Pro-inflammatory markers predict response to sequential pharmacotherapy in major depressive disorder: a CAN-BIND-1 report

Introduction
Despite replicated cross-sectional evidence of aberrant levels of peripheral inflammatory markers in individuals with major depressive disorder (MDD), there is limited literature on associations between inflammatory tone and response to sequential pharmacotherapies.
ObjectivesTo assess associations between plasma levels of pro-inflammatory markers and treatment response to escitalopram and adjunctive aripiprazole in adults with MDD.
MethodsIn a 16-week open-label clinical trial, 211 participants with MDD were treated with escitalopram 10– 20 mg daily for 8 weeks. Responders continued on escitalopram while non-responders received adjunctive aripiprazole 2–10 mg daily for 8 weeks. Plasma levels of pro-inflammatory markers – C-reactive protein, Interleukin (IL)-1β, IL-6, IL-17, Interferon gamma (IFN)-Γ, Tumour Necrosis Factor (TNF)-α, and Chemokine C–C motif ligand-2 (CCL-2) - measured at baseline, and after 2, 8 and 16 weeks were included in logistic regression analyses to assess associations between inflammatory markers and treatment response.
ResultsPre-treatment levels of IFN-Γ and CCL-2 were significantly higher in escitalopram non-responders compared to responders. Pre-treatment IFN-Γ and CCL-2 levels were significantly associated with a lower of odds of response to escitalopram at 8 weeks. Increases in CCL-2 levels from weeks 8 to 16 in escitalopram non-responders were significantly associated with higher odds of non-response to adjunctive aripiprazole at week 16.
ConclusionsPre-treatment levels of IFN-Γ and CCL-2 were predictive of response to escitalopram. Increasing levels of these pro-inflammatory markers may predict non-response to adjunctive aripiprazole. These findings require validation in independent clinical populations.

Introduction:
This study investigates the dose escalation to dominant intra-prostatic lesions (DILs) that is achievable using single-source-strength (SSS) and dual-source-strength (DSS) low-dose-rate (LDR) prostate brachytherapy and a sector-based plan approach.
Methods:Twenty patients were retrospectively analysed. Image registration and planning were undertaken using VariSeed v9·0. SSS and DSS boost plans were produced and compared to clinical plans. Dosimetric robustness to seed displacement for SSS and DSS plans was compared to clinical plans using Monte Carlo simulations.
Results:Fourteen out of 20 patients had DIL identifiable on magnetic resonance imaging. Median increase in sector D90 of 27% (p < 0·0001) and sector V150 of 31% (p < 0·0001) was achieved with SSS planning without exceeding local rectum and urethra dose constraints. DSS plans achieved dose distributions not statistically significantly different from the SSS plans with a median of eight fewer seeds and two fewer needles. SSS and DSS plan sensitivity to random seed displacement was similar to the clinical plans.
Conclusions:Treatment planning using VariSeed to produce SSS and DSS focal boost plans is feasible for LDR prostate brachytherapy to achieve a median escalation in sector D90 of 27% without exceeding local urethral and rectal constraints. SSS and DSS plan dosimetric robustness was similar to clinical plan dosimetric robustness.

Background
Reward processing has been proposed to underpin the atypical social feature of autism spectrum disorder (ASD). However, previous neuroimaging studies have yielded inconsistent results regarding the specificity of atypicalities for social reward processing in ASD.
AimsUtilising a large sample, we aimed to assess reward processing in response to reward type (social, monetary) and reward phase (anticipation, delivery) in ASD.
MethodFunctional magnetic resonance imaging during social and monetary reward anticipation and delivery was performed in 212 individuals with ASD (7.6–30.6 years of age) and 181 typically developing participants (7.6–30.8 years of age).
ResultsAcross social and monetary reward anticipation, whole-brain analyses showed hypoactivation of the right ventral striatum in participants with ASD compared with typically developing participants. Further, region of interest analysis across both reward types yielded ASD-related hypoactivation in both the left and right ventral striatum. Across delivery of social and monetary reward, hyperactivation of the ventral striatum in individuals with ASD did not survive correction for multiple comparisons. Dimensional analyses of autism and attention-deficit hyperactivity disorder (ADHD) scores were not significant. In categorical analyses, post hoc comparisons showed that ASD effects were most pronounced in participants with ASD without co-occurring ADHD.
ConclusionsOur results do not support current theories linking atypical social interaction in ASD to specific alterations in social reward processing. Instead, they point towards a generalised hypoactivity of ventral striatum in ASD during anticipation of both social and monetary rewards. We suggest this indicates attenuated reward seeking in ASD independent of social content and that elevated ADHD symptoms may attenuate altered reward seeking in ASD.

Previous studies have shown that male mink (Mustela vison) removed from their mothers at seven weeks of age develop more tail-biting than males left with their mothers until six months. Mink in the wild do not damage their own pelts in this way, and such behaviour may well be an indication of chronic stress.
The aim of this experiment was to investigate further the causes of tail-biting by considering female young as well as male, and by lowering the age at which the late-weaned ‘ mink were separated from their mother to 11 weeks, by which age their period of socialization should be complete. This was to generate results of more practical use to farmers, who cannot leave all young with their mothers until six months of age for reasons of space. Mink removed from their mothers at seven or eleven weeks of age did indeed differ in the incidence of tail-biting. Early-weaned ‘females were more likely than late-weaned females to have bitten their tails at six months of age. A similar result was evident as a trend for both sexes at ten months. Furthermore, at this age, some animals ‘ tail tips were completely bald and such animals were all early-weaned.
Where provided with plastic drinker dishes, early-weaned animals were also more likely to chew these. Thus weaning age had long-lasting effects on a number of oral behaviour patterns. These results suggest that young animals predisposed to tail-bite might be diverted by the provision of other objects to chew, and that if problems of over-crowding are avoided, leaving mink kits with their mothers until 11 weeks might improve their welfare.
Humans can identify cats’ affective states from subtle facial expressions

Although cats’ popularity as pets rivals that of dogs, cats are little studied, and people's abilities to read this apparently ‘inscrutable’ species have attracted negligible research. To determine whether people can identify feline emotions from cats’ faces, participants (n = 6,329) each viewed 20 video clips of cats in carefully operationalised positively (n =10) or negatively valenced states (n = 10) (cross-factored with low and high activity levels). Obvious cues (eg open mouths or fully retracted ears) were eliminated. Participants’ average scores were low (11.85/20 correct), but overall above chance; furthermore, 13% of participants were individually significantly successful at identifying the valence of cats’ states (scoring ≥ 15/20 correct). Women were more successful at this task than men, and younger participants more successful than older, as were participants with professional feline (eg veterinary) experience. In contrast, personal contact with cats (eg pet-owning) had little effect. Cats in positive states were most likely to be correctly identified, particularly if active rather than inactive. People can thus infer cats’ affective states from subtle aspects of their facial expressions (although most find this challenging); and some individuals are very good at doing so. Understanding where such abilities come from, and precisely how cats’ expressions change with affective state, could potentially help pet owners, animal care staff and veterinarians optimise feline care and welfare.
Micro-level de-coupling of negative affect and binge eating in relationship to macro-level outcomes in binge eating disorder treatment

Background
While negative affect reliably predicts binge eating, it is unknown how this association may decrease or ‘de-couple’ during treatment for binge eating disorder (BED), whether such change is greater in treatments targeting emotion regulation, or how such change predicts outcome. This study utilized multi-wave ecological momentary assessment (EMA) to assess changes in the momentary association between negative affect and subsequent binge-eating symptoms during Integrative Cognitive Affective Therapy (ICAT-BED) and Cognitive Behavior Therapy Guided Self-Help (CBTgsh). It was predicted that there would be stronger de-coupling effects in ICAT-BED compared to CBTgsh given the focus on emotion regulation skills in ICAT-BED and that greater de-coupling would predict outcomes.
MethodsAdults with BED were randomized to ICAT-BED or CBTgsh and completed 1-week EMA protocols and the Eating Disorder Examination (EDE) at pre-treatment, end-of-treatment, and 6-month follow-up (final N = 78). De-coupling was operationalized as a change in momentary associations between negative affect and binge-eating symptoms from pre-treatment to end-of-treatment.
ResultsThere was a significant de-coupling effect at follow-up but not end-of-treatment, and de-coupling did not differ between ICAT-BED and CBTgsh. Less de-coupling was associated with higher end-of-treatment EDE global scores at end-of-treatment and higher binge frequency at follow-up.
ConclusionsBoth ICAT-BED and CBTgsh were associated with de-coupling of momentary negative affect and binge-eating symptoms, which in turn relate to cognitive and behavioral treatment outcomes. Future research is warranted to identify differential mechanisms of change across ICAT-BED and CBTgsh. Results also highlight the importance of developing momentary interventions to more effectively de-couple negative affect and binge eating.
The Measurement Invariance of Schizotypy in Europe

The short version of the Oxford-Liverpool Inventory of Feelings and Experiences (sO-LIFE) is a widely used measure assessing schizotypy. There is limited information, however, on how sO-LIFE scores compare across different countries. The main goal of the present study is to test the measurement invariance of the sO-LIFE scores in a large sample of non-clinical adolescents and young adults from four European countries (UK, Switzerland, Italy, and Spain). The scores were obtained from validated versions of the sO-LIFE in their respective languages. The sample comprised 4190 participants (M = 20.87 years; SD = 3.71 years). The study of the internal structure, using confirmatory factor analysis, revealed that both three (i.e., positive schizotypy, cognitive disorganisation, and introvertive anhedonia) and four-factor (i.e., positive schizotypy, cognitive disorganisation, introvertive anhedonia, and impulsive nonconformity) models fitted the data moderately well. Multi-group confirmatory factor analysis showed that the three-factor model had partial strong measurement invariance across countries. Eight items were non-invariant across samples. Significant statistical differences in the mean scores of the s-OLIFE were found by country. Reliability scores, estimated with Ordinal alpha ranged from 0.75 to 0.87. Using the Item Response Theory framework, the sO-LIFE provides more accuracy information at the medium and high end of the latent trait. The current results show further evidence in support of the psychometric proprieties of the sO-LIFE, provide new information about the cross-cultural equivalence of schizotypy and support the use of this measure to screen for psychotic-like features and liability to psychosis in general population samples from different European countries.

A small suite of plant macrofossils indicates that southeastern Minnesota supported subarctic to arctic vegetation 18,700 yr B.P. Fossil tepals of Polygonum viviparum are exceptionally well preserved; they occur with more fragmentary remains of Dryas integrifolia, Vaccinium uliginosum var. alpinum, and probable species of arctic Salix, S. cf. herbacea, and S. cf. arctica. The pollen spectrum from this site is dominated by Picea, Pinus, and Cyperaceae, which are typical of midwestern full-glacial sequences. Tundra-like conditions with permafrost were present in southeastern Minnesota during full-glacial time. Local environments 18,700 yr B.P. reconstructed from both physical and paleobotanical evidence include wind-swept ridge tops with thin loess, outcrops of dolostone and sandstone on valley walls, colluvial slopes, sandy to gravelly floodplains, shallow floodplain pools, wet meadows, and peaty turfs, all in a treeless or nearly treeless environment.
A local perspective on community structure in multilayer networks

The analysis of multilayer networks is among the most active areas of network science, and there are several methods to detect dense “communities” of nodes in multilayer networks. One way to define a community is as a set of nodes that trap a diffusion-like dynamical process (usually a random walk) for a long time. In this view, communities are sets of nodes that create bottlenecks to the spreading of a dynamical process on a network. We analyze the local behavior of different random walks on multiplex networks (which are multilayer networks in which different layers correspond to different types of edges) and show that they have very different bottlenecks, which correspond to rather different notions of what it means for a set of nodes to be a good community. This has direct implications for the behavior of community-detection methods that are based on these random walks.
Botulinum Toxin-A use in Paediatric Hypertonia: Canadian Practice Patterns

Background:
This study aims to assess current practices of Canadian physicians providing botulinum toxin-A (BoNT-A) treatments for children with hypertonia and to contrast these with international “best practice” recommendations, in order to identify practice variability and opportunities for knowledge translation.
Methods:Thirteen Canadian physicians assembled to develop and analyze results of a cross-sectional electronic survey, sent to 50 physicians across Canada.
Results:Seventy-eight percent (39/50) of physicians completed the survey. The most frequently identified assessment tools were Gross Motor Function Classification System, Modified Tardieu Scale and neurological examination. Goal-setting tools were infrequently utilized. Common indications for BoNT-A injections and the muscles injected were identified. Significant variability was identified in using BoNT-A for hip displacement associated with hypertonia. The most frequent adverse event reported was localized weakness; 54% reporting this “occasionally“ and 15% “frequently”. Generalized weakness, fatigue, ptosis, diplopia, dysphagia, aspiration, respiratory distress, dysphonia and urinary incontinence were reported rarely or never. For dosage, 52% identified 16 Units/kg body weight of Botox® as maximum. A majority (64%) reported a maximum 400 Units for injection at one time. For localization, electrical stimulation and ultrasound were used infrequently (38% and 19% respectively). Distraction was the most frequently used pain-management technique (64%).
Conclusions:Canadian physicians generally adhere to international best practices when using BoNT-A to treat paediatric hypertonia. Two knowledge-translation opportunities were identified: use of individualized goal setting prior to BoNT-A and enhancing localization techniques. Physicians reported a good safety profile of BoNT-A in children.

Leek moth, Acrolepiopsis assectella, recently became established in the Ottawa Valley, where it significantly damages garlic, leek, and onion (Allium L., Liliaceae) crops. At a threshold of 7 °C, populations in eastern Ontario require 444.6 day-degrees to develop from egg to adult. Pheromone-trap data identify spring, early-summer, and late-summer flight periods of overwintered 1st- and 2nd-generation adults, respectively. Depending on ambient temperatures, the life cycle takes 3–6 weeks in the field, with three generations possible. Management strategies such as application of reduced-risk foliar insecticides and use of row covers require precise timing to target appropriate life-cycle stages. Implementation windows can be determined by incorporating pheromone-trap data and ambient air temperature into a life-cycle development model. A proposed integrated pest management program will involve the use of pesticides, mechanical barriers, and classical biological control.
Review of the species of Trichomalus (Chalcidoidea: Pteromalidae) associated with Ceutorhynchus (Coleoptera: Curculionidae) host species of European origin

Six species of Trichomalus Thomson were reared as parasitoids of Ceutorhynchinae hosts in Europe during surveys in 2000–2004. Trichomalus rusticus (Walker) is treated as a valid species, resurrected from synonymy under T. lucidus (Walker), and T. lyttus (Walker) is transferred from synonymy under T. lucidus and newly placed in synonymy with T. rusticus. Illustrated keys to females and males are given to differentiate the six species (T. bracteatus (Walker), T. campestris (Walker), T. gynetelus (Walker), T. lucidus, T. perfectus (Walker), and T. rusticus) except for males of T. bracteatus and T. gynetelus. A lectotype female is designated for T. rusticus. Trichomalus campestris is newly recorded as a parasitoid of Ceutorhynchus cardariae Korotyaev. Implications of the host-parasitoid associations recovered by the surveys are discussed relative to introduction of species to North America for classical biological control.

The ceutorhynchine weevils Hadroplontus litura (F.) and Microplontus edentulus (Schultze) (Coleoptera: Curculionidae), are established in North America as biological control agents for Canada thistle, Cirsium arvense (L.) Scop., and scentless chamomile, Tripleurospermum perforatum (Mérat) M. Lainz (Asteraceae), respectively. In North America, both weeds occur sympatrically and in similar habitats as another ceutorhynchine, Ceutorhynchus obstrictus (Marsham) (cabbage seedpod weevil), an important pest of canola, Brassica napus L., and Brassica rapa L. (Brassicaceae). Ceutorhynchinae weevils released to control weeds in cultivated crops may serve as alternate hosts if agents released for biological control of C. obstrictus are not specific to that species. Parasitoids associated with M. edentulus and H. litura inflict similar levels of mortality on their hosts, yet a single species was associated with the latter host, whereas 13 species attacked the former. The stem-mining M. edentulus appears to be at some risk but not the root-crown feeding H. litura, should the parasitoids Trichomalus perfectus (Walker) and Mesopolobus morys (Walker) (Hymenoptera: Pteromalidae) be introduced as biological control agents of the silique-feeding C. obstrictus. These findings suggest that feeding niche may be an important criterion for developing a nontarget species test list for host-range testing of potential biological control agents.

In Europe, Ceutorhynchus turbatus Schultze and Ceutorhynchus typhae (Herbst) (Coleoptera: Curculionidae) feed on seeds from hoary cress and shepherd's purse (Cardaria draba (L.) Desv. and Capsella bursa-pastoris (L.) Medik.); both plants are invasive in North America. In North America, C. turbatus is a candidate for biological control of hoary cress, C. typhae is adventive, and both are sympatric with cabbage seedpod weevil (Ceutorhynchus obstrictus (Marsham)), an invasive alien pest of canola (Brassica napus L. and Brassica rapa L., Brassicaceae). We investigated host associations among C. turbatus, C. typhae, and their parasitoids in Europe. Of particular interest was host specificity of Trichomalus perfectus (Walker) and Mesopolobus morys (Walker) (Hymenoptera: Pteromalidae), candidates for biological control of C. obstrictus in North America. We found no evidence that T. perfectus attacks C. turbatus or C. typhae; however, M. morys was the most common parasitoid associated with C. turbatus.
Organic chemistry of NH3 and HCN induced by an atmospheric abnormal glow discharge in N2-CH4 mixtures

The formation of the chemical products produced in an atmospheric glow discharge fed by a N2-CH4 gas mixture has been studied using Fourier Transform InfraRed (FTIR) and Optical Emission Spectrometry (OES). The measurements were carried out in a flowing regime at ambient temperature and pressure with CH4 concentrations ranging from 0.5% to 2%. In the recorded emission spectra the lines of the second positive system CN system and the first negative system of N2 were found to be the most intensive but atomic H$^{\alpha}$, H$^{\beta}$, and C (247 nm) lines were also observed. FTIR-measurements revealed HCN and NH3 to be the major products of the plasma with traces of C2H2. These same molecules have been detected in Titan's atmosphere and the present experiments may provide some novel insights into the chemical and physical mechanisms prevalent in Titan's atmosphere with these smaller species believed to be the precursors of heavier organic species in Titan's atmosphere and on its surface.

Negative ions formed in N2/CH4/Ar discharge – A simulation of Titan's atmosphere chemistry

The formation of negative ions produced in a negative point-to-plane corona discharge fed by a Ar/N2/CH4 gas mixture has been studied using mass spectrometry. The measurements were carried out in flowing regime at ambient temperature and a reduced pressure of 460 mbar. The CN- anion has been found to be the most dominant negative ion in the discharge and is believed to be the precursor of heavier negative ions such as C3N- and C5N-. The most likely pathway for the formation of such molecular anions is H-loss dissociative electron attachment to HCN, H3CN and H5CN formed in the discharge. These same anions have been detected in Titan's atmosphere and the present experiments may provide some novel insights into the chemical and physical mechanisms prevalent in Titan's atmosphere and hence assist in the interpretation of results from the Cassini Huygens space mission.
