Interactive impact of childhood maltreatment, depression, and age on cortical brain structure: mega-analytic findings from a large multi-site cohort

Background
Childhood maltreatment (CM) plays an important role in the development of major depressive disorder (MDD). The aim of this study was to examine whether CM severity and type are associated with MDD-related brain alterations, and how they interact with sex and age.
MethodsWithin the ENIGMA-MDD network, severity and subtypes of CM using the Childhood Trauma Questionnaire were assessed and structural magnetic resonance imaging data from patients with MDD and healthy controls were analyzed in a mega-analysis comprising a total of 3872 participants aged between 13 and 89 years. Cortical thickness and surface area were extracted at each site using FreeSurfer.
ResultsCM severity was associated with reduced cortical thickness in the banks of the superior temporal sulcus and supramarginal gyrus as well as with reduced surface area of the middle temporal lobe. Participants reporting both childhood neglect and abuse had a lower cortical thickness in the inferior parietal lobe, middle temporal lobe, and precuneus compared to participants not exposed to CM. In males only, regardless of diagnosis, CM severity was associated with higher cortical thickness of the rostral anterior cingulate cortex. Finally, a significant interaction between CM and age in predicting thickness was seen across several prefrontal, temporal, and temporo-parietal regions.
ConclusionsSeverity and type of CM may impact cortical thickness and surface area. Importantly, CM may influence age-dependent brain maturation, particularly in regions related to the default mode network, perception, and theory of mind.
Altered cortical-amygdala coupling in social anxiety disorder during the anticipation of giving a public speech

Background
Severe stress in social situations is a core symptom of social anxiety disorder (SAD). Connectivity between the amygdala and cortical regions is thought to be important for emotion regulation, a function that is compromised in SAD. However, it has never been tested if and how this connectivity pattern changes under conditions of stress-inducing social evaluative threat. Here we investigate changes in cortical-amygdala coupling in SAD during the anticipation of giving a public speech.
MethodTwenty individuals with SAD and age-, gender- and education-matched controls (n = 20) participated in this study. During the functional magnetic resonance imaging (fMRI) session, participants underwent three ‘resting-state’ fMRI scans: one before, one during, and one after the anticipation of giving a public speech. Functional connectivity between cortical emotion regulation regions and the amygdala was investigated.
ResultsCompared to controls, SAD participants showed reduced functional integration between cortical emotion regulation regions and the amygdala during the public speech anticipation. Moreover, in SAD participants cortical-amygdala connectivity changes correlated with social anxiety symptom severity.
ConclusionsThe distinctive pattern of cortical-amygdala connectivity suggests less effective cortical-subcortical communication during social stress-provoking situations in SAD.

We recently presented (Sbordone et al., 2009a) the largest sample to date of lithium abundances in extremely metal-poor (EMP) Halo dwarf and Turn-Off (TO) stars. One of the most crucial aspects in estimating Li abundances is the Teff determination, since the Li I 670.8 nm doublet is highly temperature sensitive. In this short contribution we concentrate on the Teff determination based on Hα wings fitting, and on its sensitivity to the chosen stellar gravity.

We present the largest sample available to date of lithium abundances in extremely metal poor (EMP) Halo dwarfs. Four Teff estimators are used, including IRFM and Hα wings fitting against 3D hydrodynamical synthetic profiles. Lithium abundances are computed by means of 1D and 3D-hydrodynamical NLTE computations. Below [Fe/H]~−3, a strong positive correlation of A(Li) with [Fe/H] appears, not influenced by the choice of the Teff estimator. A linear fit finds a slope of about 0.30 dex in A(Li) per dex in [Fe/H], significant to 2–3 σ, and consistent within 1 σ among all the Teff estimators. The scatter in A(Li) increases significantly below [Fe/H]~−3. Above, the plateau lies at 〈A(Li)3D, NLTE〉 = 2.199 ± 0.086. If the primordial A(Li) is the one derived from standard Big Bang Nucleosynthesis (BBN), it appears difficult to envision a single depletion phenomenon producing a thin, metallicity independent plateau above [Fe/H] = −2.8, and a highly scattered, metallicity dependent distribution below.
The systemic absorption and disposition of levobupivacaine 0.5% after epidural administration in surgical patients: a stable-isotope study

Summary
Background and objective: Absorption and disposition kinetics can be studied with a stable-isotope method. The aim of this study was to validate a stable-isotope method for levobupivacaine and to derive the relevant pharmacokinetics after epidural administration.
Methods: Eight volunteers (18–32 yr) received approximately 23 mg of both levobupivacaine and deuterium-labelled levobupivacaine simultaneously by intravenous infusion. Venous blood samples were taken for 8 h. Fifteen patients (23–85 yr) received 19 mL levobupivacaine 0.5% (including a 3 mL test dose) epidurally and, 25 min later, approximately 25 mg deuterium-labelled levobupivacaine (D3-levobupivacaine) intravenously. Arterial blood samples were collected for 24 h. Plasma concentrations were determined using liquid chromatography-mass spectrometry. Plasma concentration–time data were analysed by compartmental and non-compartmental analysis.
Results: Based on the ratio of the normalized areas under the curve of unlabelled and deuterium-labelled levobupivacaine in volunteers, as determined by both compartmental (mean ratio: 1.02, 90% CI: 1.00–1.04) and non-compartmental analysis (mean ratio: 1.02, 90% CI: 1.00–1.03) the two formulations were considered equivalent. In surgical patients the elimination half-life (mean ± SD: 196 ± 65 min), total body clearance (349 ± 114 mL min−1) and volume of distribution at steady state (56 ± 14 L), derived by compartmental analysis, were similar to those obtained by non-compartmental analysis. The absorption was bi-phasic. The fraction absorbed and half-life of the fast absorption process were 0.22 ± 0.06 and 5.2 ± 2.7 min, respectively. The values for the slow absorption process were 0.84 ± 0.14 and 386 ± 91 min, respectively.
Conclusions: D3-levobupivacaine is pharmacokinetically equivalent to unlabelled levobupivacaine and can be used to study the absorption and disposition kinetics after perineural administration of levobupivacaine in a single experiment.
