Matrix methods may be applied to the analysis of experimental data concerning group structure when these data indicate relationships which can be depicted by line diagrams such as sociograms. One may introduce two concepts, n-chain and clique, which have simple relationships to the powers of certain matrices. Using them it is possible to determine the group structure by methods which are both faster and more certain than less systematic methods. This paper describes such a matrix method and applies it to the analysis of practical examples. At several points some unsolved problems in this field are indicated.

This article analyses the challenges that online marketplaces and e-commerce pose to traditional product liability doctrines. It uses a comparative perspective to examine whether an online platform can be liable to a consumer for a defective product purchased on its platform, and the adaption of product liability law to this challenge in a series of jurisdictions. It reflects on the role of litigation and regulation, focusing on Europe and the United States, and considers reform in a number of jurisdictions in this area. It concludes with proposals for increasing the accountability of online marketplaces for products sold on their websites.

Close-range sensors are employed to observe glaciological processes that operate over short timescales (e.g. iceberg calving, glacial lake outburst floods, diurnal surface melting). However, under poor weather conditions optical instruments fail while the operation of radar systems below 17 GHz do not have sufficient angular resolution to map glacier surfaces in detail. This letter reviews the potential of millimetre-wave radar at 94 GHz to obtain high-resolution 3-D measurements of glaciers under most weather conditions. We discuss the theory of 94 GHz radar for glaciology studies, demonstrate its potential to map a glacier calving front and summarise future research priorities.

Frontal ablation, the combination of submarine melting and iceberg calving, changes the geometry of a glacier's terminus, influencing glacier dynamics, the fate of upwelling plumes and the distribution of submarine meltwater input into the ocean. Directly observing frontal ablation and terminus morphology below the waterline is difficult, however, limiting our understanding of these coupled ice–ocean processes. To investigate the evolution of a tidewater glacier's submarine terminus, we combine 3-D multibeam point clouds of the subsurface ice face at LeConte Glacier, Alaska, with concurrent observations of environmental conditions during three field campaigns between 2016 and 2018. We observe terminus morphology that was predominately overcut (52% in August 2016, 63% in May 2017 and 74% in September 2018), accompanied by high multibeam sonar-derived melt rates (4.84 m d−1 in 2016, 1.13 m d−1 in 2017 and 1.85 m d−1 in 2018). We find that periods of high subglacial discharge lead to localized undercut discharge outlets, but adjacent to these outlets the terminus maintains significantly overcut geometry, with an ice ramp that protrudes 75 m into the fjord in 2017 and 125 m in 2018. Our data challenge the assumption that tidewater glacier termini are largely undercut during periods of high submarine melting.

In hedonic theories of motivation, ‘motivational affective states’ (MASs) are typically seen as adaptations which motivate certain types of behaviour, especially in situations where a flexible or learned response is more adaptive than a rigid or reflexive one. MASs can be negative (eg unpleasant feelings of hunger or pain) or positive (eg pleasant feelings associated with eating and playing). Hedonic theories often portray negative and positive MASs as opposite ends of a one-dimensional scale.

We suggest that natural selection has favoured negative and positive affect as separate processes to solve two different types of motivational problems. We propose that negative MASs (eg thirst, fear) evolved in response to ‘need situations’ where the fitness benefit of an action has increased, often because the action is needed to cope with a threat to survival or reproductive success. We propose that these negative MASs develop in response to a change in the body (eg dehydration) or the environment (eg the approach of a predator) which creates the need for action, and that negative MASs can become intense and prolonged if the threat to fitness is high and persistent. We propose that positive MASs evolved in ‘opportunity situations ‘ where an action (eg playing, exploring) has become advantageous because the fitness cost of performing it has declined. We propose that these positive MASs occur during, or as a result of, the performance of types of behaviour which are beneficial for fitness at a variety of times, not only when they are required to meet immediate needs; and that the pleasure inherent in the behaviour motivates the animal to perform it when the cost of so doing is sufficiently low. Some behaviour (eg eating) can be motivated by both positive and negative affect. Other behaviour, such as playing or fleeing from a predator, may be motivated largely by positive or negative affect alone. Our hypothesis needs to be tested, but we suggest that it corresponds well to common human experience.

The hypothesis provides a basis for predicting whether an aspect of animal management is likely to cause strong and prolonged negative affect (‘suffering’), or to prevent animals from experiencing certain types of pleasure. This distinction is important for bringing animal welfare assessment into line with ethical concerns.

For on-farm sheep welfare assessment, a reliable, simple and robust method is required to assess the level of flock lameness. This study examined the level of test agreement for two binary lameness scoring systems for sheep. The first was a group-level lameness assessment of sheep performed on ungathered sheep at pasture and was termed group observation method (GOM). The second method of lameness assessment was performed after gathering of the sheep and involved close observation of the gait of individual sheep in a handling pen and was termed individual animal gait assessment (IAGA). Following individual gait assessment, each sheep was also examined for the presence of specific foot and limb lesions: white line lesions (WL); inter-digital dermatitis (ID); footrot (FR); contagious digital dermatitis (CODD); toe granuloma (TG); and joint swellings (JS). A total of 3,074 sheep were assessed from 40 flocks in North England and Wales by one assessor. Test agreement between the assessment methods was found to be good as judged by linear regression and Bland-Altman plots. The method of group observation identified a slightly higher proportion of lame sheep compared to the individual animal examination and also appeared to be a more feasible on-farm method of observation. Over half of the sample sheep were identified with WL but this did not appear to be associated with a high level of lameness (as assessed by IAGA) with just under 12% of sheep with WL being identified as lame. In contrast, the percentage of lame sheep was most closely associated with CODD and over 80% of animals with this lesion were scored as lame.

A critical barrier to generating cumulative knowledge in political science and related disciplines is the inability of researchers to observe the results from the full set of research designs that scholars have conceptualized, implemented, and analyzed. For a variety of reasons, studies that produce null findings are especially likely to be unobserved, creating biases in publicly accessible research. While several approaches have been suggested to overcome this problem, none have yet proven adequate. We call for the establishment of a new discipline-wide norm in which scholars post short “null results reports” online that summarize their research designs, findings, and interpretations. To address the inevitable incentive problems that earlier proposals for reform were unable to overcome, we argue that decentralized research communities can spur the broader disciplinary norm change that would bring advantage to scientific advance. To facilitate our contribution, we offer a template for these reports that incorporates evaluation of the possible explanations for the null findings, including statistical power, measurement strategy, implementation issues, spillover/contamination, and flaws in theoretical priors. We illustrate the template’s utility with two experimental studies focused on the naturalization of immigrants in the United States and attitudes toward Syrian refugees in Jordan.

ABSTRACT IMPACT: We hope to provide a more nuanced understanding of the type-III IFN system, thereby exploring its therapeutic potential in the realm of infectious diseases. OBJECTIVES/GOALS: The role of IFNLR1 receptor dynamics and plasticity in regulating the type-III IFN response is largely unknown. As a specific, powerful component of innate immunity, understanding how the type-III IFN system is regulated could lead to the development of novel therapeutic targets and strategies to face a multitude of viral illnesses. METHODS/STUDY POPULATION: To facilitate our investigation, we will generate doxycycline-inducible FLAG-tagged IFNLR1-expression plasmids representing all known transcriptional variants. These plasmids will allow us to: 1) Evaluate the effect of IFNLR1 surface abundance on the type-III IFN transcriptional profile and 2) Assess the extent of IFNLR1-FLAG co-localization with several notable intracellular structures using immunofluorescence, before and after stimulation with IFNL3. RESULTS/ANTICIPATED RESULTS: We have successfully generated three IFNLR1-FLAG transcriptional variants and confirmed inducible-expression and function in vitro. We are currently assessing the role of surface abundance, internalization, differential isoform expression, and trafficking. DISCUSSION/SIGNIFICANCE OF FINDINGS: By completing this study, we hope to provide a more nuanced understanding of the type-III IFN system, thereby exploring its therapeutic potential in the realm of infectious diseases.

At the time of European contact, Torres Strait, New Guinea and northern Australia were home to highly restricted fraternities focused on warfare, headhunting and mortuary rituals. Masked dancers, representing spirits of the dead, initiated the next generation into secrets reputedly brought by a pantheon of wandering heroes, such as Waiat. A new project explores the deep history of Islander traditions, excavating initiation places associated with Waiat. In so doing, it demonstrates the advantages of collaborative history-building using archaeology and traditional knowledge.

The interpretations of relevant interfaces (i.e. the surface and bed) in radar sounding datasets over glaciers and ice sheets are primary boundary conditions in a variety of climate studies and particularly subglacial water routing models. It is therefore necessary to ensure these interpretations are consistent and not affected by cross-track clutter. For the surface interface, interferometry and a family of methods relying on digital elevation models have been used to successfully discriminate cross-track surface clutter. Here we present how interferometry can be applied to the problem of basal clutter from cross-track bed topography. Our approach is based on a comparison of the differential phases of ambiguous reflectors that may represent bed clutter and the differential phase of a reflector in an adjacent area that appears unaffected by basal clutter. The reflector yielding the smallest interferometric phase difference relative to the unambiguous bed reflector is considered to represent its consistent continuation. We successfully demonstrate our approach using 60 MHz center frequency MARFA data collected over Devon Ice Cap in the Canadian Arctic. Finally, we investigate the effects of clutter-affected and interferometry-corrected bed interpretations on ice layer thickness estimates, basal hydraulic head gradients and the potential extent of inferred subglacial water bodies.

The COllaborative project of Development of Anthropometrical measures in Twins (CODATwins) project is a large international collaborative effort to analyze individual-level phenotype data from twins in multiple cohorts from different environments. The main objective is to study factors that modify genetic and environmental variation of height, body mass index (BMI, kg/m2) and size at birth, and additionally to address other research questions such as long-term consequences of birth size. The project started in 2013 and is open to all twin projects in the world having height and weight measures on twins with information on zygosity. Thus far, 54 twin projects from 24 countries have provided individual-level data. The CODATwins database includes 489,981 twin individuals (228,635 complete twin pairs). Since many twin cohorts have collected longitudinal data, there is a total of 1,049,785 height and weight observations. For many cohorts, we also have information on birth weight and length, own smoking behavior and own or parental education. We found that the heritability estimates of height and BMI systematically changed from infancy to old age. Remarkably, only minor differences in the heritability estimates were found across cultural–geographic regions, measurement time and birth cohort for height and BMI. In addition to genetic epidemiological studies, we looked at associations of height and BMI with education, birth weight and smoking status. Within-family analyses examined differences within same-sex and opposite-sex dizygotic twins in birth size and later development. The CODATwins project demonstrates the feasibility and value of international collaboration to address gene-by-exposure interactions that require large sample sizes and address the effects of different exposures across time, geographical regions and socioeconomic status.

Radio-echo sounding (RES) can be used to understand ice-sheet processes, englacial flow structures and bed properties, making it one of the most popular tools in glaciological exploration. However, RES data are often subject to ‘strip noise’, caused by internal instrument noise and interference, and/or external environmental interference, which can hamper measurement and interpretation. For example, strip noise can result in reduced power from the bed, affecting the quality of ice thickness measurements and the characterization of subglacial conditions. Here, we present a method for removing strip noise based on combined wavelet and two-dimensional (2-D) Fourier filtering. First, we implement discrete wavelet decomposition on RES data to obtain multi-scale wavelet components. Then, 2-D discrete Fourier transform (DFT) spectral analysis is performed on components containing the noise. In the Fourier domain, the 2-D DFT spectrum of strip noise keeps its linear features and can be removed with a ‘targeted masking’ operation. Finally, inverse wavelet transforms are performed on all wavelet components, including strip-removed components, to restore the data with enhanced fidelity. Model tests and field-data processing demonstrate the method removes strip noise well and, incidentally, can remove the strong first reflector from the ice surface, thus improving the general quality of radar data.

Although federal regulation of vehicle fuel economy is often seen as environmental policy, over 70% of the estimated benefits of the 2017–2025 federal standards are savings in consumer expenditures on gasoline. Rational-choice economists question the counting of these benefits since studies show that the fuel efficiency of a car is reflected in its price at sale and resale. We contribute to this debate by exploring why most consumers in the United States do not purchase a proven fuel-saving innovation: the hybrid-electric vehicle (HEV). A database of 110 vehicle pairs is assembled where a consumer can choose a hybrid or gasoline version of virtually the same vehicle. Few choose the HEV. A total cost of ownership model is used to estimate payback periods for the price premiums associated with the HEV choice. In a majority of cases, a rational-choice explanation is sufficient to understand consumer disinterest in the HEV. However, in a significant minority of cases, a rational-choice explanation is not readily apparent, even when non-pecuniary attributes (e.g., performance and cargo space) are considered. Future research should examine, from a behavioral economics perspective, why consumers do not choose HEVs when pricing and payback periods appear to be favorable.

Corundum xenocrysts from alkaline basalt fields differ in characteristics and hence lithospheric origins. Trace element, fluid/melt inclusion and oxygen isotope studies on two eastern Australian corundum deposits are compared to consider their origins. Sapphires from Weldborough, NE Tasmania, are magmatic (high-Ga, av. 200 ppm) and dominated by Fe (av. 3300 ppm) and variable Ti (av. 400 ppm) as chromophores. They contain Cl, Fe, Ga, Ti and CO2-rich fluid inclusions and give δ18O values (5.1–6.2‰) of mantle range. Geochronology on companion zircons suggests several sources (from 290 Ma to 47 Ma) were disrupted by basaltic melts (47 ± 0.6 Ma). Gem corundums from Barrington, New South Wales, also include magmatic sapphires (Ga av. 170 ppm; δ18O (4.6–5.8‰), but with more Fe (av. 9000 ppm) and less Ti (av. 300 ppm) as chromophores. Zircon dating suggests that gem formation preceded and was overlapped by Cenozoic basaltic melt generation (59–4 Ma). In contrast, a metamorphic sapphire-ruby suite (low-Ga, av. 30 ppm) here incorporates greater Cr into the chromophores (up to 2250 ppm). Fluid inclusions are CO2-poor, but melt inclusions suggest some alkaline melt interaction. The δ18O values (5.1–6.2‰) overlap magmatic sapphire values. Interactions at contact zones (T = 780–940°C) between earlier Permian ultramafic bodies and later alkaline fluid activity may explain the formation of rubies.
