This paper explores active wake-flow control on a notchback Ahmed body using genetically inspired optimization. Hotwire and particle image velocimetry measurements record velocity data and flow structures in the wake. Pulsed jets at four actuation slots (two at the roof trailing edge, two at the side trailing edges) dynamically control the wake to minimize aerodynamic drag. The study achieves up to 9.2 % (without consideration of energy consumption) drag reduction, primarily by manipulating vortices from the roof rear end. The paper elucidates the underlying flow mechanism and evaluates various actuation strategies, highlighting how optimal control leads to reattachment of wake separation at the rear slant, diminishing the slant bubble and promoting downstream reattachment for enhanced drag reduction.

Nonlinear compression has become an obligatory technique along with the development of ultrafast lasers in generating ultrashort pulses with narrow pulse widths and high peak power. In particular, techniques of nonlinear compression have experienced a rapid progress as ytterbium (Yb)-doped lasers with pulse widths in the range from hundreds of femtoseconds to a few picoseconds have become mainstream laser tools for both scientific and industrial applications. Here, we report a simple and stable nonlinear pulse compression technique with high efficiency through cascaded filamentation in air followed by dispersion compensation. Pulses at a center wavelength of 1040 nm with millijoule pulse energy and 160 fs pulse width from a high-power Yb:CaAlGdO4 regenerative amplifier are compressed to 32 fs, with only 2.4% loss from the filamentation process. The compressed pulse has a stable output power with a root-mean-square variation of 0.2% over 1 hour.

This paper presents a numerical investigation of the effects of the moving ground and rotating wheels on the turbulent flow around a 1/10 scaled square-back van model. A comprehensive comparison among the partially averaged Navier–Stokes (PANS), large eddy simulation (LES) and particle image velocimetry (PIV) involving the aerodynamic drag, the wake topology, the velocity and the Reynolds stress profiles in the wake region is conducted. The proper orthogonal decomposition (POD) and fast Fourier transform (FFT) are applied to the shear layers shedding from the trailing edges to comment on the coherent structures and their frequency content. The Reynolds number for both simulations and experiments is set to Re = 2.5 × 105 based on the inlet velocity ${U_{inf}} = 9\;\textrm{m}\;{\textrm{s}^{ - 1}}$ and the width of the model W = 0.17 m. The results show that PANS accurately predicts the flow field measured in experiments and predicted by a resolved LES, even with a low-resolution grid. The superiority of the PANS approach could provide good guidance for industrial research in predicting the turbulent flow around the square-back van model with affordable computational grids. The ground and wheel motion mechanism on the aerodynamic forces has been revealed by analysing the surface pressure distribution, the wheels’ surrounding flow, the underbody flow characteristics and the turbulent wake structures. The effects of the ground and wheel motion on the frequency, evolution and development characteristics of the wake shear layers are analysed, thus providing relevant insights for future experimental investigations of square-back van models.

The wake of a notchback Ahmed body presenting a bi-stable nature is investigated by performing wind tunnel experiments and large-eddy simulations. Attention is confined to the Reynolds number ($Re$) influence on the wake state instability within $5\times 10^{4}\leq Re \leq 25\times 10^{4}$. Experimental observations suggest a wake bi-stability with low-frequency switches under low $Re$. The wake becomes ‘tri-stable’ with the increase of $Re$ with the introduction of a new symmetric state. The higher presence of the symmetric state can be considered as a symmetrization of the wake bi-stability with an increasing $Re$. The wake symmetry under high $Re$ attributed to the highly frequent switches of the wake is extremely sensitive to small yaw angles, showing the feature of bi-stable flows. The wake asymmetry is confirmed in numerical simulations with both low and high $Re$. The wake asymmetries are indicated by the wake separation, the reattachment and the wake dynamics identified by the proper orthogonal decomposition. However, the turbulence level is found to be significantly higher with a higher $Re$. This leads to a higher possibility to break the asymmetric state, resulting in highly frequent switches showing symmetry.

Large-eddy simulations are used to investigate the origin of the wake asymmetry and symmetry behind notchback Ahmed bodies. Two different effective backlight angles, ${\beta _1} = 17.8\mathrm{^\circ }$ and ${\beta _2} = 21.0\mathrm{^\circ }$, are simulated resulting in wake asymmetry and symmetry in flows without external perturbations, in agreement with previous experimental observations. In particular, the asymmetric case presents a bi-stable nature showing, in a random fashion, two stable mirrored states characterized by a left or right asymmetry for long periods. A random switch and several attempts to switch between the bi-stability are observed. The asymmetry of the flow is ascribed to the asymmetric separations and reattachments in the wake. The deflection of the near-wall flow structures behind the slant counteracting the asymmetry drives the wake to be temporarily symmetric, triggering the switching process of the bi-stable wake. The consequence of deflection that forces the flow structure to form on the opposite side of the slant is the decisive factor for a successful switch. Modal analysis applying proper orthogonal decomposition is used for the exploration of the wake dynamics of the bi-stable nature observed.

The fundamental step of learning transcriptional regulation mechanism is to identify the target genes regulated by transcription factors (TFs). Despite numerous target genes identified by chromatin immunopre-cipitation followed by high-throughput sequencing technology (ChIP-seq) assays, it is not possible to infer function from binding alone in vivo. This is equally true in one of the best model systems, the nematode Caenorhabditis elegans (C. elegans), where regulation often occurs through diverse TF binding features of transcriptional networks identified in modENCODE. Here, we integrated ten ChIP-seq datasets with genome-wide expression data derived from tiling arrays, involved in six TFs (HLH-1, ELT-3, PQM-1, SKN-1, CEH-14 and LIN-11) with tissue-specific and four TFs (CEH-30, LIN-13, LIN-15B and MEP-1) with broad expression patterns. In common, TF bindings within 3 kb upstream of or within its target gene for these ten studies showed significantly elevated level of expression as opposed to that of non-target controls, indicated that these sites may be more likely to be functional through up-regulating its target genes. Intriguingly, expression of the target genes out of 5 kb upstream of their transcription start site also showed high levels, which was consistent with the results of following network component analysis. Our study has identified similar transcriptional regulation mechanisms of tissue-specific or broad expression TFs in C. elegans using ChIP-seq and gene expression data. It may also provide a novel insight into the mechanism of transcriptional regulation not only for simple organisms but also for more complex species.

Gene expression profiling of peroxisome-proliferator-activated receptor α (PPARα) has been used in several studies, but there were no consistent results on gene expression patterns involved in PPARα activation in genome-wide due to different sample sizes or platforms. Here, we employed two published microarray datasets both PPARα dependent in mouse liver and applied meta-analysis on them to increase the power of the identification of differentially expressed genes and significantly enriched pathways. As a result, we have improved the concordance in identifying many biological mechanisms involved in PPARα activation. We suggest that our analysis not only leads to more identified genes by combining datasets from different resources together, but also provides some novel hepatic tissue-specific marker genes related to PPARα according to our re-analysis.

We suggest that the broad-line regions (BLRs) of QSOs with broad emissionlines (BALs) are expanding, i.e., the clouds are undergoing radial outflow, as illustrated simply in Fig. 1. At least the following observational facts can be explained:

1. 1. Optical observations show that some moderate and high-redshift QSOs have BALs. The number of these QSOs is about 3–10% of all QSOs (Foltz et al. 1990). One can see in Fig. 2 that very low-redshift QSOs have no BALs.

2. 2. Almost all BAL QSOs exhibit zabs < zem.

3. 3. High-resolution observations reveal that the widths of the broad absorption lines are narrower than that of the corresponding emission line for all BAL QSOs, i.e. Δλ ab < Δλ em.
