Broad variations in dietary and physical activity patterns are part of nutritional transition concept. An additional nutritional transition has as main characteristic the change of consumption of processed foods for ultra-processed foods (UPF). This study aims to evaluate trends of UPF availability in Portuguese population and its association with diet-related non-communicable diseases (NCD) health indicators. This ecological study used data from the Household Budget Surveys conducted by the National Statistics Institute each 5 years within a national representative sample of households. The percentage of UPF was calculated based on the total daily amount of food and beverages available per capita (in grams). Data from the years 1990, 1995, 2000 and 2005 were used, which were retrieved from DAFNE-Anemos Software. NCD age-standardised mortality, prevalence and incidence were obtained from the Global Burden of Disease database, for the years 2000, 2005, 2010 and 2015. Between 1990 and 2005, the UPF availability increased from 3·9 % to 13·8 %. Over the years, almost all food and beverages categories increased the UPF availability contribution, mainly noticeable for milk, sugar, cereal and meat products. Positive correlations were observed between UPF availability and digestive diseases both in prevalence (r = 0·062; P = 0·037) and incidence (r = 0·005; P = 0·010) measures. Neoplasms incidence also showed positive correlation with UPF availability (r = 0·002; P = 0·012). Trends in UPF availability in Portugal increased exponentially. At the same time, there is a trend towards a decrease in unprocessed and processed food availability. The Portuguese population should be made aware of the health risks resulting from excessive consumption of UPF.

This study aims to investigate the dietary share of ultra-processed foods and its association with the non-communicable disease (NCD)-related nutrient profile of adult and elderly populations in Portugal. Cross-sectional data from the National Food, Nutrition and Physical Activity Survey (2015–2016) of the Portuguese population were analysed. Dietary intake was assessed by two non-consecutive 24-h recalls, and food items were classified according to the NOVA system. We estimated the percentage of dietary energy provided by each of the NOVA food groups and assessed the NCD-related nutrient profile of the overall diet across quintiles of ultra-processed food consumption. Weighted t tests, besides crude and adjusted linear and Poisson regressions, were performed. Ultra-processed foods contributed to around 24 and 16 % of daily energy intake for adults and elderly, respectively. In both groups, as the consumption of ultra-processed foods increased, the dietary content of free sugars, total fats and saturated fats increased, while the dietary content of protein decreased. In adults, total energy intake, dietary energy density and content of carbohydrates also increased as the consumption of ultra-processed foods increased, while the dietary content of fibre, Na and K decreased. The prevalence of those exceeding the upper limits recommended for free sugars and saturated fats increased by 544 and 153 % in adults and 619 and 60 % in elderly, when comparing the lowest with the highest quintile of ultra-processed food consumption. Such a scenario demands for effective strategies addressing food processing in the Portuguese population to improve their diet quality and prevention against diet-related NCD.

Holocene sediments from Nelson Lake, on Michigan's eastern Upper Peninsula, provide isotopic, pollen, and charcoal evidence for a two-step sequence of changes in moisture source and increased lake-effect precipitation during the late Holocene. Between 8000 and 5300 cal yr B.P., a warm, dry climate and zonal atmospheric circulation produced enriched stable oxygen and carbon isotopic values in combination with high percentages of pine pollen and sustained influx of charcoal particles. After 5300 cal yr B.P., decreasing isotopic values in marl and increasing pollen percentages of mesic hardwoods and northern white cedar indicate increased meridional air flow and precipitation from cold winter storms generated in Alberta, Canada. After 3000 cal yr B.P., abrupt declines in values of δ13C and δ18O and increased pollen representation of hemlock, American beech, spruce, and aquatic plants indicate paludification from increased lake-effect snowfall. The moisture was derived from the Great Lakes and transported by Alberta cyclonic storms that were steered across Lakes Superior and Michigan by a southward shift in the modal winter position of the polar jet stream.

Leishmaniasis is a vector-borne disease and an important public health issue. Glycosaminoglycan ligands in Leishmania parasites are potential targets for new strategies to control this disease. We report the subcellular distribution of heparin-binding proteins (HBPs) in Leishmania (Viannia) braziliensis and specific biochemical characteristics of L. (V.) braziliensis HBPs. Promastigotes were fractionated, and flagella and membrane samples were applied to HiTrap Heparin affinity chromatography columns. Heparin-bound fractions from flagella and membrane samples were designated HBP Ff and HBP Mf, respectively. Fraction HBP Ff presented a higher concentration of HBPs relative to HBP Mf, and SDS-PAGE analyses showed 2 major protein bands in both fractions (65 and 55 kDa). The 65 kDa band showed gelatinolytic activity and was sensitive to inhibition by 1,10-phenanthroline. The localization of HBPs on the promastigote surfaces was confirmed using surface plasmon resonance (SPR) biosensor analysis by binding the parasites to a heparin-coated sensor chip; that was inhibited in a dose-dependent manner by pre-incubating the parasites with variable concentrations of heparin, thus indicating distinct heparin-binding capacities for the two fractions. In conclusion, protein fractions isolated from either the flagella or membranes of L. (V.) braziliensis promastigotes have characteristics of metallo-proteinases and are able to bind to glycosaminoglycans.

Analyses of pollen and other terrestrial microfossils in sediment profiles from neotropical lakes can complement and extend archaeological studies by documenting the timing of prehistoric human disturbances within watersheds. However, assessing the scale of prehistoric human impact from sedimentary microfossil assemblages alone is often difficult. We explore here the utility of combining stable carbon isotope (δ13C) analyses of lake sediments and isotopie mixing models to improve our ability to gauge the extent of prehistoric human disturbance recorded in sediment profiles. Our test case involves the analysis o f a sediment core from Laguna Bonillita on the central Caribbean slope of Costa Rica that spans approximately the last 2,700 calendar years. Variations in the δ13C values of the Laguna Bonillita sediments suggest that human population growth and environmental impacts in the watershed were at their maximum ~cal yr 300 B.C. This finding is in keeping with archaeological evidence of rapid regional population growth at this time but differs from initial interpretations of the sediment record that were based on pollen and charcoal analyses alone. We believe that the use of stable carbon isotope data from sediment profiles can improve estimates of the scale of prehistoric human impact and in doing so improve the contributions of paleoecological research to archaeology.

Different techniques to assess bacterial community structure and diversity were evaluated in silages prepared with four different maize cultivars, three conventional and one transgenic (cv. Tundra, event Bt-176). Plants were cultivated in the greenhouse and harvested after 30 days of growth. Silage samples were collected at successive times during fermentation and analyzed for bacterial counts and by various DNA-based fingerprinting techniques. Bacterial counts were similar between cultivars for the total culturable bacteria, sporeforming, and mesophilic and thermophilic lactic acid bacteria (LAB). Further analysis of the species composition of 388 LAB strains by intergenic transcribed spacer (ITS) PCR followed by sequencing of 16S rRNA gene did not reveal differences between cultivars. In contrast, molecular fingerprinting methods targeting whole bacterial communities, such as automated ribosomal intergenic spacers analysis (ARISA) and 16S rRNA gene length heterogeneity-PCR (LH-PCR), indicated that different maize silage batches or cultivars hosted different bacterial communities. Thus, ARISA and LH-PCR fingerprinting techniques offer a fast and sensitive method to compare bacterial communities, and to detect differences in silage bacterial communities.

Paleosols Crop Out extensively in the Appalachian region of eastern North America, from the Canadian Maritime Provinces southward to the Tennessee-Alabama border along the western side of the Appalachian Orogen (Fig. 1). The paleosols occur primarily in terrigenous clastic redbed deposits ranging in age from Late Ordovician to Early Permian (Mora and Driese, 1999; Driese and Mora, in press), which encompass a time interval characterized by rapid evolution and diversification of terrestrial ecosystems (Table 1). These Appalachian paleosols formed under relatively constant sediment-source area and pedogenic environments (Table 1); thus they share generally uniform physical and chemical properties. As such, they are suitable particularly for investigating evolutionary advances in the gross morphology of plant roots and rhizomes, preserved as traces, and their relationship to physical and chemical features in the paleosols (Table 2). Important changes in plant abundance, rhizome and root systems, and adaptive capabilities are recorded in the paleosols, concurrent with a proxy record of changing environmental conditions. Paleosols represent a still-underutilized record of the terrestrial ecosystem (Retallack, 1990).
