The virulence of Campylobacter jejuni and C. coli isolated from various water sources was compared with that of clinical strains by in vitro assays of adhesion, invasion and cytotoxicity to HeLa cells. Variation in degree of attachment was observed, but this did not appear to be related to strain source, However, water strains were less invasive and less cytotoxic to HeLa cells than clinical strains as shown by immunofluorescence and electron microscopy.
These differences were particularly evident between clinical and water isolates of the same serotype and biotype implicated in an outbreak of campylobacter enteritis in a school. The enhanced virulence of the clinical isolates, possibly induced by passage, was confirmed by colonization tests on infant mice.
The speciation and subtyping of campylobacter isolates from sewage plants and waste water from a connected poultry abattoir using molecular techniques

In this study the distribution of phenotypes of campylobacter strains in sewage and surface waters was investigated by subtyping and by speciation of isolates from various aquatic environments. These environments included two municipal sewage plants (SPA and SPB) and waste water from a poultry abattoir (WWA). Both the sewage plants SPA and SPB collected domestic and industrial waste, and SPA received drain water from WWA. SPB received no waste water from any meat-processing plant. The isolates were speciated by PCR and subtyped by PCR/RFLP based on the flagellin PCR products.
From all three reservoirs, no Campylobacter lari was isolated, and approximately 80% of the isolates could be identified as C. jejuni and the rest belonged to the C. coli species. The PCR/RFLP typing technique has a high discrimination level and was reproducible between two separate laboratories. The 182 isolates tested yielded 22 distinct Dde I profiles. The results indicate that strains with profiles found in poultry are also detectable in waste water presumed to be solely from domestic and human sources. In addition some strains were unique to the known poultry-related sources, suggesting that avian-specific strains, non-pathogenic to man, may exist in the environment. In contrast some strains were unique to human waste indicating the potential importance of non-poultry sources of infection. No seasonality was observed in the profile distribution. So, at least in the Netherlands, it is unlikely that infections caused by contaminated surface waters contribute to the seasonality of human campylobacteriosis.

A 12-month abattoir study was undertaken from January 2003. We collected 7492 intestinal samples from cattle, sheep and pigs at slaughter. Rectal samples were taken from cattle and sheep and caecal samples from pigs. They were examined for verocytotoxigenic E. coli (VTEC) O157, Salmonella, thermophilic Campylobacter and Yersinia enterocolitica. Data were collected on the animal from which the sample came and this information was analysed to look at potential risk factors for carriage of these organisms. Logistic regression models were run where an adequate number of positive results were available. This revealed that VTEC O157 carriage in cattle was associated with the summer period and that age was a protective factor. Salmonella carriage in pigs was associated with lairage times >12 h, the North East and not feeding when there was no bedding available. In cattle, carriage was associated with the summer period, the Eastern region of GB and dairy animals. In sheep a spring seasonal effect was seen, which coincided with the lambing period. The carriage of thermophilic Campylobacter in cattle was associated with single-species abattoirs, with age a protective factor. In sheep, winter was a risk period with lairage management influential. For pigs, lairage times of <12 h were found to be associated with carriage. A seasonal trend for carriage of Y. enterocolitica in all species was demonstrated with the period December–May a risk. For cattle, age was also a risk factor; for sheep feeding in the lairage and for pigs being held overnight were risk factors.

An abattoir survey was undertaken to determine the prevalence of foodborne zoonotic organisms colonizing cattle, sheep and pigs at slaughter in Great Britain. The study ran for 12 months from January 2003, involved 93 abattoirs and collected 7703 intestinal samples. The design was similar to two previous abattoir surveys undertaken in 1999–2000 allowing comparisons. Samples were examined for VTEC O157, Salmonella, thermophilic Campylobacter and Yersinia enterocolitica. The prevalence of VTEC O157 faecal carriage was 4·7% in cattle, 0·7% in sheep and 0·3% in pigs. A significant decrease in sheep was detected from the previous survey (1·7%). Salmonella carriage was 1·4% in cattle, a significant increase from the previous survey of 0·2%. In sheep, faecal carriage was 1·1% a significant increase from the previous survey (0·1%). In pigs, carriage was 23·4%, consistent with the previous study. Thermophilic Campylobacter spp. were isolated from 54·6% of cattle, 43·8% of sheep and 69·3% of pigs. Y. enterocolitica was isolated from 4·5% of cattle, 8·0% of sheep and 10·2% of pigs.

The degradation of AlGaAs/GaAs diode laser performance during operation is typically due to catastrophic optical damage of the facets caused when thermal runaway occurs. These heating effects are due to the presence of non-radiative recombination sites at and near the facets. MOCVD GaS is deposited on the facets of 825-nm ridge waveguide AlGaAs/GaAs quantumwell laser diodes as an electronic passivation to reduce the number of surface states available for non-radiative recombination. For passivated devices, a peak pulsed power nearly double that of unpassivated devices was achieved. The passivated devices also exhibit a longer lifetime before degradation. The impact of the passivation process on other optical characteristics of the laser diodes will also be discussed.

A diffusion model is used to find heavy traffic approximate autocorrelation functions for several variables in queueing systems (i.e. for waiting time, system time, number in system and unfinished work). A table is given by which correlations can easily be found for each variable in any GI/G/1 queue. Further, the infinite sum or integral of the autocorrelations is also found, and the spectral density function. The sum has applications in statistical analysis of queues.
Extensive comparisons of approximate and exact correlations and their sums are reported, particularly for waiting times and system times, but also including number in system in M/M/1 queues. In general the correlations have similar accuracy to the probability distributions found by diffusion approximations. The percentage error is less for number in system than for waiting time.

The arrival rate of customers to a service facility is assumed to have the form λ(t) = λ(0) — βt2 for some constant β. Diffusion approximations show that for λ(0) sufficiently close to the service rate μ, the mean queue length at time 0 is proportional to β–1/5. A dimensionless form of the diffusion equation is evaluated numerically from which queue lengths can be evaluated as a function of time for all λ(0) and β. Particular attention is given to those situations in which neither deterministic queueing theory nor equilibrium stochastic queueing theory apply.

During a rush hour, the arrival rate λ(t) of customers to a service facility is assumed to increase to a maximum value exceeding the service rate μ, and then decrease again. In Part I it was shown that, after λ(t) has passed μ, the expected queue E{X(t)} exceeds that given by the deterministic theory by a fixed amount, (0.95)L, which is proportional to the (–1/3) power of a(t) = dλ(t)/dt evaluated at time t = 0 when λ(t) = μ. The maximum of E{X(t)}, therefore, occurs when λ(t) again is equal to μ at time t1 as predicted by deterministic queueing theory, but is larger than given by the deterministic theory by this same constant (0.95)L (provided t1 is sufficiently large). It is shown here that the maximum queue, suptX(t) is, approximately normally distributed with a mean (0.95) (L + L1) larger than predicted by deterministic theory where L, is proportional to the (–1/3) power of a(t1). We also investigate the distribution of X(t) at the end of the rush hour when the queue distribution returns to equilibrium. During the transition, the queue distribution is approximately a mixture of a truncated normal and the equilibrium distributions. These results are applied to a case where λ(t) is quadratic in t.

Suppose that the arrival rate λ(t) of customers to a service facility increases with time at a nearly constant rate, dλ(t)/dt = a, so as to pass through the saturation condition, λ(t) = μ = service capacity, at some time which we label as t = 0. The stochastic properties of the queue are investigated here through use of the diffusion approximation (Fokker-Planck equation). It is shown that there is a characteristic time Tproportional to α–2/3 such that if , then the queue distribution stays close to the prevailing equilibrium distribution associated with the λ(t) and μ, evaluated at time t. For |t| = O(T), however, the mean queue length is much less than the equilibrium mean, and is measured in units of some characteristic length L which is proportional to α–1/3. For , the queue is approximately normally distributed with a mean of the order L larger than that predicted by deterministic queueing models. Numerical estimates are given for the mean and variance of the distribution for all t. The queue distributions are also evaluated in non-dimensional units.

If on a long homogeneous highway there is no interaction between cars, then, under a wide range of conditions, an initial distribution of cars will in the course of time tend toward that of a Poisson process with statistically independent velocities for the cars in any finite interval of highway. Here we will generalize this known property to obtain the following. Suppose cars do interact in such a way as to delay a car when it passes another, but the density of cars is so low that we can neglect simultaneous interactions between three or more cars. There will again be equilibrium distributions of cars to which general classes of initial distributions will converge. These equilibrium distributions are superpositions of two statistically independent processes, one a Poisson process of single free cars with statistically independent velocities, and the other a Poisson process of interacting pairs of cars with various velocities. In the limit of zero interaction, the density of pairs vanishes leaving only the Poisson process of single cars as a special case. To the same order of approximation, including the first order effects of interactions, the headway distribution between consecutive cars will still have exponential tail outside the range of interaction.

A. J. Miller has proposed a stochastic model for the study of highway traffic in one direction along a two-lane road in which the desired speed of a vehicle is sampled from a continuous probability distribution and queues of vehicles are Poisson distributed on the highway. Miller has derived the equilibrium expression for the relation between the spacial densities of all vehicles and the spacial densities of those which are freely travelling. This relationship takes the form of a non-linear integral equation which is left unsolved. The present model differs from the original only in the assumption of a discrete distribution of desired speeds. Although the derivations of the equilibrium equations are based upon different arguments from those of Miller, these relations transform to the corresponding integral equations in the limit of a continuous speed distribution. The main body of the paper is devoted to a detailed investigation of the case of two speeds and asymptotic results are obtained for three speeds.
