A 19F Nuclear Magnetic Resonance Study of Natural Clays

A series of natural clays, including 1:1 layer silicates (serpentines, kaolin minerals), smectites, vermiculite, micas, talc, pyrophyllite, sepiolite, and palygorskite, were studied by 19F magic-angle spinning (MAS) nuclear magnetic resonance (NMR) spectroscopy. The 19F chemical shift in these layer silicates is characteristic of the structure, in particular, to the local octahedral cation occupancy. Fluoride ions bonded to three Mg octahedral cations have a chemical shift of about −177 ppm and those bonded to two Al cations and a vacancy have a chemical shift of about −134 parts per million (ppm). The shift at −182.8 ppm in hectorite is apparently associated with fluoride bonded to two Al cations and a Li cation. Surprisingly, the difference in chemical shift of the interlayer and inner fluoride in 1:1 layer silicates is insufficient to distinguish these sites. Based on trends in chemical shift, it appears that fluoride substitution for inner hydroxyls in clays with octahedral substitution is not random. Fluoride is apparently preferentially associated with Mg rather than Al in the octahedral sheet as no resonance due to a fluoride bonded to two Al cations and a vacancy is observed in clays such as SAz-1.

Two independent temporal-spatial clusters of hospital-onset Rhizopus infections were evaluated using whole-genome sequencing (WGS). Phylogenetic analysis confirmed that isolates within each cluster were unrelated despite epidemiological suspicion of outbreaks. The ITS1 region alone was insufficient for accurate analysis. WGS has utility for rapid rule-out of suspected nosocomial Rhizopus outbreaks.
A Model-Informed Drug Development Approach Supporting the Approval of a New Valbenazine Dose for Tardive Dyskinesia

Introduction
Tardive dyskinesia (TD) is a persistent and potentially disabling movement disorder associated with dopamine receptor blocking agents (eg, antipsychotics). Valbenazine is a highly selective vesicular monoamine transporter 2 inhibitor with several safe and effective dosing options now approved for once-daily (QD) treatment of TD in adults. Valbenazine 80 mg QD is the recommended dose for TD; however, 40 or 60 mg QD (newly approved dose) may be considered depending on response and tolerability. The recent approval of valbenazine 60 mg was based on results from an analysis that used the FDA’s model-informed drug development (MIDD) approach and leveraged existing data from the 6-week, phase 3 registration trial of valbenazine (KINECT 3).
MethodsA population pharmacokinetic (popPK) model was developed to describe plasma concentration-time profiles for valbenazine and its primary active metabolite, [+]-α-dihydrotetrabenazine ([+]-α-HTBZ). An exposure-response (E-R) model was developed using the area under the concentration-time curve (AUC) of [+]-α-HTBZ (exposure) and change from baseline in the Abnormal Involuntary Movement Scale total score (AIMS-CFB) (response). Stepwise E-R model development evaluated various linear and nonlinear models to describe AIMS-CFB vs [+]-α-HTBZ AUC and time. E-R relationships established with the 40 and 80 mg data were used to predict AIMS-CFB for a 60 mg dose up to week 6, accounting for study-to-study, inter-individual, and residual variabilities.
ResultsSteady-state valbenazine and [+]-α-HTBZ concentrations were well described by a joint parent-metabolite popPK model. An Emax model with asymptotic exponential delay in the maximal valbenazine effect adequately characterized the E-R relationship between AIMS-CFB and [+]-α-HTBZ AUC. The simulated confidence intervals of response were consistent with the observed KINECT 3 results, demonstrating the utility of the model to predict efficacy results. The established E-R model was subsequently used to predict AIMS-CFB for valbenazine 60 mg QD at week 6. Mean AIMS scores decreased (improved) in a dose-dependent manner, with 60 mg QD predicted to result in least-squares mean (SEM) AIMS-CFB of −2.7 0.4, which is between the reported AIMS-CFB for 40 mg (−1.9 ± 0.4) and 80 mg (−3.2 ± 0.4). All simulated trials demonstrated valbenazine 60 mg to be significantly superior to placebo in AIMS-CFB after 6 weeks of treatment.
ConclusionsThis analysis integrated and leveraged data from two previously approved valbenazine doses (40 and 80 mg QD) using an MIDD approach. The results provided key evidence that an intermediate dose (newly approved 60 mg QD) could be considered therapeutically beneficial without the need for an additional clinical trial. The availability of a valbenazine 60 mg dose to complement the previously approved doses fills an existing medical need for patients with TD who could benefit from this third effective dose.

Introduction

At the heart of this chapter is a recorded conversation between four people working at the interface between art and research. This Introduction, along with the Conclusion, is designed to situate the conversation within the wider context of arts practice and co-produced research.
Contemporary art practice has experienced a paradigm shift from the idea of art as sitting within the gallery to a more diffuse concept of art as collaborative and participatory (Bishop, 2012). These approaches involve people building knowledge together, and rely on a de-centring or de-materialising of the art object to suggest something new. This chapter sits within a tradition of dialogical approaches to knowledge production and draws on the tradition within contemporary art practice of the conversation as a form of engagement (Kester, 2004, 2011). Kester (2011, p 67) writes about such art projects:
What is at stake in these projects is not dialogue per se but the extent to which the artist is able to catalyze emancipatory insights through dialogue. This requires an acute understanding of the many ways in which these insights can be constrained and compromised.
The conversation that follows is between an artist (Steve Pool), a chief executive of a museum service (Kim Streets), an arts educator (Natalie Walton) and a researcher (David Bell). All worked on the ‘cultural’ part of the Imagine project. This dialogue reflects their concerns about how they respond as individuals to a co-produced project, Imagine, with its focus on imagining better futures.
As a conversation, it explores the dialogic form (Banks and Armstrong et al, 2014). This enables the authorship to sit with more than one person, and be composed of multiple viewpoints. This situates a community development approach to the co-production of research within a conversation that lets in multiple viewpoints and points of difference (Mouffe, 2007). The discussion is personal but also political. Reading it provides a different kind of lens. The conversation reflects the lived reality of the project, the aspirations and pragmatic decisions, and it makes visible the problems and opportunities of bringing people together from different backgrounds, histories and expectations to try and build something together.

Summary
Providing genome-informed personalized treatment is an important goal of modern medicine. Identifying new translational targets in nucleic acid characterizations is an important step toward that goal. The information tsunami produced by such genome-scale investigations is stimulating parallel developments in statistical methodology and inference, analytical frameworks, and computational tools. Within the context of genomic medicine and with a strong focus on cancer research, this book describes the integration of high-throughput bioinformatics data from multiple platforms to inform our understanding of the functional consequences of genomic alterations. This includes rigorous and scalable methods for simultaneously handling diverse data types such as gene expression array, miRNA, copy number, methylation, and next-generation sequencing data. This book is intended for statisticians who are interested in modeling and analyzing high-throughput data. It covers the development and application of rigorous statistical methods (Bayesian and non-Bayesian) in the analysis of high-throughput bioinformatics data that arise from problems in medical and cancer research and molecular and structural biology. The specific focus of the volume is to provide an overview of the current state of the art of methods to integrate novel high-throughput multiplatform bioinformatics data, for a better understanding of the functional consequences of genomic alterations. The introductory description of biological and technical principles behind multiplatform high-throughput experimentation may be helpful to statisticians who are new to this research area.
