Cannabidiol (CBD) is one of the main cannabinoids present in Cannabis sativa female flowers. Previous investigation has already provided insights into the CBD molecular mechanism; however, there is no transcriptome data for CBD effects on hippocampal subfields. Here, we investigate transcriptomic changes in dorsal and ventral CA1 of adult mice hippocampus after 100 mg/kg of CBD administration (i.p.) for one or seven consecutive days.
Methods:
C57BL/6JUnib mice were treated with either vehicle or CBD for 1 or 7 days. The collected brains were sectioned, and the hippocampal sub-regions were laser microdissected for RNA-Seq analysis.
Results:
The transcriptome analysis following 7 days of CBD administration indicates the differential expression of 1559 genes in dCA1 and 2924 genes in vCA1. Furthermore, GO/KEGG analysis identified 88 significantly enriched biological process and 26 significantly enriched pathways for dCBD7, whereas vCBD7 revealed 128 enriched BPs and 24 pathways.
Conclusion:
This dataset indicates a widespread decrease of electron transport chain and ribosome biogenesis transcripts in CA1, while chromatin modifications and synapse organization transcripts were increased following CBD administration for 7 days.
In light of Mozambique’s natural resources boom—especially its large-scale investments in mining, oil, and gas—this chapter analyses the prospects for the extractive industries to contribute to economic transformation from an institutional perspective. For this purpose, we address the institutional dynamics of the resources sector and consider the underlying causes of the identified outcomes, and we discuss the National Development Strategy, as the instrument outlining the vision for economic transformation and diversification. The chapter is based on a desk review—documental and bibliographic—and on primary data gathered by the authors as part of their research into the field of natural resources and the political economy of development. We conclude that, given Mozambique’s political patronage and clientelism, intra-ruling elite competition, limited productive base, weak state capacity, high level of poverty, and recurrent fiscal deficits, the prospects of the current resource boom leading to economic transformation, despite its considerable potential, are at best uncertain.
During the second half of the 20th century, the Antarctic Peninsula region has undergone a long and sustained warming period, followed by a shorter but also sustained cooling period, and then a very recent return to warming conditions. All of these have profoundly impacted the glaciers peripheral to the Antarctic Peninsula. This paper focuses on the analysis of the surface mass balance monitoring of such glaciers by the glaciological method, complemented by the analysis of mass-balance estimates by geodetic methods, as well as frontal ablation estimates. We aim to summarize the current knowledge and outline the main challenges faced by investigating the mass balance of such peripheral glaciers and their current contribution to sea-level rise.
Rotavirus (RV) was a common healthcare-associated infection prior to the introduction of the RV vaccine. Following widespread RV vaccination, healthcare-associated rotavirus cases are rare. We describe an investigation of a cluster of rotavirus infections in a pediatric hospital in which an uncommon genotype not typically circulating in the United States was detected.
Despite the widely recognized value of wetlands in providing vital ecosystem services, these are presently being degraded and ultimately destroyed, leading to a decrease in the biodiversity associated with these areas. Some species inextricably linked to wetlands, however, have been increasing and (re)colonizing areas across their range; a notable example being the Eurasian Spoonbill Platalea leucorodia. In this study we aimed to identify the most important habitats for juvenile spoonbills fledging from a traditional colony in Portugal, located in Ria Formosa, during the period of their life with the lowest survival rates: the first months after leaving the colony. We deployed 16 GPS/GSM tags on juveniles captured in different years (2016 to 2020) and tracked them during post-fledging dispersal and first winter (average 166.4 ± 29.2 SE days). Using Corine Land Cover data, we were able to identify which habitats were most important. Several habitats were used in variable proportions by individuals originating from the same colony, but there was a general trend towards using fewer habitats along the first months of life. Intertidal wetlands were the most used habitat, but anthropogenic habitats such as Wastewater Treatment Plants, saltpans and rice fields were identified as alternative habitats for young spoonbills, and may had contributed to the recent expansion of this species in Portugal.
Our aim was to assess the value of nintedanib for non-idiopathic progressive fibrosing interstitial lung disease (non-IPF PF-ILD) and systemic sclerosis-associated ILD (SSc-ILD) in the Spanish context, using a multi-criteria decision analysis (MCDA).
Methods
Following an adaptation of the Evidence and Value: Impact on DEcision Making (EVIDEM) MCDA methodology, the estimated value of nintedanib was obtained by means of an additive linear model that combined individual weights (100-points distribution) of criteria with the individual scoring of nintedanib in each criterion for every indication, assigned by a multidisciplinary committee of twelve clinicians, patients, pharmacists, and decision-makers. To assess the reproducibility, an alternative weighting method was applied, as well as a re-test of weights and scores at a different moment of time.
Results
The experts committee recognized nintedanib as an intervention with a positive value contribution in comparison to placebo for the treatment of non-IPF PF-ILD (0.50 ± 0.16, on a scale from −1 to 1) and SSc-ILD (0.40 ± 0.12), diseases which were considered as very severe and with high unmet needs. The drug was perceived as a treatment that provides an added therapeutic benefit for patients (0.06–0.07), given its proven clinical efficacy (0.05–0.06), slight improvements in patient-reported outcomes (0.01–0.02), and similar safety profile than placebo (−0.04–0.00), which will likely be positioned as a recommended therapy in the next clinical practice guidelines updates.
Conclusions
Under this increasingly used methodology, nintedanib has shown to provide a positive value estimate for non-IPF PF-ILD and SSc-ILD when compared to placebo in Spain.
Genetic gains (ΔG) are determined by the breeders' equation ΔG = [(ck σ2G)/(y σP)], where c, k and y are the parental control, a function of the selection intensity and number of years to complete one selection cycle, respectively, while σ2G and are σP the genetic variance and the square root of the phenotypic variance. Plant breeding programs should deliver above 1% of annual genetic gains after testing and selection. The aim of this research was to estimate genetic gains in potato breeding after testing of cultivars released in western Europe in the last 200 years under high yield potential, and stress-prone environments affected by a pest (late blight) or daylength. The annual genetic gains for tuber yield and flesh's starch content for potato breeding in Europe were about 0.3 and −0.1%, respectively, thus telling that the realized genetic gains of foreign cultivars for both traits are small or negative, respectively, in the Nordic testing sites. The national annual productivity gains in potato grown in Sweden were on average 0.7% in the last 60 years while the genetic gains for tuber yield considering only the table cultivars released after the 2nd World War were about 0.36%, thus showing that breeding contributed just above ½ of it. Furthermore, genetic gains for breeding low reducing sugars in the tuber flesh, and high host plant resistance to late blight were small (<0.2% per year). These results highlight that genetic gains are small when testing bred germplasm outside their target population of environments.
Fertility, pregnancy and childbirth are connected and inseparable realities that ultimately need to combine to result in a “baby at home”; each one of these phenomena possesses their own suite of challenges and limitations. From a biological viewpoint, the factors contributing to a successful “baby at home” are male factors (mainly sperm quality) and female factors (mainly oocyte quality and endometrial receptivity). Any failure or disability in one of these competences shall likely contribute to the reduced probability of having a “baby at home”. However, in modern times a new factor is playing a crucial role in this scenario, the so called “Artificial Reproduction Techniques” (ART). Thus, it is becoming increasingly possible for reproductive biologists and clinicians to apply a range of technologies to transform a biologically infertile couple to an assisted fertile one. To this purpose, a series of novel technologies, some of them with ethical implications at the time of their use on a routine basis, have and will continue to have a remarkable potential impact on fertility treatments [1].
Carmen made its debut in the Spanish Americas when the local networks of opera were at their apex, with a constant stream of singers from Europe and a desire for new repertoires outside the main staples of Italian opera. In this chapter, considering sources from across the region, we discuss four layers of Carmen’s reception in the Americas. First, the reception of the opera beyond the stage, in the form of vocal scores, arrangements for military bands and isolated numbers. Second, the perception of Carmen as a French opera, and the way it served as a vehicle for French opera companies in the final decades of the nineteenth century. Third, the idea of Carmen as Spanish, and how different countries considered that hispanicity as part of their own culture and theatrical expectations. Finally, we discuss how the habanera in Carmen was perceived as part of a larger contemporary debate on the transatlantic popularity of the habanera as a musical genre, its origins, ethnicity and its moral and musical character.
Several studies have independently suggested that patients with schizophrenia are more likely to have an enlarged cavum septum pellucidum (CSP) and an absent adhesio interthalamica (AI), respectively. However, neither finding has been consistently replicated and it is unclear whether there is an association between these two midline brain abnormalities. Thus, we compared the prevalence of absent AI and the prevalence, size and volume of CSP in 38 patients with schizophrenia and 38 healthy controls using magnetic resonance imaging (MRI). There were no between group differences in the presence or volume of CSP; however, an enlarged CSP was commoner among patients than controls. There was also a positive correlation between CSP ratings and volumes. No differences in the presence or absence of the AI were found between patients and controls; however, an absent AI was commoner in male patients with schizophrenia than females. There was absolutely no overlap between the presence of a large CSP and an absence of AI. In conclusion, our findings are in line with several case series and other MRI investigations that have shown a higher incidence of putatively developmental brain abnormalities in patients with schizophrenia, particularly in males, and support the neurodevelopmental model of this disorder.
In the current work, we compared the green synthesis of silver nanoparticles (AgNP) using plant extracts, a promising methodology against the use of chemical reducers, such as oleic acid and oleylamine. The advantages of green synthesis are one-step method, economic and ecological while the advantages of classic synthesis methods are high nanoparticle performance, homogeneity in size and smaller average sizes. With this work we want to demonstrate that plant extracts with specific mixtures of chemical compounds can obtain smaller average sizes with greater homogeneity in nanoparticles compared to the use of classical synthesis. Myrtillocactus geometrizans was used as a polar plant extract, which was selected by the chemical components contained in the extract. Phytosterols, oleic acid and betalains contained in Myrtillocactus geometrizans are biomolecules responsible for the reduction and stability of AgNP below 5 nm. TEM analysis of the green synthesis of nanoparticles revealed the formation of spherical particles with an average diameter of 5 nm and with preferential crystallographic directions of the silver plane [111].
A student's t-test was applied in carbon nanospheres synthesis from cis-1,4-polyisoprene considering the green chemical principles. The synthesis was carried out by Chemical Vapor Deposition method with a quartz tube reactor using an AISI 304 steel bar as catalyst. It was possible to obtain two types of different samples, one from the surface of the steel bar (catalyst) and another from the quartz tube surface (without catalyst) in the same experiment. Carbon spheres were observed in both samples by micrographs obtained by FESEM. The Raman and FTIR spectroscopies shown characteristic bands of this carbon structures (G and D). The results obtained by student's t-test proved a statistical significance between spheres means of samples collected from steel bar and quartz tube surface.
The main sequence offers a method for the systematization of quasar spectral properties. Extreme FeII emitters (or extreme Population A, xA) are believed to be sources accreting matter at very high rates. They are easily identifiable along the quasar main sequence, in large spectroscopic surveys over a broad redshift range. The very high accretion rate makes it possible that massive black holes hosted in xA quasars radiate at a stable, extreme luminosity-to-mass ratio. After reviewing the basic interpretation of the main sequence, we report on the possibility of identifying virial broadening estimators from low-ionization line widths, and provide evidence of the conceptual validity of redshift-independent luminosities based on virial broadening for a known luminosity-to-mass ratio.
People with epilepsy (PWE) are less physically active compared with the general population. Explanations include prejudice, overprotection, unawareness, stigma, fear of seizure induction and lack of knowledge of health professionals. At present, there is no consensus on the role of exercise in epilepsy. This paper reviews the current evidence surrounding the risks and benefits associated with physical activity (PA) in this group of patients. In the last decade, several publications indicate significant benefits in physiological and psychological health parameters, including mood and cognition, physical conditioning, social interaction, quality of life, as well as potential prevention of seizure presentation. Moreover, experimental studies suggest that PA provides mechanisms of neuronal protection, related to biochemical and structural changes including release of β-endorphins and steroids, which may exert an inhibitory effect on the occurrence of abnormal electrical activity. Epileptic discharges can decrease or disappear during exercise, which may translate into reduced seizure recurrence. In some patients, exercise may precipitate seizures. Available evidence suggests that PA should be encouraged in PWE in order to promote wellbeing and quality of life. There is a need for prospective randomized controlled studies that provide stronger clinical evidence before definitive recommendations can be made.
The aim of this study was to develop and to assess a specific Multi-Criteria Decision Analysis (MCDA) framework to evaluate new drugs in an hospital pharmacy and therapeutics committee (P&TC) setting.
Methods:
A pilot criteria framework was developed based on the EVIDEM (Evidence and Value: Impact on DEcisionMaking) framework, together with other relevant criteria, and assessed by a group of P&TC's members. The weighting of included criteria was done using a 5-point weighting technique. Two drugs were chosen by evaluation: an orphan-drug for Gaucher disease, and a nonorphan drug for the treatment of inflammatory bowel disease. Evidence matrices were developed, and value contribution of each drug was evaluated by P&TC's members. An agreed final framework was obtained through a discussion between the P&TC's members.
Results:
After criteria assessment, the pilot framework included eight quantitative criteria: “disease severity,” “unmet needs,” “comparative efficacy/effectiveness,” “comparative safety/tolerability,” “comparative patient-reported outcomes,” “comparative cost consequences-cost of treatment,” “comparative cost consequences-other medical costs,” and “quality of evidence”; and one contextual criterion: “opportunity costs and affordability.” The most valued criteria were: “comparative safety/tolerability,” “disease severity,” and “comparative efficacy/effectiveness.” When assessing the drugs most valued characteristics of the MCDA were the possibility that all team may contribute to drug assessment by means of scoring the matrices and the discussion to reach a consensus in drug positioning and value decision making.
Conclusions:
The reflective MCDA would integrate quantitative and qualitative criteria relevant for a P&TC setting, allowing reflective discussions based on the criteria weighting score.
Literature reports that body size can be associated with latitudinal distribution, for instance larger animals inhabit higher latitudes and colder habitats. This rule can be applied for species and populations within a species. The potential influence of body size on non-breeding distribution and habitat use at the intra-population level was investigated for southern giant petrels Macronectes giganteus (Gmelin) from Elephant Island, South Shetland Islands. The non-breeding distribution of 23 individuals was tracked, and total body length, culmen length, wing length, wing load and body mass were measured. Positions of core areas were used to estimate the latitudinal distribution of each individual. Smaller individuals were found to be associated more with lower latitudes, where warmer conditions and more coastal and productive waters prevail, whereas large males were associated more with higher latitudes, with colder conditions near sea ice caps, presumably feeding on carrion or preying on penguins. This association reflects a latitudinal gradient, with smaller individuals positioning themselves towards the north, and larger individuals towards the south. In this case, body size, individual distribution and habitat use were found to be associated, highlighting the importance of studying potential effects of individual body size on the ecology of seabirds.
Composites from carbon nanotubes and polymers have been synthesized and studied. The composites were obtained joining carbon nanotubes with polymethyl methacrylate, nylon-6 and polystyrene. The materials were observed through scanning electron microscopy to evaluate the carbon nanotubes dispersion in the polymeric matrices. FTIR and Raman spectroscopies were used to analyze the interactions among functionalized and non-functionalized multiwalled carbon nanotubes and polymers, demonstrating affinity and peculiar spectra behaviors for each composite with different carbon nanotubes loads.
PA6 and PMMA polymers with different MWCNTs addition (5, 7 and 9 wt %) were synthetized through casting solution, resulting in improvement properties in contrast to pristine polymers. SEM images showed the MWCNTs embedded into polymeric matrices. D, G and G´ bands of MWCNTs were confirmed by Raman spectroscopy and functional groups observed in both nanocomposites by FTIR demonstrated a strong interaction. A significant increasing in electrical conductivity and microhardness was observed in all the nanocomposites. Major microhardness values were obtained in MWCNTs/PA6 (50 HV) however the MWCNTs/PMMA nanocomposites showed the highest electrical conductivity value (6.4×10-4 S/cm).
Background: The importance of economic evaluation in decision making is growing with increasing budgetary pressures on health systems. Diverse economic evidence is available for a range of interventions across national contexts within Europe, but little attention has been given to identifying evidence gaps that, if filled, could contribute to more efficient allocation of resources. One objective of the Research Agenda for Health Economic Evaluation project is to determine the most important methodological evidence gaps for the ten highest burden conditions in the European Union (EU), and to suggest ways of filling these gaps.
Methods: The highest burden conditions in the EU by Disability Adjusted Life Years were determined using the Global Burden of Disease study. Clinical interventions were identified for each condition based on published guidelines, and economic evaluations indexed in MEDLINE were mapped to each intervention. A panel of public health and health economics experts discussed the evidence during a workshop and identified evidence gaps.
Results: The literature analysis contributed to identifying cross-cutting methodological and technical issues, which were considered by the expert panel to derive methodological research priorities.
Conclusions: The panel suggests a research agenda for health economics which incorporates the use of real-world evidence in the assessment of new and existing interventions; increased understanding of cost-effectiveness according to patient characteristics beyond the “-omics” approach to inform both investment and disinvestment decisions; methods for assessment of complex interventions; improved cross-talk between economic evaluations from health and other sectors; early health technology assessment; and standardized, transferable approaches to economic modeling.
