Breast cancer is a high-risk disease with a high mortality rate among women. Chemotherapy plays an important role in the treatment of breast cancer. However, chemotherapy eventually results in tumours that are resistant to drugs. In recent years, many studies have revealed that the activation of Wnt/β-catenin signalling is crucial for the emergence and growth of breast tumours as well as the development of drug resistance. Additionally, drugs that target this pathway can reverse drug resistance in breast cancer therapy. Traditional Chinese medicine has the properties of multi-target and tenderness. Therefore, integrating traditional Chinese medicine and modern medicine into chemotherapy provides a new strategy for reversing the drug resistance of breast tumours. This paper mainly reviews the possible mechanism of Wnt/β-catenin in promoting the process of breast tumour drug resistance, and the progress of alkaloids extracted from traditional Chinese medicine in the targeting of this pathway in order to reverse the drug resistance of breast cancer.

Direct numerical simulations are performed to investigate the wake transitions of the flow normal to a circular rotating disk. The diameter-thickness aspect ratio of the disk is $\chi =50$. The Reynolds number of the free stream is defined as $Re_s=U_\infty D/\nu$, with incoming flow velocity $U_\infty$, disk diameter $D$, and kinematic viscosity of the fluid $\nu$. The rotational motion of the disk is described by the Reynolds number of rotation $Re_r=\varOmega Re_s$, with non-dimensional rotation rate $\varOmega =\frac {1}{2}\omega D/U_\infty$, where $\omega$ is the angular rotation speed of the disk. Extensive numerical simulations are performed in the parameter space $50 \leqslant Re_s \leqslant 250$ and $0 \leqslant Re_r \leqslant 250$, in which six flow regimes are identified as follows: the axisymmetric state, the low-speed steady rotation (LSR) state, the high-speed steady rotation (HSR) state, the low-speed unsteady rotation (LUR) state, the rotational vortex shedding state, and the chaotic state. Although plane symmetry exists in the wake when the disk is stationary, a small rotation will immediately destroy its symmetry. However, the vortex shedding frequencies and wake patterns of the stationary disk are inherited by the unsteady rotating cases at low $Re_r$. A flow rotation rate jump is observed at $Re_s\approx 125$. The LUR state is intermediate between the LSR and HSR states. Due to the rotational motion, the wake of the disk enters the steady rotation state earlier at large $Re_r$, and is delayed into the vortex shedding state in the whole range of $Re_r$. In the steady rotation states (LSR and HSR), the steady flow rotation rate is linearly correlated with the disk rotation rate. It is found that the rotation of the disk can restrain the vortex shedding. The chaotic state can be regularized by the medium rotation speed of the disk.

We investigated the ecology and paleoecology of testate amoebae in peatlands of the Colombian páramo to assess the use of testate amoebae as paleoenvironmental indicators. Objectives were to (1) identify environmental controls on testate amoebae, (2) develop transfer functions for paleoenvironmental inference, and (3) examine testate amoebae in a Holocene peat core and compare our findings with other proxy records. Results from 96 modern samples indicate that testate amoebae are sensitive to pH and surface moisture, and cross-validation of transfer functions indicates potential for paleoenvironmental applications. Testate amoebae from the Triunfo Peatland in the Central Cordillera provided a proxy record of pH and water-table depth for the late Holocene, and inferred changes were correlated with peat C/N measurements during most of the record. Comparison with a lake-level reconstruction suggests that at least the major testate amoeba–inferred changes were driven by climate. Our work indicates that testate amoebae are useful paleoenvironmental indicators in high-elevation tropical peatlands.

Multi-proxy records of a subalpine lacustrine sequence in Daping Swamp in the western Nanling Mountains provide evidence for exploring climate variability in the past 15,400 yr. Two dry and cool (15,400–14,500 and 13,000–11,000 cal yr BP) and one humid and warm interval (14,500–13,200 cal yr BP), which we correlate to Heinrich Event 1, the Younger Dryas and the Bølling-Allerød event respectively, are revealed. The early Holocene climate (11,000–8000 cal yr BP) was characterized by less humid and warm conditions, suggesting a weaker Asian summer monsoon (ASM) intensity. Our findings indicate that the Holocene optimum occurred between 8000 and 4500 cal yr BP, and the most intensified ASM appears from 8000 to 7000 cal yr BP. After 4500 cal yr BP, climate shifted to relatively cool and dry conditions. We speculate that five short dry and cool events centered at ~ 11,000, 9000, 8400, 6000, and 3500 cal yr BP were linked to the Holocene ice-rafting events detected in the North Atlantic. Migration of the ITCZ, and the oceanic–atmospheric circulations, particularly SST changes in the tropical Pacific may play a pivotal role in climate variation of the study region.

Observational studies have suggested that the intake of flavonoids is associated with a decreased risk of CVD. However, the results of these studies remain controversial. The aim of the present study was to evaluate the association between dietary flavonoid intake and CVD risk by conducting a systematic review of prospective cohort studies. Electronic reference databases were searched to identify studies that met the pre-stated inclusion criteria. The studies were assessed for eligibility and data were extracted by two authors independently. For each study, relative risks (RR) and 95 % CI were extracted and pooled using either a fixed-effects or a random-effects model. Generalised least-squares trend estimation analysis was used to evaluate dose–response relationships. The inclusion criteria were met by fourteen prospective cohort studies. The intakes of anthocyanidins (RR 0·89, 95 % CI 0·83, 0·96), proanthocyanidins (RR 0·90, 95 % CI 0·82, 0·98), flavones (RR 0·88, 95 % CI 0·82, 0·96), flavanones (RR 0·88, 95 % CI 0·82, 0·96) and flavan-3-ols (RR 0·87, 95 % CI 0·80, 0·95) were inversely associated with the risk of CVD when comparing the highest and lowest categories of intake. A similar association was observed for flavonol intake and CVD risk. Sensitivity and subgroup analyses further supported this association. The summary RR for CVD for every 10 mg/d increment in flavonol intake was 0·95 (95 % CI 0·91, 0·99). The present systematic review suggests that the dietary intakes of six classes of flavonoids, namely flavonols, anthocyanidins, proanthocyanidins, flavones, flavanones and flavan-3-ols, significantly decrease the risk of CVD.

A nanostructured surface layer was fabricated in the surface of 316L stainless steel by a novel fast multiple rotation rolling (FMRR) technique. The microstructure and the tensile properties of the treated sample were investigated in detail. The experimental results indicate that a nanograined (NG) film was successfully obtained in the surface of the sample. Equiaxed nanograins with the average grain size of about 12 nm are achieved in the surface layer. At the sample time, deformation-induced α-martensite is produced during the FMRR treatment. The volume fraction of martensite is about 20%. The yield strength (0.2% offset) of the sample, of which one side is of NG structure and the other is coarse grained (CG), is increased by 51% in comparison with that of the CG sample. Though the plasticity is diminished slightly for the FMRR specimen, the elongation still reaches a high value of about 38% owing to the contribution of the CG structure.

The elements for the Semliki forest virus (SFV) RNA replicon were obtained from the Alphavirus genome. It was designed to overcome the poor efficacy of some current plasmid vectors. Genes coding for viral replicases are preserved while genes coding for structural proteins are replaced by foreign genes in the RNA replicon. High levels of RNA replication and expression of foreign genes in the cytoplasm are regulated by the replicases. To evaluate the effects of the SFV RNA replicon on the improvement of gene expression, a LacZ gene was inserted into pIRES-neo digested by BamHI and dephosphorylated by alkaline phosphatase to construct pIRES-neo-LacZ. The RNA replicon vector pCMV-rep-LacZ and two conventional cytomegalovirus (CMV) promoter-based vectors (pLNCX-LacZ and pIRES-neo-LacZ) were transfected, using Lipofectin, to prepared 293 cells. Growth hormone releasing hormone (GHRH) expression vectors (pCMV-Rep-GHRH, pCDNA3.1(+)-GHRH and pIRES-neo-GHRH) were also tested using the same procedure. Target gene expression was detected with radioimmunoassay (RIA) and reverse transcription-polymerase chain reaction (RT-PCR). Results showed that the expression level of the RNA replicon vector was 2–3 times higher than with normal plasmid vectors. This result will help to improve the efficiency of gene expression in eukaryotic cells.

Spindle movement, including spindle migration during first meiosis and spindle rotation during second meiosis, is essential for asymmetric divisions in mouse oocytes. Previous studies by others and us have shown that microfilaments are required for both spindle migration and rotation. In the present study, we aimed to further investigate the mechanism controlling spindle movement during mouse oocyte meiosis. By employing drug treatment and immunofluorescence microscopy, we showed that dynamic microtubule assembly was involved in both spindle migration and rotation. Furthermore, we found that the calcium/CaM/CaMKII pathway was important for regulating spindle rotation.

Early studies have shown that some mouse cumulus–oocyte complexes (COCs) stored at room temperature for 24 h still retained full developmental potential. In this study, we stored denuded mouse oocytes (DOs) at room temperature (25 °C) for 24 h and activated these oocytes with 10 mM SrCl2 or fertilized the oocytes by IVF. We found that nearly half of the DOs stored at room temperature for 1 day can be fertilized normally by IVF and that two foster mothers gave birth to seven pups. Embryos from stored oocytes were cultured in CZB medium with or without 1 μg/ml 17β-estradiol (E2). The numbers of embryo that developed to morula/blastocyst stage after parthenogenetic activation and IVF were significantly increased when E2 was added to the culture (p < 0.05). These results suggest that E2 might improve mouse embryo development in vitro. The birth of seven agouti pups and their healthy growth indicated that the storage of DOs at room temperature for 1 day may be a practical procedure for mammalian reproduction.

Following our previous work on the converse piezoelectric constant- in epitaxial ferroelectric films for MEMS actuator applications, the orientation dependence of the direct piezoelectric constants , and are generally formulated, which can help to predict and optimize the performance of piezoelectric MEMS sensor devices based on ferroelectric thin films. Numerical results are obtained and discussed for Pb(ZrxTi1-x)O3 thin films grow on Si substrates with various compositions and structures.
