Background
The severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) pandemic, with its impact on our way of life, is affecting our experiences and mental health. Notably, individuals with mental disorders have been reported to have a higher risk of contracting SARS-CoV-2. Personality traits could represent an important determinant of preventative health behaviour and, therefore, the risk of contracting the virus.
AimsWe examined overlapping genetic underpinnings between major psychiatric disorders, personality traits and susceptibility to SARS-CoV-2 infection.
MethodLinkage disequilibrium score regression was used to explore the genetic correlations of coronavirus disease 2019 (COVID-19) susceptibility with psychiatric disorders and personality traits based on data from the largest available respective genome-wide association studies (GWAS). In two cohorts (the PsyCourse (n = 1346) and the HeiDE (n = 3266) study), polygenic risk scores were used to analyse if a genetic association between, psychiatric disorders, personality traits and COVID-19 susceptibility exists in individual-level data.
ResultsWe observed no significant genetic correlations of COVID-19 susceptibility with psychiatric disorders. For personality traits, there was a significant genetic correlation for COVID-19 susceptibility with extraversion (P = 1.47 × 10−5; genetic correlation 0.284). Yet, this was not reflected in individual-level data from the PsyCourse and HeiDE studies.
ConclusionsWe identified no significant correlation between genetic risk factors for severe psychiatric disorders and genetic risk for COVID-19 susceptibility. Among the personality traits, extraversion showed evidence for a positive genetic association with COVID-19 susceptibility, in one but not in another setting. Overall, these findings highlight a complex contribution of genetic and non-genetic components in the interaction between COVID-19 susceptibility and personality traits or mental disorders.

Summary
Introduction
In this chapter we discuss predictive modeling of series time obtained by functional magnetic resonance imaging (fMRI), representing an important case of spatiotemporal data. Following its development in the early 1990s, fMRI has become a well established approach to investigating brain activity in vivo (Huettel et al. 2004), providing temporally and spatially resolved recordings of the “blood oxygen level dependent” (BOLD) signal of neural tissue. fMRI time series consist of a temporal sequence of scans of the brain and the surrounding space (discretized into voxels), such that the resulting data sets may be stored as vector time series.
The practical work with fMRI time series poses considerable challenges in many aspects, including the huge dimensionality of the data, which usually is recorded from several 10 of voxels, and the plethora of artifacts and contaminations disturbing the data (Strother 2006). Further difficulties arise from the low temporal sampling frequency, typically well below 1 Hz, and the indirect relationship between the BOLD signal and the underlying neural processes.
Currently available approaches to fMRI time series analysis may be broadly classified into three groups:
• exploratory methods, such as cluster analysis (Goutte et al. 1999), principal component analysis (PCA) (Anderson et al. 1999) and independent component analysis (ICA) (McKeown 2000);
• massively univariate (voxel-wise) regression methods, implemented in software packages such as statistical parametric mapping (SPM) (Friston et al. 1994), the FMRIB software library (FSL) (Smith et al. 2004), or the analysis of functional Neuroimages (AFNI) package (Cox 1996);
• generative dynamic models, based on specific assumptions regarding the properties of the underlying neural masses and the biophysical processes which produce the experimental data; as examples we mention dynamic causal modeling (DCM) (Friston et al. 2003) and the hemodynamic state space model (SSM) of Riera et al. (2004a).
Among these three groups of methods, the third may be interpreted as an example of predictive modeling, while for the second group this is possible only in a very limited sense, and essentially impossible for the first group.

Direct non-medical and indirect costs for families with children with congenital cardiac defects in Germany: a survey from a university centre

Introduction
Parents of children with congenital cardiac disease suffer from psychological stress and financial burdens. These costs have not yet been quantified.
Materials and methodsIn cooperation with paediatricians, social workers, and parents, a questionnaire was devised to calculate direct non-medical and indirect costs. Direct non-medical costs include all costs not directly related to medical services such as transportation. Indirect costs include lost productivity measured in lost income from wages. Parents were retrospectively queried on costs and refunds incurred during the child’s first and sixth year of life. The questionnaire was sent out to 198 families with children born between 1980 and 2000. Costs were adjusted for inflation to the year 2006. Children were stratified into five groups according to the severity of their current health status.
ResultsFifty-four families responded and could be included into the analysis (27.7%). Depending on severity, total direct non-medical and indirect costs in the first year of life ranged between an average of €1654 in children with no or mild (remaining) cardiac defects and an average €2881 in children with clinically significant (residual/remaining) findings. Mean expenses in the sixth year of life were as low as €562 (no or mild (remaining) cardiac defects) and as high as €5213 (potentially life-threatening findings). At both points in time, the highest costs were lost income and transportation; and day care/ babysitting for siblings was third.
DiscussionFamilies of children with congenital cardiac disease and major sequelae face direct non-medical and indirect costs adding up to €3000 per year on average. We should consider compensating families from low socioeconomic backgrounds to minimise under-use of non-medical services of assistance for their children.
Grip force parameters in precision grip of individuals with myelomeningocele

The aim of this study was to characterize impairments of hand function in individuals with myelomeningocele (MMC) using a quantitative method. A grip–lift task was studied in 29 individuals with MMC (17 females, 12 males; mean age 12 years 4 months, SD 5 years 6 months; range 4 to 28 years) and 29 age- and sex-matched control participants. A small object (weight 200g) was grasped and lifted with a precision grip of the dominant hand and grip forces (GF) and time intervals were measured. Sensibility was examined with a two-point discrimination test. In those with MMC, the latency between GF onset of the thumb and index finger and ensuing preload duration were both significantly prolonged (p<0.01). Fingertip forces were excessively high and variable. Several parameters differed significantly between those with MMC and control individuals: GF peak, GF at start of loading, mean GF, and the SDs of the GF during static holding. Although the two-point discrimination task indicated deficits of sensibility, these did not correlate with grip force parameters. Results demonstrate that fine motor skills in patients with MMC are impeded by slowness and inadequate adjustments of manipulative forces.
