The classical implementation for putting quantitative figures on maps to reveal elemental compositions in transmission electron microscopy is by analytical methods like X-ray and energy-loss spectroscopy. Typically, the technique in use often depends on whether lighter or heavier elements are present and—more practically—which calibrations are available or sample-related properties are known. A framework linking electron energy-loss spectroscopy (EELS) and energy-dispersive X-ray (EDX) signals such that absolute volumetric concentrations can be derived without assumptions made a priori about the unknown sample, is largely missing. In order to combine both techniques and harness their respective potentials for a light and heavy element analysis, we have set up a powerful hardware configuration and implemented an experimental approach, which reduces the need for estimates on many parameters needed for quantitative work such as densities, absolute thicknesses, theoretical ionization cross-sections, etc. Calibrations on specimens with known geometry allow the measurement of inelastic mean free paths. As a consequence, mass-thicknesses obtained from the EDX ζ-factor approach can be broken up and quantities like concentrations and partial energy-differential ionization cross-sections become accessible. ζ-factors can then be used for conversion into EELS cross-sections that are hard to determine otherwise, or conversely, connecting EDXS and EELS in a quantitative manner quite effectively.

Summary
According to the hierarchical model of love, Lee's (1973) love styles may be subdivided into passionate and companionate love. From the perspective of life-span theory of control that differentiates selective and compensatory control, new insights on the development of love styles are derived. The goal of partnership formation depends on age-graded opportunity structures. Age-normative factors influence the chances to fulfill tasks with respect to the initiation and maintenance of a partnership. Passionate love is more easily realized in early than in late adulthood. In later adulthood, disengagement from goals related to passionate love and increased emphasis on companionate love are expected. A review of empirical findings tends to support these assumptions.
Hardly anywhere is the rivalry between lay insights and scientific knowledge as strong as in the area of romantic and marital relationships. We all know or expect to know from our everyday experience what this kind of relationship means. We have our personal insights and implicit theories about what characterizes a successful marital relationship and about the conditions under which a romantic relationship is formed. For example, people tend to equate marital relationship with romantic love. In addition, they assume that romantic love emerges immediately after the first contact has been established (love at first sight; Averill, 1985).
Other implicit assumptions refer to the development of romantic and marital relationships across time. From self-observation we are convinced that love changes over time. We feel that a personal relationship has developed somewhat across, say, five or ten years.

Human RNase P recognizes a small model substrate consisting of only the 5′ leader sequence, aminoacyl acceptor stem, and T stem and loop of a tRNA precursor. It was demonstrated here that a bimolecular construct in which the T loop is opened between G57 and A58 (tRNA numbering system) is still processed by RNase P. The strand that is cleaved can be considered the target RNA, whereas the other strand serves as an external guide sequence (EGS). The nucleotides corresponding to nt 58–60 in the T loop could be deleted without affecting cleavage of the substrate. Thus, the complete T loop can be replaced by the single-stranded sequence UUCG or UUCA (nt 55–57 in the T loop). The four nucleotides UUCR possibly form a structure that resembles the uridine turn in the T loop of tRNA. Because recognition by RNase P is independent of the helical sequence, this motif can be used for targeting RNA molecules for EGS-directed cleavage by human RNase P. Chemically modified EGSs with 2′-O-methyl groups also showed activity in inducing RNase P cleavage. Several 13-mer EGSs targeted to the 2.1-kb surface antigen mRNA of hepatitis B virus (HBV) were designed and tested using a co-transcriptional cleavage assay with a 2.1-kb HBV transcript. Some of the new EGSs were capable of inducing cleavage of the HBV RNA by RNase P.
