Psilocybin in neuropsychiatry: a review of its pharmacology, safety, and efficacy

Psilocybin is a tryptamine alkaloid found in some mushrooms, especially those of the genus Psilocybe. Psilocybin has four metabolites including the pharmacologically active primary metabolite psilocin, which readily enters the systemic circulation. The psychoactive effects of psilocin are believed to arise due to the partial agonist effects at the 5HT2A receptor. Psilocin also binds to various other receptor subtypes although the actions of psilocin at other receptors are not fully explored. Psilocybin administered at doses sufficient to cause hallucinogenic experiences has been trialed for addictive disorders, anxiety and depression. This review investigates studies of psilocybin and psilocin and assesses the potential for use of psilocybin and a treatment agent in neuropsychiatry. The potential for harm is also assessed, which may limit the use of psilocybin as a pharmacotherapy. Careful evaluation of the number needed to harm vs the number needed to treat will ultimately justify the potential clinical use of psilocybin. This field needs a responsible pathway forward.

Summary
Conflicts between humans and bears have occurred since prehistory. Through time, the catalogue of human–bear conflicts (HBC) has been changing depending on the values and needs of human societies and their interactions with bears. Even today, conflict situations vary among the eight species of bears and geographically across these species’ ranges. This results in a broad range of interactions between bears and humans that may be considered as conflicts, including: (1) predation of domestic or semiwild animals, including bees, hunting dogs, and pet animals; (2) damage due to foraging on cultivated berries, fruits, agricultural products, and the tree bark in forest plantations; (3) economic loss due to destruction of beehives, fences, silos, houses, and other human property; (4) bear attacks on humans causing mild or fatal trauma; (5) bluff charges, bear intrusions into residential areas; and (6) vehicle collisions with bears and traffic accidents. In this chapter we aim to outline the principal types of HBC and geographical differences in the occurrence of conflicts and the coexistence between people and bears.
Sexual size dimorphism, spatial segregation and sex-biased bycatch of southern and northern royal albatrosses in pelagic longline fisheries

Bycatch in longline fisheries is a major contributor to the global decline of albatrosses. Sexual segregation at sea often leads to unequal overlap with different fisheries, resulting in sex-biased bycatch, exacerbating the impact on a population level. In great albatrosses (Diomedea spp.), males (the larger sex) tend to spend more time at higher latitudes than females, attributed to competitive exclusion or differences in flight performance mediated by the pronounced sexual size dimorphism (SSD). Consequently, larger numbers of females are bycaught in pelagic longline fisheries in subtropical and temperate areas. Although this has been shown for Diomedea exulans, it has not been confirmed for all great albatross species. Here we examined the degree of SSD and developed discriminant functions to determine species and sex in D. epomophora and D. sanfordi; species that are often killed in several fisheries in the Southern Hemisphere. Based on a large sample of albatrosses bycaught off Uruguay, both species showed substantial SSD. Discriminant functions assigned species and sex to otherwise indeterminate individuals with 90–100% accuracy. Based on all birds identified (n=128), bycatch in the pelagic longline fishery was female-biased, indicating sexual segregation at sea. The discriminant functions presented enable species and sex to be identified, providing critical data for future bycatch assessments.

Satellite cloud imagery is a standard tool of meteorology, and also of ground-based observational astronomy. Fast access to cloud imagery through the Internet now permits more accurate prediction of local cloud cover than was formerly the case. This is particularly useful for automatic and remotely operated telescopes, where an observer may be at a considerable distance from the telescope itself. Since satellite imagery can now be accessed through the Internet with such ease, direct reception of radio transmissions from weather satellites may seem unnecessary. However, advances in computing power and radio receiver technology permit simple automated receipt of satellite transmissions, particularly from the NOAA series of low Earth orbit satellites. This has the advantage of more up-to-date, and higher resolution, cloud imagery than can be obtained from the Internet. We have operated such a satellite receiving station since 2003 April, and have found it particularly useful when remotely operating our telescopes at Siding Spring Observatory (SSO). By automatically publishing our cloud imagery onto the World Wide Web we have established a resource that is widely used by other local observatories and by the general public.

Analysis of sky images obtained from an automated experiment at Dome C, Antarctica, at 2-hourly intervals from February to November 2001 show cloud-free conditions 74% of the time. This augurs well for the prospects of future astronomical observatories at this site.

The study of organic/inorganic hybrid materials has become an increasingly widespread research focus. The advantage derived from such materials is the combining of two very different compounds which may result in something that bridges the performance gap between the two systems. We have previously reported such hybrid systems prepared by incorporating polyhedral oligosilsesquioxanes (POSS™) into traditional organic polymers by the copolymerization of POSS™ monomers and organic monomers. This presentation will discuss a more convenient method of incorporating POSS™ into a polymer: the blending of POSS™ into organic polymers. The research discussion will focus on the development of the POSS™ macromers used in our studies as well as the POSS™ polymer blends synthesized. One important property enhancement observed is the increase in surface hardness for a POSS™/poly styrene sample.

The demand for multi-purpose, high-performance polymer systems has resulted in a need for advancing polymer properties beyond what traditional systems can offer. Only through control/alteration at the molecular level can one maximize property enhancements to meet current military and commercial needs. Over the last seven years the Air Force Research Laboratory has developed and incorporated discrete Si-O frameworks (POSS = Polyhedral Oligomeric Silsesquioxanes) into traditional organic polymer systems. This research has resulted in new hybrid inorganic-organic polymer systems with remarkable enhancements in mechanical and physical properties including dramatic increases in both glass transition and decomposition temperatures, reduced flammability, increased moduli and oxidation resistance. We have shown that these enhancements result from the chemical composition (Si-O core) and size (∼15 A in diameter) of the POSS frameworks, and can be copolymerized, grafted, or even blended using traditional processing methods. Recently, we have focused our efforts on understanding and controlling the molecular level interactions between POSS frameworks and the polymer matrix. The development of new POSS monomers has allowed us to study how functionality, size, and geometry enhances the bulk properties of these hybrid materials. For example, increasing the solubility of side groups on the POSS framework results in greater POSS-polymer matrix interactions and increased Tg and storage moduli. Recent studies with POSS blends have resulted in increases in hardness, and reductions in dielectric constants and coefficients of thermal expansion. New results in polymer synthesis, characterization, and applications will be discussed with a strong emphasis on the versatility of this new nanotechnology to many polymer systems.
