Advancements in VLBI instrumentation, driven by the geodetic community’s goal of achieving positioning accuracy of 1 mm and stability of 0.1 mm/y, have led to the development of new broadband systems. Here, we assess the potential of these new capabilities for space weather monitoring. These enhanced VLBI capabilities were used to investigate interplanetary scintillation (IPS), a phenomenon caused by the scattering of radio waves due to density irregularities in the solar wind. Compact radio sources near the Sun were observed using the AuScope VLBI array in Australia, which consists of 12-meter telescopes at Hobart, Katherine, and Yarragadee. The baseline lengths between these telescopes are approximately 3400 km (Hobart–Katherine), 3200 km (Hobart–Yarragadee), and 2400 km (Katherine–Yarragadee). The observations covered solar elongations from 6.5° to 11.3° and frequencies between 3 and 13 GHz. The study focused on phase scintillation as an indicator of turbulence in the solar wind. As the solar elongation decreased, we observed an increase in the phase scintillation index, consistent with theoretical models. Importantly, the broadband system also detected IPS using relatively weak radio sources. Additionally, the phase scintillation increased with baseline length, in agreement with Kolmogorov turbulence with an index of 11/3. These findings demonstrate the effectiveness of geodetic broadband VLBI in capturing detailed features of the solar wind. This capability enables continuous space weather monitoring and advances our understanding of solar and interplanetary dynamics.

The radio telescopes of the European VLBI Network (EVN) and the University of Tasmania (UTAS) conducted an extensive observation campaign of the European Space Agency’s (ESA) Mars Express (MEX) spacecraft between 2013 and 2020. The campaign, carried out under the Planetary Radio Interferometry and Doppler Experiment (PRIDE) framework, aimed to study interplanetary phase scintillation and assess the noise budget in the closed-loop Doppler observations. The average closed-loop Doppler noise was determined to be approximately 10 mHz at a 10-second integration time, reaffirming the technique’s suitability for radio science experiments. We evaluated how different observational parameters such as the solar elongation, antenna size, and elevation angle impact the Doppler noise. A key part of the analysis involved comparing results from co-located telescopes to investigate system noise effects. Co-located telescopes at both Wettzell and Hobart provided highly consistent results, with any deviations serving as diagnostic tools to identify station-dependent issues. Additionally, the use of phase calibration tones during spacecraft tracking showed that the instrumental noise contribution is of the order of 5% of the total noise. This study provides a detailed noise budget for closed-loop Doppler observations with VLBI telescopes while emphasizing the effectiveness of the co-location method in isolating system-level noise. These findings are important for optimizing future radio science and VLBI tracking missions using stations outside the the Deep Space Network (DSN) and European Space Tracking (ESTRACK) network.

Objectives/Goals: The goal of this work is to understand the physiological profile of phage susceptibility and identify candidate phage defense mechanisms. Additionally, it aims to determine the host receptors targeted by bacteriophages to infect E. coli O157:H7 through random bar code transposon-site sequencing (RB-TnSeq). Methods/Study Population: A collection of 109 E. coli O157:H7 strains from environmental, food, and animal sources were analyzed, representing phylogenetic lineages corresponding to clades 2, 3, 5, 6, 7, and 8. Phage susceptibility profiles were determined using 23 bacteriophages, assessing plaque morphology. Using the O157:H7 genomes, a genomic analysis was conducted with the Prokaryotic Antiviral Defense Locator (PADLOC), which identified putative phage defense systems through sequence homology. Additionally, 5 RB-TnSeq libraries were generated in representative strains to study loss-of-function mutations. These libraries will be screened against a subset of diverse phage to identify the receptors involved in phage adsorption. Results/Anticipated Results: The phage resistance patterns showed susceptibility varied across clades, suggesting distinct mechanisms. Several defense systems were identified using PADLOC, including restriction-modification, Cas, Lamassu, and Druantia. Phage defense candidate (PDC) systems were identified, showing homology to known systems, though their specific function remains unknown. Clade 7.2 exhibited higher phage resistance and a greater presence of PDCs compared to the other clades. Five saturated RB-TnSeq libraries were constructed in O157:H7, achieving 84.5–89% gene coverage. These libraries will facilitate the identification of receptors involved in phage adsorption and resistance. Discussion/Significance of Impact: This study deepens our understanding of phage resistance in E. coli O157:H7 by identifying key defense systems and receptors. The discovery of novel antiviral mechanisms offers promising targets for phage-based interventions, potentially enhancing strategies for controlling this dangerous pathogen.

Quantum field theory predicts a nonlinear response of the vacuum to strong electromagnetic fields of macroscopic extent. This fundamental tenet has remained experimentally challenging and is yet to be tested in the laboratory. A particularly distinct signature of the resulting optical activity of the quantum vacuum is vacuum birefringence. This offers an excellent opportunity for a precision test of nonlinear quantum electrodynamics in an uncharted parameter regime. Recently, the operation of the high-intensity Relativistic Laser at the X-ray Free Electron Laser provided by the Helmholtz International Beamline for Extreme Fields has been inaugurated at the High Energy Density scientific instrument of the European X-ray Free Electron Laser. We make the case that this worldwide unique combination of an X-ray free-electron laser and an ultra-intense near-infrared laser together with recent advances in high-precision X-ray polarimetry, refinements of prospective discovery scenarios and progress in their accurate theoretical modelling have set the stage for performing an actual discovery experiment of quantum vacuum nonlinearity.

Jansen and Roskam (1986) discussed the compatibility of the unidimensional polytomous Rasch model with dichotomization of the response continuum. They derived a rather strict condition in which dichotomization of multicategory data that fit the unidimensional polytomous Rasch model, results in dichotomous data which fit the dichotomous Research model with effectively the same subject parameter. In this paper a more general dichotomization condition is derived for the polytomous Rasch model, which appears less restrictive, but upholds that the intrinsic logic of the unidimensional polytomous Rasch model defies dichotomization in general. The robustness of dichotomous analysis investigated in a simulation study. It shows a close relation with the two-parameters (Birnbaum) model. Theoretical and methodological implications are discussed.

This paper discusses the compatibility of the polychotomous Rasch model with dichotomization of the response continuum. It is argued that in the case of graded responses, the response categories presented to the subject are essentially an arbitrary polychotomization of the response continuum, ranging for example from total rejection or disagreement to total acceptance or agreement of an item or statement. Because of this arbitrariness, the measurement outcome should be independent of the specific polychotomization applied, for example, presenting a specific multicategory response format should not affect the measurement outcome. When such is the case, the original polychotomous model is called “compatible” with dichotomization.

A distinction is made between polychotomization or dichotomization “before the fact,” that is, in constructing the response format, and polycho- or dichotomization “after the fact,” for example in dichotomizing existing graded response data.

It is shown that, at least in case of dichotomization after-the-fact, the polychotomous Rasch model is not compatible with dichotomization, unless a rather special condition of the model parameters is met. Insofar as it may be argued that dichotomization before the fact is not essentially different from dichotomization after the fact, the value of the unidimensional polychotomous Rasch model is consequently questionable. The impact of our conclusion on related models is also discussed.

Previous studies in rodents suggest that mismatch between fetal and postnatal nutrition predisposes individuals to metabolic diseases. We hypothesized that in nonhuman primates (NHP), fetal programming of maternal undernutrition (MUN) persists postnatally with a dietary mismatch altering metabolic molecular systems that precede standard clinical measures. We used unbiased molecular approaches to examine response to a high fat, high-carbohydrate diet plus sugar drink (HFCS) challenge in NHP juvenile offspring of MUN pregnancies compared with controls (CON). Pregnant baboons were fed ad libitum (CON) or 30% calorie reduction from 0.16 gestation through lactation; weaned offspring were fed chow ad libitum. MUN offspring were growth restricted at birth. Liver, omental fat, and skeletal muscle gene expression, and liver glycogen, muscle mitochondria, and fat cell size were quantified. Before challenge, MUN offspring had lower body mass index (BMI) and liver glycogen, and consumed more sugar drink than CON. After HFCS challenge, MUN and CON BMIs were similar. Molecular analyses showed HFCS response differences between CON and MUN for muscle and liver, including hepatic splicing and unfolded protein response. Altered liver signaling pathways and glycogen content between MUN and CON at baseline indicate in utero programming persists in MUN juveniles. MUN catchup growth during consumption of HFCS suggests increased risk of obesity, diabetes, and cardiovascular disease. Greater sugar drink consumption in MUN demonstrates altered appetitive drive due to programming. Differences in blood leptin, liver glycogen, and tissue-specific molecular response to HFCS suggest MUN significantly impacts juvenile offspring ability to manage an energy rich diet.

The interaction of water with synthetically prepared goethite (α-FeOOH) and amorphous hyd-rated ferric oxide surfaces was studied using i.r. absorption and water vapor adsorption measurements. I.R. results show that the last traces of physically adsorbed water are removed from the amorphous material by outgassing at 25°C. In contrast, goethite retains approximately a monolayer of physically adsorbed water with similar outgassing. This monolayer of water on goethite, which is presumably hydrogen-bonded at least in part with structural hydroxyls, is readily exchangeable with D2O.

Integral entropies of adsorption were evaluated from water vapor adsorption isotherms at 15, 25 and 35°C and compared with values for mobile and immobile layers calculated through application of statistical mechanics (McCafferty and Zettlemoyer, 1970). Entropy values for both the first physically adsorbed monolayer of water on the amorphous material and the second monolayer on goethite were about the same as or greater than those calculated for an immobile layer, indicating strong hydrogen bonding of water by both surfaces. The larger deviation between the entropy values for goethite and those calculated for the immobile layer may be associated with changes in the structure of the first as well as the second physically adsorbed water layers. Surface areas, calculated using the BET method, were 320 and 32 m2 g-1 for the amorphous material and goethite respectively. Since the unit surface activity is probably about the same for the two materials, it follows that as the amorphous material crystallizes to form goethite, there would be a reduction in total surface activity in proportion to the reduction in surface area.

The nature of freshly-precipitated and aged hydrated ferric oxides prepared by the addition of ferric chloride to KOH was investigated by the use of scanning and transmission electron microscopy, X-ray diffraction, i.r. absorption, and pH 3·0 ammonium oxalate extraction. The results show the fresh material to be essentially non-crystalline hydrated ferric oxide, which when aged at 60°C C and high pH rapidly crystallizes as goethite, without any indication of coexisting hematite. The various methods were evaluated as indices of crystallinity for aging materials. The acid ammonium oxalate method was shown to extract selectively only the non-crystalline portion of such mixtures. The use of X-ray diffraction analysis for estimating aging stage requires elimination of the preferred orientation of the goethite crystals. While both the oxalate and X-ray methods can detect as little as 2 per cent crystallinity, the oxalate method is probably superior for quantitative determinations as it depends directly on an inherent difference in the solubility of the crystalline and non-crystalline materials, rather than on a technique dependent intensity measurement. The use of the intensity of the O-H bending vibrations of the infrared absorption spectra can also potentially detect as little as 2 per cent crystallinity, but the procedure is probably less useful for quantitative determinations than the oxalate or X-ray methods because of the problem of evaluating the area under the peaks.

During the antebellum period and American Civil War, ‘puritan’ was a contested identity, fraught with layers of meaning and interpretation. Historians have charted the ways Southern intellectuals cast the differences between North and South as an outplaying of the old conflict between Cavalier and puritan. This article highlights the ways Southern ministers claimed the puritan identity for the South and accused the North of hypocrisy, for having fallen far from the theological ideals of their puritan forebears. Furthermore, Southern ministers noted the hypocrisy of Northern puritans for having escaped religious tyranny only to impose it upon those who did not conform to their form of Christianity; they had thus fallen into the very sin which they had decried. This came from Southern ministers whose attempt to appropriate the memory of puritanism as liberty-loving revealed their own hypocrisy in fighting for the ‘liberty’ to maintain a system of racial slavery.

Northern Arizona University, Flagstaff, Arizona, USA, recently installed a MIni CArbon DAting System (MICADAS) with a gas interface system (GIS) for determining the 14C content of CO2 gas released by the acid dissolution of biogenic carbonates. We compare 48 paired graphite, GIS, and direct carbonate 14C determinations of individual mollusk shells and echinoid tests. GIS sample sizes ranged between 0.5 and 1.5 mg and span 0.1 to 45.1 ka BP (n = 42). A reduced major axis regression shows a strong relationship between GIS and graphite percent Modern Carbon (pMC) values (m = 1.011; 95% CI [0.997–1.023], R2 = 0.999) that is superior to the relationship between the direct carbonate and graphite values (m = 0.978; 95% CI [0.959-0.999], R2 = 0.997). Sixty percent of GIS pMC values are within ±0.5 pMC of their graphite counterparts, compared to 26% of direct carbonate pMC values. The precision of GIS analyses is approximately ±70 14C yrs to 6.5 ka BP and decreases to approximately ±130 14C yrs at 12.5 ka BP. This precision is on par with direct carbonate and is approximately five times larger than for graphite. Six Plio-Pleistocene mollusk and echinoid samples yield finite ages when analyzed as direct carbonate but yield non-finite ages when analyzed as graphite or as GIS. Our results show that GIS 14C dating of biogenic carbonates is preferable to direct carbonate 14C dating and is an efficient alternative to standard graphite 14C dating when the precision of graphite 14C dating is not required.

We report the discovery of a bow-shock pulsar wind nebula (PWN), named Potoroo, and the detection of a young pulsar J1638$-$4713 that powers the nebula. We present a radio continuum study of the PWN based on 20-cm observations obtained from the Australian Square Kilometre Array Pathfinder (ASKAP) and MeerKAT. PSR J1638$-$4713 was identified using Parkes radio telescope observations at frequencies above 3 GHz. The pulsar has the second-highest dispersion measure of all known radio pulsars (1 553 pc cm$^{-3}$), a spin period of 65.74 ms and a spin-down luminosity of $\dot{E}=6.1\times10^{36}$ erg s$^{-1}$. The PWN has a cometary morphology and one of the greatest projected lengths among all the observed pulsar radio tails, measuring over 21 pc for an assumed distance of 10 kpc. The remarkably long tail and atypically steep radio spectral index are attributed to the interplay of a supernova reverse shock and the PWN. The originating supernova remnant is not known so far. We estimated the pulsar kick velocity to be in the range of 1 000–2 000 km s$^{-1}$ for ages between 23 and 10 kyr. The X-ray counterpart found in Chandra data, CXOU J163802.6$-$471358, shows the same tail morphology as the radio source but is shorter by a factor of 10. The peak of the X-ray emission is offset from the peak of the radio total intensity (Stokes $\rm I$) emission by approximately 4.7$^{\prime\prime}$, but coincides well with circularly polarised (Stokes $\rm V$) emission. No infrared counterpart was found.
