Pebrine disease, caused by Nosema bombycis (Nb) infection in silkworms, is a severe and long-standing disease that threatens sericulture. As parasitic pathogens, a complex relationship exists between microsporidia and their hosts at the mitochondrial level. Previous studies have found that the translocator protein (TSPO) is involved in various biological functions, such as membrane potential regulation, mitochondrial autophagy, immune responses, calcium ion channel regulation, and cell apoptosis. In the present study, we found that TSPO expression in silkworms (BmTSPO) was upregulated following Nb infection, leading to an increase in cytoplasmic calcium, adenosine triphosphate, and reactive oxygen species levels. Knockdown and overexpression of BmTSPO resulted in the promotion and inhibition of Nb proliferation, respectively. We also demonstrated that the overexpression of BmTSPO promotes host cell apoptosis and significantly increases the expression of genes involved in the immune deficiency and Janus kinase-signal transducer and the activator of the transcription pathways. These findings suggest that BmTSPO activates the innate immune signalling pathway in silkworms to regulate Nb proliferation. Targeting TSPO represents a promising approach for the development of new treatments for microsporidian infections.
The purpose of this study was to explore the electroencephalogram (EEG) features sensitive to situation awareness (SA) and then classify SA levels. Forty-eight participants were recruited to complete an SA standard test based on the multi-attribute task battery (MATB) II, and the corresponding EEG data and situation awareness global assessment technology (SAGAT) scores were recorded. The population with the top 25% of SAGAT scores was selected as the high-SA level (HSL) group, and the bottom 25% was the low-SA level (LSL) group. The results showed that (1) for the relative power of $\beta$1 (16–20Hz), $\beta$2 (20–24Hz) and $\beta$3 (24–30Hz), repeated measures analysis of variance (ANOVA) in three brain regions (Central Central-Parietal, and Parietal) × three brain lateralities (left, midline, and right) × two SA groups (HSL and LSL) showed a significant main effect for SA groups; post hoc comparisons revealed that compared with LSL, the above features of HSL were higher. (2) for most ratio features associated with $\beta$1 ∼ $\beta$3, ANOVA also revealed a main effect for SA groups. (3) EEG features sensitive to SA were selected to classify SA levels with small-sample data based on the general supervised machine learning classifiers. Five-fold cross-validation results showed that among the models with easy interpretability, logistic regression (LR) and decision tree (DT) presented the highest accuracy (both 92%), while among the models with hard interpretability, the accuracy of random forest (RF) was 88.8%, followed by an artificial neural network (ANN) of 84%. The above results suggested that (1) the relative power of $\beta$1 ∼ $\beta$3 and their associated ratios were sensitive to changes in SA levels; (2) the general supervised machine learning models all exhibited good accuracy (greater than 75%); and (3) furthermore, LR and DT are recommended by combining the interpretability and accuracy of the models.
Previous studies have suggested that the habenula (Hb) may be involved in the mechanism of obsessive-compulsive disorder (OCD). However, the specific role of Hb in OCD remains unclear. This study aimed to explore the structural and functional abnormalities of Hb in OCD and their relationship with the clinical symptoms.
Methods
Eighty patients with OCD and 85 healthy controls (HCs) were recruited as the primary dataset. The grey matter volume, resting-state functional connectivity (FC), and effective connectivity (EC) of the Hb were calculated and compared between OCD group and HCs. An independent replication dataset was used to verify the stability and robustness of the results.
Results
Patients with OCD exhibited smaller Hb volume and increased FC of right Hb-left hippocampus than HCs. Dynamic causal model revealed an increased EC from left hippocampus to right Hb and a less inhibitory causal influence from the right Hb to left hippocampus in the OCD group compared to HCs. Similar results were found in the replication dataset.
Conclusions
This study suggested that abnormal structure of Hb and hippocampus-Hb connectivity may contribute to the pathological basis of OCD.
Knowledge of clay mineralogy is essential for understanding the source areas and weathering environments of fluvial sediments, particularly in large reservoirs facing serious problems with sediment deposition, such as the Three Gorges Reservoir (TGR) in east-central China. The purpose of the present study was to identify the sediment provenances and weathering regimes contributing to the sediment load in the TGR by determining the clay-mineral and geochemical compositions of surface sediments during various seasons. X-ray diffractometry and scanning electron microscopy (SEM) were used to identify the clay minerals. The results showed that illite was the dominant mineral, followed in order by kaolinite, chlorite, and montmorillonite. From a mineralogical perspective, distal sources were the main contributors to the TGR sediments, and regional sources (surrounding tributaries) also contributed much during the three seasons, while proximal sources (hillslope soils) supplied sediment in the flood season but not in the other two seasons. The geochemical and hydrological data generally supported the mineralogical results. In the flood season, the chemical indices of the TGR sediments were >0.4, showing that the sediments contained Al-rich illite minerals and experienced intense hydrolysis. In the other two seasons the TGR sediments were enriched in Fe- and Mg-rich illite minerals, resulting from strong physical weathering. Furthermore, precipitation, rather than air temperature or latitude, was the factor that controlled weathering intensity. These findings provide deep insights into the sediment cycle and chemical weathering in this large reservoir basin.
Increasing evidence shows that maternal hyperglycemia inhibits cardiomyocyte (CM) proliferation and promotes cell apoptosis during fetal heart development, which leads to cardiac dysplasia. Accumulating evidence suggests that the overexpression of miR-21 in CMs has a protective role in cardiac function. Therefore, we investigated whether miR-21 can rescue CM injury caused by high glucose. First, we performed biological function analysis of miR-21-5p overexpression in H9c2 cells treated with high glucose. We found that the proliferation of H9c2 cells treated with high glucose decreased significantly and was rescued after overexpression of miR-21-5p. CCK-8 and EdU incorporation assays were performed to assess cell proliferation. The cell proliferation of the miR-21-5p mimic transfection group was improved compared with that of the NC mimic group (*p < 0.05, miR-21-5p mimics vs. NC mimics) when the proliferation of H9c2 cells was reduced by high glucose (****p < 0.0001, high glucose (HG) vs. normal glucose (NG)). Then, we verified the targeted and negative regulation of miR-21-5p on Rhob using a dual-luciferase activity assay and RT-qPCR, respectively. We further demonstrated that miR-21-5p regulates Rhob to rescue the inhibition of CM proliferation induced by high glucose. The CCK-8 results showed that the cell proliferation of the siRNA-Rhob group was higher than that of the NC mimic group (***p < 0.001) and that of the cotransfection group with Up-Rhob plasmids and miR-21-5p mimics was lower than that of the miR-21-5p mimic group (*p < 0.05). Conclusion: Overexpression of miR-21-5p rescues the inhibition of high glucose-induced CM proliferation through regulation of Rhob.
The western Mongolian Lake Zone was a Neoproterozoic to early Paleozoic volcanic arc where tuffs, lavas, fossiliferous siliciclastics, and carbonates accumulated during the early Cambrian. An uppermost Cambrian Series 2 (upper Stage 4) trilobite assemblage is described here from the Burgasutay Formation representing a continuous lower Cambrian succession at the Seer Ridge of the Great Lake Depression. The new assemblage is dominated by dorypygids and consists of 13 trilobite genera belonging to nine families including Catinouyia heyunensis new species. These fossils comprise the youngest and richest lower Cambrian trilobite assemblage in Mongolia. The composition of the Lake Zone fauna suggests its biogeographic affinity with the Siberian Platform and Altay-Sayan Foldbelt, but the presence of inouyiids also implies a connection of this region with East Gondwana.
The anti-corruption campaign launched by General Secretary Xi Jinping has been one of the most far-reaching bureaucratic overhauls in modern Chinese history. How has Xi's crackdown on corruption shaped bureaucratic selection at the sub-provincial level? In this paper, we find that the purge has influenced how local ties are weighed in the appointment of prefecture city leaders. While it is common for provincial Party chiefs to appoint locally embedded officials to govern localities without high-profile corruption cases, they tend to appoint outside officials without local experience and connections to manage cities whose ex-leaders have recently been prosecuted for corruption. We argue that the provincial leaders’ objective of installing non-local officials is to exert hierarchical control and oversight in localities affected by corruption. Using an original dataset of all Party secretaries from China's 287 prefecture-level cities between 2013 and 2020, we find a significant divergence in the local embeddedness of officials who are appointed to replace corrupt ex-leaders and the embeddedness of those who fill the vacancies of transferred or retired predecessors. Our study sheds light on how Xi's anti-corruption campaign has reshaped the central–local relations and the logic of political control in China.
In this study, aluminium fluoride trihydrate (AlF3⋅3H2O) was used to inhibit the sintering of calcined coal-series kaolin (CCSK). In addition, the oil absorption properties of CCSK were studied. The particle-size distribution, specific surface area and porosity of the samples were investigated as a function of calcination temperature and the addition of AlF3⋅3H2O. Moreover, the ability of AlF3⋅3H2O to improve the oil absorption of CCSK was explored. The morphology, structure and phase composition of the specimens were investigated using scanning electron microscopy and X-ray diffraction. The phase transition during heating of the samples was studied using thermogravimetric analysis. The preparation with 10 wt.% AlF3⋅3H2O had the optimal sintering inhibition effect on CCSK at 1000°C. The release of SiF4 gas during heating and the formation of mullite whiskers on the particle surface caused by AlF3⋅3H2O moderated the formation of molten-phase liquid bridges between particles and inhibited sintering of the raw material powder. Furthermore, the formation of submicron mullite whiskers on the surface of the CCSK particles enhanced the oil-absorption properties of the sample significantly.
Temporal contrast directly affects the interaction between ultraintense and ultrashort pulse lasers with matter. Seed laser sources with broad bandwidth and high temporal contrast are significant for overall temporal contrast enhancement. The technique of cascaded nonlinear processes with optical parametric amplification and second-harmonic generation is demonstrated for high temporal contrast seed source generation. Within 40 ps before the main pulse, the temporal contrast reaches over 1011. The pulse energy and duration of the high-contrast pulse are 112 μJ and 70 fs, respectively. Considering its high beam quality and stability, this laser source can serve as a high-quality seed for Nd:glass-based ultraintense and ultrashort pulse laser facilities.
Infrared femtosecond optical vortices open up many new research fields, such as optical micro–nano manipulation, time-resolved nonlocal spectroscopy in solids, vortex secondary radiation and particle generations. In this article, we demonstrate a femtosecond optical vortex laser system based on a two-stage optical parametric amplifier. In our experiment, 1.45 μm vortex signal pulses with energy of 190 μJ and 1.8 μm vortex idler pulses with energy of 158 μJ have been obtained, and the pulse durations are 51 and 48 fs, respectively. Both the energy fluctuations of the signal and idler pulses are less than 0.5% (root mean square), and the spectral fluctuations are less than 1.5% within 1 hour. This type of highly stable femtosecond optical vortex laser has a wide range of applications for vortex strong-field physics.
We have grown intermetallic ErPd2Si2 single crystals employing laser diodes with the floating-zone method. The temperature dependence of the unit-cell parameters was determined using synchrotron and in-house X-ray powder diffraction measurements from 20 to 500 K. The diffraction patterns fit well with the tetragonal I4/mmm space group (No. 139) with two chemical formulae within the unit cell. The synchrotron powder diffraction study shows that the refined unit-cell parameters are a = 4.10320(2) Å, c = 9.88393(5) Å at 298 K and a = 4.11737(2) Å, c = 9.88143(5) Å at 500 K, resulting in the unit-cell volume V = 166.408(1) Å3 (298 K) and 167.517(2) Å3 (500 K). In the whole studied temperature range, no structural phase transition was observed. Upon cooling, the unit-cell parameters a and c are shortened and elongated, respectively.
T long-term effects of cognitive therapy and behavior therapy (CTBT) for menopausal symptoms are unknown, and whether the effects are different between natural menopause and treatment-induced menopause are currently unclear. Therefore, we sought to conduct an accurate estimate of the efficacy of CTBT for menopausal symptoms.
Methods
We conducted searches of Cochrane Library, EMBASE, PsycINFO, PubMed, and Web of Science databases for studies from 1 January 1977 to 1 November 2021. Randomized controlled trials (RCTs) comparing intervention groups to control groups for menopausal symptoms were included. Hedge's g was used as the standardized between-group effect size with a random-effects model.
Results
We included 14 RCTs comprising 1618 patients with a mean sample size of 116. CTBT significantly outperformed control groups in terms of reducing hot flushes [g = 0.39, 95% confidence interval (CI) 0.23–0.55, I2 = 45], night sweats, depression (g = 0.50, 95% CI 0.34–0.66, I2 = 51), anxiety (g = 0.38, 95% CI 0.23–0.54, I2 = 49), fatigue, and quality of life. Egger's test indicated no publication bias.
Conclusions
CTBT is an effective psychological treatment for menopausal symptoms, with predominantly small to moderate effects. The efficacy is sustained long-term, although it declines somewhat over time. The efficacy was stronger for natural menopause symptoms, such as vasomotor symptoms, than for treatment-induced menopause symptoms. These findings provide support for treatment guidelines recommending CTBT as a treatment option for menopausal symptoms.
Schizophrenia is a severely debilitating psychiatric disorder with high heritability and polygenic architecture. A higher polygenic risk score for schizophrenia (SzPRS) has been associated with smaller gray matter volume, lower activation, and decreased functional connectivity (FC). However, the effect of polygenic inheritance on the brain white matter microstructure has only been sparsely reported.
Methods
Eighty-four patients with first-episode schizophrenia (FES) patients and ninety-three healthy controls (HC) with genetics, diffusion tensor imaging (DTI), and resting-state functional magnetic resonance imaging (rs-fMRI) data were included in our study. We investigated impaired white matter integrity as measured by fractional anisotropy (FA) in the FES group, further examined the effect of SzPRS on white matter FA and FC in the regions connected by SzPRS-related white matter tracts.
Results
Decreased FA was observed in FES in many commonly identified regions. Among these regions, we observed that in the FES group, but not the HC group, SzPRS was negatively associated with the mean FA in the genu and body of corpus callosum, right anterior corona radiata, and right superior corona radiata. Higher SzPRS was also associated with lower FCs between the left inferior frontal gyrus (IFG)–left inferior temporal gyrus (ITG), right IFG–left ITG, right IFG–left middle frontal gyrus (MFG), and right IFG–right MFG in the FES group.
Conclusion
Higher polygenic risks are linked with disrupted white matter integrity and FC in patients with schizophrenia. These correlations are strongly driven by the interhemispheric callosal fibers and the connections between frontotemporal regions.
Nutritional Risk Screening index is a standard tool to assess nutritional risk, but epidemiological data are scarce on controlling nutritional status (CONUT) as a prognostic marker in acute haemorrhagic stroke (AHS). We aimed to explore whether the CONUT may predict a 3-month functional outcome in AHS. In total, 349 Chinese patients with incident AHS were consecutively recruited, and their malnutrition risks were determined using a high CONUT score of ≥ 2. The cohort patients were divided into high-CONUT (≥ 2) and low-CONUT (< 2) groups, and primary outcomes were a poor functional prognosis defined as the modified Rankin Scale (mRS) score of ≥ 3 at post-discharge for 3 months. Odds ratios (OR) with 95 % confidence intervals (CI) for the poor functional prognosis at post-discharge were estimated by using a logistic analysis with additional adjustments for unbalanced variables between the high-CONUT and low-CONUT groups. A total of 328 patients (60·38 ± 12·83 years; 66·77 % male) completed the mRS assessment at post-discharge for 3 months, with 172 patients at malnutrition risk at admission and 104 patients with a poor prognosis. The levels of total cholesterol and total lymphocyte counts were significantly lower in high-CONUT patients than low-CONUT patients (P = 0·012 and < 0·001, respectively). At 3-month post discharge, there was a greater risk for the poor outcome in the high-CONUT compared with the low-CONUT patients at admission (OR: 2·32, 95 % CI: 1·28, 4·17). High-CONUT scores independently predict a 3-month poor prognosis in AHS, which helps to identify those who need additional nutritional managements.
The disease burden of infectious diarrhea cannot be underestimated. Its seasonal patterns indicate that weather patterns may play an important role and have an important effect on it. The objective of this study was to clarify the relationship between temperature and infectious diarrhea, and diarrhea-like illness.
Methods:
Distributed lag non-linear model, which was based on the definition of a cross-basis, was used to examine the effect.
Results:
Viral diarrhea usually had high incidence in autumn-winter and spring with a peak at -6°C; Norovirus circulated throughout the year with an insignificant peak at 8°C, while related bacteria usually tested positive in summer and peaked at 22°C. The lag-response curve of the proportion of diarrhea-like cases in outpatient and emergency cases revealed that at -6°C, with the lag days increasing, the proportion increased. Similar phenomena were observed at the beginning of the curves of virus and bacterial positive rate, showing that the risk increased as the lag days increased, peaking on days 16 and 9, respectively. The shape of lag-response curve of norovirus positive rate was different from others, presenting m-type, with 2 peaks on day 3 and day 18.
Conclusion:
Weather patterns should be taken into account when developing surveillance programs and formulating relevant public health intervention strategies.
To determine which set of BMI cut-offs is the most appropriate to define child and adolescent obesity in urban China.
Design:
A cross-sectional study was carried out between 1 November and 31 December in 2017.
Setting:
Community Healthcare Center in Minhang District, Shanghai, China.
Participants:
A total of 12 426 children and adolescents aged 7–17 years were selected by cluster random sampling. Bioelectrical impedance analysis was the gold standard to measure body composition.
Results:
Comparisons of three sets of BMI cut-offs by sensitivity and κ value revealed that the Working Group on Obesity in China (WGOC) (sensitivity 39·9–84·0 %; κ 0·51–0·79) and WHO (sensitivity 25·5–74·5 %; κ 0·35–0·78) cut-offs were not superior to the International Obesity Task Force (IOTF) (sensitivity 47·9–92·4 %; κ 0·58–0·85) cut-offs across all subgroups. The WGOC and WHO cut-offs yielded higher misclassification rates, in the worst case, categorising 11·2 % of girls with high adiposity as normal and 44·4 % of them as overweight, while the IOTF cut-offs categorised 2·3 % as normal and 30·7 % as overweight. Individuals who were classified by the IOTF cut-offs as overweight had the lowest ratios of high adiposity (4·2–41·6 %) than by the BMI cut-offs for each subgroup. Among pubertal girls, none of the BMI-based cut-offs indicated excellent agreement with body fat percentage, and κ value of the WHO cut-offs (0·35 (95 % CI 0·29, 0·41)) was lower than the other two sets of BMI cut-offs (all P < 0·001).
Conclusions:
The IOTF cut-offs for Asian should be recommended for child obesity screening in urban China. Pubertal individuals need a more accurate indicator of obesity screening.
Synaptotagmin 1 (Syt1) is an abundant and important presynaptic vesicle protein that binds Ca2+ for the regulation of synaptic vesicle exocytosis. Our previous study reported its localization and function on spindle assembly in mouse oocyte meiotic maturation. The present study was designed to investigate the function of Syt1 during mouse oocyte activation and subsequent cortical granule exocytosis (CGE) using confocal microscopy, morpholinol-based knockdown and time-lapse live cell imaging. By employing live cell imaging, we first studied the dynamic process of CGE and calculated the time interval between [Ca2+]i rise and CGE after oocyte activation. We further showed that Syt1 was co-localized to cortical granules (CGs) at the oocyte cortex. After oocyte activation with SrCl2, the Syt1 distribution pattern was altered significantly, similar to the changes seen for the CGs. Knockdown of Syt1 inhibited [Ca2+]i oscillations, disrupted the F-actin distribution pattern and delayed the time of cortical reaction. In summary, as a synaptic vesicle protein and calcium sensor for exocytosis, Syt1 acts as an essential regulator in mouse oocyte activation events including the generation of Ca2+ signals and CGE.
A magnetic dipole quasi-Yagi antenna based on a dielectric resonator (DR) is proposed in this letter. The dominate TE1δ1 mode of the rectangular DR is used as a magnetic dipole which can be differentially fed by the coplanar strip line (CPS). Thanks to the DR employment, the proposed antenna has several advantages such as compact size and flexible design which means the 3-D dimensions of the DR can be easily adjusted to cater for various applications. Meanwhile, the gain of the proposed DR quasi-Yagi antenna is higher than that of traditional electric dipole counterparts. Furthermore, since the DR driver is horizontally polarized, both the metal strip and DR can be used as a director for enhancing the end-fire gain. To verify the design concept, a prototype operating at the X-band is fabricated and measured. Good agreement between the simulated and measured results can be observed.
Electrospun coaxial fibers are used to create core/sheath fiber structures to act as growth-promoting scaffolds for in vitro dorsal root ganglia (DRG) cell cultures. The core was a conducting polymer, poly(3,4-ethylenedioxythiophene):poly(styrene sulfonate) (PEDOT:PSS), and the sheath was poly-L-lactic acid (PLLA), which created coaxial fibers with a conductive core and an insulating sheath. SEM analysis confirmed the conductivity of the core and insulation of the sheath. Several coaxial spinneret designs were tested with the best results obtained by using various annular spinning needle combinations. Using a 22G/16G and 22G/17G combination, fibers with diameters of 6.1 ± 2.4 µm and 3.3 ± 0.9 µm were spun, respectively. The fibers showed a Young’s modulus and hardness of 0.16 ± 0.13 and 0.02 ± 0.01 GPa for the larger diameters, and 0.7 ± 0.4 and 0.03 ± 0.03 GPa for the smaller diameter fibers. In vitro test cultures showed the fibers successfully directed chick DRG axonal outgrowth with low biotoxicity.
We show that chief executive officers (CEOs) exhibit a hometown bias in acquisitions. Firms are over twice as likely to acquire targets located in the states of their CEOs’ childhood homes than similar targets domiciled elsewhere. Small, private home-state deals underperform other small, private deals, and the bias is stronger when acquirer governance is lax, suggesting that CEOs acquire private home-state targets for their own benefits. In contrast, large, public home-state acquisitions are value enhancing. CEOs create value in public home-state acquisitions by avoiding extremely poor deals and through deals with higher synergies. Thus, both agency issues and hometown advantages drive home-state acquisitions.
