband and her son harboured a Gly716Arg sequence variant of the β-myosin heavy chain. Neither the proband’s father nor the mother were carriers of this sequence variant; thus, the mutation was classified as “de novo”. Further survival analysis revealed that female patients appear to have a longer life expectancy compared with males. Our study may provide an effective approach for the genetic diagnosis of hypertrophic cardiomyopathy.

China is home to three subspecies of tiger Panthera tigris but there are no estimates of the size of any of the populations. We detected a population of the Endangered Amur tiger Panthera tigris altaica in Hunchun Nature Reserve in Jilin Province using both mitochondrial DNA and nuclear microsatellite loci. Four male and one female tigers were detected, indicating the potential for a small breeding group. However, genetic diversity was low overall, with six loci showing a heterozygote deficiency and a mean of 2.55 alleles per locus. This study is the first estimate of the wild Amur tiger population in China to use non-invasive techniques, and the presence of a female tiger indicates this is a potentially viable population. We provide baseline genetic diversity estimates to support monitoring of the population. The small number of tiger scats located indicates the importance of continuing the current conservation efforts for this tiger subspecies in Hunchun Nature Reserve. Such efforts include reducing poaching of tigers and their prey, and implementation of management plans to encourage the persistence and recovery of tigers in this area.

In this paper, we demonstrate a thin film Cu–Ni–Nb alloy deposited directly on silicon, without a designated barrier, showing very high thermal stability at a temperature up to 700 °C for 1 h. Thin [Nb–Ni12]Cux films were sputter deposited and annealed, and their material and electrical properties were studied. The results can be explained by the “cluster-plus-glue atom” model for stable solid solutions, where [Nb–Ni12] cuboctahedral clusters are embedded in a Cu matrix. In this model, the clusters are congruent with the Cu minimizing atomic interactions allowing a good stability. The properties of the films were found to be affected by the Ni/Nb ratios. Especially, the (Nb1.2/13.2Ni12/13.2)0.3Cu99.7 film annealed at 500 °C for 1 h had the lowest electrical resistivity of about 2.7 μΩ cm. And even after 40 h annealing at 500 °C, it maintained a low resistivity of about 2.8 μΩ cm, demonstrating extremely high stabilities against silicide formation.

Protein tyrosine phosphatase 1B (PTP1B) is implicated in the negative regulation of the insulin signalling pathway by dephosphorylating the insulin receptor (IR) and IR substrates. Ganodermalucidum has traditionally been used for the treatment of diabetes in Chinese medicine; however, its anti-diabetic potency and mechanism in vivo is still unclear. Our previously published study reported a novel proteoglycan PTP1B inhibitor, named Fudan-Yueyang-Ganoderma lucidum (FYGL) from G. lucidum, with a half-maximal inhibitory concentration (IC50) value of 5·12 (sem 0·05) μg/ml, a protein:polyglycan ratio of 17:77 and 78 % glucose in polysaccharide, and dominant amino acid residues of aspartic acid, glycine, glutamic acid, alanine, serine and threonine in protein. FYGL is capable of decreasing plasma glucose in streptozotocin-induced diabetic mice with a high safety of median lethal dose (LD50) of 6 g/kg. In the present study, C57BL/6 db/db diabetic mice were trialed further using FYGL as well as metformin for comparison. Oral treatment with FYGL in db/db diabetic mice for 4 weeks significantly (P < 0·01 or 0·05) decreased the fasting plasma glucose level, serum insulin concentration and the homeostasis model assessment of insulin resistance. FYGL also controlled the biochemistry indices relative to type 2 diabetes-accompanied lipidaemic disorders. Pharmacology research suggests that FYGL decreases the plasma glucose level by the mechanism of inhibiting PTP1B expression and activity, consequently, regulating the tyrosine phosphorylation level of the IR β-subunit and the level of hepatic glycogen, thus resulting in the improvement of insulin sensitivity. Therefore, FYGL is promising as an insulin sensitiser for the therapy of type 2 diabetes and accompanied dyslipidaemia.

In this paper, we study the one-dimensional wave equation with Boltzmann damping. Two different Boltzmann integrals that represent the memory of materials are considered. The spectral properties for both cases are thoroughly analyzed. It is found that when the memory of system is counted from the infinity, the spectrum of system contains a left half complex plane, which is sharp contrast to the most results in elastic vibration systems that the vibrating dynamics can be considered from the vibration frequency point of view. This suggests us to investigate the system with memory counted from the vibrating starting moment. In the latter case, it is shown that the spectrum of system determines completely the dynamic behavior of the vibration: there is a sequence of generalized eigenfunctions of the system, which forms a Riesz basis for the state space. As the consequences, the spectrum-determined growth condition and exponential stability are concluded. The results of this paper expositorily demonstrate the proper modeling the elastic systems with Boltzmann damping.

The DagA gene and DagA(▽), which is a DagA gene encoding sequence without signal peptide, were cloned from genome DNA of Pseudoalteromonas atlantica 19262 by polymerase chain reaction (PCR). After ligation with pET21 vector, DagA and DagA(▽) were respectively expressed in Escherichia coli ER2566 using molecular chaperones DsbC and FkpA. A strain of ER2566-pET21a-DagA(▽)-DsbC was screened as a highly effective expressing system in the form of an inclusion body that had the target protein with up to 60% total bacterial protein. DagA protein was renatured and purified by dissolving it in 8 mol/l of urea, using Ni-NTA resin affinity chromatography and refolding using the urea gradient method. DagA with a molecular weight of ~30.8 kDa was identified by sodium dodecyl sulphate polyacrylamide gel electrophoresis (SDS-PAGE) and had the ability to digest agarose. In a pH range of 4.8–6.8, DagA maintained a bioactivity greater than 60%, with 5.8 being the optimum pH, and it exhibited activity at temperatures from 37°C to 60°C, with 55°C being the optimum temperature.

Radar, aerial netting and ground sampling were used to study the autumn migration of Nilaparvata lugens (Stål) in Jiangsu Province in east central China. Emigration of macropterous adults increased from late August until late September as the main rice crop matured and was harvested. In early and mid September, the resulting windborne migrations carried the planthoppers mainly towards the west, although the migration directions ranged (within the western sector) from south to north. By late September, however, displacements were predominantly to the south-west on the then prevailing north-east monsoon winds: migration was particularly rapid when the north-easterlies were reinforced by typhoons. Although in late September such movements to more southerly latitudes are essential for the survival of the planthoppers' progeny, we found no definite evidence for preferential emigration on winds blowing towards the south. There were, however, indications that when winds towards the north occurred, the duration of migratory flight was curtailed. Irrespective of any possible preference for migration on northerlies, a large proportion of the N. lugens population would normally be carried in an adaptive southwards direction, because the advent of the north-east monsoon occurs at a time when the number of flight-ready planthoppers approaches its peak.
