Objectives/Goals: Early childhood obesity is a major concern for Latin American children in the U.S., with gut barrier dysfunction as a key risk factor. Diet plays a role in gut development, but few studies have focused on Latin American infants. Our objective is to identify culturally relevant introductory foods that promote in vitro gut barrier development and function. Methods/Study Population: Pooled human milk (2.5 mL) from 6-month postpartum Hispanic mothers was combined with fruit and vegetable baby food products (2.5 g) and subjected to a 3-phase in vitro digestion system that simulates oral, gastric, and intestinal digestion. Digesta products were then anaerobically fermented for 24-hours using human stool inoculum, centrifuged, and filter sterilized. Intestinal epithelial cells (Caco-2, ATCC) were grown to confluence on 0.4 μm polystyrene transwell inserts using a DMEM + 10% FBS medium and allowed to differentiate for 21-days. Highly differentiated monolayers were treated with a 1:4 dilution of fermenta with medium in triplicate. The cell experiment was conducted twice. Cell layer integrity was measured using transepithelial electrical resistance (TEER) 24- and 48-hours after treatment. Results/Anticipated Results: Dietary intake data from the What We Eat in America database indicated that the top 3 fruit and vegetable exposures for infants with Mexican or Hispanic ethnicity were banana, apple, and carrot. Commercial baby food purees of these fruits and vegetables, in addition to baby foods with blueberry and spinach (Natural for Baby, Gerber Products Company) were acquired for digestion and fermentation experiments. Caco-2 cell experiments with these foods are ongoing. We expect Caco-2 monolayer incubated with fermenta from human milk and fruit or vegetables will have greater TEER values due to increased integrity of the cell layer as compared to those with breast milk alone. We also expect that exposure to fruit and vegetable fermenta will increase gene expression of tight junctions compared to exposure to media and human milk. Discussion/Significance of Impact: Using an in vitro digestion and fermentation system coupled with cell culture studies, we are identifying cellular mechanisms that link individual fruits and vegetables to gut barrier function. This will support translational work focused on mitigating obesity development in vulnerable populations.

Glacier ice flux is a key indicator of mass balance; therefore, accurate monitoring of ice dynamics is essential. Satellite-based methods are widely used for glacier velocity measurements but are limited by satellite revisit frequency. This study explores using seismic station internal GPS data to track glacier movement. While less accurate than differential GPS, this method offers high-temporal resolution as a by-product where seismic stations are deployed. Using a seismic station on Borebreen, Svalbard, we show that internal GPS provides reliable surface velocity measurements. When compared with satellite-inferred velocities, the results show a strong correlation, suggesting that the internal GPS, despite its inherent uncertainty, can serve as an efficient tool for glacier velocity monitoring. The high-temporal sampling reveals short-term dynamics of speed-up events and underscores the role of meltwater in driving these processes. This approach augments glacier observation networks at no additional cost.

Residual herbicides are primarily degraded in the soil through microbial breakdown. Any practices that result in increased soil biological activity, such as cover cropping (between cash crop seasons), could lead to a reduced persistence of herbicides in the soil. Furthermore, cover crops can also interfere with herbicide fate by interception. Field trials were conducted between 2020 and 2023, in a corn-soybean rotation, to investigate the influence of cover crop [cereal rye (Secale cereale L.) and crimson clover (Trifolium incarnatum L.) use on soil enzyme activities [β-glucosidase (BG) and dehydrogenase (DHA)], its effect on the concentration of residual herbicides (sulfentrazone, s-metolachlor, cloransulam-methyl, atrazine, and mesotrione) in the soil, and the interception of herbicides by cover crop residue. The use of cover crops occasionally resulted in increased BG and DHA activities relative to the fallow treatment. However, even when there was an increase in the activity of these two enzymes, increased degradation of the residual herbicides was not observed. The initial concentrations of all residual herbicides in the soil were significantly reduced due to interception by cereal rye biomass. Nevertheless, significant reductions in early season weed biomass were observed when residual herbicides were included in the tank mixture applied at cover crop termination relative to the application of glyphosate plus glufosinate. Results from this research suggests that the use of cereal rye or crimson clover as cover crops (between cash crop seasons) do not impact the persistence of residual herbicides in the soil nor reduce their efficacy in controlling weeds early in the growing season.

Low vitamin D associated with high parathyroid hormone (PTH) is common in HIV infection. We determined the association between total 25(OH)D and PTH in adolescents living with HIV, in Zambia and Zimbabwe. Adolescents (11–19 years) perinatally infected with HIV and established on antiretroviral therapy for ≥ 6 months were recruited into a cross-sectional study. Socio-demographic and clinical characteristics were recorded, anthropometry measured and fasted serum concentrations of 1,25(OH)2D, total 25(OH)D and intact PTH measured. The association between total 25(OH)D and PTH was examined using natural cubic spline regression. 842 participants (female: 53·2%) with a median age of 15·5 (IQR: 13·2–17·9) years were enrolled. Median antiretroviral therapy duration was 9·8 (IQR: 6·3–12·3) years, and 165/841 had an HIV viral-load >60 copies/ml. Stunting (height-for-age z-score <–2) and underweight (weight-for-age z-score <–2) were observed in 29·9 and 30·0%, respectively. Three-quarters reported daily Ca intakes <150 mg/d. The mean (sd) concentrations of total 25(OH)D and 1,25(OH)2D were 66·1(16·5) nmol/l and 210·6 (70·4) pmol/l, respectively, and median PTH level was 4·3 (IQR: 3·3–5·5) pmol/l. There was an inverse non-linear relationship between total 25(OH)D and PTH, 25(OH)D levelling off at 74·6 nmol/l (95 % CI: 74·5, 75·2). Results were consistent in those taking tenofovir disoproxil fumarate and virally unsuppressed participants. In this population with extremely low habitual Ca intakes, the lack of association between 25(OH)D and PTH when 25(OH)D exceeded 75 nmol/l potentially suggests that levels of 25(OH)D >75 nmol/l may need to be achieved to improve bone health; investigation is needed in future research studies.

The recommended first-line treatment for insomnia is cognitive behavioral therapy for insomnia (CBTi), but access is limited. Telehealth- or internet-delivered CBTi are alternative ways to increase access. To date, these intervention modalities have never been compared within a single study. Further, few studies have examined a) predictors of response to the different modalities, b) whether successfully treating insomnia can result in improvement of health-related biomarkers, and c) mechanisms of change in CBTi. This protocol was designed to compare the three CBTi modalities to each other and a waitlist control for adults aged 50-65 years (N = 100). Participants are randomly assigned to one of four study arms: in-person- (n=30), telehealth- (n=30) internet-delivered (n=30) CBTi, or 12-week waitlist control (n=10). Outcomes include self-reported insomnia symptom severity, polysomnography, circadian rhythms of activity and core body temperature, blood- and sweat-based biomarkers, cognitive functioning, and magnetic resonance imaging.

Field studies were conducted on certified organic land in Lafayette and Vincennes, IN, in 2023 to determine the impact of different between-row weed control methods on weed suppression and sweetpotato yield. Between-row treatments consisted of organic buckwheat (108 kg ha−1) broadcast seeded immediately after sweetpotato transplanting followed by silage tarping from 3 wk after transplanting (WATr) through harvest, organic buckwheat (108 kg ha−1) broadcast seeded 3 WATr and terminated 7 WATr, and cultivation as a grower standard. Weed density at 6 WATr was 0, 184, and 162 plants m−2 for the silage tarping, living mulch buckwheat, and cultivation treatments, respectively. Total yield was 11,048 kg ha−1 for the living mulch buckwheat, 19,792 kg ha−1 for the cultivation, and 17,814 kg ha−1 for the tarping treatments. Tarping effectively suppressed weeds and produced sweetpotato yields comparable to cultivation, indicating the potential for use by organic growers. When buckwheat was grown between rows 3 to 7 WATr, sweetpotato yield was lower than it was with tarping and cultivation. These results suggest that researchers should be evaluating tarps for small-acreage farmers as a weed management strategy.

Phylogenetic analysis demonstrates that Kuamaia lata, a helmetiid euarthropod from the lower Cambrian (Series 2, Stage 3) Chengjiang Konservat-Lagerstätte, nests robustly within Artiopoda, the euarthropod clade including trilobitomorphs. Microtomography of new specimens of K. lata reveals details of morphology, notably a six-segmented head and raptorial frontal appendages, the latter contrasting with filiform antennae considered to be a diagnostic character of Artiopoda. Phylogenetic analyses demonstrate that a raptorial frontal appendage is a symplesiomorphy for upper stem-group euarthropods, retained across a swathe of tree space, but evolved secondarily in K. lata from an antenna within Artiopoda. The phylogenetic position of K. lata adds support to a six-segmented head being an ancestral state for upper stem- and crown-group euarthropods.

The battles over official secrecy have to be fought on many different fronts. In the United Kingdom—whose ministers and public servants allegedly have a “passion for secrecy”—this was more than usually evident during 1967. Two pieces of legislation—namely, the Parliamentary Commissioner Act, 1967 (U.K.) and the Public Records Act, 1967 (Eng.)—represented a modest triumph for those who would wish to see more light shed upon the affairs of the nation. Elsewhere the picture was rather more familiar. The Security Commission recommended a further tightening of documentary security in the Cabinet Office; this followed upon the conviction under the Official Secrets Acts, 1911 to 1939 (U.K.) of a young typist in the Cabinet Office, “the first known occasion on which classified papers have been extracted from” this inner sanctum of government.

A nonparametric test of dispersion with paired replicates data is described which involves jackknifing logarithmic transformations of the ratio of variance estimates for the pre- and post-treatment populations. Results from a Monte Carlo simulation show that the test performs well under Ho and has good power properties. Examples are given of applying the procedure on psychiatric data.

The use of U-statistics based on rank correlation coefficients in estimating the strength of concordance among a group of rankers is examined for cases where the null hypothesis of random rankings is not tenable. The studentized U-statistics is asymptotically distribution-free, and the Student-t approximation is used for small and moderate sized samples. An approximate confidence interval is constructed for the strength of concordance. Monte Carlo results indicate that the Student-t approximation can be improved by estimating the degrees of freedom.

Indexes of skewness and kurtosis for a test-score distribution are expressed in terms of item parameters. Both are shown to depend, in part, on item means, variances, and covariances. The index of skewness depends also on trivariances. A trivariance is a product moment involving first powers of deviation scores for three items. The index of kurtosis depends on quadrivariances, as well as trivariances. A quadrivariance is a product moment involving first powers of deviation scores for four items. Empirical data are presented for responses of groups of subjects to 25 triads and 25 tetrads of items from five tests.

We present a re-discovery of G278.94+1.35a as possibly one of the largest known Galactic supernova remnants (SNRs) – that we name Diprotodon. While previously established as a Galactic SNR, Diprotodon is visible in our new Evolutionary Map of the Universe (EMU) and GaLactic and Extragalactic All-sky MWA (GLEAM) radio continuum images at an angular size of $3{{{{.\!^\circ}}}}33\times3{{{{.\!^\circ}}}}23$, much larger than previously measured. At the previously suggested distance of 2.7 kpc, this implies a diameter of 157$\times$152 pc. This size would qualify Diprotodon as the largest known SNR and pushes our estimates of SNR sizes to the upper limits. We investigate the environment in which the SNR is located and examine various scenarios that might explain such a large and relatively bright SNR appearance. We find that Diprotodon is most likely at a much closer distance of $\sim$1 kpc, implying its diameter is 58$\times$56 pc and it is in the radiative evolutionary phase. We also present a new Fermi-LAT data analysis that confirms the angular extent of the SNR in gamma rays. The origin of the high-energy emission remains somewhat puzzling, and the scenarios we explore reveal new puzzles, given this unexpected and unique observation of a seemingly evolved SNR having a hard GeV spectrum with no breaks. We explore both leptonic and hadronic scenarios, as well as the possibility that the high-energy emission arises from the leftover particle population of a historic pulsar wind nebula.
