Recombination between F42lac and λplac5 is typically 20- to 50-fold more efficient than recombination between chromosomal lac and λplac5. This enhancement of recombination is recBCD-dependent and requires the expression of genes from the tra regulon of the F factor. Also required is oriT, the origin of F factor conjugational transfer, which must be located in-cis to the cellular copy of lac. In this study we show that enhanced recombination is not supported by an oriT point mutant that reduces oriT function in conjugation. We also present evidence that the activation of oriT for recombination enhancement involves the same strand-specific nick that is required for conjugal DNA transfer. Although it is thought that the role of oriT in recombination enhancement is related to the facilitated entry of RecBCD enzyme into the DNA duplex, we were unable to detect any double-strand breakage at oriT.

Cerium oxide is widely used for chemomechanical polishing (CMP) of silicate glasses. Uses include finishing of optical elements and planarizing dielectrics in the semiconductor industry. This study is designed to investigate the fundamentals of the cerium oxide/silica CMP process by measuring the interaction force between silicate glasses and cerium oxide. Surface forces involved in the polishing of glass by a cerium oxide abrasive can be studied in a controlled manner by measuring sample-tip interactions between a glass substrate and a cerium oxide tip in an atomic force microscope (AFM). Commercially available AFM tips have been coated with thin, uniform films of cerium oxide. By using a square pyramid tip as a template for the shape of the cerium oxide film, challenges related to irregular or blunt tip shape can be overcome. However, complete characterization of structure, shape and chemical composition is required before useful information can be obtained using the AFM.

Grain boundary migration (GBM) during the sintering and densification of a polycrystalline ceramic material occurs as a result of mass transport across an interface. When there is a liquid film present, either due to additives used for liquid-phase sintering or unavoidable impurities in the material, the mass transport can be visualized in terms of dissolving material from one grain and precipitating it on another. in order to study the effects of crystallography on GBM in the presence of a liquid phase, alumina bicrystals have been fabricated with anorthite (CaA12Si208) glass films at the interface. The alumina-anorthite system in a bicrystal geometry is used because the pseudo-binary phase diagram of the system is well known, the bicrystal geometry allows for control over the original interface misorientation, and the glassy phase of anorthite is the most commonly occurring glass in commercially used alumina.
Fig. 1 is a secondary-electron SEM image of an alumina bicrystal recorded using a field-emission SEM (Hitachi S900) operating at 5 kV.

The sintering process of ceramics involves grain-boundary migration (GBM) that is accompanied by mass transport across an interface. In this study, electron backscatter diffraction (EBSD) has been used to examine grain-boundary migration in alumina bicrystals with liquid films at the interface. EBSD patterns, taken near the sintered interface, have been used to study the effects of crystallography on GBM and to study the orientation relationships within the migrated regions of the crystal. Results indicate that the direction of migration is not always the same as that predicted by the current theories on GBM. It was also found that there may be small-angle misorientations in the migrated regions.
Effects of Electrode Spacing on Reactive Ion Etching of 4H-SiC

4H-SiC was selectively etched in a Reactive Ion Etch (RIE) system using a nickel mask. The power, pressure, and electrode spacing were varied within a RF generated SF6:O2 (1:2) plasma. Peak etch rates of up to 2600 Aring;/min. were obtained at a pressure of 350 mT, power of 90 W (2 W/cm2), and electrode spacing of 3.180 cm. Etches were all residue-free, although power levels above 60 W (1.36 W/cm2) resulted in the SiC surface being roughened, which limited smooth surface etch capability to 2000 Aring;/min. When comparing electrode spacing from 3.180 cm to 1.270 cm, the 3.180 cm spacing was found to have the highest etch rate at pressures ranging from 250 mT to 500 mT.
