OBJECTIVES/SPECIFIC AIMS: The off-target and organ-specific toxicities observed in cancer immunotherapy present an obstacle to T-cell-based therapeutics. A recent clinical trial underscored the need for improved methods to define TCR specificity after melanoma patients treated with TCR engineered T-cells suffered from fatal cardiovascular toxicity arising from the unpredicted recognition of a muscle-specific peptide. METHODS/STUDY POPULATION: To address this drawback to T-cell-based immunotherapies, we developed a novel protein engineering approach to define the peptide specificity of a given TCR. Here, directed evolution in a yeast display system produced a large scale peptide library, where recognition by the melanoma reactive DMF5 TCR acted as the guiding selective pressure. After this technique identified a panel of putative cross reactive peptides, sequence analysis and computational modeling followed by kinetic binding experiments and structural analysis determined the DMF5 TCR recognizes 2 distinct classes of peptides through chemically distinct mechanisms. RESULTS/ANTICIPATED RESULTS: This information led to the rational, structure-based design of additional cross reactive peptides and introduced a unique approach to screen the human proteome and identify the TCR targets which triggered undesired autoimmunity when this molecule was used in clinical trials. DISCUSSION/SIGNIFICANCE OF IMPACT: The distinct chemical nature of the 2 peptide classes suggest TCRs are more cross reactive than previously thought, presenting an obstacle to cell-based immunotherapy. Defining the peptide specificity of TCRs is of high interest to the immunology community, and will lead to improved approaches to designing engineered TCRs for cell therapy.

Background
Long-acting injectable formulations of antipsychotics are treatment alternatives to oral agents.
AimsTo assess the efficacy of aripiprazole once-monthly compared with oral aripiprazole for maintenance treatment of schizophrenia.
MethodA 38-week, double-blind, active-controlled, non-inferiority study; randomisation (2:2:1) to aripiprazole once-monthly 400 mg, oral aripiprazole (10–30 mg/day) or aripiprazole once-monthly 50mg (a dose below the therapeutic threshold for assay sensitivity). (Trial registration: clinicaltrials.gov, NCT00706654.)
ResultsA total of 1118 patients were screened, and 662 responders to oral aripiprazole were randomised. Kaplan–Meier estimated impending relapse rates at week 26 were 7.12% for aripiprazole once-monthly 400mg and 7.76% for oral aripiprazole. This difference (−0.64%, 95% CI −5.26 to 3.99) excluded the predefined non-inferiority margin of 11.5%. Treatments were superior to aripiprazole once-monthly 50mg (21.80%, P⩽0.001).
ConclusionsAripiprazole once-monthly 400mg was non-inferior to oral aripiprazole, and the reduction in Kaplan–Meier estimated impending relapse rate at week 26 was statistically significant v. aripiprazole once-monthly 50 mg.

Weed management is a major concern for organic farmers. Strategies and techniques used by organic farmers vary according to crops, soils, climate and other factors. Organic farmers in upstate New York experienced in weed management were interviewed about what weed–crop complexes they face, how they manage weeds, and what priorities they have for research. The interviews were complemented with an all-day focus group conducted with the farmers who were interviewed. All of the farmers interviewed relied heavily on cultural practices, such as crop rotation, fertility management, high-density planting, transplants and the use of cover crops. Mechanical practices were also important for the farmers interviewed. They emphasized selection of the appropriate tools for the situation and techniques for using tools effectively. Most of the farmers interviewed relied on intensive tillage to control perennial weeds. Differences in practices were notable among the farmers interviewed. Several farmers shared innovations both in practices and equipment modification. Farmers identified soil and nutrient management; tillage and cultivation tools and practices; natural herbicides; flame weeding and no-till systems without herbicides as research priorities. Some of the topics suggested for research have already received substantial research attention; identification of these topics by the farmers indicated a need to disseminate research findings through extension materials and demonstration of viable practices.

The heat of binding the serine protease, porcine pancreatic elastase, by the inhibitor, turkey ovomucoid third domain, is dependent on the presence of inorganic phosphate. This dependence is saturable and can be accurately modeled as the phosphate binding to a single site on the protease–inhibitor complex; thus, the elastase–ovomucoid system provides a unique opportunity to study phosphate–protein interactions. We have used isothermal titration calorimetry to investigate this binding, thereby providing one of the few complete thermodynamic characterizations of phosphate interacting with proteins. The binding is characterized by a small favorable ΔG°, a large unfavorable ΔH°, and a positive ΔCp, thermodynamics consistent with the release of water being linked to phosphate binding. These measurements provide insight into the binding of phosphotyrosine containing peptides to SH2 domains by suggesting the energetic consequences of binding phosphate free from other interactions.

Interaction between farmers and researchers has benefits for both groups. Researchers gain an understanding of farmers' needs and the production system into which new techniques must fit; farmers are helped in adopting new techniques, and get insights into why some techniques are more successful than others. However, communication between farmers and researchers usually has been just from the university to the farmer. The recovery of information from farmers, integration of techniques into whole production systems, and the research priorities of farmers have not been very important to researchers.
An exception has been in sustainable agriculture, where some researchers have been asking farmers what they are doing and what they want the university to do, and spending time observing existing practices before they design research trials. Ideally, farmers and researchers interact as equals, sharing information and expertise. We present examples from California and New York that show that this approach is highly effective, and recommend policie s to expand on this approach in researchfunding, training of research and extension personnel, and the dissemination ofinformation. We also discuss practical ways to carry out those policies, drawn from the experiences of both university and private non-profit organizations.
Fibxtem— Focussed Ion Beam Milling for TEM Sample Preparation

In the semiconductor industry, shrinking geometries and increasing process complexity have greatly increased the demand for TEM analysis of specific submicron regions. Until recently, samples of this nature have been difficult if not impossible to prepare. We have combined cross-sectional TEM sample preparation (XTEM) and the precise material sputtering of focussed ion beam milling (FIB) to thin samples to electron transparency. We call this sample preparation technique FIBXTEM.
Three advantages of this technique are: 1) The area of interest can be analyzed in the scanning electron microscope before final thinning; 2) Any specific defect area becomes a candidate for TEM analysis, including failed sub-micron structures; and 3) Samples are generally artifact-free and of uniform thickness.
Key elements of the FIBXTEM technique include precision planar polishing, unique holders for mounting and transferring samples between systems, and the FIB-induced deposition of a sacrificial protective layer over the area of interest during ion thinning.
This technique extends the use of TEM analysis into new areas of semiconductor process development and failure analysis. Recent applications for materials problem solving and failure analysis are discussed.
Groundwater Protection Policy and Agricultural Production: A Recursive Stochastic Analysis

Contamination of groundwater by agricultural practices presents a dilemma between protecting a vital resource and maintaining a valuable part of the economy. Policies to balance these objectives are presented. In addition to an historical baseline, policies that ban certain pesticides, taxes and subsidies, and control cultural practices are also considered. A model is developed to reflect the current state of agriculture in Eastern Suffolk County. This model consists of a recursive programming component, which has input for it generated by a stochastic model of Colorado potato beetle pest dynamics and management strategies to control those pests. While income is reduced by banning pesticides, the reduction is small when compared with the improvement in environmental quality. Further efforts to reduce pesticide use resulted in a reduction in potato acreage and incomes, as well as yields. Analysis concludes that improvements in both farm income and environmental quality could be achieved through the adoption of subsidies for low-input conservation crops.

A survey of organic farmers in New York State identified problems in need of university research. Weed management was the most frequently mentioned problem by far, identified as significant by two-thirds of the organic farmers. Only a few other problems were listed as significant, including insufficient time for farm work, lack of markets, low prices, and lack of appropriate tools. These were cited by more than a third of the farmers. Drought, insect management, and a lack of a dependable supply of labor were cited by about one-third of the respondents. The survey also examined organic farmers' information sources. They do not use conventional sources of agricultural information, such as the extension service and conventional agricultural media, as much as books, magazines, and newsletters on organic f arming, other organic f armers, and on-farm experiments. Many respondents noted that local extension agents did not know very much about non-chemical solutions to organic production problems. They considered University Extension to be accessible, but not very useful in solving problems specific to organic farming, and had many suggestions to improve Land Grant research in organic agriculture.
