Background: Liquid biopsy represents a major development in cancer research, with significant translational potential. Similarly, the integration of multiple molecular platforms has yielded novel insights into disease biology and heterogeneity. We hypothesise that applying contemporary multi-omic approaches to liquid biopsies will improve the power of current models. Methods: We have compiled a cohort of 51 patients with glioblastoma, brain metastasis, and primary CNS lymphoma who underwent CSF sampling as part of clinical care. Cell free methylated DNA and shotgun proteomic profiling was obtained from the CSF of each patient and used to build tumour-specific classifiers. Integrated classifiers were compared with single platform classifiers using multiple approaches. Results: In this study, we show that the DNA methylation and protein profiles of cerebrospinal fluid can be combined to fully discriminate lymphomas from their major diagnostic counterparts with perfect AUCs of 1 (95% confidence interval 1-1) and 100% specificity. Each integrated lymphoma classifier significantly outperforms single-platform classifiers, suggesting synergistic biology is obtained using multiple molecular platforms. Conclusions: We present the most specific and accurate CNS lymphoma classifier to date by integrating the methylome and proteome of CSF. This has important implications for the future of cancer diagnostics and generates immediate utility for patients with CNS lymphoma.
Three-dimensional textural investigation of sulfide mineralisation from the Loolekop carbonatite–phoscorite polyphase intrusion in the Phalaborwa Igneous Complex (South Africa), with implications for ore-forming processes

Copper-sulfides within carbonatites and phoscorites of the Phalaborwa Igneous Complex, South Africa, have been investigated since the middle of the 20th Century. However, aspects of ore formation have remained unclear. This study examines the mechanisms involved in Cu-sulfide mineralisation by micro-focus X-ray computed tomography as applied to sulfide-rich drill core samples. Several texturally distinct assemblages of magmatic sulfides can be identified, including: (1) <500 μm rounded bornite and chalcopyrite grains disseminated within the gangue; (2) elongated mm-scale assemblages of chalcopyrite and bornite; and (3) mm-to-cm thick chalcopyrite cumulates. Chalcopyrite veins were also observed, as well as late-stage valleriite, documenting late-stage fluid circulation within the pipe, and alteration of magmatic and hydrothermal sulfides along fractures within the gangue, respectively. The results of micro-focus X-ray computed tomography indicate that magmatic sulfides are sub-vertically aligned. Spatial variability of the sulfide assemblages suggests that textural changes within sulfide layers reflect fluctuating magma flow rate during emplacement of carbonatite–phoscorite magmas, through coalescence or breakup of sulfide liquid droplets during ascent. Modal sulfide abundances, especially for disseminated assemblages, differ from one carbonatite–phoscorite layer to another, suggesting a strong control of the mechanical sorting in the formation of Cu-sulfide textures within the Loolekop carbonatite. The alternation of carbonatite and phoscorite within the intrusion suggest that the Loolekop Pipe was emplaced through a series of successive magma pulses, which differentiated into carbonatite and phoscorite by melt immiscibility/progressive fractional crystallisation and pressure drop. Three-dimensional textural analysis represents an effective tool for the characterisation of magma flow and is useful for the understanding of magmatic processes controlling sulfide liquid-bearing phoscorite–carbonatite magmas.

The widespread use of herbicides in cropping systems has led to the evolution of resistance in major weeds. The resultant loss of herbicide efficacy is compounded by a lack of new herbicide sites of action, driving demand for alternative weed control technologies. While there are many alternative methods for control, identifying the most appropriate method to pursue for commercial development has been hampered by the inability to compare techniques in a fair and equitable manner. Given that all currently available and alternative weed control methods share an intrinsic energy consumption, the aim of this review was to compare methods based on energy consumption. Energy consumption was compared for chemical, mechanical, and thermal weed control technologies when applied as broadcast (whole-field) and site-specific treatments. Tillage systems, such as flex-tine harrow (4.2 to 5.5 MJ ha−1), sweep cultivator (13 to 14 MJ ha−1), and rotary hoe (12 to 17 MJ ha−1) consumed the least energy of broadcast weed control treatments. Thermal-based approaches, including flaming (1,008 to 4,334 MJ ha−1) and infrared (2,000 to 3,887 MJ ha−1), are more appropriate for use in conservation cropping systems; however, their energy requirements are 100- to 1,000-fold greater than those of tillage treatments. The site-specific application of weed control treatments to control 2-leaf-stage broadleaf weeds at a density of 5 plants m−2 reduced energy consumption of herbicidal, thermal, and mechanical treatments by 97%, 99%, and 97%, respectively. Significantly, this site-specific approach resulted in similar energy requirements for current and alternative technologies (e.g., electrocution [15 to 19 MJ ha−1], laser pyrolysis [15 to 249 MJ ha−1], hoeing [17 MJ ha−1], and herbicides [15 MJ ha−1]). Using similar energy sources, a standardized energy comparison provides an opportunity for estimation of weed control costs, suggesting site-specific weed management is critical in the economically realistic implementation of alternative technologies.

Plant height and lodging resistance can affect rice yield significantly, but these traits have always conflicted in crop cultivation and breeding. The current study aimed to establish a rapid and accurate plant type evaluation mechanism to provide a basis for breeding tall but lodging-resistant super rice varieties. A comprehensive approach integrating plant anatomy and histochemistry was used to investigate variations in flexural strength (a material property, defined as the stress in a material just before it yields in a flexure test) of the rice stem and the lodging index of 15 rice accessions at different growth stages to understand trends in these parameters and the potential factors influencing them. Rice stem anatomical structure was observed and the lignin content the cell wall was determined at different developmental stages. Three rice lodging evaluation models were established using correlation analysis, multivariate regression and artificial radial basis function (RBF) neural network analysis, and the results were compared to identify the most suitable model for predicting optimal rice plant types. Among the three evaluation methods, the mean residual and relative prediction errors were lowest using the RBF network, indicating that it was highly accurate and robust and could be used to establish a mathematical model of the morphological characteristics and lodging resistance of rice to identify optimal varieties.

During the five years of the mission, the Gaia spectrograph, the Radial Velocity Spectrometer (RVS) will repeatedly survey the celestial sphere down to magnitude V ~ 17–18. This talk presents: (i) the system which is currently developed within the Gaia Data Processing and Analysis Consortium (DPAC) to reduce and calibrate the spectra and to derive the radial and rotational velocities, (ii) the RVS expected performances and (iii) scientific returns.

Background
Depression is associated with neural abnormalities in emotional processing.
AimsThis study explored whether these abnormalities underlie risk for depression.
MethodWe compared the neural responses of volunteers who were at high and low-risk for the development of depression (by virtue of high and low neuroticism scores; high-N group and low-N group respectively) during the presentation of fearful and happy faces using functional magnetic resonance imaging (fMRI).
ResultsThe high-N group demonstrated linear increases in response in the right fusiform gyrus and left middle temporal gyrus to expressions of increasing fear, whereas the low-N group demonstrated the opposite effect. The high-N group also displayed greater responses in the right amygdala, cerebellum, left middle frontal and bilateral parietal gyri to medium levels of fearful v. happy expressions.
ConclusionsRisk for depression is associated with enhanced neural responses to fearful facial expressions similar to those observed in acute depression.

Background
Cognitive theories associate depression with negative biases in information processing. Although negatively biased cognitions are well documented in depressed patients and to some extent in recovered patients, it remains unclear whether these abnormalities are present before the first depressive episode.
MethodHigh neuroticism (N) is a well-recognized risk factor for depression. The current study therefore compared different aspects of emotional processing in 33 high-N never-depressed and 32 low-N matched volunteers. Awakening salivary cortisol, which is often elevated in severely depressed patients, was measured to explore the neurobiological substrate of neuroticism.
ResultsHigh-N volunteers showed increased processing of negative and/or decreased processing of positive information in emotional categorization and memory, facial expression recognition and emotion-potentiated startle (EPS), in the absence of global memory or executive deficits. By contrast, there was no evidence for effects of neuroticism on attentional bias (as measured with the dot-probe task), over-general autobiographical memory, or awakening cortisol levels.
ConclusionsThese results suggest that certain negative processing biases precede depression rather than arising as a result of depressive experience per se and as such could in part mediate the vulnerability of high-N subjects to depression. Longitudinal studies are required to confirm that such cognitive vulnerabilities predict subsequent depression in individual subjects.

The classical NDE techniques of periodical maintenance are just now evolving towards the continuous health monitoring of materials and structures. Taking account of the relation of this approach with the bioinspired notion of smart materials, it seems useful to extend this passive concept to a more active one, especially in the case of materials systems as composite materials. Effectively, if sensors and actuators are embedded in a composite structure before curing in order to monitor and improve the processing parameters, they are able, remaining in the structure, to assume the health monitoring and some optimization of the usage in the next stages of the life. Moreover, in slightly damaged systems, it becomes reasonable to use the results collected by the sensors for improvements of safety of materials and structures. Finally in the last stage of the life, when the material is deeply damaged, it is sometimes possible to slow down, and more rarely, to heal the damage.

Twin Differences and Similarities of Birthweight and Term in the French Romulus Population

This study was performed to examine the main characteristics of the French Romulus twin population: zygosity, chorionicity, sex, term and birthweight. A sample of 104 pairs of twins was distinguished by zygosity, chorionicity and sex, and divided into concordant and discordant birthweight groups. Fifty-three % (n = 55) of the fetus pairs studied were born at “ideal term” (35-38 weeks), and 25% (n = 26) were delivered at “preterm” (28-34 weeks). The very preterm (< 28 weeks), and “postterm” (> 38 weeks) represented altogether 22% (n = 23) of the sample. Of the 104 twin pairs, 68% (n = 71) differ by less than 15% in birthweight, and 32% (n = 33) have a birthweight difference higher than 15%. In dizygotic (DZ) pairs females had more tendency to be in the discordant group (p = 0.01) while in monochorionic-monozygotic (MC-MZ) pairs males were more discordant (p = 0.07). We found a significant interaction between sex and zygosity type (p = 0.02). Males had a birthweight difference significantly weaker than that of females in dichorionic-monozygotic (DC-MZ) and DZ twins whereas it was higher than that of females in MC-MZ twins. There were no MZ twin pairs with DC placentation over than 15% birthweight difference. Log linear analysis demonstrated a three-way interaction (p < 0.05) between term type, zygosity type and hypotrophy. Our data indicate that in the group of twins born between 35 and 38 weeks' gestation the crucial question still remains unsettled on how term and birhweight are related to zygotism and/or chorionicity.

Parents of a sample of 76 same sexed pairs of twins aged 3 to 9 months completed a mailed similarity questionnaire. It included the Bonnelykke et al.'s questionnaire and a four anthropological variable scale. To improve each of these two methods, three other combined methods were carried out and results were compared with the biological zygosity diagnosis. The Bonnelykke et al.'s classification combined with anthropological scale (method 4) gave only 1.2% misclassified in the whole sample. It is concluded that zygosity diagnosis using this type of procedure to distinguish MZ and DZ pairs would be important not only for epidemiological study but also for pediatricians and parents.
