The first detection of deuterated water toward extragalactic hot cores with ALMA

We discuss the first detection of deuterated water (HDO) in extragalactic hot cores. The HDO 211–212 line has been detected with the Atacama Large Millimeter/submillimeter Array (ALMA) toward hot cores N 105–2 A and 2 B in the N 105 star-forming region in the low-metallicity Large Magellanic Cloud (LMC), the nearest star-forming galaxy. We compared the HDO line luminosity (LHDO) measured toward two hot cores in N 105 to those observed toward a sample of 17 Galactic hot cores and found that the observed values of LHDO for the LMC hot cores fit very well into the LHDO trends with Lbol and metallicity observed toward the Galactic hot cores. Our results indicate that LHDO seems to be largely dependent on the source luminosity, but metallicity also plays a role. We provide a rough estimate of the H2O column density and abundance ranges toward N 105–2 A and 2 B by assuming that HDO/H2O toward the LMC hot cores is the same as that observed in the Milky Way; the obtained values are systematically lower than those measured in the Galactic hot cores. The spatial distribution and velocity structure of the HDO emission in N 105–2 A is consistent with HDO being the product of the low-temperature dust grain chemistry.
Mid-IR cosmological spectrophotometric surveys from space: Measuring AGN and star formation at the cosmic noon with a SPICA-like mission

We use the SPace Infrared telescope for Cosmology and Astrophysics (SPICA) project as a template to demonstrate how deep spectrophotometric surveys covering large cosmological volumes over extended fields (1– $15\, \rm{deg^2}$ ) with a mid-IR imaging spectrometer (17– $36\, \rm{\rm{\upmu m}}$ ) in conjunction with deep $70\, \rm{\rm{\upmu m}}$ photometry with a far-IR camera, at wavelengths which are not affected by dust extinction can answer the most crucial questions in current galaxy evolution studies. A SPICA-like mission will be able for the first time to provide an unobscured three-dimensional (3D, i.e. x, y, and redshift z) view of galaxy evolution back to an age of the universe of less than $\sim$ 2 Gyrs, in the mid-IR rest frame. This survey strategy will produce a full census of the Star Formation Rate (SFR) in the universe, using polycyclic aromatic hydrocarbons (PAH) bands and fine-structure ionic lines, reaching the characteristic knee of the galaxy luminosity function, where the bulk of the population is distributed, at any redshift up to $z \sim 3.5$ . Deep follow-up pointed spectroscopic observations with grating spectrometers onboard the satellite, across the full IR spectral range (17– $210\, \rm{\rm{\upmu m}}$ ), would simultaneously measure Black Hole Accretion Rate (BHAR), from high-ionisation fine-structure lines, and SFR, from PAH and low- to mid-ionisation lines in thousands of galaxies from solar to low metallicities, down to the knee of their luminosity functions. The analysis of the resulting atlas of IR spectra will reveal the physical processes at play in evolving galaxies across cosmic time, especially its heavily dust-embedded phase during the activity peak at the cosmic noon ( $z \sim 1$ –3), through IR emission lines and features that are insensitive to the dust obscuration.

The conclusion of Zaffiro et al. (2019; Constraints on the Equations of State of stiff anisotropic minerals: rutile, and the implications for rutile elastic barometry. Mineralogical Magazine, 83, 339–347) that the Mie–Grüneisen–Debye (MGD) Equation of State (EoS) cannot fit the available data for rutile is shown to be incorrect, even though rutile exhibits significant anisotropic thermal pressure which invalidates the quasi-harmonic approximation used as the basis for the MGD EoS. The refined parameters for the MGD EoS of rutile are: KTR0= 205.05(25) GPa, $K_{TR0}^{\prime} $ = 7.2(5), θD = 399(20) K, γ0= 1.40(2) and q = 1.5(7). This EoS predicts volumes, bulk moduli and volume thermal expansion coefficients for rutile at metamorphic conditions that are statistically indistinguishable from those predicted by the ‘isothermal’ type of EoS reported previously.

Astrophysics Telescope for Large Area Spectroscopy Probe is a concept for a National Aeronautics and Space Administration probe-class space mission that will achieve ground-breaking science in the fields of galaxy evolution, cosmology, Milky Way, and the Solar System. It is the follow-up space mission to Wide Field Infrared Survey Telescope (WFIRST), boosting its scientific return by obtaining deep 1–4 μm slit spectroscopy for ∼70% of all galaxies imaged by the ∼2 000 deg2 WFIRST High Latitude Survey at z > 0.5. Astrophysics Telescope for Large Area Spectroscopy will measure accurate and precise redshifts for ∼200 M galaxies out to z < 7, and deliver spectra that enable a wide range of diagnostic studies of the physical properties of galaxies over most of cosmic history. Astrophysics Telescope for Large Area Spectroscopy Probe and WFIRST together will produce a 3D map of the Universe over 2 000 deg2, the definitive data sets for studying galaxy evolution, probing dark matter, dark energy and modifications of General Relativity, and quantifying the 3D structure and stellar content of the Milky Way. Astrophysics Telescope for Large Area Spectroscopy Probe science spans four broad categories: (1) Revolutionising galaxy evolution studies by tracing the relation between galaxies and dark matter from galaxy groups to cosmic voids and filaments, from the epoch of reionisation through the peak era of galaxy assembly; (2) Opening a new window into the dark Universe by weighing the dark matter filaments using 3D weak lensing with spectroscopic redshifts, and obtaining definitive measurements of dark energy and modification of General Relativity using galaxy clustering; (3) Probing the Milky Way’s dust-enshrouded regions, reaching the far side of our Galaxy; and (4) Exploring the formation history of the outer Solar System by characterising Kuiper Belt Objects. Astrophysics Telescope for Large Area Spectroscopy Probe is a 1.5 m telescope with a field of view of 0.4 deg2, and uses digital micro-mirror devices as slit selectors. It has a spectroscopic resolution of R = 1 000, and a wavelength range of 1–4 μm. The lack of slit spectroscopy from space over a wide field of view is the obvious gap in current and planned future space missions; Astrophysics Telescope for Large Area Spectroscopy fills this big gap with an unprecedented spectroscopic capability based on digital micro-mirror devices (with an estimated spectroscopic multiplex factor greater than 5 000). Astrophysics Telescope for Large Area Spectroscopy is designed to fit within the National Aeronautics and Space Administration probe-class space mission cost envelope; it has a single instrument, a telescope aperture that allows for a lighter launch vehicle, and mature technology (we have identified a path for digital micro-mirror devices to reach Technology Readiness Level 6 within 2 yr). Astrophysics Telescope for Large Area Spectroscopy Probe will lead to transformative science over the entire range of astrophysics: from galaxy evolution to the dark Universe, from Solar System objects to the dusty regions of the Milky Way.
2074: Comparing the properties of human umbilical cord-derived mesenchymal stromal cells from preterm Versus full-term infants

OBJECTIVES/SPECIFIC AIMS: To compare functional differences in WJ-MSCs-derived from term Versus preterm infants. METHODS/STUDY POPULATION: WJ-MSCs were enzymatically digested from umbilical cord tissue from Term (gestational age ≥37 wk, n=4) and Preterm (gestational age ≤32 wk, n=5) neonates. Cells were characterized by (1) surface antigen markers using flow cytometry, (2) ability to differentiate into adipogenic, chondrogenic, and osteogenic lineages following in vitro stimulation, (3) colony forming unit efficiency, (4) proliferation rates, and (5) cell motility assay. RESULTS/ANTICIPATED RESULTS: WJ-MSCs were successfully isolated from both Preterm and Term groups. Cells adhered to plastic and displayed characteristic spindle-shaped morphology when cultured under standard conditions. WJ-MSCs from both groups expressed surface antigen markers CD73, CD90, and CD146 (≥90%) and did not express hematopoietic markers HLA-DR, CD79, or CD117 (<5%). Preterm and Term cells were capable of differentiating into osteogenic, chondrogenic, and adipogenic lineages. There were no significant differences between the groups when evaluated by colony forming efficiency, proliferation rates, or cell motility. DISCUSSION/SIGNIFICANCE OF IMPACT: These preliminary findings suggest that WJ-MSCs derived from full-term or preterm neonates have similar functional characteristics. Future studies will focus on the regenerative potential of WJ-MSCs from preterm and term infants following changes in the microenvironment (eg, oxygen tension, inflammatory stimulation).

OBJECTIVES/SPECIFIC AIMS: To assess the efficacy of exogenous administration of MSCs in animal models of HIE. METHODS/STUDY POPULATION: Adhering to the Systematic Review Protocol for Animal Intervention Studies, a systematic search of English articles was performed using MEDLINE, Web of Science, CINAHL, and Google Scholar. Search term items included mesenchymal stem/stromal cell, hypoxic ischemic encephalopathy, asphyxia, cerebral ischemia, and neonatology. We selected randomized and nonrandomized studies that examined in vivo neonatal models of induced HIE. We excluded studies that combined MSCs with an adjunct therapy. Data were collected on study specifics, MSC characteristics, and outcome measurements. The primary outcome was efficacy of MSC treatment, assessed by functional neurologic measures (cognitive, motor, sensory). Risk of bias was assessed using the SYRCLE’s risk of bias tool and we used the ARRIVE guidelines to describe the quality of study reporting. RESULTS/ANTICIPATED RESULTS: A total of 17 preclinical publications focusing on MSC therapy for HIE met our inclusion criteria. Fifteen of the studies (88%) induced HIE in rodents by ligating the common carotid artery followed by a period of hypoxic exposure. Nine (53%) studies derived their MSCs from rodent bone marrow, whereas the other investigators provided xenografts from human bone marrow or umbilical cord-derived MSCs. Range of MSC dose was between 0.25 and 3.5× 106 cells with 71% of the experiments transplanting the MSCs intranasally or intracerebral. Three studies (18%) administered multiple doses. The cylinder rearing test was the most common (73%) sensorimotor functional outcome performed in the first month following the establishment of HIE. All studies demonstrated a reduction in asymmetrical paw preference after receiving MSC therapy. Lesional size was assessed, using neuroimaging or histologic evaluations, and the majority of studies showed a decreased insult following MSC therapy. DISCUSSION/SIGNIFICANCE OF IMPACT: MSC treatment demonstrates improved functional and structural outcomes that are encouraging for future translational studies.

OBJECTIVE
Surgical site infections (SSIs) are responsible for significant morbidity and mortality. Preadmission skin antisepsis, while controversial, has gained acceptance as a strategy for reducing the risk of SSI. In this study, we analyze the benefit of an electronic alert system for enhancing compliance to preadmission application of 2% chlorhexidine gluconate (CHG).
DESIGN, SETTING, AND PARTICIPANTSFollowing informed consent, 100 healthy volunteers in an academic, tertiary care medical center were randomized to 5 chlorhexidine gluconate (CHG) skin application groups: 1, 2, 3, 4, or 5 consecutive applications. Participants were further randomized into 2 subgroups: with or without electronic alert. Skin surface concentrations of CHG (μg/mL) were analyzed using a colorimetric assay at 5 separate anatomic sites.
INTERVENTIONPreadmission application of chlorhexidine gluconate, 2%
RESULTSMean composite skin surface CHG concentrations in volunteer participants receiving EA following 1, 2, 3, 4, and 5 applications were 1,040.5, 1,334.4, 1,278.2, 1,643.9, and 1,803.1 µg/mL, respectively, while composite skin surface concentrations in the no-EA group were 913.8, 1,240.0, 1,249.8, 1,194.4, and 1,364.2 µg/mL, respectively (ANOVA, P<.001). Composite ratios (CHG concentration/minimum inhibitory concentration required to inhibit the growth of 90% of organisms [MIC90]) for 1, 2, 3, 4, or 5 applications using the 2% CHG cloth were 208.1, 266.8, 255.6, 328.8, and 360.6, respectively, representing CHG skin concentrations effective against staphylococcal surgical pathogens. The use of an electronic alert system resulted in significant increase in skin concentrations of CHG in the 4- and 5-application groups (P<.04 and P<.007, respectively).
CONCLUSIONThe findings of this study suggest an evidence-based standardized process that includes use of an Internet-based electronic alert system to improve patient compliance while maximizing skin surface concentrations effective against MRSA and other staphylococcal surgical pathogens.

Putting logic-based distributed systems on stable grounds

In the Declarative Networking paradigm, Datalog-like languages are used to express distributed computations. Whereas recently formal operational semantics for these languages have been developed, a corresponding declarative semantics has been lacking so far. The challenge is to capture precisely the amount of nondeterminism that is inherent to distributed computations due to concurrency, networking delays, and asynchronous communication. This paper shows how a declarative, model-based semantics can be obtained by simply using the well-known stable model semantics for Datalog with negation. We show that the model-based semantics matches previously proposed formal operational semantics.
