Introduction: Transcranial Direct current stimulation (tDCS) and repetitive transcranial magnetic stimulation (rTMS) are neuromodulatory techniques capable of modifying the altered cerebral hyperexcitability in amnestic mild cognitive impairment (aMCI). Cortical excitability can be estimated with motor evoked potentials (MEPs) and synaptic plasticity can be induced with a 5 Hz rTMS paradigm applied to the motor cortex (M1). An increase in MEP amplitude reflects a better capacity for plasticity in M1, and these measures can, in turn, be associated with cognitive performance. Cognitive stimulation (CS) and tDCS in aMCI can modify excitability and improve cognition.

Objectives: Study the effect of the combination of CS and tDCS (real vs. placebo) on cognitive performance and cortical excitability.

Methods: Randomized, double-blind, placebo-controlled clinical trial in aMCI. The diagnosis was established through a clinical evaluation by a psychogeriatrician and a neuropsychological assessment. To determine the effect of the interventions, evaluations were conducted at two time points: before (T0) and after administering 9 sessions of CS and 15 sessions of tDCS over three weeks (T1). The evaluations included: MEP amplitude, Montreal Cognitive Assessment (MoCA), and Screening for Cognitive Impairment in Psychiatry (SCIP-S). For data analysis, ARTool in RStudio was used to perform aligned rank transformation for non-parametric analysis of variance in factorial models with fixed and random effects, applying a factorial ANOVA for each response variable.

Results: A total of 18 participants were enrolled (real n = 8 and placebo n = 10). Comparing T0 and T1, differences were found in both groups in MEP amplitude after applying the paradigm (F = 5.479; p = 0.032) as well as in the total MoCA score (F = 4.808; p = 0.043). When comparing the groups, differences were found in the delayed verbal learning domain assessed with SCIP-S (F = 6.038; p = 0.025) and in MEP amplitude (F = 6.165; p = 0.024). No differences were found in any of the evaluations when studying the effect of the GroupxTimeinteraction.

Conclusions: Both groups benefit from cognitive stimulation, and the use of tDCS does not appear to enhance the cognitive effect or the MEPs. It seems that cognitive stimulation alone is capable of modifying cortical excitability and improving cognitive performance.

Control over the legislative messaging agenda has important political, electoral and policy consequences. Existing models of congressional agenda-setting suggest that national polarization drives the agenda. At the same time, models of home style and formal models of leadership hypothesize that legislators shift their messaging as they balance coordination and information problems. We say the coordination problem dominates when conditions incentivize legislators to agree on the same message rather than fail to reach consensus. Conversely, the information problem is said to dominate in circumstances where legislators prefer to say nothing at all rather than reach consensus on the wrong political message. Formal theories predict that when coordination problems are pressing, legislative members follow the policy positions of party leaders. When their party’s information problem is acute, party members instead rely on the wisdom of the caucus to set the party’s agenda. To test these theories, we analyze the Twitter accounts of U.S. House members with a Joint Sentiment Topic model, generating a new understanding of House leadership power. Our analyses reveal complex leader-follower relationships. Party leaders possess the power to substantially affect the propensity of rank-and-file members to discuss topics, especially when the coordination problem dominates; these effects are pronounced even when coordination problems are pressing. That said, when the underlying politics are unclear, rank-and-file members exert influence on the discussion of a topic because the information problem is more acute. At the same time and for these uncertain topics, leadership influence decreases, consistent with theory. We show these results are robust to the underlying dynamics of contemporary political discussion and context, including leading explanations for party leadership power, such as national polarization.

The in vitro culture of domestic cat embryos without the zona pellucida affects their implantation capacity. MicroRNAs (miRNAs) have an important role in embryo–maternal communication and implantation. The objective of this study was to evaluate the expression of specific miRNAs in domestic cat blastocysts cultured without the zona pellucida. Two experimental groups were done: (1) domestic cat embryos cultured with the zona pellucida (zona intact control group, ZI); and (2) cultured without the zona pellucida (zona free group, ZF). The cleavage, morula and blastocyst rates were evaluated. The blastocysts and their spent medium were used for miRNA expression analysis using RT-qPCR (miR-21, miR-24, mi25, miR-29, miR-96, miR-98, miR-103, miR-191, miR-196, miR-199, miR-130, miR-155 and miR-302). The pre-mature microRNAs (pre-miRNAs) and miRNAs were evaluated in the blastocysts and only miRNAs were evaluated in the spent medium. No differences were observed in the cleavage, morula and blastocyst rates between the ZF and ZI groups (P > 0.05). For miRNAs analysis, miR-103 and miR-191 had the most stable expression and were selected as internal controls. ZF blastocysts had a higher expression of miR-21, miR-25, miR-29 and miR-199 and a lower expression of miR-96 than their ZI counterparts (P < 0.05). Furthermore, higher levels of miR-21, miR-25 and miR-98 were detected in the spent medium of ZF blastocysts (P < 0.05). In conclusion, in vitro culture of domestic cat embryos without the zona pellucida modifies the expression of miR-21, miR-25, miR-29, miR-199 and miR-96 at the blastocyst stage and the release of miR-21, miR-25 and miR-98.

Most human cystic echinococcosis (CE) cases worldwide are attributed to Echinococcus granulosus sensu stricto (s.s), followed by the G6 and G7 genotypes. While E. granulosus s.s. has a cosmopolitan distribution, the G6 genotype is restricted to areas where camels and goats are present. Goats are the primary livestock in the Neuquén province in Argentina where the G6 genotype has been reported to be responsible for a significant percentage of CE human cysts genotyped. In the present study, we genotyped 124 Echinococcus cysts infecting 90 CE-confirmed patients. Echinococcus granulosus s.s. was identified in 51 patients (56.7%) with 81 cysts and the G6 genotype in 39 patients (43.3%) harbouring 43 cysts. Most CE cases ≤18 years were male suggesting pastoral work could be a risk factor for the infection. Echinococcus granulosus s.s. was significantly found more frequently in the liver (32/51 patients) and the G6 genotype in the lungs and extrahepatic localizations (27/39). The patients infected with E. granulosus s.s., presented up to 6 cysts while patients infected with G6 presented a maximum of 2. The diameter of lung cysts attributed to E. granulosus s.s. was significantly larger compared to lung cysts from G6. Following the WHO ultrasound classification of liver cysts, we observed inactive cysts in 55.6% of G6 cysts and only 15.3% of E. granulosus s.s cysts. In conclusion, we provide evidence of differences in clinical aspects of CE caused by E. granulosus s.s. and the G6 genotype of E. granulosus s.l. complex infecting humans.

Domestic cat embryos generated by in vitro fertilization (IVF) and cultured without the zona pellucida have a reduced implantation capacity after embryo transfer at the blastocyst stage. The objective of this study was to evaluate the expression of trophectoderm markers in domestic cat blastocysts cultured without the zona pellucida. Two experimental groups were selected: (1) domestic cat embryos generated by IVF and cultured in vitro normally (zona intact group, ZI); and (2) domestic cat embryos generated by IVF and cultured in vitro without a zona pellucida (zona-free group, ZF). In the ZF group, the zona pellucida of the presumptive zygote was removed and these were cultured using the well of the well (WOW) system. In vitro culture was carried out for 7 days. The cleavage, morula and blastocyst rates were estimated. Finally, the relative expression levels of the trophectoderm markers TEAD4, YAP1, CDX2 and EOMES, the cell adhesion marker E-cadherin and the apoptosis marker CASP3 were evaluated by RT-qPCR in the blastocysts. The Wilcoxon test was used to evaluate differences (P < 0.05). No differences were observed in the cleavage, morula and blastocyst rates between the ZF and ZI groups. No differences were found in the expression of TEAD4, CDX2, E-cadherin and CASP3 between groups. The expression of YAP1 and EOMES was higher in ZF blastocysts than in ZI blastocysts. In conclusion, the in vitro culture without the zona pellucida generates an overexpression of YAP1 and EOMES in the domestic cat blastocysts. More studies are needed to confirm if this overexpression might affect in vivo development.

The COVID-19 pandemic has led to significant disruptions in Canada's public school system as provincial and territorial governments have enacted sudden and prolonged school closures. We compare the different school closure decisions in Alberta and Ontario during spring 2021, using official public briefings and publicly available data about rates of COVID-19 cases. We ask if provincial policy decisions can be explained by different epidemiological contexts and risks. We find that key epidemiological indicators such as the rate of cases were not directly linked to school closure decisions. This is important for policy makers and experts: it problematizes the assumption of a straight line between evidence and decisions and has implications for transparency and public trust in pandemic policy choices. A systematic description of the gap between evidence and policy is an important starting point for asking, What does drive decisions to close schools?

ABSTRACT IMPACT: The importance of this study is to evaluate the responses following the exposure of opioid drugs in young adults to address the current opioid epidemics OBJECTIVES/GOALS: To compare the proportion of patients between 18 and 25 years of age, who develop an opioid abuse disorder following dental surgery, to those following other surgical procedures, when an opioid drug is used for acute postoperative pain control. METHODS/STUDY POPULATION: We fashioned an IRB-sponsored retrospective cohort study of patients, ages 18 to 25 years old, who presented for dental surgery or other medical surgical procedures, at Strong Memorial Hospital Medical Center, at the University of Rochester and received opioid drug treatments, for acute surgical pain management. Patients with the diagnosis of acute surgical pain were included in the study. However, those with chronic pain or other related abnormalities were excluded, even if a diagnosis of acute surgical pain was present in the electronic chart. The clinical data were retrieved from electronic medical records and NYS-iStop records. All statistics were significant at the level of <0.1 RESULTS/ANTICIPATED RESULTS: We identified 167 subjects, of whom, only 150 subjects met inclusion criteria (n=100) in dentistry and (n=50) in other medical specialties. Patients were followed up in a 7-year period. Most of the subjects were females (n=91), Caucasian (n=80), and lived in a suburban location (n=78). The most frequently prescribed opioid included hydrocodone (n=119). A significantly higher proportion (8.7%) of patients developed opioid abuse disorders in the control group, compared to 1% of subjects in the experimental group (p 0.02). Those in the control group received marginally significant higher doses of MMEs 447.2 +/-644.8 mg vs 306.2 +/-354.7mg in the experimental group (p 0.086). Those in the control group had significantly longer periods of opioid treatment 10 (+/-12) compared to 6(+/-6) days in the experimental group DISCUSSION/SIGNIFICANCE OF FINDINGS: It is paramount to evaluate the morphine milligram equivalents, and duration of opioid treatment given to the young population with acute surgical pain, to prevent opioid abuse disorders in this vulnerable cohort.

Human embryos generated in vitro have a high incidence of chromosomal abnormalities that negatively affect pregnancy rate. Embryos generated in vitro secrete extracellular vesicles (EVs) into the culture medium that could be used potentially as indicators of embryo competence. This research aimed to evaluate the concentration and size of EVs and their gDNA content as an indicator of developmental competence in human embryos. Human embryos generated by intracytoplasmic sperm injection (ICSI) were classified morphologically as of either TOP, FAIR or POOR quality. Culture medium and developmentally arrested embryos (which were not able to be used for embryo transfer) were collected. Microvesicles, exosomes (MV/Exo) and apoptotic bodies (ABs) were isolated from culture medium. Nanoparticle tracking analysis (NTA) and array comparative genomic hybridization (aCGH) analysis were performed to evaluate EVs and their gDNA content. From NTA, the diameter (mean) of MVs/Exo from TOP quality embryos was higher (112.17 nm) compared with that of FAIR (108.02) and POOR quality embryos (102.78 nm) (P < 0.05). aCGH analysis indicated that MVs/Exo and ABs carried gDNA with the presence of 23 chromosome pairs. However, when arrested embryos were compared with their respective MVs/Exo and ABs, the latter had an increased rate of chromosomal abnormalities (24.9%) compared with embryos (8.7%) (P < 0.05). In conclusion, the size of EVs from culture medium might be an alternative for evaluating competence of human embryos, however more studies are needed to validate the use of gDNA from EVs as an indicator of embryo competence.

Identifying benthic substrates is important to researchers studying aquatic organisms in fresh and salt water systems. Benthic substrates are often not visible from the surface making it necessary to find another method to gather these data. Previous research has demonstrated that low cost side-scan sonar is a reliable way to identify hard substrates, such as rock and gravel, in a small, freshwater stream. In this study, the reliability of the side-scan sonar to accurately identify softer substrates such as grass and mud was tested in a large, brackish lagoon system. A total area of 11.55 km2 was surveyed with the sonar. Videos and pictures were taken at various points to groundtruth the sonar images and provide a measure of accuracy. Five substrate types were identified: dense seagrass, sparse seagrass, mangrove soil, mangrove soil with rock, and silt. Unidentifiable substrates were classified as unknown. A manually zoned benthic substrate map was created from the sonar recordings. Dense seagrass was most accurately identified. Sparse seagrass was the least accurately identified. A bathymetric map was also created from the sonar recordings.

The anthropogenic modification of natural landscapes, and the consequent changes in the environmental conditions and resources availability at multiple spatial scales can affect complex species interactions involving key-stone species such as bat–parasite interactions. In this study, we aimed to identify the drivers potentially influencing host–bat fly interactions at different spatial scales (at the host, vegetation stand and landscape level), in a tropical anthropogenic landscape. For this purpose, we mist-netted phyllostomid and moormopid bats and collected the bat flies (streblids) parasitizing them in 10 sites representing secondary and old growth forest. In general, the variation in fly communities largely mirrored the variation in bat communities as a result of the high level of specialization characterizing host–bat fly interaction networks. Nevertheless, we observed that: (1) bats roosting dynamics can shape bat–streblid interactions, modulating parasite prevalence and the intensity of infestation; (2) a degraded matrix could favor crowding and consequently the exchange of ectoparasites among bat species, lessening the level of specialization of the interaction networks and promoting novel interactions; and (3) bat–fly interaction can also be shaped by the dilution effect, as a decrease in bat diversity could be associated with a potential increase in the dissemination and prevalence of streblids.

Riparian forests (RF) provide shelter, feeding resources and commuting areas and are fundamental for local fauna during long drought periods in tropical dry forests. Nevertheless, information regarding vertebrate ecology in tropical RF is scarce. The studies of bats in riparian habitats have been mostly focused on insectivorous assemblages in non-tropical regions. We contrasted the occurrence of frugivorous bats between RF and upland dry forest (UDF) in a Neotropical dry forest region and evaluated how vegetation structure modulates frugivorous bat presence and abundance. We sampled bat ensembles and vegetation structure in three UDF and three RF sites in Jalisco, Mexico. Bat ensembles and populations were analysed with generalized linear models, including as independent factors: (1) habitat and (2) vegetation attributes. In 72 nights we captured 840 bats belonging to seven species. No effect of habitat type or vegetation structure on species richness was found, but habitat type did affect bat species composition and abundance. Moreover, vegetation structural traits affected frugivorous bat abundance. Frugivore abundance was higher in RF than in UDF probably because RF provides greater resources and more roosting sites. Riparian forests also present more canopy cover and accessible flight spaces. Our results support the importance of RF for frugivorous bats in terms of abundance and composition. Additionally, the lack of differences in species richness between UDF and RF highlights the importance of both habitats for maintaining frugivorous bats, as well as the related ecosystem processes and services in tropical dry forest landscapes.

About three years after Gaia was set into orbit, the first release of Gaia data, DR1, has been published by ESA and DPAC. Gaia’s first archive contains the results from the analysis of the initial 14 months of mission data. Outstandingly, it includes TGAS, Gaia-Tycho2 astrometric solution for about 2 million stars up to visible magnitude around 11.5.

In addition to the five parameter astrometric solution for the sky (positions, parallaxes and proper motions), future Gaia data releases will provide the spectral energy distributions from 330-1050 nm, together with radial velocities, for sources brighter than visible magnitude 16. Within this context, the relevance of Gaia data for the study of PN is be briefly presented.
