OBJECTIVES/GOALS: We aimed to conduct an updated genome-wide meta-analysis of keloids in expanded populations, including those most afflicted by keloids. Our overall objective was to improve understanding of keloid development though the identification and further characterization of keloid-associated genes with genetically predicted gene expression (GPGE). METHODS/STUDY POPULATION: We used publicly available summary statistics from several large-scale DNA biobanks, including the UK Biobank, FinnGen, and Biobank Japan. We also leveraged data from the Million Veterans Program and performed genome-wide association studies of keloids in BioVU and eMERGE. For each of these datasets, cases were determined from ICD-9/ICD-10 codes and phecodes. With these data we conducted fixed effects meta-analysis, both across ancestries and stratified by broad ancestry groups. This approach allowed us to consider cumulative evidence for genetic risk factors for keloids and explore potential ancestry-specific components of risk. We used FUMA for functional annotation of results and LDSC to estimate ancestry-specific heritability. We performed GPGE analysis using S-PrediXcan with GTEx v8 tissues. RESULTS/ANTICIPATED RESULTS: We detected 30 (23 novel) genomic risk loci in the cross-ancestry analysis. Major risk loci were broadly consistent between ancestries, with variable effects. Keloid heritability estimates from LDSC were 6%, 21%, and 34% for European, East Asian, and African ancestry, respectively. The top hit (P = 1.7e-77) in the cross-ancestry analysis was at a replicated variant (rs10863683) located downstream of LINC01705. GPGE analysis identified an association between decreased risk of keloids and increased expression of LINC01705 in fibroblasts (P = 3.6e10-20), which are important in wound healing. The top hit in the African-ancestry analysis (P = 5.5e-31) was a novel variant (rs34647667) in a conserved region downstream of ITGA11. ITGA11 encodes a collagen receptor and was previously associated with uterine fibroids. DISCUSSION/SIGNIFICANCE: This work significantly increases the yield of discoveries from keloid genetic association studies, describing both common and ancestry-specific effects. Stark differences in heritability support a potential adaptive origin for keloid disparities. Further work will continue to examine keloids in the broader context of other fibrotic diseases.

Non-technical summary
We summarize some of the past year's most important findings within climate change-related research. New research has improved our understanding about the remaining options to achieve the Paris Agreement goals, through overcoming political barriers to carbon pricing, taking into account non-CO2 factors, a well-designed implementation of demand-side and nature-based solutions, resilience building of ecosystems and the recognition that climate change mitigation costs can be justified by benefits to the health of humans and nature alone. We consider new insights about what to expect if we fail to include a new dimension of fire extremes and the prospect of cascading climate tipping elements.
Technical summaryA synthesis is made of 10 topics within climate research, where there have been significant advances since January 2020. The insights are based on input from an international open call with broad disciplinary scope. Findings include: (1) the options to still keep global warming below 1.5 °C; (2) the impact of non-CO2 factors in global warming; (3) a new dimension of fire extremes forced by climate change; (4) the increasing pressure on interconnected climate tipping elements; (5) the dimensions of climate justice; (6) political challenges impeding the effectiveness of carbon pricing; (7) demand-side solutions as vehicles of climate mitigation; (8) the potentials and caveats of nature-based solutions; (9) how building resilience of marine ecosystems is possible; and (10) that the costs of climate change mitigation policies can be more than justified by the benefits to the health of humans and nature.

In this paper we define almost gentle algebras, which are monomial special multiserial algebras generalizing gentle algebras. We show that the trivial extension of an almost gentle algebra by its minimal injective co-generator is a symmetric special multiserial algebra and hence a Brauer configuration algebra. Conversely, we show that any almost gentle algebra is an admissible cut of a unique Brauer configuration algebra and, as a consequence, we obtain that every Brauer configuration algebra with multiplicity function identically one is the trivial extension of an almost gentle algebra. We show that a hypergraph is associated with every almost gentle algebra A, and that this hypergraph induces the Brauer configuration of the trivial extension of A. Among other things, this gives a combinatorial criterion to decide when two almost gentle algebras have isomorphic trivial extensions.

Randomized experiments commonly compare subjects receiving a treatment to subjects receiving a placebo. An alternative design, frequently used in field experimentation, compares subjects assigned to an untreated baseline group to subjects assigned to a treatment group, adjusting statistically for the fact that some members of the treatment group may fail to receive the treatment. This article shows the potential advantages of a three-group design (baseline, placebo, and treatment). We present a maximum likelihood estimator of the treatment effect for this three-group design and illustrate its use with a field experiment that gauges the effect of prerecorded phone calls on voter turnout. The three-group design offers efficiency advantages over two-group designs while at the same time guarding against unanticipated placebo effects (which would undermine the placebo-treatment comparison) and unexpectedly low rates of compliance with the treatment assignment (which would undermine the baseline-treatment comparison).

We develop a theory of group actions and coverings on Brauer graphs that parallels the theory of group actions and coverings of algebras. In particular, we show that any Brauer graph can be covered by a tower of coverings of Brauer graphs such that the topmost covering has multiplicity function identically one, no loops, and no multiple edges. Furthermore, we classify the coverings of Brauer graph algebras that are again Brauer graph algebras.

The CSBN meeting held in Rio de Janeiro on August 11 was attended by just six members, including Pam Kilmartin as the acting chair, and several visitors. Since there was not a quorum of members, it was not possible to make any decisions. But there was a good discussion on many topics, from which several points emerged that should be more fully discussed by the whole committee during the next few months:

A total of 701 comets received names between July 2005 and June 2008. Comets observed only from the SOHO and STEREO missions, as well as further comets recognized from the long-defunct SOLWIND satellite, accounted for 520 of these names.

Abstract
In this paper we survey, without proof, the main structural results of Koszul algebras. We also survey the extension of this theory to semiperfect Noetherian rings. Applications to algebras of global dimensions 1 and 2 are discussed as well as applications to Auslander algebras and to preprojective algebras.
Introduction and Definitions
Koszul algebras have played an importart role in commutative algebra and algebraic topology [22, 16, 17, 3, 4, 24, 25]. Recently there have been important applications of noncommutative Koszul algebras to algebraic topology, Lie theory and quantum groups [8, 7, 23, 5]. This paper begins with some terminology and definitions in this section. In section 2, we survey the main results about Koszul algebras. Definition of a Koszul algebra follows in this section. The proofs of the results in this section can be found in [14, 15]. In section 3, we survey the main results about Koszul modules stressing the duality between the category of Koszul modules of a Koszul algebra and the category of Koszul modules over the Yoneda algebra. Again, proofs may be found in [14, 15]. The cited references, the concept of a Koszul algebra and module is extended outside the graded case. The notion of quasi-Koszul algebra is introduced.

This paper studies the construction of Frobenius algebras. We begin with a description of when a graded -algebra has a Frobenius algebra as a homomorphic image. We then turn to the question of actual constructions of Frobenius algebras. We give a, method for constructing Frobenius algebras as factor rings of special tensor algebras. Since the representation theory of special tensor algebras has been studied intensively ([6], see also [2; 3; 4]), our results permit the construction of Frobenius algebras which have representations with prescribed properties. Such constructions were successfully used in [9].

Let ∧ denote a basic artin ring and r its radical. In most of this paper we assume that r2 = 0 and that Λ is a trivial extension Λ/r ⋉ r (see Section 1 for definition). Let P1 …, Pn be the non-isomorphic indecomposable projective (left) Λ-modules, and consider triples (Pi, Mi, ui), where the Mi, are (left) Λ-modules and ui:rPi → Mi/rMi isomorphisms. From this data we construct a new ring Г, which in “nice cases” has the property that r′3 = 0, Г/r′2 ≅ Λ, and r′ Qi ≅ Mi as (left) Λ-modules, where the Qi are the indecomposable projective (left) Г-modules and r′ is the radical of Г.
