This study aimed to analyze the clinical effects of microdissection testicular sperm extraction (micro-TESE) surgery combined with an intracytoplasmic sperm injection (ICSI) regimen in the treatment of non-obstructive azoospermia (NOA) patients with different etiologies. In total, 128 NOA patients participated in this study, in which they received infertility treatment by micro-TESE surgery combined with an ICSI regimen, and all patients were divided into three groups [the Klinefelter syndrome (KS), the idiopathic and the secondary NOA groups]. In addition, the sperm retrieval rate (SRR), fertilization rate, embryo development status and clinical treatment effects were analyzed. Among the 128 NOA patients, the SRR of KS NOA patients was 48.65%, those of idiopathic and the secondary patients were 33.82% and 73.91%, respectively. Regardless of etiologies, there was no correlation with age, hormone value or testicular volume. Further analysis showed that the SRR of the KS group was positively related with testosterone (T) values, and the SRR of the secondary group had a positive relationship with follicle-stimulating hormone or luteinizing hormone values. In the subsequent clinical treatment, the retrieved sperm was subjected to ICSI and achieved good treatment effects, especially in the secondary group, and the implantation rate (55.56%) and clinical pregnancy rate (68.42%) were both higher than those of the idiopathic group (28.75% and 40.00%) and KS group (22.05% and 30.77%). Micro-TESE surgery combined with ICSI insemination is the most effective treatment regimen for NOA patients. The SRR of NOA patients with different etiologies are related to certain specific factors, and micro-TESE surgery seems to be the ideal and only way to have biological children.

The aim of this study was to explore the status of thyroid peroxidase antibody (TPOAb) and thyroglobulin antibody (TGAb) in three areas with differing water iodine concentrations; and to discuss the relationships between these two thyroid antibodies and thyroid diseases in the three areas. We investigated 2503 adults from three areas. Urinary iodine concentrations, thyroid stimulating hormone (TSH), free thyroxine (FT4), free triiodothyronine (FT3), TPOAb, TGAb and thyroid volume (TV) were measured, and thyroid ultrasonography was performed. The positivity rates of TGAb(+), TPOAb(+) and TGAb(+) and TPOAb(+) or TGAb(+) were significantly higher in iodine fortification (IF) areas than iodine adequate (IA) areas (all P < 0·05). In IF and iodine excess areas, the positivity rates of TPOAb(+), TGAb(+) and TPOAb(+) or TGAb(+) significantly increased with age (all P for trend < 0·05). The levels of TSH, TV and the prevalence of overt hypothyroidism, subclinical hypothyroidism and goitre were significantly elevated in the thyroid antibody-positive groups in the three areas, but the FT3 was diminished (all P < 0·010). Positivity for TPOAb and TGAb was associated with an increased risk of subclinical hypothyroidism in the three areas. In areas with different median water iodine, positivity for both TPOAb and TGAb was associated with elevated TSH values. Notably, with the increased levels of TPOAb, the frequency of abnormally elevated TSH increased dramatically in the three areas.

The effects of wall-normal vibration on bypass transition in a boundary-layer flow over a flat plate with roughness elements in the form of circular cylinders are investigated using direct numerical simulations (DNS), linear Floquet analyses and dynamic mode decompositions (DMD). The vibration of the plate strengthens the streamwise vorticity, consequently enhancing the velocity streaks and reducing the critical Reynolds number for transition. A map is constructed to identify the coupling effect of the vibration amplitude and Reynolds number on transition. Among all investigated combinations of height and diameter of roughness, the critical Reynolds numbers at different vibration amplitude $A$ can be unified by a scaling function of $(1-10A)$. Two instability modes are identified in the vibration-induced transition process: a wake mode immediately downstream of the roughness, related to the inviscid Kelvin–Helmholtz instability of the wall-normal shear in the wake; and a streak mode, in response to the spanwise shear of the streaky flow, occurring further downstream. The first one results in the generation of central hairpin vortices which then feed the second one. Further development of the streak instability leads to two arrays of hairpin vortices. Results from linear Floquet analyses and DMD further confirm the two modes of instability observed in DNS. A quantitative study suggests that the amplification of vibration-induced disturbance by the base shear dominates the production of streamwise vorticity and subsequently the hairpin vortices.

Bypass transition in flow over a flat plate triggered by a pair of dielectric-barrier-discharge plasma actuators mounted on the plate surface and aligned in the streamwise direction is investigated. A four-species plasma–fluid model is used to model the electrohydrodynamic force generated by the plasma actuation. A pair of counter-rotating streamwise vortices is created downstream of the actuators, leading to the formation of a high-speed streak in the centre and two low-speed streaks on each side. As the length of actuators increases, more momentum is added to the boundary layer and eventually a turbulent wedge is generated at an almost fixed location. With large spanwise distance between the actuators (wide layout), direct numerical simulations indicate that the low-speed streaks on both sides lose secondary stability via an inclined varicose-like mode simultaneously, leaving a symmetric perturbation pattern with respect to the centre of the actuators. Further downstream, the perturbations are tilted by the mean shear of the high- and low-speed streaks and consequently a ‘W’-shaped pattern is observed. When the pair of plasma actuators is placed closer (narrow layout) in the spanwise direction, the mean shear in the centre becomes stronger and secondary instability first occurs on the high-speed streak with an asymmetric pattern. Inclined varicose-like and sinuous-like instabilities coexist in the following breakdown of the negative streaks on the side and the perturbations remain asymmetric with respect to the centre. Here the tilting of disturbances is dominated by the mean shear in the centre and the perturbations display a ‘V’ shape. Linear analysis techniques, including biglobal stability and transient growth, are performed to further examine the fluid physics; the aforementioned phenomena at narrow and wide layouts, such as the secondary instabilities, the ‘V’ and ‘W’ shapes, and the symmetric and asymmetric breakdown, are all observed.

The development of free-stream disturbances in flow over a vertically vibrating flat plate with a slender leading edge is investigated. The evolution of the optimal inflow perturbation that results in the maximum amplification is computed to investigate the effect of the plate vibration on the development of free-stream disturbance, secondary instability of streaks and subsequently the bypass transition to turbulence. It is observed that the plate vibration leads to periodic change of the angle of attack, shifting the free-stream disturbance to the upper or lower side of the plate. Therefore, the development of steady inflow perturbations, which receive the largest amplification, is interrupted by the vibration, and the perturbation amplification via the lift-up mechanism is weakened. The vibration brings a second peak of perturbation growth at the vibration frequency, leading to high-frequency free-stream perturbations penetrating into the base boundary layer, which is not observed in flow over a stationary plate owing to the sheltering mechanism. This resonance of the flow perturbation and the vibrating plate is explained by the staggering effect of the leading edge. Further, the direct numerical simulations with the optimal inflow perturbation imposed on the inflow boundary show that the vertical vibration of the plate leads to streamwise periodic vorticity near the edge of the boundary layer. This inhomogeneity of the streamwise vorticity brings about streamwisely localized distortion of the low-speed streaks and, thus, an intermittent secondary instability. Therefore, before the streaks break down to turbulence, they undergo several rounds of secondary instabilities, resulting in an elongated bypass transition process.

Lipopolysaccharides (LPS) could induce milk fat depression via regulating the body and blood fat metabolism. However, it is not completely clear how LPS might regulate triglyceride synthesis in dairy cow mammary epithelial cells (DCMECs). DCMECs were isolated and purified from dairy cow mammary tissue and treated with LPS. The level of triglyceride synthesis, the expression and activity of the liver X receptor α (LXRα), enzymes related to de novo fatty acid synthesis, and the expression of the fatty acid transporters were investigated. We found that LPS decreased the level of triglyceride synthesis via a down-regulation of the transcription, translation, and nuclear translocation level of the LXRα. The results also indicated that the transcription level of the LXRα target genes, sterol regulatory element binding protein 1 (SREBP1), fatty acid synthetase (FAS), acetyl-CoA carboxylase-1 (ACC1), were significantly down-regulated in DCMECs after LPS treatment. Our data may provide new insight into the mechanisms of milk fat depression caused by LPS.

The first case of 2019-nCoV pneumonia infection occurred in Wuhan, Hubei Province, South China Seafood Market in December 2019. As a group with a high probability of infection, health workers are faced with a certain degree of psychological challenges in the process of facing the epidemic. This study attempts to evaluate the impact of 2019-nCoV outbreak on the psychological state of Chinese health workers and to explore the influencing factors. During the period from 31 January 2020 to 4 February 2020, the ‘Questionnaire Star’ electronic questionnaire system was used to collect data. The 2019-nCoV impact questionnaire and The Impact of Event Scale (IES) were used to check the psychological status of health workers in China. A total of 442 valid data were collected in this study. Seventy-four (16.7%) male and 368 (83.3%) female individuals participated in this study. The average score of high arousal dimension was 5.15 (s.d. = 4.71), and the median score was 4.0 (IQR 2.0, 7.0). The average score of IES was 15.26 (s.d. = 11.23), and the median score was 13.5 (IQR 7.0, 21.0). Multiple regression analysis showed that there were critical statistical differences in high arousal scores among different gender groups (male 3.0 vs. female 5.0, P = 0.075). Whether being quarantined had significant statistical differences of IES scores (being quarantined 16.0 vs. not being quarantined 13.0, P = 0.021). The overall impact of the 2019-nCoV outbreak on health workers is at a mild level. Chinese health workers have good psychological coping ability in the face of public health emergencies.

Japanese foxtail (Alopecurus japonicus Steud.) is an invasive grass weed that severely threatens the production of wheat (Triticum aestivum L.) and canola (Brassica napus L.) crops in eastern Asia. Mesosulfuron-methyl is a highly efficient acetolactate synthase (ALS)-inhibiting herbicide widely used for control of this species in China. However, in recent years, some A. japonicus populations have evolved resistance to mesosulfuron-methyl by different amino acid substitutions (AASs) within the ALS gene. In the current study, 11 populations of A. japonicus were collected from Anhui Province, China, where the wheat fields were severely infested with this weed. Based on single-dose screening, eight of these populations evolved resistance to mesosulfuron-methyl, and gene sequencing revealed three AASs located in codon 197 or 574 of the ALS gene in the different resistant populations. Subsequently, three typical populations, AH-1, AH-4, and AH-10 with Trp-574-Leu, Pro-197-Thr, and Pro-197-Ser mutations, respectively, in ALS genes were selected to characterize their cross-resistance patterns to ALS inhibitors. Compared with the susceptible population AH-S, AH-1 showed broad-spectrum cross-resistance to sulfonylureas (SUs), imidazolinones (IMIs), triazolopyrimidines (TPs), and sulfonyl-aminocarbonyl-triazolinones (SCTs); whereas AH-4 and AH-10 were resistant to SUs, TPs, and SCTs but sensitive to IMIs. Moreover, all three resistant populations were sensitive to both photosystem II inhibitor isoproturon and 4-hydroxyphenylpyruvate dioxygenase inhibitor QYM201 (1-(2-chloro-3-(3-cyclopropyl-5-hydroxy-1-methyl-1H-pyrazole-4-carbonyl)-6-(trifluoromethyl)phenyl)piperidin-2-one). Based on the current state of knowledge, this study is the first report of A. japonicus evolving cross-resistance to ALS-inhibiting herbicides due to a Pro-197-Ser mutation in the ALS gene.

The aim of this study is to analyse the efficacy rate of folate for the treatment of hyperhomocysteinaemia (HHcy) and to explore how folate metabolism-related gene polymorphisms change its efficacy. This study also explored the effects of gene–gene and gene–environment interactions on the efficacy of folate. A prospective cohort study enrolling HHcy patients was performed. The subjects were treated with oral folate (5 mg/d) for 90 d. We analysed the efficacy rate of folate for the treatment of HHcy by measuring homocysteine (Hcy) levels after treatment. Unconditioned logistic regression was conducted to analyse the association between SNP and the efficacy of folic acid therapy for HHcy. The efficacy rate of folate therapy for HHcy was 56·41 %. The MTHFR rs1801133 CT genotype, TT genotype and T allele; the MTHFR rs1801131 AC genotype, CC genotype and C allele; the MTRR rs1801394 GA genotype, GG genotype and G allele; and the MTRR rs162036 AG genotype and AG+GG genotypes were associated with the efficacy of folic acid therapy for HHcy (P<0·05). No association was seen between other SNP and the efficacy of folic acid. The optimal model of gene–gene interactions was a two-factor interaction model including rs1801133 and rs1801394. The optimal model of gene–environment interaction was a three-factor interaction model including history of hypertension, history of CHD and rs1801133. Folate supplementation can effectively decrease Hcy level. However, almost half of HHcy patients failed to reach the normal range. The efficacy of folate therapy may be genetically regulated.

A callus induction and plantlet regeneration system for croftonweed was developed by studying the influence of explant type (leaf, stem, and nodal segment) and different concentrations of plant growth regulators. The leaf was a better explant for callogenesis compared to the stem. The highest callus induction frequency (87.2%) was obtained from leaf segments on Murashige and Skoog's medium (MS medium) supplemented with 0.5 mg/L (2,4-dichlorophenoxy)acetic acid and 2.0 mg/L 6-benzylaminopurine (BA), and 71.6% differentiation along with a multiplication rate of 4.1 adventitious shoots per callus was achieved with a combination of 0.5 mg/L 1-naphthaleneacetic acid (NAA) and 1.0 mg/L BA. In addition, MS medium supplemented with 0.5 mg/L NAA and 1.0 mg/L BA was the best medium for axillary shoot regeneration from nodal segments. Rhizogenesis of cultured shoots was satisfactorily obtained in half-strength MS without any growth regulators. The regenerated rooted plantlets were successfully acclimatized in soil where they grew normally without showing any morphological variation. These studies provide the prerequisite system for the development of genetic engineering in the future and propagating croftonweed rapidly for further study.

Areas with low, adequate and excessive I content in water co-exist in China. Limited data are currently available on I nutrition and thyroid disease in lactating women and their breast-fed infants with different I intakes. This study aimed to evaluate I nutrition in both lactating women and their infants and the prevalence of thyroid disease in areas with different levels of I in water. From January to June 2014, a total of 343 healthy lactating women (excluding those taking anti-thyroid drugs or I supplements within a year of the study, consuming seafood at the time of the study or those diagnosed with congenital thyroid disease) from Beihai in Guangxi province and Jiajiazhuang, Yangcheng, Jicun and Pingyao townships in Shanxi province were selected. Compared with the I-sufficient group, median urinary I concentrations in both lactating women and infants as well as breast milk I levels were significantly lower in the I-deficient group (P<0·001). The prevalence of thyroid disease in lactating women, particularly subclinical hypothyroidism, was higher in the I-excess group than in the I-sufficient group (P<0·05). In areas with excessive water I content, high thyroid peroxidase antibody and high thyroglobulin levels were risk factors for abnormal thyroid-stimulating hormone levels. Our data collectively suggest that excessive I intake potentially causes subclinical hypothyroidism in lactating women. Moreover, enhanced monitoring of I status is important to avoid adverse effects of I deficiency or excess, particularly in susceptible populations such as pregnant or lactating women and infants.

Titanium dioxide (TiO2), a widely used inorganic semiconductor owing to its superb photoelectric properties, has frequently been fabricated into composites to reduce its relatively large band gap and overcome its limited visible light absorption. In this article, a “layer-by-layer” method has been developed to prepare the composite structure of nitrogen (N)-doped graphene quantum dots (GQDs)-sensitized TiO2 nanofibers. The as-prepared structure shows considerable luminescence and exhibits excellent photoelectric properties. Various factors including the crystalline phase of TiO2, amount of N in GQDs, and irradiation wavelength were investigated to find the optimal conditions for enhanced photoelectric activity. It is demonstrated that the combination of highest N amount GQDs with TiO2 nanofibers of mixed phases (750 °C-sintered TiO2 nanofibers) possess the best photoelectric properties. The enhancement of properties using TiO2 nanofibers with mixed phases mainly contributes to the transfer of electrons between conduction bands of different phases in TiO2 and the distinctive photoluminescence (PL) property of N-GQDs. Furthermore, this enhancement can be achieved in most areas of the visible light range. The general mechanism of the electron generation and transfer of the structure is based on the normal PL and upconversion PL property of N-GQDs which serve as the sensitizer. We consider it a feasible method to improve the photoelectric conversion efficiency in photovoltaic devices.

Lymphatic stomata are small openings of lymphatic capillaries on the surface of the mesothelium that lines the serous cavity and have the function of active absorption. They play an important role in physiological and pathological conditions. The cavity of the tunica vaginalis is a typical serous cavity of the testis, but the lymphatic stomata of the tunica vaginalis of humans have never been reported. Here, we studied their ultrastructure by scanning and transmission electron microscopy. The submesothelial connective tissue with foramina was investigated after the mesothelial cells were digested using NaOH solution. We found the lymphatic stomata in cuboidal mesothelial cell regions of the parietal layer of the tunica vaginalis of humans with a diameter of about 1–2 μm. Sometimes, closed lymphatic stomata could be observed. Our study is the first to report the existence of lymphatic stomata of the tunica vaginalis of humans. We found that the tunica vaginalis cavity is connected with the lymphatic system through the stomata, which might provide a morphological basis for the drainage of hydrocele and tumor metastasis of the tunica vaginalis of humans.

Since the discovery of submarine hot vents in the late 1970s, it has been postulated that submarine hydrothermal environments would be suitable for emergence of life on Earth. To simulate warm spring conditions, we designed a series of microwave-assisted amino acid synthesis involving direct reactions between succinic acid and ammonia in the presence of the magnetite catalyst. These reactions which generated aspartic acid and glycine were carried out under mild temperatures and pressures (90–180 °C, 4–19 bar). We studied this specific reaction inasmuch as succinic acid and ammonia were traditionally identified as prebiotic compounds in primitive deep-sea hydrothermal systems on Earth. The experimental results were discussed in both biochemical and geochemical context to offer a possible route for abiotic amino acid synthesis. With extremely diluted starting materials (0.002 M carboxylic acid and 0.002 M ammonia) and catalyst loading, an obvious temperature dependency was observed in both cases [neither product was detected at 90 °C in comparison with 21.08 μmol L−1 (aspartic acid) and 70.25 umol L−1 (glycine) in 180 °C]. However, an opposite trend presented for reaction time factor, namely a positive correlation for glycine, but a negative one for aspartic acid.
