Changes in vocal fold gene expression and histology after injection augmentation in a recurrent laryngeal nerve injury model

Objective
To investigate changes in neuroregenerative pathways with vocal fold denervation in response to vocal fold augmentation.
MethodsEighteen Yorkshire crossbreed swine underwent left recurrent laryngeal nerve transection, followed by observation or augmentation with carboxymethylcellulose or calcium hydroxyapatite at two weeks. Polymerase chain reaction expression of genes regulating muscle growth (MyoD1, MyoG and FoxO1) and atrophy (FBXO32) were analysed at 4 and 12 weeks post-injection. Thyroarytenoid neuromuscular junction density was quantified using immunohistochemistry.
ResultsDenervated vocal folds demonstrated reduced expression of MyoD1, MyoG, FoxO1 and FBXO32, but overexpression after augmentation. Healthy vocal folds showed increased early and late MyoD1, MyoG, FoxO1 and FBXO32 expression in all animals. Neuromuscular junction density had a slower decline in augmented compared to untreated denervated vocal folds, and was significantly reduced in healthy vocal folds contralateral to augmentation.
ConclusionInjection augmentation may slow neuromuscular degeneration pathways in denervated vocal folds and reduce compensatory remodelling in contralateral healthy vocal folds.

The COllaborative project of Development of Anthropometrical measures in Twins (CODATwins) project is a large international collaborative effort to analyze individual-level phenotype data from twins in multiple cohorts from different environments. The main objective is to study factors that modify genetic and environmental variation of height, body mass index (BMI, kg/m2) and size at birth, and additionally to address other research questions such as long-term consequences of birth size. The project started in 2013 and is open to all twin projects in the world having height and weight measures on twins with information on zygosity. Thus far, 54 twin projects from 24 countries have provided individual-level data. The CODATwins database includes 489,981 twin individuals (228,635 complete twin pairs). Since many twin cohorts have collected longitudinal data, there is a total of 1,049,785 height and weight observations. For many cohorts, we also have information on birth weight and length, own smoking behavior and own or parental education. We found that the heritability estimates of height and BMI systematically changed from infancy to old age. Remarkably, only minor differences in the heritability estimates were found across cultural–geographic regions, measurement time and birth cohort for height and BMI. In addition to genetic epidemiological studies, we looked at associations of height and BMI with education, birth weight and smoking status. Within-family analyses examined differences within same-sex and opposite-sex dizygotic twins in birth size and later development. The CODATwins project demonstrates the feasibility and value of international collaboration to address gene-by-exposure interactions that require large sample sizes and address the effects of different exposures across time, geographical regions and socioeconomic status.

In North America, terrestrial records of biodiversity and climate change that span Marine Oxygen Isotope Stage (MIS) 5 are rare. Where found, they provide insight into how the coupling of the ocean–atmosphere system is manifested in biotic and environmental records and how the biosphere responds to climate change. In 2010–2011, construction at Ziegler Reservoir near Snowmass Village, Colorado (USA) revealed a nearly continuous, lacustrine/wetland sedimentary sequence that preserved evidence of past plant communities between ~140 and 55 ka, including all of MIS 5. At an elevation of 2705 m, the Ziegler Reservoir fossil site also contained thousands of well-preserved bones of late Pleistocene megafauna, including mastodons, mammoths, ground sloths, horses, camels, deer, bison, black bear, coyotes, and bighorn sheep. In addition, the site contained more than 26,000 bones from at least 30 species of small animals including salamanders, otters, muskrats, minks, rabbits, beavers, frogs, lizards, snakes, fish, and birds. The combination of macro- and micro-vertebrates, invertebrates, terrestrial and aquatic plant macrofossils, a detailed pollen record, and a robust, directly dated stratigraphic framework shows that high-elevation ecosystems in the Rocky Mountains of Colorado are climatically sensitive and varied dramatically throughout MIS 5.

A trend toward greater body size in dizygotic (DZ) than in monozygotic (MZ) twins has been suggested by some but not all studies, and this difference may also vary by age. We analyzed zygosity differences in mean values and variances of height and body mass index (BMI) among male and female twins from infancy to old age. Data were derived from an international database of 54 twin cohorts participating in the COllaborative project of Development of Anthropometrical measures in Twins (CODATwins), and included 842,951 height and BMI measurements from twins aged 1 to 102 years. The results showed that DZ twins were consistently taller than MZ twins, with differences of up to 2.0 cm in childhood and adolescence and up to 0.9 cm in adulthood. Similarly, a greater mean BMI of up to 0.3 kg/m2 in childhood and adolescence and up to 0.2 kg/m2 in adulthood was observed in DZ twins, although the pattern was less consistent. DZ twins presented up to 1.7% greater height and 1.9% greater BMI than MZ twins; these percentage differences were largest in middle and late childhood and decreased with age in both sexes. The variance of height was similar in MZ and DZ twins at most ages. In contrast, the variance of BMI was significantly higher in DZ than in MZ twins, particularly in childhood. In conclusion, DZ twins were generally taller and had greater BMI than MZ twins, but the differences decreased with age in both sexes.

For over 100 years, the genetics of human anthropometric traits has attracted scientific interest. In particular, height and body mass index (BMI, calculated as kg/m2) have been under intensive genetic research. However, it is still largely unknown whether and how heritability estimates vary between human populations. Opportunities to address this question have increased recently because of the establishment of many new twin cohorts and the increasing accumulation of data in established twin cohorts. We started a new research project to analyze systematically (1) the variation of heritability estimates of height, BMI and their trajectories over the life course between birth cohorts, ethnicities and countries, and (2) to study the effects of birth-related factors, education and smoking on these anthropometric traits and whether these effects vary between twin cohorts. We identified 67 twin projects, including both monozygotic (MZ) and dizygotic (DZ) twins, using various sources. We asked for individual level data on height and weight including repeated measurements, birth related traits, background variables, education and smoking. By the end of 2014, 48 projects participated. Together, we have 893,458 height and weight measures (52% females) from 434,723 twin individuals, including 201,192 complete twin pairs (40% monozygotic, 40% same-sex dizygotic and 20% opposite-sex dizygotic) representing 22 countries. This project demonstrates that large-scale international twin studies are feasible and can promote the use of existing data for novel research purposes.

Deep water waves in two-dimensional flow can have curvature singularities on the surface profile; for example, the limiting Stokes wave has a corner of radians and the limiting standing wave momentarily forms a corner of radians. Much less is known about the possible formation of curvature singularities in general. A novel way of exploring this possibility is to consider the curvature as a complex function of the complex arclength variable and to seek the existence and nature of any singularities in the complex arclength plane. Highly accurate boundary integral methods produce a Fourier spectrum of the curvature that allows the identification of the nearest singularity to the real axis of the complex arclength plane. This singularity is in general a pole singularity that moves about the complex arclength plane. It approaches the real axis very closely when waves break and is associated with the high curvature at the tip of the breaking wave. The behaviour of these singularities is more complex for standing waves, where two singularities can be identified that may collide and separate. One of them approaches the real axis very closely when a standing wave forms a very narrow collapsing column of water almost under free fall. In studies so far, no singularity reaches the real axis in finite time. On the other hand, the surface elevation has square-root singularities in the complex plane that do reach the real axis in finite time, the moment when a wave first starts to break. These singularities have a profound effect on the wave spectra.

The instability of an initially flat vortex sheet to a sinusoidal perturbation of the vorticity is studied by means of high-order Taylor series in time t. All finite-amplitude corrections are retained at each order in t. Our analysis indicates that the sheet develops a curvature singularity at t = tc < ∞. The variation of tc with the amplitude a of the perturbation vorticity is in good agreement with the asymptotic results of Moore. When a is O(1), the Fourier coefficient of order n decays slightly faster than predicted by Moore. Extensions of the present prototype of Kelvin-Helmholtz instability to other layered flows, such as Rayleigh-Taylor instability, are indicated.

The motion of free surfaces in incompressible, irrotational, inviscid layered flows is studied by evolution equations for the position of the free surfaces and appropriate dipole (vortex) and source strengths. The resulting Fredholm integral equations of the second kind may be solved efficiently in both storage and work by iteration in both two and three dimensions. Applications to breaking water waves over finite-bottom topography and interacting triads of surface and interfacial waves are given.

Fink & Soh (1978) reported a technique to calculate numerically the motion of vortex sheets. They claim it gives reliable results. This paper re-examines the error in the calculation of the velocity of mesh points representing the sheet and shows that the test case used by Fink & Soh is not an adequate one. Instead the roll-up of a vortex sheet shed by a ring wing is studied. The results obtained proved unreliable. (Although several possibilities are discussed) the reason for the breakdown in results remains unknown.

The energetics of a linear array of hollow or stagnant-cored vortices of finite cross-section in an ideal fluid is studied in this paper. The results are useful in a discussion of the amalgamation of vortex structures in a turbulent mixing layer.

The motion of vortex sheets is susceptible to the onset of the Kelvin–Helmholz instability. There is now a large body of evidence that the instability leads to the formation of a curvature singularity in finite time. Vortex blob methods provide a regularization for the motion of vortex sheets. Instead of forming a curvature singularity in finite time, the curves generated by vortex blob methods form spirals. Theory states that these spirals will converge to a classical weak solution of the Euler equations as the blob size vanishes. This theory assumes that the blob method is the result of a convolution of the sheet velocity with an appropriate choice of a smoothing function. We consider four different blob methods, two resulting from appropriate choices of smoothing functions and two not. Numerical results indicate that the curves generated by these methods form different spirals, but all approach the same weak limit as the blob size vanishes. By scaling distances and time appropriately with blob size, the family of spirals generated by different blob sizes collapse almost perfectly to a single spiral. This observation is the next step in developing an asymptotic theory to describe the nature of the weak solution in detail.
