In most Arctic and subarctic regions, Trichinella nativa is a common zoonotic pathogen circulating among wild carnivores. The polar bear (Ursus maritimus) is one of the most important reservoirs for T. nativa in frigid zones. In Iceland, Trichinella infection has never been detected in the local wildlife, despite the presence of one of the host species, the arctic fox (Alopex lagopus). In 2008, one of two polar bears that had swum to Iceland's coast was found to have been infected with Trichinella sp. (8.5 larvae/g in the tongue, 6.8 larvae/g in the masseter and 4.4 larvae/g in the diaphragm); the larvae were identified as T. nativa. This is the second report of Trichinella infection in polar bears that reached the Icelandic coast. In the present work, we describe this case of infection and discuss the epidemiological features that have allowed T. nativa to spread in Arctic regions.

The monogenean Pseudodactylogyrus bini parasitizes the gills of eels belonging to the genus Anguilla. Circumstantial evidence suggests that the parasite has been spread accidentally from the Pacific area (East Asia) to Europe by the intercontinental eel trade. This is based on early descriptions of the parasites from Asian regions and the lack of records of the parasites in Europe before 1977. In addition, the susceptibility of European eels to infections with the parasite is significantly higher compared to that of Japanese eels, which could indicate that the European eel had not undergone co-evolution with this parasite. The present study was undertaken to elucidate the origin of the parasite by using molecular tools. Parasite samples were obtained from Europe (Germany), Asia (Taiwan) and Nova Scotia, the latter of which is the first record of P. bini in Canada. Sequencing of rDNA comprising part of the internal transcribed spacer 1 (ITS1) gene, 5.8S and part of ITS2 (1323 bp) showed that P. bini isolates from the first two regions showed high variability. One sequence was found both in a number of Asian parasites and with one to a few transitions in European parasites, which could indicate that they were split recently into the two regions. Other sequence variations suggested that one or a few genotypes of P. bini were imported on one occasion from Asia to Europe and that the two geographic isolates subsequently developed differently in the two regions. The Nova Scotian/Canadian isolates showed no variation and were found to be unique compared to the European and Taiwanese forms, indicating that this population is independent in origin. This could indicate that the Canadian parasites were introduced to North America on another occasion and independently of the European colonization.

Infected hosts progressively decrease egg output during the chronic phase of Schistosoma mansoni infection. Ageing may be a factor that results in a progressive decrease in the ability to reproduce. This study was performed to gain insight into the effects of ageing on adult schistosomes, using confocal laser scanning microscopy. Adult worms were recovered from experimentally infected Nectomys squamipes (water rat). Viable eggs were voided in the faeces for 65 weeks and, thereafter, only unviable eggs were eliminated. The rat died after 70 weeks in captivity. Recovered worms (six males and one female) were prepared for confocal microscopy and images were obtained with an LSM 510–ZETA laser confocal microscope. The overall morphology of the adult worms (suckers and tegument) and reproductive organs were the focus of this work. The suckers, ovary, vitellaria and oocytes appeared to be apparently normal. The ootype was formed by flattened cells and unclear nuclei, suggesting that the mechanism for eggshell production by the ootype during organogenesis might have been impaired. Testicular lobes showed empty areas around the germinative cells. Male adult worms showed flaccid musculature of the dorsal surface. Taken together, the present results provided several indications of infertility in older male and female schistosomes.

Experimental infections of pre-adult Galba truncatula (two populations) using one, two, three or five miracidia of Fasciola hepatica per snail were carried out to determine the total number of cercariae produced by snails when they were sacrificed at day 70 post-exposure (at 24°C). When the number of miracidia used for each snail increased at exposure, significant numerical augmentations of live rediae and of intraredial differentiating cercariae were noted. In contrast, only insignificant differences between mean numbers of free cercariae were found. The number of metacercariae produced by each snail (after shedding and/or after dissection) increased in infections with up to three miracidia per snail in each population and strongly decreased in infections with five miracidia per snail. Compared to the other types of infections, the delay of cercarial differentiation noted in the five-miracidia groups was probably due to the volume of the snail host, which would be too small to assure the complete development of five sporocysts.

Parasitism of fish in fully bunded storage reservoirs has rarely been studied, while the impact of a rapid decline in a fish population on its parasite fauna is poorly understood. The present paper investigates the helminth fauna of perch (Perca fluviatilis) over a 5-year period in the Queen Mary reservoir, a large (290 ha) completely artificial water storage impoundment, which forms a unique and challenging habitat for its resident fish population. After 3 years of study, the perch population suffered a significant reduction due to a disease outbreak caused by the pathogenic bacterium Flavobacterium columnare, ‘columnaris disease’, and the subsequent effects on the helminth fauna over the next 2 years are evaluated. Conditions in the reservoir favour the development of large populations of zooplankton, which act as intermediate hosts for the majority of helminths in the dominant perch community. The prevalence and intensity of helminth infections showed much variation over the period prior to the perch mortality. These were likely to be due to changes in the zooplankton biomass, which was exposed to biotoxins released from periodic algal blooms and the application of copper to control them. The outbreak of F. columnare resulted in a significant decrease in the size and condition coefficient of the perch population. Changes also occurred in the composition of helminth parasites, with many species demonstrating either an increase or decrease in infection levels. These changes may be partly associated with an increase in the zooplankton biomass and altered population structure, probably caused by a decrease in fish predation pressure, which would influence the parasite population dynamics, although other factors directly associated with changes in the perch population are also likely to be influential. The long-term affects on the levels of helminth infections in the fish and associated intermediate hosts in the Queen Mary reservoir are discussed.

In a parasitological survey of free-ranging Cerdocyon thous (Carnivora: Canidae) from Brazilian Caatinga shrubland, a new species of Pterygodermatites (Multipectines) was recovered from the small intestine of this host. Morphological analysis showed that P. (Multipectines) pluripectinata n. sp. is distinguished from all other congeneric species mainly by the numerous plate-like projections and male caudal morphology and spicular length. There are few records on the occurrence of this genus in Neotropical regions.

The aim of this study was to test the hypothesis of the presence of hybrids between Schistosoma guineensis and S. haematobium in the Estuaire province (Western Gabon). Egg morphometry and single-stranded conformational polymorphism (SSCP) analysis on adult worms were used in order to characterize the schistosome populations of two sites. The morphology of the eggs showed three morphotypes: S. haematobium, S. guineensis and intermediate morphotypes, but the eggs of the morphotype S. guineensis were smaller compared to the values found in the literature. Furthermore, the SSCP analysis of the adult schistosomes showed that all the patterns corresponded to that of S. haematobium and gave evidence that hybrids were absent from our samples.

A study of nematode (Heligmosomum costellatum) prevalence in the common vole (Microtus arvalis) population was performed in 2002 and 2003 in an agricultural region of southern Moravia, Czech Republic. The influence of the trapping date, body weight, age, gender and reproduction of the common vole on the prevalence of the larvae of H. costellatum encapsulated in the stomach wall was examined. Of 503 common voles examined, 27.6% were H. costellatum positive. The prevalence of H. costellatum in the common vole population was influenced mainly by the weight, trapping date, year and reproductive state, as well as by the interactions between year and gender and between year and reproductive state. The influence of age was weak. The most infected common vole groups were the older, heavier and already reproducing females captured from April to August and all voles captured in 2002. In general, parasite prevalence seems to be dependent on the population parameters of the host species. However, the H. costellatum prevalence study in a single common vole population was not sufficient to fully describe and explain the nematode–host interactions.

Two invasive fish species, the round goby Apollonia melanostoma syn. Neogobius melanostomus (Pallas 1814) and the bighead goby Neogobius kessleri (Günther, 1861), have established a firm population in Austrian waters during the past 15 years. As there have been no records of the parasite fauna from these populations, a total of 79 specimens of A. melanostoma and 12 specimens of N. kessleri were examined for parasites between May and October 2007 from three different sampling sites from the Danube River in Austria. In total 12 parasite taxa were recovered. The protozoans Trichodina sp. and Ichthyophthirius multifiliis from the gills and skin; two crustacean species, Paraergasilus brevidigitus and Ergasilus sieboldi, from the gills; and the two monogeneans Gyrodactylus sp. and Dactylogyrus sp., from the skin and gills respectively, all occurred at low prevalence and intensities. Furthermore, cystacanths of the acanthocephalan Acanthocephalus lucii were found in the body cavity. Metacercariae of the digeneans Diplostomum sp. and Tylodelphys clavata were found in the lens of the eye and the vitreous humour, respectively. Adults of two digeneans, Nicolla skrjabini and Bunodera nodulosa, were found in the intestine. In addition, during this survey metacercariae of the Holarctic digenean Bucephalus polymorphus, encysted in the skin and fins, with prevalence up to 78%, were recorded for the first time in Austria.

The blue mussel Mytilus edulis is described as second intermediate host for Echinostephilla patellae from the common limpet Patella vulgata. Mussels were infected with metacercariae of E. patellae under laboratory conditions. Average infection rates increased with increasing temperature, whereas numbers of cercariae, to which individual mussels were exposed, had no effect on relative infection success. The round to slightly oval metacercariae with an average cyst diameter of 208 μm (range 186–243 μm) encysted exclusively in the foot tissue of M. edulis. Morphologically similar metacercariae were found in naturally infected mussels at sites where parasitized P. vulgata and M. edulis are sympatric. This is the first report of E. patellae in blue mussels. The detection of M. edulis being a second intermediate host is of particular interest with regard to the abundance of the parasite and host organisms in intertidal rocky shore ecosystems. The potential role of the common limpet P. vulgata as an alternative secondary host is discussed.

The production of cercariae from their snail host is a fundamental component of transmission success in trematodes. The emergence of Echinoparyphium recurvatum (Trematoda: Echinostomatidae) cercariae from Lymnaea peregra was studied under natural sunlight conditions, using naturally infected snails of different sizes (10–17 mm) within a temperature range of 10–29°C. There was a single photoperiodic circadian cycle of emergence with one peak, which correlated with the maximum diffuse sunlight irradiation. At 21°C the daily number of emerging cercariae increased with increasing host snail size, but variations in cercarial emergence did occur between both individual snails and different days. There was only limited evidence of cyclic emergence patterns over a 3-week period, probably due to extensive snail mortality, particularly those in the larger size classes. Very few cercariae emerged in all snail size classes at the lowest temperature studied (10°C), but at increasingly higher temperatures elevated numbers of cercariae emerged, reaching an optimum between 17 and 25°C. Above this range emergence was reduced. At all temperatures more cercariae emerged from larger snails. Analysis of emergence using the Q10 value, a measure of physiological processes over temperature ranges, showed that between 10 and 21°C ( ≈ 15°C) Q10 values exceeded 100 for all snail size classes, indicating a substantially greater emergence than would be expected for normal physiological rates. From 14 to 25°C ( ≈ 20°C) cercarial emergence in most snail size classes showed little change in Q10, although in the smallest size class emergence was still substantially greater than the typical Q10 increase expected over this temperature range. At the highest range of 21–29°C ( ≈ 25°C), Q10 was much reduced. The importance of these results for cercarial emergence under global climate change is discussed.

One hundred and three samples of faeces of reared grey wolves from four locations (Stobnica Park and Zoological Gardens in Bydgoszcz, Wrocław and Cracow) and twenty-six samples of faeces from two free-roaming packs of grey wolf (Canis lupus L.) in Piła (Forest Divisions: Borne Sulinowo, Czarnobór, Jastrowo) and Zielona Góra (Forest Divisions: Torzym, Krosno Odrzańskie) were collected between 2005 and 2007. Helminth eggs were detected in 78.6% of faecal samples of reared grey wolves and in 88.4% of those of free-roaming wolves. The trematode Alaria alata (80.1%) and nematodes Eucoleus aerophilus (23.1%) and Spirocerca lupi (11.5%) were only detected from wild packs of wolves and the nematodes Ancylostoma caninum (35.9%), Trichuris vulpis (15.5%) and Toxocara canis (3.9%) were only detected from reared wolves. Differences were observed in the prevalence and composition of helminth fauna between reared and wild grey wolves and our results are compared with those from studies within Poland and elsewhere in Europe.

An experimental infection with echinostomatid miracidia in sympatric or ‘local’ vs. allopatric or ‘away’ snail combinations, as a model to examine parasite compatibility, was carried out. We employed Euparyphium albuferensis miracidia to infect Gyraulus chinensis snails, from three different natural parks: Albufera (Valencia, Spain); the Ebro Delta (Tarragona, Spain) and Coto de Doñana (Huelva, Spain). Insignificant differences between the three snail strains were noted for the infection rate and the rhythm of daily cercarial production. However, a significantly higher total cercarial production per snail, patent period and life span were observed in local snails. The different infection characteristics in the three G. chinensis strains considered reveal that E. albuferensis miracidia demonstrate local adaptation.

The present study aimed to investigate the seasonal epidemiological prevalence of gastro-intestinal nematodes (GINs) of goats with respect to sex and age of the host in the Kashmir valley from 1 February 2005 to 31 January 2007. A total of 1267 goats were examined [faecal examination: 938 (year 1: 470; year 2: 468); gastro-intestinal (GIT) examination: 329 (year 1: 175; year 2: 154)]. The overall prevalence of GIN infection in these animals was 54.3% (year 1: 54.8%; year 2: 53.8%; P = 0.842). The different parasites reported with their respective prevalences (%) were: Haemonchus contortus (48.3); Bunostomum trigonocephalum (30.1); Chabertia. ovina (29.8); Ostertagia circumcincta (29.8); Nematodirus spathiger (25.2); Trichostrongylus spp. (25.1); Oesophagostomum columbianum (23.5); Trichuris ovis (19.0); and Marshallagia marshalli (16.6). The mean maximum prevalence of GIN infection (faecal examination: 75.6 ± 0.20; GIT examination: 85.3 ± 0.95), faecal egg counts (2552 ± 85.7) and average worm burden (333.25 ± 2.25) were found in the summer and they were lowest in winter (prevalence: faecal examination, 23.2 ± 0.95; GIT examination, 12.7 ± 0.20; faecal egg counts: 134.15 ± 9.15; and average worm burden: 79.8 ± 52.2), with significant differences between the seasons (P < 0.05). The sex of the hosts was not an important factor influencing the prevalence of GIN infection. With the increase in host age, prevalence of infection decreased significantly (P ≥ 0.05). Thus seasonal dynamics and age of the host animals significantly influenced the prevalence of GIN infection. The above findings will be helpful in devising the appropriate control strategies for GINs of goats reared under the traditional husbandry system in temperate agro-climatic conditions in the Kashmir valley as well as in similar climatic zones of other parts of the world.

Endoparasites associated with the small bromelicolous lizard Bogertia lutzae, a poorly studied phyllodactylid inhabitant of north-eastern Brazil, were studied. Fifty-seven specimens collected from the Atlantic Forest of Alagoas state were dissected. Only one species of parasite, the nematode Spauligodon oxkutzcabiensis, was found, with a prevalence of 22.8%. The intensity of infection was 2.62 ± 1.19, and neither the prevalence nor mean intensity differed between the sexes. There was no correlation between lizard body size and intensity of infection. An aggregated pattern of distribution (D = 0.813) of S. oxkutzcabiensis was found in this lizard host population. Bogertia lutzae represents a new host recorded for S. oxkutzcabiensis, a parasite reported for the first time for Brazil.

Larval stages (cercariae) of schistosomatid flukes represent the causative agents of swimmer's itch (cercarial dermatitis), a waterborne allergic disease. Cercariae of bird schistosomes are the most frequently reported agent. Recent studies on parasite behaviour in mammals showed that infections by cercariae can be linked to more than skin syndromes. Despite the failure of complete development in mammals, bird schistosomes can escape from the skin and migrate transitorily in the hosts. These findings brought novel insights into the fate and potential pathogenic effect of the parasites in non-compatible hosts, including humans. Cercarial dermatitis occurs globally and recently is considered to be re-emerging; however, there are no data on the number of afflicted persons per year. This might be explained by a relatively low interest in human skin infections arising after bathing in fresh water. In addition, the real occurrence of bird schistosomes in the field is known only for a few areas. The paucity of epidemiological/biogeographical data is probably caused by difficulties associated with detection of the parasites in intermediate and definitive hosts. Therefore, based on personal experience and data available in the literature, we have summarized methodological approaches enabling the detection of bird schistosomes in various hosts and environments.

Praziquantel (PZQ) is now widely used for the treatment of human schistosomiasis. However, in recent years, there has been a growing concern about the resistance of Schistosoma to PZQ. The mechanisms of PZQ action against Schistosoma and resistance of Schistosoma to PZQ are poorly understood. Here, we report differential susceptibilities to PZQ between male and female cercariae in the PZQ-susceptible and PZQ-resistant isolates of Schistosoma mansoni, using tail loss as a measurement of PZQ action. The miracidia were collected by hatching eggs collected from faeces of infected mice. Single-sex cercaria lines were made by infecting a single Biomphalaria glabrata snail with a single miracidium. The sex of each single-sex cercaria line was identified by a direct W1-specific polymerase chain reaction (PCR) technique. Single-sex cercariae of two isolates were exposed to four different concentrations of PZQ, respectively. The tail shedding of cercariae was observed under a dissecting microscope for five time points up to 100 min after adding PZQ. The results showed that male cercariae have higher tail-shedding rates than that of female cercariae when PZQ-susceptible isolates of S. mansoni are exposed to the same concentration of PZQ. But this phenomenon was not observed in the PZQ-resistant isolates. This sexual differential resistance phenomenon of S. mansoni suggests that resistance to PZQ is induced by decreasing the PZQ susceptibility of male worms. The experiment described here may also be useful for developing tests to detect PZQ resistance in the field.

Extracts prepared from the microbivorous nematode Panagrellus redivivus and the plant-parasitic nematode Meloidogyne incognita were used to provide general protease activities for peptide substrate screening and species comparisons. Each extract was evaluated for its ability to degrade a broad range of nematode FMRFamide-like peptides (FLPs), key regulatory messengers governing nematode growth and development. Clear quantitative differences between the two extracts were observed using FMRFamide as a substrate. Extract potency assessed at EC50 (μg/μ l extract protein for 50% substrate digestion) was 1.8-fold greater for P. redivivus than for M. incognita, and potency assessed at EC90 was 2.5-fold greater. An overall potency difference was also present when screening the digestion of 17 nematode FLPs, but it was not universal. The mean percentage digestion of eight of the 17 FLPs was greater (P < 0.02) with P. redivivus extract (76.3 ± 8.2) than with M. incognita extract (38.1 ± 8.7), but the means for the other nine FLPs were not different. Three FLPs (KPSFVRFa, AQTFVRFa, RNKFEFIRFa) were degraded extensively by the extracts of both species, and two FLPs (SAPYDPNFLRFa, SAEPFGTMRFa) were degraded 2.9-fold and 5.3-fold greater, respectively, with M. incognita extract than with P. redivivus extract. The ability of each extract to degrade FMRFa and KSAYMRFa was significantly reduced by using peptide analogues containing single d-amino acid substitutions, and the substitution effects were positional. Both FMRFa and KSAYMRFa were competitive substrates for aminopeptidases in each extract, but only the competitive ability of FMRFa was reduced by d-amino acid substitution. The variety and complexity of nematode FLP degradation by preparations representing phylogenetically and developmentally different nematode sources are discussed.

Molecular analyses such as polymerase chain reaction (PCR) and sequencing are very useful for taxon identification, especially when morphological characters useful for identifying taxa are lacking. However, the use of molecular tools can be the source of taxon misidentification if they are not correctly applied and the results are not critically evaluated and compared with the literature and GenBank data. We describe a case of misidentification of a taxon of the genus Trichinella due to sequencing mistakes, lack of reference material and selection of a single molecular marker. A Trichinella sp. isolate from an Iranian wild boar (Sus scrofa) was identified as belonging to the Nearctic species Trichinella murrelli, through the molecular analysis of the 5S rRNA intergenic spacer region. A successive molecular identification of the same isolate was performed by the International Trichinella Reference Centre in Rome, Italy, using the 5S rRNA intergenic spacer region, the LSU rDNA expansion segment five, and the internal transcribed spacers 1 and 2. According to these analyses, the Iranian isolate belonged to Trichinella britovi, a Palaearctic species already described in Iran.

Estimates of parasite intensity within host populations are essential for many studies of host–parasite relationships. Here we evaluated the seasonal, age- and sex-related variability in faecal water content for two wild ungulate species, springbok (Antidorcas marsupialis) and plains zebra (Equus quagga). We then assessed whether or not faecal water content biased conclusions regarding differences in strongyle infection rates by season, age or sex. There was evidence of significant variation in faecal water content by season and age for both species, and by sex in springbok. Analyses of faecal egg counts demonstrated that sex was a near-significant factor in explaining variation in strongyle parasite infection rates in zebra (P = 0.055) and springbok (P = 0.052) using wet-weight faecal samples. However, once these intensity estimates were re-scaled by the percent of dry matter in the faeces, sex was no longer a significant factor (zebra, P = 0.268; springbok, P = 0.234). These results demonstrate that variation in faecal water content may confound analyses and could produce spurious conclusions, as was the case with host sex as a factor in the analysis. We thus recommend that researchers assess whether water variation could be a confounding factor when designing and performing research using faecal indices of parasite intensity.
