Genetic liability to suicidal thoughts and behaviors and risk of suicide attempt in US military veterans: moderating effects of cumulative trauma burden

Background
Little is known about environmental factors that may influence associations between genetic liability to suicidality and suicidal behavior.
MethodsThis study examined whether a suicidality polygenic risk score (PRS) derived from a large genome-wide association study (N = 122,935) was associated with suicide attempts in a population-based sample of European-American US military veterans (N = 1664; 92.5% male), and whether cumulative lifetime trauma exposure moderated this association.
ResultsEighty-five veterans (weighted 6.3%) reported a history of suicide attempt. After adjusting for sociodemographic and psychiatric characteristics, suicidality PRS was associated with lifetime suicide attempt (odds ratio 2.65; 95% CI 1.37–5.11). A significant suicidality PRS-by-trauma exposure interaction emerged, such that veterans with higher levels of suicidality PRS and greater trauma burden had the highest probability of lifetime suicide attempt (16.6%), whereas the probability of attempts was substantially lower among those with high suicidality PRS and low trauma exposure (1.4%). The PRS-by-trauma interaction effect was enriched for genes implicated in cellular and developmental processes, and nervous system development, with variants annotated to the DAB2 and SPNS2 genes, which are implicated in inflammatory processes. Drug repurposing analyses revealed upregulation of suicide gene-sets in the context of medrysone, a drug targeting chronic inflammation, and clofibrate, a triacylglyceride level lowering agent.
ConclusionResults suggest that genetic liability to suicidality is associated with increased risk of suicide attempt among veterans, particularly in the presence of high levels of cumulative trauma exposure. Additional research is warranted to investigate whether incorporation of genomic information may improve suicide prediction models.

Three Modes of Metal-Enriched Star Formation at High Redshift

It is generally accepted that the very first stars in the universe were significantly more massive and formed much more in isolation than stars observed today. This suggests that there was a transition in star formation modes that was most likely related to the metallicity of the star-forming environment. We study how the addition of heavy elements alters the dynamics of collapsing gas by performing a series of numerical simulations of primordial star formation with various levels of pre-enrichment, using the adaptive mesh refinement, hydrodynamic + N-body code, Enzo. At high redshifts, the process of star formation is heavily influenced by the cosmic microwave background (CMB), which creates a temperature floor for the gas. Our results show that cloud-collapse can follow three distinct paths, depending on the metallicity. For very low metallicities (log10(Z/Z⊙) < −3.5), star formation proceeds in the primordial mode, producing only massive, singular objects. For high metallicities (log10(Z/Z⊙) > −3), efficient cooling from the metals cools the gas to the CMB temperature when the core density is still very low. When the gas temperature reaches the CMB temperature, the core becomes very thermally stable, and further fragmentation is heavily suppressed. In our simulations with log10(Z/Z⊙) > −3, only a single object forms with a mass-scale of a few hundred M⊙. We refer to this as the CMB-regulated star formation mode. For metallicities between these two limits (−3.5 < log10(Z/Z⊙) < −3), the gas cools efficiently, but never reaches the CMB temperature. In this mode, termed the metallicity-regulated star formation mode, the minimum gas temperature is reached at much higher densities, allowing the core to fragment and form multiple objects with mass-scales of only a few M⊙. Our results imply that the stellar initial mass function was top-heavy at very high redshift due to stars forming in the CMB-regulated mode. As the CMB temperature lowers with time, the metallicity-regulated star formation mode (producing multiple low-mass stars) operates at higher metallicities and eventually becomes the sole mode of star formation.

For more than a century, archaeologists have frequently been drawn to understand the human past in broadly evolutionary terms, applying Darwinian thinking to the development of human societies. The unilinear models of human development that often result typically regard the state as the culmination of human progress, the end-point of a journey through intervening stages of bands, tribes and chiefdoms. Neo-evolutionary thinking was especially prevalent from the 1940s onwards, in the work of Julian Steward and others writing on the origins of the state. In the volume reviewed, Norman Yoffee challenges the former dominance of the neo-evolutionary approach, arguing that over the past half century it has stifled rather than stimulated our understanding of early state development.
Yoffee contests the idea that states develop through a series of programmatic stages from less complex kinds of society. Instead, he stresses the diversity of the archaic state, drawing heavily on his specialist knowledge (drawn from texts as well as archaeology) of early Mesopotamia. Here we see city-state societies in which heterarchies play a role alongside hierarchies, and in which the varieties of lived experience varied considerably from place to place, even though all may at some level be considered to have been part of a shared Mesopotamian civilization.
Yoffee's book is not, however, concerned solely with Mesopotamia; far from it, he draws comparative evidence from Egypt, South and East Asia and Central and South America to demonstrate the diversity and fluidity of the entities he is describing. Few of them conform to models that might be drawn from ethnography, and each state may in many ways be considered unique. Yet in a broader perspective, all states arise through a widespread pattern of change that has taken place in human society since the end of the Pleistocene in which individuals and groups have competed for control of resources.
Yoffee concludes that ‘The central myth about the study of the earliest states ... is that there was something that could be called the archaic state, and that all of the earliest states were simply variations on this model’. The methodological alternative is to consider each society (of whatever type) as individual and unique, and constantly in a state of flux. In this review feature we invite a series of archaeologists specializing in the study of early states to address this and other issues raised by this important book. We begin, however, with an opening statement from the author himself.

1. The milk and blood iodine of a herd of thirty Guernsey and thirty Holstein cows has been determined, previous to and during prolonged increased iodine feeding to one-half of the herd. The increased amount of iodine was fed as potassium iodide which was mixed uniformly with a grain feed.
2. The blood iodine of the iodized cows was greatly and uniformly increased.
3. The milk iodine was increased. This increase was principally dependent on the amount of iodine which was fed. During late spring, however, the milk iodine from both the iodized and control cows was unusually low. During early autumn the milk iodine from the iodized cows was low.
4. The milk from the iodized cows contained 7–26 times as much iodine as that from the control cows. An average milk iodine of 80μg. % was obtained from the iodized cows throughout a mid-year period of 5 months.
5. This experiment reveals that a milk of increased iodine content can be readily produced by increased iodine feeding.
