Background: Currently there are no disease modifying treatment for Synucleinopathies including Parkinson’s disease Dementia (PDD). Carrying a mutation in the GBA gene (beta-glucocerebrosidase/ GCAse) is a leading risk factor for synucleinopathies. Raising activity GCAse lowers α-synuclein levels in cells and animal models. Ambroxol is a pharmacological chaperone for GCAse and can raise GCAse levels. Our goal is to test Ambroxol as a disease-modifying treatment in PDD. Methods: We randomized fifty-five individuals with PDD to Ambroxol 1050mg/day, 525mg/day, or placebo for 52 weeks. Primary outcome measures included safety, Alzheimer’s disease Assessment Scale-cognitive (ADAS-Cog) subscale and the Clinician’s Global Impression of Change (CGIC). Secondary outcomes included pharmacokinetics, cognitive and motor outcomes and and plasma and CSF biomarkers. Results: Ambroxol was well tolerated. There were 7 serious adverse events (SAEs) none deemed related to Ambroxol. GCase activity was increased in white blood cells by ~1.5 fold. There were no differences between groups on primary outcome measures. Patients receiving high dose Ambroxol appeared better on the Neuropsychiatric Inventory. GBA carriers appeared to improve on some cognitive tests. pTau 181 was reduced in CSF. Conclusions: Ambroxol was safe and well-tolerated in PDD. Plastics pollute all environmental compartments because of human activities and mismanagement. Public perceptions and knowledge about plastic pollution differ among individuals and across different jurisdictions. Targeted survey-based research tools can help measure consumer awareness about the impacts of mismanaged plastics and help identify trends and solutions to reduce plastic use and plastic pollution. This review primarily focused on survey-based research from presenters at the scientific track session TS-2.15 Plastic Pulse of the Public at the 7th International Marine Debris Conference (www.7imdc.org) and supplemented by contemporary literature. Survey-based research helps provide new insights about public opinions related to the pervasiveness of plastic pollution. This review includes results about consumer use and perceptions of plastic pollution impacts from diverse studies from nine countries including Ghana, Kenya, Bangladesh, Pakistan, United States, Canada, Norway, Germany, and United Kingdom. Overwhelmingly, public perceptions and consumer awareness of the negative impacts of plastic pollution were extremely high, regardless of geographic location. Awareness about the environmental impacts of plastic waste and plastic pollution was highest within younger, white, female, and well-educated demographic groups. However, differences were observed in public attitudes toward willingness to pay for sustainable alternatives, end-of-life plastic uses, unintended consequences, recycling, and mismanagement.

Wild badgers (Meles meles) in Wytham woods, Oxfordshire, are routinely trapped, transported to a central field laboratory, studied and released as part of an on-going population study. These procedures have been carefully developed to minimise impact on the badgers' welfare; however they are potentially stressful, and, as part of our on-going welfare refinements, and our exploration to develop methods for quantifying stress in wild mammals, we studied the effects of transport stress on neutrophil activation in wild trapped badgers. Blood samples were obtained from 28 badgers. We compared three transport regimes: transported (n = 9), transported and rested for at least 30 mins (n = 11), and not transported (n = 8). Total and differential white cell counts were carried out and neutrophil activation was measured by the nitroblue tetrazolium test. Our goal was primarily to validate neutrophil activity as an indicator of stress, on the basis that the transport treatment was expected to be more stressful than the non-transport treatment. There were significant increases in % activated circulating neutrophils in response to transport. This study supports the proposition that stress affects circulating neutrophil numbers and the state of their activation, as determined by the nitroblue tetrazolium reduction assay, and therefore adds weight to the idea that neutrophil activation is a potential measure of stress in wild animals.

We report on the efficacy of body weight change as a measure of trapping and handling stress in two species of wild small mammal: bank voles (Clethrionomys glareolus) and wood mice (Apodemus sylvaticus). We tested two hypotheses: (1) that weight change after capture and handling is related to the intensity of the trapping and handling regime, and (2) that weight change after an intensive handling regime is related to an individual's current pattern of energy expenditure. Trapped wood mice that were subjected to intensive handling (intensive stressor) lost more weight than did animals that were handled minimally (less intensive stressor), but this was not the case for bank voles. Patterns and factors related to body weight change in response to intensive handling also differed between the two species: heavier and non-breeding bank voles were more likely to lose weight, but this was not true for wood mice, and none of the factors we measured was found to affect weight loss in this species. Our results were broadly consistent with the predictions of the biological cost hypothesis. We discuss the limitations and benefits of weight loss as a measure of stress.
