Bombyx mori Tryptophanyl-tRNA synthetase (BmWARS) belongs to the family of Ic-like aminoacyl-tRNA synthetases (aaRSs), whose specific recognition of the substrate Trp, tRNA, maintains the fidelity of protein synthesis. In this study, BmWARS was cloned and characterized from the midgut of the silkworm, Bombyx mori, resulting in an open reading frame (ORF) with a full length of 1,149 bp, which can encode 382 Aa. BmWARS is localized in the cytoplasm, and is expressed in all tissues of the silkworm, with higher expression in the testis, ovary, silk gland and malpighian tubule. The expression of BmWARS was significantly up-regulated in the midgut and silk gland after infection with Bombyx mori nuclear polyhedrosis virus (BmNPV). In addition, overexpression of BmWARS inhibited BmNPV infection and replication extremely significantly, while interference with BmWARS expression promoted BmNPV infection and replication. Analysis of the immune pathways in which BmWARS may be involved revealed that the expression of the key genes of the PI3K-Akt pathway, BmPI3K, BmAkt, BmPDK1, BmeIF4E, BmS6, and p-Akt protein was significantly reduced, whereas the expression of BmPTEN, BmFoxO, and BmCaspase9 was significantly increased in the cells that overexpressed BmWARS and were infected with BmNPV. Meanwhile, the results of the study interfering with the expression of BmWARS were completely opposite to those of the study overexpressing BmWARS. This is the first report that BmWARS has antiviral effects in Bombyx mori. Moreover, BmWARS inhibits BmNPV infection and replication in Bombyx mori cells by promoting apoptosis and inhibiting cell proliferation.

The demand for separating and analysing rare target cells is increasing dramatically for vital applications such as cancer treatment and cell-based therapies. However, there remains a grand challenge for high-throughput and label-free segregation of lesion cells with similar sizes. Cancer cells with different invasiveness usually manifest distinct deformability. In this work, we employ a hydrogel microparticle system with similar sizes but varied stiffness to mimic cancer cells and examine in situ their deformation and focusing under microfluidic flow. We first demonstrate the similar focusing behaviour of hydrogel microparticles and cancer cells in confined flow that is dominated by deformability-induced lateral migration. The deformation, orientation and focusing position of hydrogel microparticles in microfluidic flow under different Reynolds numbers are then systematically observed and measured using a high-speed camera. Linear correlations of the Taylor deformation and tilt angle of hydrogel microparticles with the capillary number are revealed, consistent with theoretical predictions. Detailed analysis of the dependence of particle focusing on the flow rate and particle stiffness enables us to identify a linear scaling between the equilibrium focusing position and the major axis of the deformed microparticles, which is uniquely determined by the capillary number. Our findings provide insights into the focusing and dynamics of soft beads, such as cells and hydrogel microparticles, under confined flow, and pave the way for applications including the separation and identification of circulating tumour cells, drug delivery and controlled drug release.

This study aimed to assess the concordance between different anthropometric indexes in the Global Leaders Initiated Malnutrition Standards (GLIM) and the Geriatric Risk Index (GNRI) for evaluating muscle mass, while also exploring performance-based criteria for GLIM muscle content suitable for elderly patients with intermediate and advanced tumors. A total of 312 patients admitted to Shanghai Tenth People’s Hospital between September 2022 and June 2023 were retrospectively included. Nutritional assessments were conducted using the GLIM framework, employing grip strength, upper arm circumference, and calf circumference as criteria for muscle content evaluation. The diagnostic value of these tools was compared against the GNRI as a reference standard. Among the participants, 127 (40.71%) were diagnosed as malnourished by GNRI, while the GLIM assessments yielded 138 (44.23%), 128 (41.03%), and 162 (51.92%) malnutrition diagnoses based on grip strength, calf circumference, and upper arm circumference, respectively. Both GNRI and GLIM-grip strength were significantly associated with complications and length of hospital stays. Notably, using GNRI as a reference, GLIM-grip strength demonstrated good consistency in diagnosing malnutrition (K value = 0.692, P < 0.001), with calf circumference having the highest diagnostic value. In conclusion, grip strength is a practical and effective performance-based criterion within the GLIM standards and has the potential to enhance malnutrition diagnosis in elderly patients with advanced malignancies, highlighting its relevance in nutritional science.

This study investigates the stability and instability of the language control network in bilinguals using longitudinal resting-state functional magnetic resonance imaging (rs-fMRI) data. We compared the language control network of Chinese university students majoring in English with those not, using three other functional networks as controls. Results indicate that the English major group exhibits reduced stability and increased instability in the language control network compared with the non-English major group. This suggests that second language (L2) learning experience may induce adaptive neural changes. Moreover, the coexistence of stability and instability in the language control network appears less modular in the English major group, implying a more integrated response to language experience. Notably, these results were not observed in the control networks. Overall, these findings enhance the understanding of bilingual language control and the impact of L2 learning on neural plasticity.

Determining the insdel distance of linear codes is a very challenging problem. Recently, Ji et al. [‘Strict half-Singleton bound, strict direct upper bound for linear insertion-deletion codes and optimal codes’, IEEE Trans. Inform. Theory 69(5) (2023), 2900–2910] proposed their strict half-Singleton bound for linear codes that do not include the all-one vector. A natural question asks for linear codes with both large Hamming distance and insdel distance. Almost maximum distance separable (MDS) codes are a special kind of linear code with good Hamming error-correcting capability. We present a sufficient condition for the insdel distance of almost MDS codes to be bounded by the strict half-Singleton bound. In addition, we construct a class of two-dimensional near MDS codes that achieve the strict half-Singleton bound using twisted Reed–Solomon codes. Finally, we present a construction of optimal almost MDS insdel codes from Reed–Solomon codes for large dimensions.

Stimulated Raman scattering is a third-order nonlinear optical effect that is not only effective for wavelength converting laser output, but also for single longitudinal-mode output due to the absence of spatial hole burning. Diamond is a prominent Raman-active medium that has significant potential for linewidth narrowing and wavelength converting lasers at high power levels due to its high thermal conductivity, long Raman frequency shift and wide spectral transmission range. In this work we utilize diamond in a resonantly mode-matched external cavity to achieve cascaded Raman conversion of a 1064 nm laser. By fine-tuning the length of this external cavity, we can obtain narrow linewidth emission at 1240 and 1485 nm. When operating at maximum power, the measured linewidths were more than twofold narrower than the linewidth of the fundamental field. In addition, the noise levels of the Stokes fields are lower than that of the fundamental field throughout the entire noise frequency range, and the intrinsic linewidth of the second Stokes field, which is expressed at the hertz level (~3.6 Hz), is decreased by approximately three orders of magnitude compared to that of the pump. This work represents the first measurement and analysis of the linewidth and noise characteristics of cascaded diamond Raman lasers and, significantly, offers a new means by which high-power, narrow linewidth laser output can be produced from wavelength-converted laser systems.

Large-aperture gratings have significant applications in inertial confinement fusion, immersion lithography manufacturing and astronomical observation. Currently, it is challenging and expensive to manufacture sizable monolithic gratings. Therefore, tiled multiple small-aperture gratings are preferred. In this study, the impact of seam phase discontinuity on the modulation of the laser beam field was explored based on the measurement results of the Shenguang-II laser large-aperture multi-exposure-tiled grating. An innovative method for accurately calculating the phase jump of multi-exposure-tiled grating seams was proposed. An intensive electromagnetic field analysis was performed by applying rigorous coupled-wave analysis to a reasonably constructed micrometer-level periodic grating seam structure, and the phase jump appearing in millimeter-scale seams of large-aperture tiled gratings was obtained accurately.

Humans depend heavily on nature. Drylands are home to 2.5 billion people, but the extent to which nature contributes to people (NCP) in drylands has been little explored. We examined the global contribution of nature to people, aiming to compare drylands and non-drylands. We predicted a lower contribution in drylands than non-drylands, largely because of the sparser population densities (peoples’ needs) and more degraded status of natural resources (lower potential contribution). Consistent with expectation, nature’s contribution was about 30% lower in drylands, with significantly lower values for drylands in Asia, Oceania, Africa and South America, but no difference for Europe and North America. Differences were due mainly to lower contributions from material and regulating contributions, i.e., the regulation of air quality, climate, water quantity and flow, soil protection and the supply of woody material, and potentially, lower use by people in drylands. Predicted declines in rainfall and increasing temperature are likely to place increasing pressure on nature to contribute to human well-being in drylands. A better understanding of nature’s contributions to people would improve our ability to allocate limited resources and achieve sustainable development in drylands.

Background: Ventilator associated pneumonia (VAP) is associated with significant rates of morbidity and all-cause mortality. Active VAP surveillance can identify risk factors for which targeted preventive measures can be implemented. However, surveillance efforts are complicated by challenges associated with accurate VAP diagnosis. We aimed to improve the accuracy and automation of existing VAP diagnostic algorithms to better identify patients at risk. Methods: The study was conducted at NYU Langone Health from June 2022 through December 2023. We created a semi-automated VAP surveillance system using the Centers for Disease Control & Prevention (CDC) ventilator associated event (VAE) definition as a base framework (Figure 1). We modified this definition to include additional elements, such as having a sputum culture ordered within 48 hours of worsening oxygen status, regardless of culture result. Using this algorithm—followed by manual clinician reviews—we retrospectively assessed possible VAP cases to determine the ability of our surveillance system to correctly identify VAP. Results: Of the 123 possible VAP cases identified through our automated system, 75 (61%) were correctly diagnosed as VAP after clinical review. This reflects a rate of 1.5 infections per 1000 ventilation days across the system and 1.85 infections per 100 patients ventilated for greater than 2 days. Of the 48 remaining patients without VAP after clinical review, 25% (n=12) were characterized as having hospital-acquired pneumonia, 21% (n=10) as acute respiratory distress syndrome or infection at another site and 10% (n=5) as pulmonary embolism/infarction. Among all patients identified through this automated system (VAP and non-VAP), 53% experienced in-hospital death. Discussion: Our automated VAP surveillance algorithm identified 123 cases of potential VAP, 61% of which were consistent with a clinical diagnosis of VAP upon manual chart review. Our VAP rate of 1.5 infections per 1000 ventilation days was similar to published rates at other North American hospital systems. The high in-hospital mortality rate among these patients highlights the need for improved surveillance systems and earlier interventions to reduce the risk of VAP. There are several limitations to the CDC’s VAE definition, including its requirement of a positive microbiologic culture and focus on sputum quality. This potentially misses cases of culture-negative VAP in patients receiving antibiotics prior to sputum collection. Our goal is to continue to validate and improve our algorithm’s ability to correctly identify patients with clinical VAP, so that targeted prevention efforts can be focused upon the patients with the highest risk for poor outcomes.

Nontuberculous mycobacteria (NTM) is a large group of mycobacteria other than the Mycobacterium tuberculosis complex and Mycobacterium leprae. Epidemiological investigations have found that the incidence of NTM infections is increasing in China, and it is naturally resistant to many antibiotics. Therefore, studies of NTM species in clinical isolates are useful for understanding the epidemiology of NTM infections. The present study aimed to investigate the incidence of NTM infections and types of NTM species. Of the 420 samples collected, 285 were positive for M. tuberculosis, 62 samples were negative, and the remaining 73 samples contained NTM, including 35 (8.3%) only NTM and 38 (9%) mixed (M. tuberculosis and NTM). The most prevalent NTM species were Mycobacterium intracellulare (30.1%), followed by Mycobacterium abscessus (15%) and M. triviale (12%). M. gordonae infection was detected in 9.5% of total NTM-positive cases. Moreover, this study reports the presence of Mycobacterium nonchromogenicum infection and a high prevalence of M. triviale for the first time in Henan. M. intracellulare is the most prevalent, accompanied by some emerging NTM species, including M. nonchromogenicum and a high prevalence of M. triviale in Henan Province. Monitoring NTM transmission and epidemiology could enhance mycobacteriosis management in future.

We study the statistically steady states of the forced dissipative three-dimensional homogeneous isotropic turbulence at scales larger than the forcing scale in real separation space. The probability density functions (p.d.f.s) of longitudinal velocity difference at large separations are close to, but deviate from, Gaussian, measured by their non-zero odd parts. The analytical expressions of the third-order longitudinal structure functions derived from the Kármán–Howarth–Monin equation prove that the odd-part p.d.f.s of velocity differences at large separations are small but non-zero. Specifically, when the forcing effect in the displacement space decays exponentially as the displacement tends to infinity, the odd-order longitudinal structure functions have a power-law decay with an exponent of $-$2, implying a significant coupling between large and small scales. Under the assumption that forcing controls the large-scale dynamics, we propose a conjugate regime to Kolmogorov's inertial range, independent of the forcing scale, to capture the odd parts of p.d.f.s. Thus, dynamics of large scales departs from the absolute equilibrium, and we can partially recover small-scale information without explicitly resolving small-scale dynamics. The departure from the statistical equilibrium is quantified and found to be viscosity-independent. Even though this departure is small, it is significant and should be considered when studying the large scales of the forced three-dimensional homogeneous isotropic turbulence.

Trauma is a significant health issue that not only leads to immediate death in many cases but also causes severe complications, such as sepsis, thrombosis, haemorrhage, acute respiratory distress syndrome and traumatic brain injury, among trauma patients. Target protein identification technology is a vital technique in the field of biomedical research, enabling the study of biomolecular interactions, drug discovery and disease treatment. It plays a crucial role in identifying key protein targets associated with specific diseases or biological processes, facilitating further research, drug design and the development of treatment strategies. The application of target protein technology in biomarker detection enables the timely identification of newly emerging infections and complications in trauma patients, facilitating expeditious medical interventions and leading to reduced post-trauma mortality rates and improved patient prognoses. This review provides an overview of the current applications of target protein identification technology in trauma-related complications and provides a brief overview of the current target protein identification technology, with the aim of reducing post-trauma mortality, improving diagnostic efficiency and prognostic outcomes for patients.

In this article, we introduce a hierarchy on the class of non-archimedean Polish groups that admit a compatible complete left-invariant metric. We denote this hierarchy by $\alpha $-CLI and L-$\alpha $-CLI where $\alpha $ is a countable ordinal. We establish three results:

1. (1) G is $0$-CLI iff $G=\{1_G\}$;

2. (2) G is $1$-CLI iff G admits a compatible complete two-sided invariant metric; and

3. (3) G is L-$\alpha $-CLI iff G is locally $\alpha $-CLI, i.e., G contains an open subgroup that is $\alpha $-CLI.

Subsequently, we show this hierarchy is proper by constructing non-archimedean CLI Polish groups $G_\alpha $ and $H_\alpha $ for $\alpha <\omega _1$, such that:

1. (1) $H_\alpha $ is $\alpha $-CLI but not L-$\beta $-CLI for $\beta <\alpha $; and

2. (2) $G_\alpha $ is $(\alpha +1)$-CLI but not L-$\alpha $-CLI.

The poor environmental stability of natural anthocyanin hinders its usefulness in various functional applications. The objectives of the present study were to enhance the environmental stability of anthocyanin extracted from Lycium ruthenicum by mixing it with montmorillonite to form an organic/inorganic hybrid pigment, and then to synthesize allochroic biodegradable composite films by incorporating the hybrid pigment into sodium alginate and test them for potential applications in food testing and packaging. The results of X-ray diffraction, Fourier-transform infrared spectroscopy, and use of the Brunauer–Emmett–Teller method and zeta potential demonstrated that anthocyanin was both adsorbed on the surface and intercalated into the interlayer of montmorillonite via host–guest interaction, and the hybrid pigments obtained allowed good, reversible, acid/base behavior after exposure to HCl and NH3 atmospheres. The composite films containing hybrid pigments had good mechanical properties due to the uniform dispersion of the pigments in a sodium alginate substrate and the formation of hydrogen bonds between them. Interestingly, the composite films also exhibited reversible acidichromism. The as-prepared hybrid pigments in composite films could, therefore, serve simultaneously as a reinforced material and as a smart coloring agent for a polymer substrate.
