Maternal nutrition during gestation provides the nutrients for fetal growth and development. Herein, we investigated the transcriptomic responses of fetal brain and muscle tissues at day 83 of gestation (the end of the first trimester) in female fetuses from heifers assigned in a 2 × 2 factorial design to one of four treatments. The treatments consisted of the main effects of vitamin and mineral supplementation (VTM vs. NoVTM; 113 g·heifer−1·d−1 of mineral premix) and two maternal body weight gain rates (low [LG; 0.28 kg d−1] vs. moderate [MG; 0.79 kg d−1]). RNA sequencing of 31 fetuses revealed 399 unique differentially expressed genes (DEGs) in the brain and 1,273 unique DEGs in the muscle across the six contrasts (false discovery rate ≤ 0.1). Overall, brain DEGs were enriched in tissue morphogenesis and Wnt/phosphatidylinositol 3-kinase–protein kinase B signaling pathways, including WNT2B, WNT5A, and WNT7B (upregulated in VTM_LG vs. NoVTM_LG). In muscle, maternal gain and VTM supplementation modulated the expression of myogenic and epigenetic regulators, including PAX7, MSTN, FOXO3, and DNA demethylation enzymes TET1, TET2, and TET3, highlighting the potential epigenetic programming. These findings demonstrate that maternal diet during the first trimester of gestation alters fetal brain and muscle molecular profiles through genes involved in nutrient-sensing and epigenetic pathways. Together, these results highlight the potential for targeted maternal vitamin and mineral supplementation during early gestation to modulate brain and muscle development. Further research is needed to determine the long-term effects of supplementation on offspring behavior, growth, and efficiency in beef production systems.
We argue for the existence of functionally complex constructions whose elements compositionally impose discourse-functional constraints on the use of the whole. In particular, we examine th-clefts (as in That's John who wrote the book), equatives with epistemic would and demonstrative subjects (as in That would be John), and simple equatives with demonstrative subjects (as in That's John). We show that, contra previous approaches, the latter two constructions need not be analyzed as truncated clefts. Rather, the properties that these constructions share with th-clefts can be straightforwardly accounted for as the sum of the constraints on their shared elements—that is, the equative construction, the demonstrative subject, and the presence of a contextually salient open proposition. The convergence of these elemental properties in each of these three constructions results in the possibility of the demonstrative being used to refer to the instantiation of the variable in the open proposition, which in turn predicts a complex of distributional behaviors shared by precisely the constructions that share these properties. Because these distributional behaviors can be straightforwardly explained in terms of this functional compositionality, the motivation for a truncated-cleft analysis disappears. These results support the view that not all functional properties must be learned on a construction-by-construction basis; instead, the discourse functions of an utterance are built up compositionally from those of its parts.
In Behemoth, Thomas Hobbes identified the ‘great purse of the City of London’ as crucial for the Parliamentary cause during the English Civil Wars and Revolution. Hobbes’ analysis has influenced subsequent generations of historians of the 1640s. This essay demonstrates that wealthy Londoners did not draw only from their own pockets. Instead, many members of the metropolitan financial elite, in their role as livery company officers, chose to allocate company funds in ways that deflected financial risk away from themselves and onto some of the most vulnerable members of their society. London merchants often tied up their wealth in illiquid assets, such as land or working capital. This liquidity crisis affected livery companies as well as their members, forcing the companies to borrow money and to sell assets in order to meet their assessments. The debt the companies assumed in the early 1640s impaired their ability to support philanthropy and the social customs that promoted philanthropic activity for well over a decade. As a result, Hobbes and his followers have overlooked the fact that the ‘great purse of the City’ depended heavily on the ability of civic institutions to deprive the many small purses of poor Londoners.
Strongyloides stercoralis is an intestinal nematode capable of infecting humans and dogs. Occurrence in dogs from temperate, traditionally non-endemic regions remains poorly characterized, often due to the absence of accessible veterinary diagnostic tests. Recent reports of infections from temperate metropolitan areas have raised concerns about the extent of S. stercoralis prevalence in dogs in these regions. This study investigated the presence of S. stercoralis DNA in canine faecal samples from Sydney, New South Wales, Australia, using a real-time quantitative PCR (qPCR) targeting the 18S rRNA gene, with a limit of detection of 2 DNA copies. Archived faecal DNA (n = 448) collected between 2014 and 2024 from 2 university veterinary hospitals were screened. Of all samples, 1 (0.02%) was positive for S. stercoralis DNA, corresponding to approximately 6.8 × 103 18S rDNA copies, equivalent to 3.2 of S. ratti third-stage larvae, per 250 mg of dog faeces. Partial cox1 and 18S rDNA loci (HVR-I and HVR-IV) deep amplicon sequencing confirmed that S. stercoralis circulates between dogs and humans. The positive sample originated from a Border Collie puppy presenting with gastrointestinal signs. Although detection was rare, this confirms the parasite’s presence in companion dogs within a temperate urban environment. The results highlight the diagnostic utility of 18S rDNA-based qPCR for retrospective surveillance and support the inclusion of S. stercoralis in molecular diagnostic panels for dogs with gastrointestinal disease. Expanded, targeted molecular and coproscopic surveillance is warranted to clarify the prevalence, distribution and zoonotic potential of S. stercoralis in dogs across Australia’s non-endemic regions.
Callous-unemotional (CU) traits, characterized by lack of empathy, guilt, and deficient affect, are linked to facial emotion recognition (FER) deficits in children. While anxiety is also associated with FER anomalies, these relationships are often examined in isolation despite co-occurrence. This study aims to concurrently investigate unique contributions of CU traits and anxiety on children’s FER patterns. We recruited 107 children aged 6 to 11 from community settings, assessing CU traits through caregiver reports and anxiety via caregiver and child reports. FER performance was evaluated using a computer-based task. Results indicate that CU traits negatively impact overall FER accuracy, particularly when controlling for parent-reported anxiety. CU traits were inversely related to total FER accuracy for children self-reporting high anxiety levels. These findings enhance our understanding of how CU traits and anxiety interact to influence FER deficits, suggesting that interventions targeting CU traits should consider anxiety symptoms as a critical factor in emotional processing challenges among children.
Biological control programmes often involve releasing natural enemies to control pests that are mobile, patchily distributed, or both. Determining the optimal dispersion of release points for biological control agents is challenging, particularly when knowledge of prey locations is imprecise or information on agent dispersal capacity is lacking. Chrysoperla rufilabris Burmeister (Neuroptera: Chrysopidae) is a commercially available green lacewing species that is often released in the larval stage as an augmentative biological control in various agroecosystems. We characterised the dispersal capacity of C. rufilabris using approximately 5-minute laboratory assays and then used our laboratory data to simulate dispersal over increasing windows of time. Larvae crawled a median of 0.5 m per 5 minutes, corresponding to median (95% confidence interval) simulated dispersal distances (metres) of 2 (0.1–10.3), 12 (0.4–59.6), and 25 (2.2–107.1) within 1, 6, and 12 hours, respectively. Our simulations suggest that larval C. rufilabris should be physiologically capable of dispersing several metres in search of prey following their release, although field dispersal capacity and behaviour could differ markedly from our laboratory observations. These findings will help to inform the dispersion of release points and underscore the ability of larval lacewings to move long distances after shipment.
Hand, foot, and mouth disease (HFMD) shows spatiotemporal heterogeneity in China. A spatiotemporal filtering model was constructed and applied to HFMD data to explore the underlying spatiotemporal structure of the disease and determine the impact of different spatiotemporal weight matrices on the results. HFMD cases and covariate data in East China were collected between 2009 and 2015. The different spatiotemporal weight matrices formed by Rook, K-nearest neighbour (KNN; K = 1), distance, and second-order spatial weight matrices (SO-SWM) with first-order temporal weight matrices in contemporaneous and lagged forms were decomposed, and spatiotemporal filtering model was constructed by selecting eigenvectors according to MC and the AIC. We used MI, standard deviation of the regression coefficients, and five indices (AIC, BIC, DIC, R2, and MSE) to compare the spatiotemporal filtering model with a Bayesian spatiotemporal model. The eigenvectors effectively removed spatial correlation in the model residuals (Moran’s I < 0.2, p > 0.05). The Bayesian spatiotemporal model’s Rook weight matrix outperformed others. The spatiotemporal filtering model with SO-SWM was superior, as shown by lower AIC (92,029.60), BIC (92,681.20), and MSE (418,022.7) values, and higher R2 (0.56) value. All spatiotemporal contemporaneous structures outperformed the lagged structures. Additionally, eigenvector maps from the Rook and SO-SWM closely resembled incidence patterns of HFMD.
While many children in Africa face notable psychological problems, the majority do not receive needed mental health services. The My FRIENDS Youth Program, a universal cognitive-behavioral intervention for anxiety prevention and resilience enhancement, has demonstrated effectiveness across cultures in children and adolescents. This study explores whether the program’s effectiveness extends to Zambian children. Participants were 75 children and adolescents (53% female, ages 10–15) attending low-income schools in Zambia. Four schools were randomly assigned to an intervention (n = 44) or waitlist control (n = 31). The intervention consisted of 10 weekly sessions plus two booster sessions administered in group format. Assessments were conducted at pre-intervention, immediately post-intervention and 3-month follow-up. Data were analyzed using longitudinal multilevel modeling and controlled for child and parent sociodemographic characteristics. Intervention participation did not lead to reductions in anxiety, depression or parent-child relationship conflict but was associated with reductions in parent-reported internalizing and externalizing symptoms, attention problems and increases in positive parent-child relationships. However, both the intervention and control groups exhibited lower anxiety symptoms from Post-Intervention to 3-Month Follow-Up, suggesting potentially delayed effects. Future research may need to adapt this intervention to meet the needs of children in Zambia.
We aimed to estimate the overall apparent prevalence, true prevalence, and the spatial, temporal, and test-specific burden of bovine tuberculosis in Bangladesh. PubMed, Web of Science, Scopus, Google Scholar, and BanglaJOL were searched for bovine tuberculosis publications in Bangladesh from 1 January 1970 to 23 June 2023. Of 142 articles screened, systematic review and meta-analysis were performed on 22 (15.5%) articles. The apparent estimated bovine tuberculosis prevalence was 7%. The apparent Bayesian pooled mean bovine tuberculosis prevalences based on caudal fold test and single intradermal comparative tuberculin test were 7.83% and 9.89%, respectively, and the true pooled mean prevalences were 10.39% and 10.48%, respectively. Targeted interventions are recommended for districts with higher prevalence to effectively reduce the bovine tuberculosis burden in those areas. Current diagnostic practices employed in Bangladesh may not accurately reflect the bovine tuberculosis burden. Our findings highlight the need for better diagnostic tools and supplemental testing methods to ensure accurate diagnosis and surveillance. Efforts should prioritize obtaining ‘true’ prevalence estimates corrected for misclassification bias, rather than relying solely on apparent prevalence. Underestimating the bovine tuberculosis burden could result in inadequate resource allocation and hinder the implementation of effective control measures.
This study delves into Industry 5.0's Human Centric Manufacturing and Systems (HCM and HCS), emphasizing worker welfare and sustainability. Industry 5.0 advocates a human-centric approach, built upon three foundational pillars safety, inclusivity, and empowerment. The paper highlights the successful integration of Design and Systems Thinking in HCM and proposes a workshop at MADE COMPETENCE CENTRE proving the effectiveness in raising awareness and promoting Human-Centric principles throughout the system life cycle and in achieving Human-Centric Systems (HCS).
Marine litter poses a complex challenge in Indonesia, necessitating a well-informed and coordinated strategy for effective mitigation. This study investigates the seasonality of plastic concentrations around Sulawesi Island in central Indonesia during monsoon-driven wet and dry seasons. By using open data and methodologies including the HYCOM and Parcels models, we simulated the dispersal of plastic waste over 3 months during both the southwest and northeast monsoons. Our research extended beyond data analysis, as we actively engaged with local communities, researchers and policymakers through a range of outreach initiatives, including the development of a web application to visualize model results. Our findings underscore the substantial influence of monsoon-driven currents on surface plastic concentrations, highlighting the seasonal variation in the risk to different regional seas. This study adds to the evidence provided by coarser resolution regional ocean modelling studies, emphasizing that seasonality is a key driver of plastic pollution within the Indonesian archipelago. Inclusive international collaboration and a community-oriented approach were integral to our project, and we recommend that future initiatives similarly engage researchers, local communities and decision-makers in marine litter modelling results. This study aims to support the application of model results in solutions to the marine litter problem.
Schizophrenia (SCZ) is a neuropsychiatric disorder with strong genetic heritability and predicted genetic heterogeneity, but limited knowledge regarding the underlying genetic risk variants. Classification into phenotype-driven subgroups or endophenotypes is expected to facilitate genetic analysis. Here, we report a teen boy with chronic psychosis and cerebellar hypoplasia (CBLH) and analyze data on 16 reported individuals with SCZ or chronic psychosis not otherwise specified associated with cerebellar hypoplasia to look for shared features.
Participants and Methods:
We evaluated an 18-year-old boy with neurodevelopmental deficits from early childhood and onset of hallucinations and other features of SCZ at 10 years who had mild vermis-predominant CBLH on brain imaging. This prompted us to review prior reports of chronic psychosis or SCZ with cerebellar malformations using paired search terms including (1) cerebellar hypoplasia, Dandy-Walker malformation, Dandy-Walker variant, or mega-cisterna magna with (2) psychosis or SCZ. We found reports of 16 affected individuals from 13 reports. We reviewed clinical features focusing on demographic information, prenatal-perinatal history and neuropsychiatric and neurodevelopmental phenotypes, and independently reviewed brain imaging features.
Results:
All 17 individuals had classic psychiatric features of SCZ or chronic psychosis as well as shared neurodevelopmental features not previously highlighted including a downward shift in IQ of about 20 points, memory impairment, speech-language deficits, attention deficits and sleep disturbances. The brain imaging findings among these individuals consistently showed posterior vermis predominant CBLH with variable cerebellar hemisphere hypoplasia and enlarged posterior fossa (a.k.a. mega-cisterna magna). None had features of classic DWM.
Conclusions:
In 17 individuals with chronic psychosis or SCZ and cerebellar malformation, we found a high frequency of neurodevelopmental disorders, a consistent brain malformation consisting of posterior vermis-predominant (and usually symmetric) CBLH, and no evidence of prenatal risk factors. The consistent phenotype and lack of prenatal risk factors for CBLH leads us to hypothesize that psychosis or schizophrenia associated with vermis predominant CBLH comprises a homogeneous subgroup of individuals with chronic psychosis/schizophrenia that is likely to have an underlying genetic basis. No comprehensive targeted gene panel for CBLH has yet been defined, leading us to recommend trio-based exome sequencing for individuals who present with this combination of features.
Assess turnaround time (TAT) and cost-benefit of on-site C. auris screening and its impact on length of stay (LOS) and costs compared to reference laboratories.
Design:
Before-and-after retrospective cohort study.
Setting:
Large-tertiary medical center.
Methods:
We validated an on-site polymerase chain reaction-based testing platform for C. auris and retrospectively reviewed hospitalized adults who screened negative before and after platform implementation. We constructed multivariable models to assess the association of screening negative with hospital LOS/cost in the pre and postimplementation periods. We adjusted for confounders such as demographics and indwelling device use, and compared TATs for all samples tested.
Results:
The sensitivity and specificity of the testing platform were 100% and 98.11%, respectively, compared to send-out testing. The clinical cohort included 287 adults in the pre and 1,266 postimplementation period. The TAT was reduced by more than 2 days (3 (interquartile range (IQR): 2.0, 7.0) vs 0.42 (IQR: 0.24, 0.81), p < 0.001). Median LOS was significantly lower in the postimplementation period; however, this was no longer evident after adjustment. In relation to total cost, the time period had an effect of $6,965 (95% CI: −$481, $14,412); p = 0.067) on reducing the cost. The median adjusted total cost per patient was $7,045 (IQR: $3,805, $13,924) less in the post vs the preimplementation period.
Conclusions:
Our assessment did not find a statistically significant change in LOS, nevertheless, on-site testing was not cost-prohibitive for the institution. The value of on-site testing may be supported if an institutional C. auris reduction strategy emphasizes faster TATs.
Three polyphagous pest Liriomyza spp. (Diptera: Agromyzidae) have recently invaded Australia and are damaging horticultural crops. Parasitic wasps are recognized as effective natural enemies of leafmining species globally and are expected to become important biocontrol agents in Australia. However, the hymenopteran parasitoid complex of agromyzids in Australia is poorly known and its use hindered due to taxonomic challenges when based on morphological characters. Here, we identified 14 parasitoid species of leafminers based on molecular and morphological data. We linked DNA barcodes (5′ end cytochrome c oxidase subunit I (COI) sequences) to five adventive eulophid wasp species (Chrysocharis pubicornis (Zetterstedt), Diglyphus isaea (Walker), Hemiptarsenus varicornis (Girault), Neochrysocharis formosa (Westwood), and Neochrysocharis okazakii Kamijo) and two braconid species (Dacnusa areolaris (Nees) and Opius cinerariae Fischer). We also provide the first DNA barcodes (5′ end COI sequences) with linked morphological characters for seven wasp species, with three identified to species level (Closterocerus mirabilis Edwards & La Salle, Trigonogastrella parasitica (Girault), and Zagrammosoma latilineatum Ubaidillah) and four identified to genus (Aprostocetus sp., Asecodes sp., Opius sp. 1, and Opius sp. 2). Phylogenetic analyses suggest C. pubicornis, D. isaea, H. varicornis, and O. cinerariae are likely cryptic species complexes. Neochrysocharis formosa and Aprostocetus sp. specimens were infected with Rickettsia. Five other species (Cl. mirabilis, D. isaea, H. varicornis, Opius sp. 1, and Opius sp. 2) were infected with Wolbachia, while two endosymbionts (Rickettsia and Wolbachia) co-infected N. okazakii. These findings provide background information about the parasitoid fauna expected to help control the leafminers.
Healthcare personnel (HCP) may encounter unfamiliar personal protective equipment (PPE) during clinical duties, yet we know little about their doffing strategies in such situations.
Objective:
To better understand how HCP navigate encounters with unfamiliar PPE and the factors that influence their doffing strategies.
Setting:
The study was conducted at 2 Midwestern academic hospitals.
Participants:
The study included 70 HCP: 24 physicians and resident physicians, 31 nurses, 5 medical or nursing students, and 10 other staff. Among them, 20 had special isolation unit training.
Methods:
Participants completed 1 of 4 doffing simulation scenarios involving 3 mask designs, 2 gown designs, 2 glove designs, and a full PPE ensemble. Doffing simulations were video-recorded and reviewed with participants during think-aloud interviews. Interviews were audio-recorded and analyzed using thematic analysis.
Results:
Participants identified familiarity with PPE items and designs as an important factor in doffing. When encountering unfamiliar PPE, participants cited aspects of their routine practices such as designs typically used, donning and doffing frequency, and design cues, and their training as impacting their doffing strategies. Furthermore, they identified nonintuitive design and lack of training as barriers to doffing unfamiliar PPE appropriately.
Conclusion:
PPE designs may not be interchangeable, and their use may not be intuitive. HCP drew on routine practices, experiences with familiar PPE, and training to adapt doffing strategies for unfamiliar PPE. In doing so, HCP sometimes deviated from best practices meant to prevent self-contamination. Hospital policies and procedures should include ongoing and/or just-in-time training to ensure HCP are equipped to doff different PPE designs encountered during clinical care.
This article provides a historiographical survey of significant African American historians researching, writing, and interpreting Black people's education history. At the heart of this article are the following questions: Who were the African American historians of education who produced this work? What has been the significant scholarship of African American education historians from the late nineteenth century to the present? Although much scholarship has been published on African American education, its history remains underrepresented in the study of educational history. Nonetheless, this historiography is burgeoning thanks to Black educational historians’ scholarship.
Marine megafauna occurrence was recorded in the deep-sea region bordering the abyssal plain ~400 km north-west of Luanda, Angola. The survey took place during an Environmental Baseline Study (EBS), prior to drilling exploration activities, with the goal of characterizing the habitat and biodiversity of the region. Offshore shipboard surveys were conducted during September 2018 in water depths ranging from 2350–3850 m. We recorded daytime sightings of marine mammals and sea turtles and at night made audio recordings using passive acoustic monitoring (PAM) methods focused on capturing the sounds of vocalizing marine mammals. A variety of species were visually detected, including the humpback whale (Megaptera novaeangliae), sperm whale (Physeter macrocephalus), common dolphin (Delphinus spp.), striped dolphin (Stenella coeruleoalba), Atlantic spotted dolphin (S. frontalis), and olive ridley turtle (Lepidochelys olivacea). Acoustic click bouts similar to those made by several odontocete species, possibly including beaked whales, were recorded within the 25–48 kHz range. The humpback whale was the most frequently sighted species, accounting for 56% of mammal sightings, indicating a potential far offshore migratory habitat in this region. Most notably, right whales (probable Eubalaena australis) were visually observed. This is the first confirmed record of right whales in Angolan waters since the early 1900s. As development expands in this offshore region, these data can usefully inform future monitoring and mitigation strategies focused on minimizing impacts to wildlife.
Reasoning biases, specifically jumping to conclusions and belief inflexibility, may play a causal role in persistent paranoia. SlowMo, a new digitally supported blended cognitive-behavioural therapy, targets these biases. Adopting the terms ‘fast’ and ‘slow thinking’ as a heuristic to support therapy, SlowMo encourages people to notice a tendency to fast thinking, and to slow down for a moment to reduce paranoia. SlowMo therapy is the first digital blended therapy for paranoia, employing face to face therapy sessions with interactive digital content, and using mobile technology to promote generalisation to daily life. We report a randomised controlled trial with N=362 participants with distressing and persistent (3+months) paranoia, comparing 8 sessions of SlowMo plus Treatment as Usual (TAU) with TAU alone. We examined SlowMo’s effectiveness in reducing paranoia and improving reasoning biases; its mechanisms of action; usability; and acceptability (Garety et al., 2021). Outcomes: SlowMo was beneficial for paranoia: 10 /11 paranoia measures at 12 weeks and 8/11 at 24 weeks demonstrated significant effects, and sustained moderate effects were observed on all observer-rated measures of persecutory delusions. Improvements in self-esteem, worry, wellbeing and quality of life were also reported. Mediation: Consistent with the theory-driven design and treatment rationale, improvements in slower thinking were found to mediate change in paranoia at 12- and 24-week follow-ups. However contrary to hypothesis, reduced fast thinking did not mediate change in paranoia, whereas worry did. These findings highlight the potential therapeutic mechanisms of action of SlowMo which which are discussed further. Garety P, Ward T, Emsley R, et al.
