Summary
This chapter reviews decision-making in insanity defense trials. The chapter begins with an overview of the variety of legal definitions of insanity in the United States, discussing how these rules provide parameters and shape (or fail to shape) insanity decisions. Various factors related to decision-making in insanity defense cases are discussed, including attitudes toward the insanity defense itself (and how these reflect myths about the insanity defense and its implications), prototypes of insanity, and individual differences of both jurors and defendants. The chapter examines misconceptions of mental disorder and how these might relate to decision-making in these cases and considers the role of decision-makers’ perceptions of punishment in this context. The chapter also reflects on the role of intersecting identities in insanity judgments, provides an overview and synthesis of the current body of research on legal decision-making in insanity cases, discusses limitations to the current literature, provides future directions, and considers legal and policy implications.

Epistasis is an important feature of the genetic architecture of quantitative traits, but the dynamics of epistatic interactions in natural populations and the relationship between epistasis and pleiotropy remain poorly understood. Here, we studied the effects of epistatic modifiers that segregate in a wild-derived Drosophila melanogaster population on the mutational effects of P-element insertions in Semaphorin-5C (Sema-5c) and Calreticulin (Crc), pleiotropic genes that affect olfactory behaviour and startle behaviour and, in the case of Crc, sleep phenotypes. We introduced Canton-S B (CSB) third chromosomes with or without a P-element insertion at the Crc or Sema-5c locus in multiple wild-derived inbred lines of the Drosophila melanogaster Genetic Reference Panel (DGRP) and assessed the effects of epistasis on the olfactory response to benzaldehyde and, for Crc, also on sleep. In each case, we found substantial epistasis and significant variation in the magnitude of epistasis. The predominant direction of epistatic effects was to suppress the mutant phenotype. These observations support a previous study on startle behaviour using the same D. melanogaster chromosome substitution lines, which concluded that suppressing epistasis may buffer the effects of new mutations. However, epistatic effects are not correlated among the different phenotypes. Thus, suppressing epistasis appears to be a pervasive general feature of natural populations to protect against the effects of new mutations, but different epistatic interactions modulate different phenotypes affected by mutations at the same pleiotropic gene.

Summary
Introduction: defining substance abuse
Drug abuse is a serious medical and social problem affecting millions of people each year. Substance abuse is usually defined as the misuse of drugs, including taking a drug for purposes other than those prescribed or intended. Dependent or addicted individuals engage in compulsive drug seeking and drug taking despite the consequences to self and others. A recent national survey (in the US) on drug abuse estimated 16.6 million individuals classified as substance abuser and/or dependent, with 1.1 million seeking treatment for substance abuse disorders. Both conditions are recognized for their deleterious consequences to individuals as well as society. Therefore, the primary goal of clinicians is to optimize therapies for maladaptive drug-taking behaviors.
This chapter provides an overview of the role of noradrenergic mechanisms involved in the development and maintenance of substance abuse, as well as the integration of the mechanisms mediating the potential adverse consequences of maladaptive drug-taking behavior (i.e., tolerance, dependence, and withdrawal). A wide array of commonly abused substances is discussed. This chapter focuses on the stimulants (i.e., amphetamine and cocaine), but also includes opiates, alcohol, and nicotine. Although some studies to date do not support the clinical efficacy of noradrenergic-based therapies for the treatment of stimulant abuse, others demonstrate a therapeutic potential of noradrenergic compounds for their ability to alleviate the adverse consequences of other abused drugs such as nicotine and opiates.
A substantial literature implicates the role of dopamine in the subjective “rewarding” properties of abused drugs (as defined below).
