Clonorchis sinensis (C. sinensis), a trematode parasite that invades the hypoxic hepatobiliary tract of vertebrate hosts requires a considerable amount of oxygen for its sexual reproduction and energy metabolism. However, little is known regarding the molecular mechanism of C. sinensis involved in the adaptation to the hypoxic environments. In this study, we investigated the molecular structures and induction patterns of hypoxia-inducible factor-1α (HIF-1α) and other basic helix–loop–helix and Per–Arnt–Sim (bHLH–PAS) domain-containing proteins such as HIF-1β, single-minded protein and aryl hydrocarbon receptor, which might prompt adaptive response to hypoxia, in C. sinensis. These proteins possessed various bHLH–PAS family-specific domains. Expression of C. sinensis HIF-1α (CsHIF-1α) was highly induced in worms which were either exposed to a hypoxic condition or co-incubated with human cholangiocytes. In addition to oxygen, nitric oxide and nitrite affected the CsHIF-1α expression depending on the surrounding oxygen concentration. Treatment using a prolyl hydroxylase-domain protein inhibitor under 20%-oxygen condition resulted in an increase in the CsHIF-1α level. Conversely, the other bHLH–PAS genes were less responsive to these exogenous stimuli. We suggest that nitrite and nitric oxide, as well as oxygen, coordinately involve in the regulation of HIF-1α expression to adapt to the hypoxic host environments in C. sinensis.

Hemiwicking refers to the spreading of a liquid on a rough hydrophilic surface driven by capillarity. Here, we construct scaling laws to predict the velocity of hemiwicking on a rough substrate and experimentally corroborate them with various arrangements and dimensions of micropillar arrays. At the macroscopic scale, where the wetting front appears parallel to the free surface of the reservoir, the wicking distance is shown to grow diffusively, i.e. like $t^{1/2}$ with $t$ being time. We show that our model is consistent with pillar arrays of a wide range of pitch-to-height ratios, either square or skewed. At the microscopic scale, where the meniscus extension from individual pillars at the wetting front is considered, the extension distance begins to grow like $t$ but the spreading slows down to behave like $t^{1/3}$ when the meniscus is far from the pillar. Our microscopic flow modelling allows us to find pillar spacing conditions under which the assumption of densely spaced pillars is valid.

Despite recent reports regarding the biology of cytosine methylation in Schistosoma mansoni, the impact of the regulatory machinery remains unclear in diverse platyhelminthes. This ambiguity is reinforced by discoveries of DNA methyltransferase 2 (DNMT2)-only organisms and the substrate specificity of DNMT2 preferential to RNA molecules. Here, we characterized a novel DNA methyltransferase, named CsDNMT2, in a liver fluke Clonorchis sinensis. The protein exhibited structural properties conserved in other members of the DNMT2 family. The native and recombinant CsDNMT2 exhibited considerable enzymatic activity on DNA. The spatiotemporal expression of CsDNMT2 mirrored that of 5-methylcytosine (5 mC), both of which were elevated in the C. sinensis eggs. However, CsDNMT2 and 5 mC were marginally detected in other histological regions of C. sinensis adults including ovaries and seminal receptacle. The methylation site seemed not related to genomic loci occupied by progenies of an active long-terminal-repeat retrotransposon. Taken together, our data strongly suggest that C. sinensis has preserved the functional DNA methylation machinery and that DNMT2 acts as a genuine alternative to DNMT1/DNMT3 to methylate DNA in the DNMT2-only organism. The epigenetic regulation would target functional genes primarily involved in the formation and/or maturation of eggs, rather than retrotransposons.

A novel nano-scale manipulator capable of handling low-dimensional materials with three-dimensional linear motion, gripping action, and push–pull action of the gripper was developed for an in situ experiment in transmission electron microscopy. X-Y-Z positioning and push–pull action were accomplished by a piezotubing system, combined with a specially designed assembly stage that consisted of a lever-action gripping tip backed by a push–pull piezostack. The gripper tip consisted of tungsten wire fabricated by electrochemical etching followed by a focused ion beam process. Performance of the nano-scale manipulator was demonstrated in a grab-and-pick test of a single silver nanowire and in an in situ tensile test of a pearlitic steel sample with a specific orientation.

Extreme wetting properties of solids, either superhydrophobic or superhydrophilic, provide versatile methods to achieve unusual liquid deposit morphologies, such as liquid pearls or polygonal films. Here we report the dynamics of liquid drops that impact on solid surfaces where the extreme wetting properties are coupled in such a way that a superhydrophilic annulus is patterned on a superhydrophobic background. The drop that initially spreads on the inner superhydrophobic region is arrested by the hydrophilic annulus. The liquid deposit gets destabilized because of the strong water repellence of the inner region, exhibiting the burst and disengagement of the liquid film. This process leads to the formation of a liquid ring defined by the annulus pattern, which has practical implications in rapid printing of functional liquids. We visualize such drop dynamics with a high-speed camera and characterize their salient features by combining experimental measurements and theoretical considerations.

When a drop is deposited on a superhydrophilic micropillar array, the upper part of the drop (referred to as the bulk) collapses while the bottom part penetrates into the gaps of the array, forming a fringe film. Here we quantify the early stage dynamics of this process using a combination of experiment and theory. We show that the circular front of the fringe film spreads like t1/2, t being time, when coupled to the bulk flow. However, the film is found to advance like t1/3 through faceted zippering in the absence of the bulk. We then show that the spreading of the bulk and the entire drop footprint follows a power law (t1/4) that is different from Washburn's law. This work can be a starting point to completely understand the spreading of liquids on superhydrophilic surfaces and opens questions specific to superwetting behaviour including the criteria to determine whether the fringe film will expand through lateral zipping or advance radially outwards.

We present a method to realize nanoelectromechanical systems (NEMS) resonator structures from carbon nanotubes (CNT)/metal layered composite structures. The method utilizes a self aligning process over a near 2-D CNT network on GaAs to realize resonator structures and a highly selective etchant, a standard citric acid/hydrogen peroxide solution, to achieve release of NEMS resonators from the substrate. We find this method along with critical point drying (CPD) to yield robust hybrid CNT/metal resonator structures with fundamental resonant frequencies in the 10 MHz range. With high reflectivity, conductivity, and bio-compatibility of metals, this fabrication method along with possible benefits of CNT have many application possibilities.

During the course of a survey of sapstaining fungi in Korea, a Leptographium was isolated from Pinus densiflora and P. koraiensis logs, infested with the bark beetle Tomicus piniperda. The fungus grew optimally at 25 °C on 2% malt extract agar and showed a high level of tolerance to cycloheximide. The Leptographium has unusually short conidiophores and is morphologically similar to L. pini-densiflorae, L. lundbergii, L. yunnanense, and the Leptographium anamorph of Ophiostoma crassivaginatum. Comparisons of DNA sequence data for three gene regions, as well as morphological characteristics, confirmed that this fungus represents an undescribed taxon. We consequently provide the name Leptographium koreanum sp. nov. for it here.

We present accurate BV light curves and continuum energy curves of the EROS eclipsing binaries in the Large Magellanic Cloud to find accurate binary parameters as well as their distances. The observations have been carried out using the 2.1 meter telescope in CASLEO Argentina during Feb. 1-10, 2003. We have concentrated CCD direct observations upon EROS field 1 and 2 to improve the accuracy of light curves of eclipsing binaries in the fields. The spectroscopic observations have been also carried out using the simple dispersion method to get continuum energy curves between wavelengths of 4000-8000 Å for several EROS eclipsing binaries. At first, we determined a combined temperature of both components of each binary system using the continuum. Then, the combined temperature was resolved using the first estimation of the light curve solution. Finally, we determined the photometric solutions of several binaries in the Large Magellanic Cloud.

This paper describes the growth kinetics of an interfacial MgO layer as well as those of an MgB2 layer during ex situ annealing of the evaporated amorphous boron (a-B) film under Mg vapor overpressure. A thin MgO layer is formed at the interface between a-B and Al2O3 substrate before the formation of crystalline MgB2 layer and the interfacial layer is epitaxially related with Al2O3 substrate (MgO (111)[110] // Al2O3 (0001)[1100]). The interfacial MgO layer continues to grow during the annealing, and its apparent growth rate is about 0.1 nm/min. The analysis of MgB2 layer growth kinetics using cross-sectional transmission electron microscopy reveals that there exist two distinct growth fronts at both sides of an MgB2 layer. The growth kinetics of the lower MgB2 layer obeys the parabolic rate law during the entire annealing time. The growth of the upper MgB2 layer is controlled by the surface reaction between out-diffused boron and Mg vapor up to 10 min, resulting in a rough surface morphology of MgB2 layer. By considering the mass balance of Mg and boron during ex situ annealing, we obtained the diffusivities of Mg and boron in MgB2 layer which were in the same order range of approximately 10−12 cm2/s.

Radiata pine (Pinus radiata) lumber from Australia, Chile, and New Zealand is imported into Korea where it represents one of the most important sources of timber. Blue stain of this timber is a serious problem for which an integrated control strategy is being developed. One of the fungi that has been isolated from stained radiata pine sapwood in Korea is a Leptographium sp. that has a distinct Sporothrix synanamorph. The aim of this study was to classify this fungus. Morphological comparisons showed that this fungus is distinct from all other species of Leptographium and especially L. elegans and L. francke-grosmanniae that also have Sporothrix synanamorphs. Comparisons of sequence data for the ITS2 and part of the 28S rDNA genes as well as the β-tubulin gene also confirmed that this fungus represents an undescribed taxon, described here as Leptographium bistatum sp. nov.

Silver nanoparticles dispersed in a silica matrix were made by the consolidation of a Ag-attached silica colloid, which was synthesized via the electrolysis of a pure Ag electrode, the reduction of Ag+ ions by H2, and the nucleation and growth of Ag particles on the silica nanoparticles in water. This simple process produced Ag/silica nanocomposites with a high concentration and narrow size distribution of nanoparticles, which was confirmed by transmission electron microscopy and x-ray diffraction. As estimated by Raman and photoluminescence measurements, the quantity of broken oxygen bonds was increased with increasing Ag concentration due to the intervention of Ag ions as structural modifiers in the silica network structure. Ag ions in the matrix are probably a residue of the Ag+ ions that could not be reduced by H2 during the electrolysis/reduction reaction. The optical-absorption spectra and the HCl-soaking test suggested that a chemical-interface damping effect, which was caused by electron transfer from the metal particles to the oxide matrix, dominates the optical-absorption properties in this system.

Variations of metal concentrations with body size were investigated in kidney, digestive gland and gill of the Antarctic clam, Laternula elliptica, collected from a near-shore habitat at King George Island. Positive relationships for metal concentrations with body size were common among the three organs for most of the metals, while an inverse relationship was found only in the digestive gland for Cu, Zn and Mn. In the gill, concentrations of eight out of the ten metals increased linearly with body size, whilst in the kidney concentrations of Cd, Zn, Pb, Co and Ni were best fitted by quadratic regression curves, viz. increasing continuously until reaching a peak at around an 80 mm shell length. This may reflect specific regulatory activities for the extremely high accumulation of these metals in the kidney, particularly Cd (28–354 μg g−1 tissue dry weight). The overall prevalence of positive relationships indicate net accumulation of these metals throughout life, which may be at least in part associated with the slow growth and long life span of this cold-water bivalve species. The study demonstrates distinct body size-metal concentration relationships in the three organs of L. elliptica, and suggests they may be useful as biomonitors for assessing changes in metal concentrations in Antarctic waters.

The reaction sequence and microstructure evolution of a crystalline MgB2 layer were examined during ex situ annealing of evaporated amorphous boron (a-B) with Mg vapor. Mg was found to migrate rapidly into the a-B layer in the initial stage of reaction with a uniform concentration of about 12 at.%. A thin layer of crystalline MgO was observed at the interface between a-B and the Al2O3 substrate. It was identified that an MgB2 layer started to form at the surface by the nucleation and growth process in polycrystalline form. It appears that there exists two distinct growth fronts in the MgB2 layer: one lying at the surface and the other lying at the interface between the MgB2 layer and the a-B. The microstructural evolution of this layer showed significant differences depending on the location of these two growth fronts.
