Sperm capacitation involves biochemical and functional changes necessary for fertilization. The ubiquitin–proteasome system (UPS) regulates protein turnover, but the roles of proteasomal receptors during capacitation remain unclear. This study investigates the regulation and functional impact of proteasomal subunits RPN13 (ADRM1) and RPN2 (PSMD1) during in vitro capacitation of boar sperm. Semen samples (n = 6) were assigned to four groups: non-capacitating control (Non-Cap, 4 h in medium lacking bovine serum albumin (BSA)), capacitating control (Cap, 4 h with BSA) and groups treated with vehicle (CM + dimethyl sulfoxide (DMSO)) or the RPN13 inhibitor RA190. We measured RPN2/RPN13 levels, RPN13-associated ubiquitin signal and RPN13–RPN2 interactions (Western blot, Co-IP) and assessed sperm kinematics, intracellular Ca2+, protein tyrosine phosphorylation (pTyr), mitochondrial membrane potential (ΔΨm), ATP, ZP binding and early embryo cleavage after IVF. The Cap group showed successful capacitation, confirmed by increases in pTyr, sperm hyperactivation and Ca2+-associated Fluo-4 fluorescence. During capacitation, RPN2 and RPN13 abundance decreased, while RPN2–RPN13 association increased. RA190 treatment increased RPN13 levels, inhibited RPN13 turnover and decreased its interaction with RPN2 and the associated ubiquitin signal. Functionally, RA190 attenuated sperm hyperactivation, Ca2+-associated Fluo-4 fluorescence, protein tyrosine phosphorylation, mitochondrial membrane potential and ATP levels and was associated with reduced fertilization-related outcomes, including ZP binding and early embryo cleavage after IVF. These findings highlight the role of RPN13 in sperm capacitation and fertilization.
This real-world study aimed to characterize patients with schizophrenia who achieve sustained good functional outcomes after antipsychotic discontinuation and to develop the Functional Remission in Schizophrenia after Antipsychotic Discontinuation (FURSAD) predictive model.
Methods
We retrospectively identified individuals aged 18–65 years with schizophrenia (ICD-10) from the Shanghai Mental Health Center discharge database. Patients who discontinued antipsychotics for ≥1 year were classified as functional remission (FR) or functional non-remission (FNR) based on functioning assessments. Sociodemographic, clinical, and treatment-related data were extracted blindly from hospital records and structured interviews.
Results
Among 4,166 discharged patients screened, 180 met the inclusion criteria (FR: 116; FNR: 64). Six independent predictors were identified: total disease course, Clinical Global Impression-Severity (CGI-S) score, Positive and Negative Syndrome Scale (PANSS) emotional distress subscale score, use of first-generation antipsychotics, discontinuation due to treatment benefits, and discontinuation due to lack of insight. The logistic regression model showed strong predictive performance (AUC = 0.867, 95% CI 0.813–0.921), with 82.8% sensitivity and 81.9% specificity. Internal validation was performed via 10-fold cross-validation.
Conclusion
Discontinuation motives and illness trajectory are relevant in predicting long-term functional outcomes. A limitation is that a substantial number of patients could not be recontacted or declined participation, which may introduce selection bias. The FURSAD nomogram may help clinicians estimate the probability of FR 4.5 years post-antipsychotic discontinuation in patients previously on antipsychotics for ≥3 years.
Grasslands are one of the major ecosystem types in drylands. Encroachment of shrubs into grasslands affects the functioning of drylands by altering community structure, with impacts exacerbated under greater intensity of encroachment. Yet, we have a limited understanding of how ecosystem structure responds to the degree of shrub encroachment. Here, we describe a field-based study designed to examine changes in ecosystem structure beneath shrub patches (patch condition) and between patches (spatial distribution pattern of patches) along a gradient in encroachment in a semiarid grassland in Inner Mongolia, China. We found that greater encroachment was associated with wider and taller shrubs with more branches. As shrub encroachment intensified, the area beneath shrubs had more litter and was less exposed to grazing. The landscape was characterized by more discontinuous patches of vegetation and more bare ground as encroachment intensified. Either the patch condition or the patch spatial pattern was shaped mainly by the magnitude of shrub encroachment rather than by the structure of individual shrubs (e.g., height and canopy width). Our study highlights the idiosyncratic response of ecosystem structure (patch condition and patch spatial pattern) to intensifying encroachment, reinforcing the importance of considering the degree of shrub encroachment when managing encroached grasslands.
Ultra-thin liquid sheets generated by impinging two liquid jets are crucial high-repetition-rate targets for laser ion acceleration and ultra-fast physics, and serve widely as barrier-free samples for structural biochemistry. The impact of liquid viscosity on sheet thickness should be comprehended fully to exploit its potential. Here, we demonstrate experimentally that viscosity significantly influences thickness distribution, while surface tension primarily governs shape. We propose a thickness model based on momentum exchange and mass transport within the radial flow, which agrees well with the experiments. These results provide deeper insights into the behaviour of liquid sheets and enable accurate thickness control for various applications, including atomization nozzles and laser-driven particle sources.
Using quantitative analysis and qualitative in-depth interviews conducted in China in 2022–2024, this study explores a new form of grievance expression and dispute resolution adopted by the Chinese Communist Party (CCP): the online petition. While digital technology can receive and handle public complaints more efficiently, local officials under intense top-down pressure and increased accountability often only address citizens’ demands performatively to satisfy the political needs of their superiors. The study of “mechanical responsiveness” reveals that the CCP’s reinforced scrutiny of petitioners as well as local officials not only fails to prevent and resolve disputes at the source but also may lead to increased social discontent and pent-up resentments in the long run when the government fails to live up to its promises. Citizens will also lose faith in the institution and become politically disengaged if grassroots officials are always trying to curry favour with their superiors and avoid punishment rather than address public demands.
Asian corn borer, Ostrinia furnacalis Guenée (Lepidoptera: Crambidae), is a major pest in corn production, and its management remains a significant challenge. Current control methods, which rely heavily on synthetic chemical pesticides, are environmentally detrimental and unsustainable, necessitating the development of eco-friendly alternatives. This study investigates the potential of the entomopathogenic nematode Steinernema carpocapsae as a biological control agent for O. furnacalis pupae, focusing on its infection efficacy and the factors influencing its performance. We conducted a series of laboratory experiments to evaluate the effects of distance, pupal developmental stage, soil depth, and light conditions on nematode attraction, pupal mortality and sublethal impacts on pupal longevity and oviposition. Results demonstrated that S. carpocapsae exhibited the highest attraction to pupae at a 3 cm distance, with infection declining significantly at greater distances. Younger pupae (<12 h old), were more attractive to nematodes than older pupae, and female pupae were preferred over males. Nematode infection was highest on the head and thorax of pupae, with a significant reduction in infection observed after 24 h. Infection caused 100% mortality in pupae within 2 cm soil depth, though efficacy was reduced under light conditions. Sublethal effects included a significant reduction in the longevity of infected adults and a decrease in the number of eggs laid by infected females compared to controls. These findings underscore the potential of S. carpocapsae as an effective biocontrol agent for sustainable pest management in corn production, offering a viable alternative to chemical pesticides.
Caused by multiple risk factors, heavy burden of major depressive disorder (MDD) poses serious challenges to public health worldwide over the past 30 years. Yet the burden and attributable risk factors of MDD were not systematically known. We aimed to reveal the long-term spatio-temporal trends in the burden and attributable risk factors of MDD at global, regional and national levels during 1990–2019.
Methods
We obtained MDD and attributable risk factors data from Global Burden of Disease Study 2019. We used joinpoint regression model to assess the temporal trend in MDD burden, and age–period–cohort model to measure the effects of age, period and birth cohort on MDD incidence rate. We utilized population attributable fractions (PAFs) to estimate the specific proportions of MDD burden attributed to given risk factors.
Results
During 1990–2019, the global number of MDD incident cases, prevalent cases and disability-adjusted life years (DALYs) increased by 59.10%, 59.57% and 58.57%, respectively. Whereas the global age-standardized incidence rate (ASIR), age-standardized prevalence rate (ASPR) and age-standardized DALYs rate (ASDR) of MDD decreased during 1990–2019. The ASIR, ASPR and ASDR in women were 1.62, 1.62 and 1.60 times as that in men in 2019, respectively. The highest age-specific incidence, prevalence and DALYs rate occurred at the age of 60–64 in women, and at the age of 75–84 in men, but the maximum increasing trends in these age-specific rates occurred at the age of 5–9. Population living during 2000–2004 had higher risk of MDD. MDD burden varied by socio-demographic index (SDI), regions and nations. In 2019, low-SDI region, Central sub-Saharan Africa and Uganda had the highest ASIR, ASPR and ASDR. The global PAFs of intimate partner violence (IPV), childhood sexual abuse (CSA) and bullying victimization (BV) were 8.43%, 5.46% and 4.86% in 2019, respectively.
Conclusions
Over the past 30 years, the global ASIR, ASPR and ASDR of MDD had decreased trends, while the burden of MDD was still serious, and multiple disparities in MDD burden remarkably existed. Women, elderly and populations living during 2000–2004 and in low-SDI regions, had more severe burden of MDD. Children were more susceptible to MDD. Up to 18.75% of global MDD burden would be eliminated through early preventing against IPV, CSA and BV. Tailored strategies-and-measures in different regions and demographic groups based on findings in this studywould be urgently needed to eliminate the impacts of modifiable risk factors on MDD, and then mitigate the burden of MDD.
To meet the demands of laser-ion acceleration at a high repetition rate, we have developed a comprehensive diagnostic system for real-time and in situ monitoring of liquid sheet targets (LSTs). The spatially resolved rapid characterizations of an LST’s thickness, flatness, tilt angle and position are fulfilled by different subsystems with high accuracy. With the help of the diagnostic system, we reveal the dependence of thickness distribution on collision parameters and report the 238-nm liquid sheet generated by the collision of two liquid jets. Control methods for the flatness and tilt angle of LSTs have also been provided, which are essential for applications of laser-driven ion acceleration and others.
Childhood maltreatment is an established risk factor for psychopathology. However, it remains unclear how childhood traumatic events relate to mental health problems and how the brain is involved. This study examined the serial mediation effect of brain morphological alterations and emotion-/reward-related functions on linking the relationship from maltreatment to depression. We recruited 156 healthy adolescents and young adults and an additional sample of 31 adolescents with major depressive disorder for assessment of childhood maltreatment, depressive symptoms, cognitive reappraisal and anticipatory/consummatory pleasure. Structural MRI data were acquired to identify maltreatment-related cortical and subcortical morphological differences. The mediation models suggested that emotional maltreatment of abuse and neglect, was respectively associated with increased gray matter volume in the ventral striatum and greater thickness in the middle cingulate cortex. These structural alterations were further related to reduced anticipatory pleasure and disrupted cognitive reappraisal, which contributed to more severe depressive symptoms among healthy individuals. The above mediating effects were not replicated in our clinical group partly due to the small sample size. Preventative interventions can target emotional and reward systems to foster resilience and reduce the likelihood of future psychiatric disorders among individuals with a history of maltreatment.
As the southernmost part of the central segment of the Central Asian Orogenic Belt, the northern Alxa area is characterized by abundant Permian magmatism and records key information on the geological evolution of the Palaeo-Asian Ocean. This study reports new zircon U–Pb and Lu–Hf isotopic and whole-rock geochemical data of the early Permian (285–286 Ma) Huisentala gabbro and Huodonghaer diorites from the Zhusileng–Hangwula Belt in the northern Alxa area. The gabbro is characterized by high Al, Ca, Mg# and light rare-earth elements, and low K, P and high field strength elements (e.g., Ti, Nb and Ta). Furthermore, the gabbro shows heterogeneous zircon ϵHf(t) value (−2.5 to +2.6). The Huodonghaer diorites show high MgO (3.46–6.32 wt%), Mg# (49–58), Sr (408–617 ppm) and Ba (223–419 ppm), and low FeOT/MgO (1.27–1.83) and TiO2 (0.48–0.90 wt%), with geochemical features similar to the high-Mg andesite/diorite. They show radiogenic zircon ϵHf(t) values of +1.2 to +4.9 and high Th/Nb ratios. These features suggest that the Huisentala gabbro and the Huodonghaer diorites were derived from the partial melting of mantle peridotite that was metasomatized by subduction-related fluids and by subducted sediment-derived melts, respectively.
The authors at times propose that robots are mere depictions of social agents (a philosophical claim) and at other times that people conceive of social robots as depictions (an empirical psychological claim). We evaluate each claim's accuracy both now and in the future and, in doing so, we introduce two dangerous misperceptions people have, or will have, about social robots.
The question of whether narcissists are more creative than peers has attracted much scholarly attention in both psychology and organizational management sciences. Drawing from social cognitive theory, we theorized that the relationship between narcissism and creativity could be explained by individual creative self-efficacy, which depends on one's direct and vicarious experiences of creativity. Drawing from trait activation theory, we further proposed organizational valuing of creativity as a key contextual moderator that determines whether narcissism facilitates or inhibits creative self-efficacy and, in turn, creativity. We suggest that high organizational valuing of creativity will energize narcissists to put their attention and effort into both direct and vicarious experiences of creativity, enhancing their creative self-efficacy and creativity. We tested our conceptual model through a field study with data collected from 269 full-time employees working in 86 work teams. The empirical results provided support for the social cognitive explanation for the positive relationship between narcissism and creativity in the context of high organizational valuing of creativity. Our study not only resolved prior debates on the relationship between narcissism and creativity but also provided direct empirical support for social cognitive theory and the person-in-context interactionist perspective of creativity research.
Lamprophyre dikes and quartz veins in the Sizhuang gold deposit are used to date and unravel the Early Cretaceous stress regime in the Jiaodong Peninsula, North China Craton. The lamprophyre dikes are grouped into two major sets, trending NNW–SSE and NNE–SSW, respectively, and a subsidiary one, trending WNW–ESE, whereas the quartz veins trend mainly NNE–SSW. The age of lamprophyre dikes’ intrusion was robustly calculated at c. 119 Ma by phlogopite 40Ar–39Ar dating. The fuzzy clustering technique defined four stress states, which were grouped into three stress regimes in Sizhuang. Furthermore, these stress regimes were interrelated with two regional far-field stress regimes, ST1 and ST2, defined by the available published fault-slip data near Sizhuang by applying the separation and stress inversion TR method (TRM). The palaeostress reconstruction indicates that extension and strike-slip tectonics (i.e. transtension) affect the emplacement of the lamprophyre dikes and quartz veins. More precisely, the dike emplacement occurred under unstable stress conditions related to the shift from E–W pure extension–transtension to WNW–ESE transtension, whereas the quartz veins formed under relatively stable WNW–ESE transtension with the variant and increasing fluid pressure (${P_{\rm{f}}})$ giving rise to the strike-slip against dip-slip kinematics along the faults. The change from the ST1 to ST2 stress regimes reflects the significant clockwise rotation in the palaeo-Pacific plate subduction direction and defines the switching time at c. 119 Ma as precisely obtained by the lamprophyre dating.
Trypanosoma musculi is a, globally distributed, mouse-specific haemoflagellate, of the family Trypanosomatidae, which shares similar characteristics in morphology with Trypanosoma lewisi. The kinetoplast (mitochondrial) DNA of Trypanosomatidae flagellates is comprised of catenated maxicircles and minicircles. However, genetic information on the T. musculi kinetoplast remains largely unknown. In this study, the T. musculi maxicircle genome was completely assembled, with PacBio and Illumina sequencing, and the size was confirmed at 34 606 bp. It consisted of 2 distinct parts: the coding region and the divergent regions (DRs, DRI and II). In comparison with other trypanosome maxicircles (Trypanosoma brucei, Trypanosoma cruzi and T. lewisi), the T. musculi maxicircle has a syntenic distribution of genes and shares 73.9, 78.0 and 92.7% sequence identity, respectively, over the whole coding region. Moreover, novel insertions in MURF2 (630 bp) and in ND5 (1278 bp) were found, respectively, which are homologous to minicircles. These findings support an evolutionary scenario similar to the one proposed for insertions in Trypanosoma cruzi, the pathogen of American trypanosomiasis. These novel insertions, together with a deletion (281 bp) in ND4, question the role of Complex I in T. musculi. A detailed analysis of DRII indicated that it contains numerous repeat motifs and palindromes, the latter of which are highly conservative and contain A5C elements. The comprehensively annotated kinetoplast maxicircle of T. musculi reveals a high degree of similarity between this parasite and the maxicircle of T. lewisi and suggests that the DRII could be a valuable marker for distinguishing these evolutionarily related species.
The southeastern Central Asian Orogenic Belt (CAOB) records the assembly process between several micro-continental blocks and the North China Craton (NCC), with the consumption of the Paleo-Asian Ocean (PAO), but whether the S-wards subduction of the PAO beneath the northern NCC was ongoing during Carboniferous–Permian time is still being debated. A key issue to resolve this controversy is whether the Carboniferous magmatism in the northern NCC was continental arc magmatism. The Alxa Block is the western segment of the northern NCC and contiguous to the southeastern CAOB, and their Carboniferous–Permian magmatism could have occurred in similar tectonic settings. In this contribution, new zircon U–Pb ages, elemental geochemistry and Sr–Nd isotopic analyses are presented for three early Carboniferous granitic plutons in the southwestern Alxa Block. Two newly identified aluminous A-type granites, an alkali-feldspar granite (331.6 ± 1.6 Ma) and a monzogranite (331.8 ± 1.7 Ma), exhibit juvenile and radiogenic Sr–Nd isotopic features, respectively. Although a granodiorite (326.2 ± 6.6 Ma) is characterized by high Sr/Y ratios (97.4–139.9), which is generally treated as an adikitic feature, this sample has highly radiogenic Sr–Nd isotopes and displays significantly higher K2O/Na2O ratios than typical adakites. These three granites were probably derived from the partial melting of Precambrian continental crustal sources heated by upwelling asthenosphere in lithospheric extensional setting. Regionally, both the Alxa Block and the southeastern CAOB are characterized by the formation of early Carboniferous extension-related magmatic rocks but lack coeval sedimentary deposits, suggesting a uniform lithospheric extensional setting rather than a simple continental arc.
Narrowband microwave generation with tuneable frequency is demonstrated by illuminating a photoconductive semiconductor switch (PCSS) with a burst-mode fibre laser. The whole system is composed of a high-power linearly polarized burst-mode pulsed fibre laser and a linear-state PCSS. To obtain a high-performance microwave signal, a desired envelope of burst is necessary and a pulse pre-compensation technique is adopted to avoid envelope distortion induced by the gain-saturation effect. Resulting from the technique, homogenous peak power distribution in each burst is ensured. The maximum energy of the laser burst pulse reaches 200 μJ with a burst duration of 100 ns at the average power of 10 W, corresponding to a peak power of 4 kW. When the PCSS is illuminated by the burst-mode fibre laser, narrowband microwave generation with tuneable frequency (0.80–1.12 GHz) is obtained with a power up to 300 W. To the best of the authors’ knowledge, it is the first demonstration of frequency-tuneable narrowband microwave generation based on a fibre laser. The high-power burst-mode fibre laser reported here has great potential for generating high-power arbitrary microwave signals for a great deal of applicable demands such as smart adaptive radar and intelligent high-power microwave systems.
Anaemia is a global public health problem affecting women worldwide, and reproductive-age women are at increased risk. We conducted a population-based cross-sectional study analysing the prevalence of overall anaemia and anaemia according to severity in Chinese pre-pregnant women to update current knowledge on anaemia epidemiology. Based on the National Free Preconception Check-up Projects supported by the Chinese government, 5 679 782 women participating in this project in 2017 were included in the present study. The cyanmethemoglobin method was applied to assess Hb concentrations. Univariate and multivariate logistic regressions were applied for associated factors. The prevalence of anaemia among Chinese pre-pregnant women was 21·64 % (mild: 14·10 %, moderate: 7·17 % and severe : 0·37 %). The prevalence of overall and severe anaemia was the highest in Tibet and the lowest in Beijing among thirty-one provinces. Women’s age, region, ethnic origin, educational level, occupation and pregnancy history were all correlated with anaemia. Women with B blood type (adjusted OR (aOR) = 0·89), higher BMI (overweight: aOR = 0·84; obesity: aOR = 0·70) and alcohol consumption (aOR = 0·69) were less likely to have anaemia, while those with rhesus negative blood type (aOR = 1·10), history of anaemia (aOR = 2·60), older age at menarche (aOR = 1·19), heavy menstrual blood loss (aOR = 1·39), longer menstrual period (aOR = 1·09) and shorter menstrual cycle (aOR = 1·08) were more likely to suffer from anaemia. Meat or egg eaters were not significantly associated with severe anaemia. Anaemia is of moderate public health significance among Chinese pre-pregnant women. Interventions should be considered to prevent anaemia to the greatest extent possible to avoid potential harm in this population.
Fullerene dimers have attracted extensive attention due to their unique structures and fascinating properties. Here, fullerene dimer derivatives with four to six carbon atoms in the esters are designed and synthesized. The property differences that caused by the carbon number in the esters of the fullerene dimers are investigated by performing their electrochemical, optical, and photoelectric measurements. As the carbon atom numbers in the esters increase from four to five and six, the absorption intensities increase to 1.6- and 4.4-folds. The intensities of the fluorescence spectra increase to 1.8- and 5.2-folds. Their photocurrent increases to 2- and 7-folds under the irradiation of a 405-nm laser. The LUMO energy levels move downward slightly from −3.89 to −3.90 and −3.92 eV, respectively. Our results indicate that as the carbon number increases, the carbon chain lengths in the ester structures increase, very slight effects produced on the energy levels of the fullerene dimers, but strongly contribute to their chemical activities and thus the photoelectronic efficiencies.
