Alcohol milestones and internalizing, externalizing, and executive function: longitudinal and polygenic score associations

Background
Although the link between alcohol involvement and behavioral phenotypes (e.g. impulsivity, negative affect, executive function [EF]) is well-established, the directionality of these associations, specificity to stages of alcohol involvement, and extent of shared genetic liability remain unclear. We estimate longitudinal associations between transitions among alcohol milestones, behavioral phenotypes, and indices of genetic risk.
MethodsData came from the Collaborative Study on the Genetics of Alcoholism (n = 3681; ages 11–36). Alcohol transitions (first: drink, intoxication, alcohol use disorder [AUD] symptom, AUD diagnosis), internalizing, and externalizing phenotypes came from the Semi-Structured Assessment for the Genetics of Alcoholism. EF was measured with the Tower of London and Visual Span Tasks. Polygenic scores (PGS) were computed for alcohol-related and behavioral phenotypes. Cox models estimated associations among PGS, behavior, and alcohol milestones.
ResultsExternalizing phenotypes (e.g. conduct disorder symptoms) were associated with future initiation and drinking problems (hazard ratio (HR)⩾1.16). Internalizing (e.g. social anxiety) was associated with hazards for progression from first drink to severe AUD (HR⩾1.55). Initiation and AUD were associated with increased hazards for later depressive symptoms and suicidal ideation (HR⩾1.38), and initiation was associated with increased hazards for future conduct symptoms (HR = 1.60). EF was not associated with alcohol transitions. Drinks per week PGS was linked with increased hazards for alcohol transitions (HR⩾1.06). Problematic alcohol use PGS increased hazards for suicidal ideation (HR = 1.20).
ConclusionsBehavioral markers of addiction vulnerability precede and follow alcohol transitions, highlighting dynamic, bidirectional relationships between behavior and emerging addiction.

Summary
The Coral Triangle is the world's most biologically diverse marine environment, and the origins of this diversity have been the subject of debate for decades. Biogeographic studies have alternatively argued that this region is a center of (1) origin, (2) overlap, (3) accumulation, or (4) survival. Genetic studies focused on the pattern, distribution, and age of lineage divergence across a broad array of marine fishes and other groups show alternating support for all of these hypotheses, indicating pluralistic origins for the Coral Triangle biodiversity hotspot. While acknowledging multiple causations, the relative contribution of each model is still uncertain. Given the social and economic importance of marine fishes in the Coral Triangle combined with sharp declines in populations and coral reef habitat in this region, understanding the ecological and evolutionary processes shaping biodiversity in the Coral Triangle is more important now than ever. New genomic tools as well as standardized sampling methodologies hold great promise for refining our understanding of the processes driving lineage divergence in the Coral Triangle, providing information essential for effective conservation planning in this region.
THE CORAL TRIANGLE, FROM PATTERN TO PROCESS
Although the name and geographic scope of the Coral Triangle has changed greatly over the past 60 years [1141], it is unquestionably the most diverse marine region on the planet. Defined by the presence of >500 hard coral species [2566] and spanning six Southeast Asian nations, species diversity in a broad range of marine taxa reaches a global maximum in the Coral Triangle with steep declines in diversity moving away from this biodiversity hotspot [200,205,747,1141,2123,2415]. In reef fishes, diversity peaks in the Birds Head region of West Papua, Indonesia [41], with 1511 species in 451 genera and 111 families, 26 species, and 14 families being restricted to this region. Species diversity in fishes can be one to two orders of magnitude higher in the Coral Triangle than peripheral reefs of the Pacific or the Caribbean.

Different lifestyle patterns across Europe may influence plasma concentrations of B-vitamins and one-carbon metabolites and their relation to chronic disease. Comparison of published data on one-carbon metabolites in Western European regions is difficult due to differences in sampling procedures and analytical methods between studies. The present study aimed, to compare plasma concentrations of one-carbon metabolites in Western European regions with one laboratory performing all biochemical analyses. We performed the present study in the European Prospective Investigation into Cancer and Nutrition (EPIC) cohort among 5446 presumptively healthy individuals. Quantile regression was used to compare sex-specific median concentrations between Northern (Denmark and Sweden), Central (France, Germany, The Netherlands and United Kingdom) and Southern (Greece, Spain and Italy) European regions. The lowest folate concentrations were observed in Northern Europe (men, 10·4 nmol/l; women, 10·7 nmol/l) and highest concentrations in Central Europe. Cobalamin concentrations were slightly higher in Northern Europe (men, 330 pmol/l; women, 352 pmol/l) compared with Central and Southern Europe, but did not show a clear north–south gradient. Vitamin B2 concentrations were highest in Northern Europe (men, 22·2 nmol/l; women, 26·0 nmol/l) and decreased towards Southern Europe (Ptrend< 0·001). Vitamin B6 concentrations were highest in Central Europe in men (77·3 nmol/l) and highest in the North among women (70·4 nmol/l), with decreasing concentrations towards Southern Europe in women (Ptrend< 0·001). In men, concentrations of serine, glycine and sarcosine increased from the north to south. In women, sarcosine increased from Northern to Southern Europe. These findings may provide relevant information for the study of regional differences of chronic disease incidence in association with lifestyle.

Gross and microscopic visceral anatomy of the male Cape fur seal, Arctocephalus pusillus pusillus (Pinnipedia: Otariidae), with reference to organ size and growth

The gross and microscopic anatomy of the Cape fur seal heart, lung, liver, spleen, stomach, intestine and kidneys (n = 31 seals) is described. Absolute and relative size of organs from 30 male seals are presented, with histological examination conducted on 7 animals. The relationship between log body weight, log organ weight and age was investigated using linear regression. Twenty five animals were of known age, while 6 were aged from counts of incremental lines observed in the dentine of tooth sections. For the range of ages represented in this study, body weight changes were accurately described by the exponential growth equation, weight = wort, with body weight increasing by 23% per annum until at least 9–10 y of age. Organ weight increased at a rate of between 25% and 33% per annum until at least 9–10 y of age, with the exception of the intestines, where exponential increase appeared to have ceased by about 7 y. The relationship between body weight and organ weight was investigated using logarithmic transformations of the allometric equation, y = axb, where the exponent b is 1 if organ weight is proportional to body weight. Most organs increased in proportion to the body. However, the heart, liver and spleen had exponents b > 1, suggesting that these organs increased at a faster rate than the body. The basic anatomical features of the viscera were similar to those of other pinnipeds, with some exceptions, including the arrangement of the multilobed lung and liver. Apart from the large liver and kidneys, relative size of the organs did not differ greatly from similar sized terrestrial carnivores. The histological features of the organs were generally consistent with those previously described for this species and other otariids. The heart, as in other pinnipeds, was unlike that of cetacea in not having unusually thick endocardium or prominent Purkinje cells. Notable histological features of the lungs included prominent fibrous septa, prominent smooth muscle bundles, cartilage extending to the level of the alveolar sacs and ample lymphoid tissue. The spleen had a thick capsule, well developed trabeculae and plentiful plasma cells. Abundant parietal cells were present in the fundic glands and lymphoid follicles were present in the gastric lamina propria, particularly in the pyloric region. Small intestinal villi were very long but this could have resulted from underlying chronic inflammation. Lymphoid follicles were prominent in the colon. The kidney reniculi each had a complete cortex, medulla and calyx, but a sportaperi medullaris musculosa was not identified.
Voltaire and Hume as Historians: A Comparative Study of the Essai sur les Moeurs and the History of England

Seventeen years ago, in a highly provocative article in this journal, E. C. Mossner rose to the defense of David Hume's historical writings, which had been treated with considerable severity by scholars such as J. B. Black, who, in his Art of History, had compared them unfavorably with those of his contemporaries, in particular Voltaire. Mossner, however, in trying to bring out the qualities of Hume's History of England, confronted it almost exclusively with Voltaire's Siècle de Louis XIV, and in this instance it must be admitted that Hume labored under a grave disadvantage, since his far more comprehensive subject was harder to master from the artistic as well as the scientific point of view than Voltaire's. In the present article we shall transfer the comparison from the Siècle to Voltaire's longest and most mature historical effort, the Essai sur les moeurs et l'esprit des nations, and this considerably changes our perspective, for here Voltaire is dealing with a similar and, if anything, more difficult theme. In this way a parallel may prove highly instructive in bringing out the excellences and weaknesses of the two historians respectively in their approach to comparable subjects and in the formulation of their philosophy of history. What makes comparisons particularly inviting is the circumstance that Voltaire's topic also includes Hume's, so that in numerous instances it is possible to examine the reaction of both writers to the same events and personalities. While our investigation of the resemblances and differences in the thought of these two outstanding representatives of the Enlightenment will stress some of the traits shared by most historians of their age, we shall also attempt to underline the more individual elements that emerge from their works.
