The Variables and Slow Transients Survey (VAST) on the Australian Square Kilometre Array Pathfinder (ASKAP) is designed to detect highly variable and transient radio sources on timescales from 5 s to $\sim\!5$ yr. In this paper, we present the survey description, observation strategy and initial results from the VAST Phase I Pilot Survey. This pilot survey consists of $\sim\!162$ h of observations conducted at a central frequency of 888 MHz between 2019 August and 2020 August, with a typical rms sensitivity of $0.24\ \mathrm{mJy\ beam}^{-1}$ and angular resolution of $12-20$ arcseconds. There are 113 fields, each of which was observed for 12 min integration time, with between 5 and 13 repeats, with cadences between 1 day and 8 months. The total area of the pilot survey footprint is 5 131 square degrees, covering six distinct regions of the sky. An initial search of two of these regions, totalling 1 646 square degrees, revealed 28 highly variable and/or transient sources. Seven of these are known pulsars, including the millisecond pulsar J2039–5617. Another seven are stars, four of which have no previously reported radio detection (SCR J0533–4257, LEHPM 2-783, UCAC3 89–412162 and 2MASS J22414436–6119311). Of the remaining 14 sources, two are active galactic nuclei, six are associated with galaxies and the other six have no multi-wavelength counterparts and are yet to be identified.

In the Cordillera Darwin, southernmost South America, we used 10Be and 14C dating, dendrochronology, and historical observations to reconstruct the glacial history of the Dalla Vedova valley from deglacial time to the present. After deglacial recession into northeastern Darwin and Dalla Vedova, by ~16 ka, evidence indicates a glacial advance at ~13 ka coeval with the Antarctic Cold Reversal. The next robustly dated glacial expansion occurred at 870 ± 60 calendar yr ago (approximately AD 1150), followed by less-extensive dendrochronologically constrained advances from shortly before AD 1836 to the mid-twentieth century. Our record is consistent with most studies within the Cordillera Darwin that show that the Holocene glacial maximum occurred during the last millennium. This pattern contrasts with the extensive early- and mid-Holocene glacier expansions farther north in Patagonia; furthermore, an advance at 870 ± 60 yr ago may suggest out-of-phase glacial advances occurred within the Cordillera Darwin relative to Patagonia. We speculate that a southward shift of westerlies and associated climate regimes toward the southernmost tip of the continent, about 900–800 yr ago, provides a mechanism by which some glaciers advanced in the Cordillera Darwin during what is generally considered a warm and dry period to the north in Patagonia.

Mount Achernar moraine is a terrestrial sediment archive that preserves a record of ice-sheet dynamics and climate over multiple glacial cycles. Similar records exist in other blue ice moraines elsewhere on the continent, but an understanding of how these moraines form is limited. We propose a model to explain the formation of extensive, coherent blue ice moraine sequences based on the integration of ground-penetrating radar (GPR) data with ice velocity and surface exposure ages. GPR transects (100 and 25 MHz) both perpendicular and parallel to moraine ridges at Mount Achernar reveal an internal structure defined by alternating relatively clean ice and steeply dipping debris bands extending to depth, and where visible, to the underlying bedrock surface. Sediment is carried to the surface from depth along these debris bands, and sublimates out of the ice, accumulating over time (>300 ka). The internal pattern of dipping reflectors, combined with increasing surface exposure ages, suggest sequential exposure of the sediment where ice and debris accretes laterally to form the moraine. Subsurface structure varies across the moraine and can be linked to changes in basal entrainment conditions. We speculate that higher concentrations of debris may have been entrained in the ice during colder glacial periods or entrained more proximal to the moraine sequence.

OBJECTIVES/SPECIFIC AIMS: Clostridium difficile infection (CDI) is the most common cause of antibiotic-associated diarrhea and an increasingly common infection in children in both hospital and community settings. Between 20% and 30% of pediatric patients will have a recurrence of symptoms in the days to weeks following an initial infection. Multiple recurrences have been successfully treated with fecal microbiota transplantation (FMT), though the body of evidence in pediatric patients is limited primarily to case reports and case series. The goal of our study was to better understand practices, success, and safety of FMT in children as well as identify risk factors associated with a failed FMT in our pediatric patients. METHODS/STUDY POPULATION: This multicenter retrospective analysis included 373 patients who underwent FMT for CDI between January 1, 2006 and January 1, 2017 from 18 pediatric centers. Demographics, baseline characteristics, FMT practices, C. difficile outcomes, and post-FMT complications were collected through chart abstraction. Successful FMT was defined as no recurrence of CDI within 60 days after FMT. Of the 373 patients in the cohort, 342 had known outcome data at two months post-FMT and were included in the primary analysis evaluating risk factors for recurrence post-FMT. An additional six patients who underwent FMT for refractory CDI were excluded from the primary analysis. Unadjusted analysis was performed using Wilcoxon rank-sum test, Pearson χ2 test, or Fisher exact test where appropriate. Stepwise logistic regression was utilized to determine independent predictors of success. RESULTS/ANTICIPATED RESULTS: The median age of included patients was 10 years (IQR; 3.0, 15.0) and 50% of patients were female. The majority of the cohort was White (89.0%). Comorbidities included 120 patients with inflammatory bowel disease (IBD) and 14 patients who had undergone a solid organ or stem cell transplantation. Of the 336 patients with known outcomes at two months, 272 (81%) had a successful outcome. In the 64 (19%) patients that did have a recurrence, 35 underwent repeat FMT which was successful in 20 of the 35 (57%). The overall success rate of FMT in preventing further episodes of CDI in the cohort with known outcome data was 87%. Unadjusted predictors of a primary FMT response are summarized. Based on stepwise logistic regression modeling, the use of fresh stool, FMT delivery via colonoscopy, the lack of a feeding tube, and a lower number of CDI episodes before undergoing FMT were independently associated with a successful outcome. There were 20 adverse events in the cohort assessed to be related to FMT, 6 of which were felt to be severe. There were no deaths assessed to be related to FMT in the cohort. DISCUSSION/SIGNIFICANCE OF IMPACT: The overall success of FMT in pediatric patients with recurrent or severe CDI is 81% after a single FMT. Children without a feeding tube, who receive an early FMT, FMT with fresh stool, or FMT via colonoscopy are less likely to have a recurrence of CDI in the 2 months following FMT. This is the first large study of FMT for CDI in a pediatric cohort. These findings, if confirmed by additional prospective studies, will support alterations in the practice of FMT in children.

Stable isotopes of water (δ18O and δ2H) were measured in the debris-laden ice underlying an Antarctic blue ice moraine, and in adjoining Law Glacier in the central Transantarctic Mountains. Air bubble content and morphology were assessed in shallow ice core samples. Stable isotope measurements plot either on the meteoric waterline or are enriched from it. The data cluster in two groups: the ice underlying the moraine has a δ2H:δ18O slope of 5.35 ± 0.92; ice from adjoining portions of Law Glacier has a slope of 6.69 ± 1.39. This enrichment pattern suggests the moraine's underlying blue ice entrained sediment through refreezing processes acting in an open system. Glaciological conditions favorable to warm-based sediment entrainment occur 30–50 km upstream. Basal melting and refreezing are further evidenced by abundant vapor figures formed from internal melting of the ice crystals. Both the moraine ice and Law Glacier are sufficiently depleted of heavy isotopes that their ice cannot be sourced locally, but instead must be derived from far-field interior regions of the higher polar plateau. Modeled ice flow speeds suggest the ice must be at least 80 ka old, with Law Glacier's ice possibly dating to OIS 5 and moraine ice older still.

A large ice sheet still covered almost all of Maine and eastern New England until ca. 15 cal ka BP, reaching south of 45 °S, despite rising summer insolation intensity and major ice recession elsewhere outside the North Atlantic region. Furthermore, the well-studied moraine belt along eastern coastal Maine, including the prominent Pineo Ridge delta/moraine complex and Pond Ridge moraine, indicates repeated readvances and stillstands between ca. 16 and 15 cal ka BP. This moraine belt reflects a considerable ice sheet response over eastern North America during this time period, coeval with the latter half of the European Oldest Dryas period. Moraine deposition was concurrent with reduction or elimination of North Atlantic meridional overturning, starting with the earlier onset of peak IRD and Heinrich Event 1 (HE-1). The existing 14C chronology suggests that the coastal moraine belt and the persistence of the ice sheet until ∼ 15 cal ka BP was a response to the severe cooling of the North Atlantic region after ∼ 17 cal ka BP.

At Lago Buenos Aires, Argentina, 10Be, 26Al, and 40Ar/39Ar ages range from 190,000 to 109,000 yr for two moraines deposited prior to the last glaciation, 23,000�16,000 yr ago. Two approaches, maximum boulder ages assuming no erosion, and the average age of all boulders and an erosion rate of 1.4 mm/103 yr, both yield a common estimate age of 150,000�140,000 yr for the two moraines. The erosion rate estimate derives from 10Be and 26Al concentrations in old erratics, deposited on moraines that are >760,000 yr old on the basis of interbedded 40Ar/39Ar dated lavas. The new cosmogenic ages indicate that a major glaciation during marine oxygen isotope stage 6 occurred in the mid-latitude Andes. The next five youngest moraines correspond to stage 2. There is no preserved record of a glacial advance during stage 4. The distribution of dated boulders and their ages suggest that at least one major glaciation occurred between 760,000 and >200,000 yr ago. The mid-latitude Patagonian glacial record, which is well preserved because of low erosion rates, indicates that during the last two glacial cycles major glaciations in the southern Andes have been in phase with growth and decay of Northern Hemisphere ice sheets, especially at the 100,000 yr periodicity. Thus, glacial maxima are global in nature and are ultimately paced by small changes in Northern Hemisphere insolation.

Prominent moraines deposited by the Laurentide Ice Sheet in northern New England document readvances, or stillstands, of the ice margin during overall deglaciation. However, until now, the paucity of direct chronologies over much of the region has precluded meaningful assessment of the mechanisms that drove these events, or of the complex relationships between ice-sheet dynamics and climate. As a step towards addressing this problem, we present a cosmogenic 10Be surface-exposure chronology from the Androscoggin moraine complex, located in the White Mountains of western Maine and northern New Hampshire, as well as four recalculated ages from the nearby Littleton–Bethlehem moraine. Seven internally consistent 10Be ages from the Androscoggin terminal moraines indicate that advance culminated ~ 13.2 ± 0.8 ka, in close agreement with the mean age of the neighboring Littleton–Bethlehem complex. Together, these two datasets indicate stabilization or advance of the ice-sheet margin in northern New England, at ~ 14–13 ka, during the Allerød/Greenland Interstadial I.

Sediments from Qipisarqo Lake provide a continuous Holocene paleoenvironmental record from southern Greenland. Following deglaciation and glacio-isostatic emergence of the basin from the sea ∼9100 cal yr B.P., proxies of lake paleoproductivity, including biogenic silica and organic matter, increased markedly until 6000 cal yr B.P. and thereafter remained stable over the ensuing warm three millennia. Subsequent decreases in these proxies, most dramatically between 3000 and 2000 cal yr B.P., show the lake's responses to initial Neoglacial cooling. Intervals of ameliorated limnological conditions occurred between 1300 and 900 and between 500 and 280 cal yr B.P., briefly interrupting the decreasing trend in productivity that culminated in the Little Ice Age. Increased lake productivity during the latter half of the 20th century, which reflects the limnological response to circum-arctic warming, still has not reached peak Holocene values. The Neoglacial climate of the last 2000 yr includes the most rapid high-amplitude environmental changes of the past nine millennia. The Norse thus migrated around the North Atlantic Ocean region in the most environmentally unstable period since deglaciation. Lacustrine sediment records provide a context with which to consider future environmental changes in the Labrador Sea region. In particular, any future warming will be superposed on a regional climate system that is currently exhibiting highly unstable behavior at submillennial timescales.

The Women in Materials (WIM) program is an on-going collaboration between Simmons College and the Cornell Center for Materials Research (CCMR). Beginning in 2001, during the initial four years of the project, materials-related curricula were developed, a new joint research project was begun, and nearly 1/2 of Simmons College science majors participated in materials-related research during their first two years as undergraduates. We have previously reported the student outcomes as a result of this initial stage of the project, demonstrating a successful partnership between a primarily undergraduate women's college and a federally funded Materials Research Science and Engineering Center. Here, we report the evolution and impact of this project over the last three years, subsequent to the initial seed funding from the National Science Foundation. The Women in Materials project is now a key feature of the undergraduate science program at Simmons College and has developed into an organizing structure for materials-related research at the College. Initially, three faculty members were involved and now eight faculty members from all three laboratory science departments participate (biology, chemistry, and physics). The program now involves research related to optoelectronics, polymer synthesis, biomaterials, and green chemistry, and each semester about 80% of the students who participate in these projects are 1st and 2nd year science majors. This structure has led to enhanced funding within the sciences, shared instrumentation facilities, a new minor in materials science, and a spirit of collaboration among science faculty and departments. It has also spawned a new, innovative curricular initiative, the Undergraduate Laboratory Renaissance, now in its second year of implementation, involving all three laboratory science departments in incorporating actual, on-going research projects into introductory and intermediate science laboratories. Most importantly, the Women in Materials program has embedded materials-related research into our science curriculum and has deepened and broadened the educational experience for our students; the student outcomes speak to the program's success. Approximately 70% of our science majors go on to graduate school within two years of completing their undergraduate degree. Our students also have a high acceptance rate at highly competitive summer research programs, such as Research Experience for Undergraduates (REU) programs funded by the National Science Foundation.

The early Holocene climate of the North Atlantic region was influenced by two boundary conditions that were fundamentally different from the present: the presence of the decaying Laurentide Ice Sheet (LIS) and higher than present summer solar insolation. In order to assess spatial and temporal patterns of Holocene climate evolution across this region, we collated quantitative paleotemperature records at sub-millennial resolution and synthesized their temporal variability using principal components analysis (PCA). The analysis reveals considerable spatial variability, most notably in the time-transgressive expression of the Holocene thermal maximum (HTM). Most of the region, but especially areas peripheral to the Labrador Sea and hence closest to the locus of LIS disintegration, experienced maximum Holocene temperatures that lagged peak summer insolation by 1000–3000 years. Many sites from the northeastern North Atlantic sector, including the Nordic Seas and Scandinavia, either warmed in phase with maximum summer insolation (11,000–9000 years ago) or were less strongly lagged than the Baffin Bay–Labrador Sea region. These spatially complex patterns of Holocene climate development, which are defined by the PCA, resulted from the interplay between final decay of the LIS and solar insolation forcing.

Background. Previous studies of personality characteristics in women with eating disorders primarily have focused on women who are acutely ill. This study compares personality characteristics among women who are ill with eating disorders, recovered from eating disorders, and those without eating or other Axis I disorder pathology.

Method. Female participants were assessed for personality characteristics using the Temperament and Character Inventory (TCI): 122 with anorexia nervosa (AN; 77 ill, 45 recovered), 279 with bulimia nervosa (BN; 194 ill, 85 recovered), 267 with lifetime histories of both anorexia and bulimia nervosa (AN+BN; 194 ill, 73 recovered), 63 with eating disorder not otherwise specified (EDNOS; 31 ill, 32 recovered), and 507 without eating or Axis I disorder pathology.

Results. Women ill with all types of eating disorders exhibited several TCI score differences from control women, particularly in the areas of novelty-seeking, harm avoidance, self-directedness, and cooperativeness. Interestingly, women recovered from eating disorders reported higher levels of harm avoidance and lower self-directedness and cooperativeness scores than did normal control women.

Conclusions. Women with eating disorders in both the ill and recovered state show higher levels of harm avoidance and lower self-directedness and cooperativeness scores than normal control women. Although findings suggest that disturbances may be trait-related and contribute to the disorders' pathogenesis, additional research with more representative community controls, rather than our pre-screened, normal controls, is needed to confirm these impressions.
