In this paper, we investigate a sink-driven three-layer flow in a radial Hele-Shaw cell. The three fluids are of different viscosities, with one fluid occupying an annulus-like domain, forming two interfaces with the other two fluids. Using a boundary integral method and a semi-implicit time stepping scheme, we alleviate the numerical stiffness in updating the interfaces and achieve spectral accuracy in space. The interaction between the two interfaces introduces novel dynamics leading to rich pattern formation phenomena, manifested by two typical events: either one of the two interfaces reaches the sink faster than the other (forming cusp-like morphology), or they come very close to each other (suggesting a possibility of interface merging). In particular, the inner interface can be wrapped by the other to have both scenarios. We find that multiple parameters contribute to the dynamics, including the width of the annular region, the location of the sink, and the mobilities of the fluids.

Background: We evaluated vorasidenib (VOR), a dual inhibitor of mIDH1/2, in patients with mIDH1/2 glioma (Phase 3; NCT04164901). Methods: Patients with residual/recurrent grade 2 mIDH1/2 oligodendroglioma or astrocytoma were enrolled (age ≥12; Karnofsky Performance Score ≥80; measurable non-enhancing disease; surgery as only prior treatment; not in immediate need of chemoradiotherapy). Patients were stratified by 1p19q status and baseline tumor size and randomized 1:1 to VOR 40 mg or placebo (PBO) daily in 28-day cycles. Endpoints included imaging-based progression-free survival (PFS), time to next intervention (TTNI), tumor growth rate (TGR), health-related quality of life (HRQoL), neurocognition and seizure activity. Results: 331 patients were randomized (VOR, 168; PBO, 163). The median age was 40.0 years. 172 and 159 patients had histologically confirmed oligodendroglioma and astrocytoma, respectively. Treatment with VOR significantly improved PFS and TTNI. Median PFS: VOR, 27.7 mos; PBO, 11.1 mos (P=0.000000067). Median TTNI: VOR, not reached; PBO, 17.8 mos (P=0.000000019). Treatment with VOR resulted in shrinkage of tumor volume. Post-treatment TGR: VOR, -2.5% (95% CI: -4.7, -0.2); PBO, 13.9% (95% CI: 11.1, 16.8). HRQoL and neurocognition were preserved and seizure control was maintained. VOR had a manageable safety profile. Conclusions: VOR was effective in mIDH1/2 diffuse glioma not in immediate need of chemoradiotherapy.

Background: A key efficacy indicator in generalized myasthenia gravis (gMG) treatment is improvement in MG-ADL score. Minimal symptom expression (MSE, MG-ADL total score of 0 or 1) is explored as a novel proposed treatment target in gMG in the phase 3 study of intravenous efgartigimod, ADAPT, and its open-label extension, ADAPT+. Methods: Post hoc analyses of acetylcholine receptor antibody positive participants in ADAPT (n=129) and ADAPT+ (n=111) were performed. Results: In ADAPT, 44.6% receiving efgartigimod achieved MSE vs 10.9% of participants given placebo. Despite less frequent assessment during ADAPT+, 40.5% of participants achieved MSE. Eighty-one percent of participants treated with efgartigimod who achieved MSE in ADAPT also achieved MSE during ADAPT+; 23% who had not achieved MSE in ADAPT did in ADAPT+. Achieving MSE was associated with substantial improvements in QMG, MGC, MG-QoL15r, and EQ-5D-5L mean scores of 11.4, 16.0, 12.4, and 0.3 points, respectively, from baseline to best score (across all visits). These drastic improvements resulted in quality of life (QoL) comparable to that of healthy populations. MSE achievement also resulted in sustained improvements in these disease-specific and QoL measures. Conclusions: Participants who achieved MSE showed substantial and consistent improvements across multiple disease measures and experienced QoL comparable to that of healthy populations.

While there is a recognised role of optimising lifestyle behaviours such as diet and physical activity in the management of infertility, the best practice for lifestyle management of infertility remains unknown, and factors influencing the lifestyle behaviours of people with infertility are not well understood. The aim of this systematic review is to evaluate the barriers and enablers to a healthy lifestyle in people with infertility, from the perspectives of people with infertility and health professionals, in order to inform optimal behavioural change strategies for lifestyle management of infertility. Ovid MEDLINE(R), PsycINFO, EMBASE, EBM Reviews, and CINAHL Plus were searched from inception to 12th September 2022. Eligible studies were qualitative, quantitative or mixed-methods primary studies which explored barriers and/or enablers to lifestyle for infertility management, from the perspectives of people with infertility and/or health professionals. Two independent reviewers performed quality assessment, using the Centre for Evidence-Based Management Critical Appraisal of a Survey Tool (quantitative and mixed-methods studies) and the Critical Appraisal Skills Programme Qualitative Checklist (qualitative and mixed-methods studies). Data were analysed by inductive thematic analysis with themes mapped to the Capability, Opportunity, Motivation and Behaviour (COM-B) model(1) and Theoretical Domains Framework (TDF)(2). Relevant behaviour change techniques (BCTs)(3) to target the identified enablers and barriers were suggested. After screening 10703 citations and 82 full-texts, 22 studies were included (12 quantitative, 7 mixed-methods and 3 qualitative) with 18 studies including women with infertility (n = 2442), 10 including men with infertility (n = 1372) and 6 including health professionals (n = 261). From the perspectives of people with infertility, themes related to capability (e.g. strategies for behaviour change), opportunity (e.g. limited time, resources and money) and motivation (e.g. interplay between lifestyle and emotional state); themes mapped to 8 TDF domains. From the perspectives of health professionals, themes related to capability (e.g. identification of patients appropriate for lifestyle intervention), opportunity (e.g. mode of delivery) and motivation (e.g. professional responsibility); themes mapped to 6 TDF domains. 34 BCTs were identified across the suggested interventions. This systematic review found that several interacting factors influence lifestyle in people with infertility as well as health professional behaviour with regards to provision of lifestyle interventions for infertility. These factors can be targeted for optimisation of interventions. In light of the limited number of qualitative studies, there is a need for more qualitative research to gain deeper insights into the perspectives of people with infertility and health professionals for further exploration of the complex and interacting factors which shape lifestyle during the fertility journey.

We first sequenced and characterised the complete mitochondrial genome of Toxocara apodeme, then studied the evolutionary relationship of the species within Toxocaridae. The complete mitochondrial genome was amplified using PCR with 14 specific primers. The mitogenome length was 14303 bp in size, including 12 PCGs (encoding 3,423 amino acids), 22 tRNAs, 2 rRNAs, and 2 NCRs, with 68.38% A+T contents. The mt genomes of T. apodemi had relatively compact structures with 11 intergenic spacers and 5 overlaps. Comparative analyses of the nucleotide sequences of complete mt genomes showed that T. apodemi had higher identities with T. canis than other congeners. A sliding window analysis of 12 PCGs among 5 Toxocara species indicated that nad4 had the highest sequence divergence, and cox1 was the least variable gene. Relative synonymous codon usage showed that UUG, ACU, CCU, CGU, and UCU most frequently occurred in the complete genomes of T. apodemi. The Ka/Ks ratio showed that all Toxocara mt genes were subject to purification selection. The largest genetic distance between T. apodemi and the other 4 congeneric species was found in nad2, and the smallest was found in cox2. Phylogenetic analyses based on the concatenated amino acid sequences of 12 PCGs demonstrated that T. apodemi formed a distinct branch and was always a sister taxon to other congeneric species. The present study determined the complete mt genome sequences of T. apodemi, which provide novel genetic markers for further studies of the taxonomy, population genetics, and systematics of the Toxocaridae nematodes.

In this paper, we investigate the attitude manoeuver planning and tracking control of the flexible satellite equipped with a coilable mast. Due to its flexible beamlike structure, the coilable mast experiences bending and torsional modal vibrations in multi-direction. The complex nonlinear coupling and other external disturbances significantly impact the achievement of high-precision attitude control. To overcome these challenges, a robust attitude tracking controller is proposed for easy implementation by the Attitude Determination and Control System (ADCS). The controller consists of a disturbance compensator, feedforward controller and output feedback controller. The compensator, based on a Nonlinear Disturbance Observer (NDO), effectively compensates for the cluster disturbances caused by vibrations, environmental factors and parameter perturbations. The feedforward controller tracks the desired path in the nominal satellite model. Furthermore, the output feedback controller enables large-angle manoeuver control of the satellite and evaluates the suppression effect of the controlled output on the observation error of cluster disturbances used the ${L_2}$-gain. Simulation results demonstrate that the proposed controller successfully achieves high-precision attitude tracking control during large-angle manoeuvering.

Siltstones from the Precambrian Freda Formation in Wisconsin have been studied by scanning, scanning-transmission and analytical electron microscopy (SEM, STEM and AEM), and X-ray diffraction (XRD). XRD data for drill core samples show a change from smectite-rich Illite-Smectite (I-S) in shallow samples to illite in deeper samples, implying a transition during burial diagenesis.

Transmission electron microscopy (TEM) observations of shallow samples reveal the presence of three clearly distinguishable kinds of dioctahedral clay minerals: (1) detrital grains of micrometer-sized, mature muscovite; (2) small packets in the matrix consisting of dominant (Reichweite) R1 I-S or (3) small packets of illite. The illite and I-S stacks occur separately and have similar textures, with packet thicknesses averaging ∼400 Å, within the range of anchizonal illite. Illite and detrital muscovite commonly display strain features typical of the effects of tectonic stress. Void space within I-S or illite packets is inferred to be a strain feature, and to have served as pathways for fluids. Detrital muscovite shows abundant alteration features including (001) boundaries which are continuous with parallel packets of I-S; individual layers commonly show along-layer transitions of muscovite to smectite or I-S. Trioctahedral clays consist primarily of detrital chlorite which commonly shows direct alteration to R1 I-S and smectite, as with detrital muscovite.

Deep samples contain only unaltered, coarse detrital muscovite, and thin packets of illite forming stacks and comprising most of the matrix. The texture of the illite appears to be identical to that of shallow samples, with characteristics such as packet size typical of anchizonal illite. Trioctahedral clays consist almost entirely of detrital grains of chlorite and corrensite. They occur as separate grains with rather constant composition, without signs of alteration.

The data imply that all of the studied rocks have been subjected to a uniform anchizonal grade of metamorphism in which detrital grains were largely unchanged but matrix clays were transformed to packets of illite. The unusually abrupt transition with depth from highly expandable I-S to illite is inferred to actually be the result of subsequent alteration of authigenic illite and detrital chlorite and muscovite to R1 I-S and smectite in shallow rocks. This late overprinting of the anchimetamorphic clay mineral assemblage is inferred to have been locally caused by fluids with temperatures less than those of peak metamorphism. This process, called “retrograde diagenesis”, gave rise to a sequence of dioctahedral I-S and illite which mimics classic prograde sequences. Interpretations of such sequences as being prograde, especially in cases of ancient rocks, should be interpreted with caution when high-resolution images of textures are not available.

Caustic nuclear wastes have leaked from tanks at the US Department of Energy’s Hanford site in Washington State (USA) causing hundreds of thousands of gallons of waste fluids to migrate into the underlying sediments. In this study, four simulant tank waste (STW) solutions, which are high in NaOH (1.4 and 2.8 mol/kg), NaNO3 (3.7 mol/kg) and NaAlO2 (0.125 and 0.25 mol/kg), were prepared and reacted with reference kaolinite KGa-1 and KGa-2 at 50 and 80°C for up to 2 months. The structure and morphology of the resulting products were characterized using X-ray diffraction, scanning electron microscopy, and Fourier transform infrared spectroscopy. The products were also examined for cation exchange and Cs+ sorption as a function of ionic strength and types of cations in the background solutions. Cancrinite and sodalite were the only new minerals observed in all of the conditions tested in this experiment. Two major chemical processes were involved in the reactions: dissolution of kaolinite and precipitation of cancrinite and sodalite. Increasing NaOH concentration and temperature, and decreasing NaAlO2 concentration increased the transformation rate. Both cancrinite and sodalite appeared stable thermodynamically under the experimental conditions. The newly formed feldspathoids were vulnerable to acid attack and pronounced dissolution occurred at pH below 5.5. Cancrinite and sodalite can incorporate NaNO3 ion pairs in their cages or channels. Sodium in cancrinite and sodalite was readily exchangeable by K+, but less easily by Cs+ or Ca2+. The feldspathoid products sorb nearly an order of magnitude more Cs+ than the unaltered kaolinite. The Cs adsorption is reduced by competing cations in the background solutions. At low ionic strength (0.01 M NaNO3 or 0.005 M Ca(NO3)2), Ca2+ was more competitive than Na+. When the concentration of the background solution was increased 10 times, Na+ was more competitive than Ca2+.

Transient electromagnetic field plays very important roles in the evolution of high-energy-density matter or laser plasma. Now, a new design is proposed in this paper to diagnose the transient magnetic field, using relativistic electron bunch as a probe based on high-energy electron radiography. And based on this scheme, the continuous distribution of magnetic strength field can be snapshotted. For 1 mm thick quadrupole magnet model measured by 50 MeV probe electron beams, the simulation result indicates that this diagnosis has spatial resolution better than 4 microns and high measurement accuracy for strong magnetic strength and high magnetic gradient field no matter whether the magnetic interaction is focusing or defocusing for the range from -510 T∗ μm to 510 T∗ μm.

We argue that i-frame interventions can and do increase support for systemic reforms, and s-frame interventions should be pursued in parallel to address key societal issues. Without accompanying i-frame interventions, s-frame interventions can fail. We offer an operant conditioning framework to generate positive spillover effects. Behavioral scientists should develop i-frame interventions that enhance, rather than compete with, s-frame interventions.

In China, government at all levels relies on the specially selected graduates (SSG) scheme to recruit elite university students as future political leaders. This article examines the mechanism of the SSG scheme and the relationship between elite university education and political selection in China. We show that elite education is increasingly stratified, such that graduates from top elite universities have significant selection advantages in the SSG competition and are more likely to be offered incentives and preferences. We argue that taking elite university education as a hard eligibility criterion reinforces the homophily effects in selection of future political elites and strengthens the political influence of top elite universities on China's politics. Further, because poor and lower-class students have little chance of entering elite universities, the SSG does not provide an effective route of upward mobility for non-elite classes. Merit-based political recruitment as a channel of upward mobility for non-elite classes is largely an illusion.

OBJECTIVES/GOALS: Although gut fungi have been implicated in the immunopathogenesis of inflammatory bowel disease, the fungal microbiome has not been deeply explored across endo-histologic activity and treatment-exposure in ulcerative colitis. METHODS/STUDY POPULATION: Our retrospective cohort was derived from the Study of a Prospective Adult Research Cohort with Inflammatory Bowel Disease. We evaluated the fungal composition of fecal samples from 98 ulcerative colitis patients across endoscopic activity (n=43), endo-histologic activity (n=41), and biologic-exposure (n=98). Across all subgroups, we assessed fungal diversity and differential abundance of specific taxonomic groups. RESULTS/ANTICIPATED RESULTS: We identified 504 unique fungal amplicon sequence variants across the cohort of 98 patients, dominated by phylum Ascomycota. Compared to endoscopic remission, patients with endoscopic activity had an increased global fungus load (p DISCUSSION/SIGNIFICANCE: Endoscopic inflammation in ulcerative colitis is associated with altered fungal diversity driven by expansion of Saccharomyces and Candida compared to remission. The role of these fungal taxa as potential biomarkers and targets for personalized approaches to therapeutics in ulcerative colitis should be evaluated.

This study aimed to investigate the relationship between bone quality in terms of metabolism, homeostasis of elements, bone mineral density (BMD), and microstructure and keel-bone fractures in laying hens (Gallus gallus domesticus). One hundred and twenty 17 week old Lohmann White laying hens with normal keel bones were individually housed in furnished cages for 25 weeks. Birds were then euthanased and dissected to assess keel-bone status at 42 weeks. Serum and keel-bone samples from normal keel (NK) and fractured keel (FK) hens were collected to determine the previously mentioned bone quality parameters. The results showed FK hens to have higher levels of the components of osteocalcin, greater alkaline phosphatase activity in serum and keel bones, and greater tartrate-resistant acid phosphatase (TRAP) activity in keel bones, compared to NK hens. Additionally, FK hens also had higher concentrations of Li, B, K, Cu, As, Se, Sn, Hg, and Pb, but lower concentrations of Na, P, and Ca. Moreover, FK hens showed decreased bone microstructural parameters including bone volume/tissue volume, trabecular number, degree of anisotropy, connectivity density, and BMD, but increased trabecular separation. Meanwhile, no differences were detected in serum TRAP activity, trabecular thickness, bone surface, or bone surface/bone volume. Results showed laying hens with keel-bone fractures to have differences in bone metabolism, elements of home-ostasis, bone microstructure parameters, and BMD. These results suggest that keel-bone fractures may be associated with bone quality.
