OBJECTIVES/SPECIFIC AIMS: Characterize the expression kinetics of HIV-1 Envelope and their relationship to virus production at the cellular level. METHODS/STUDY POPULATION: In vitro and ex vivo laboratory analyses. RESULTS/ANTICIPATED RESULTS: Initial studies addressing the kinetics of cell surface. Envelope (Env) expression reveal that Env expression to peaks on day 2 post infection. Next steps include a series of experiments to compare the kinetics of Env cell surface expression with broadly neutralizing antibody (bNAb)-mediated ADCC and the characterization of virus production kinetics in this same context. To be maximally effective, ADCC elimination of infected cells should occur before peak Env expression. DISCUSSION/SIGNIFICANCE OF IMPACT: Potent bNAbs to HIV-1 recognize vulnerable sites on the HIV-1 Envelope (Env) protein and are of great clinical interest due to their potential use in the prevention and treatment of HIV-1 infection. Their effectiveness depends not only on the neutralization of viral infectivity, but also on the elimination of productively infected cells via antibody-dependent cellular cytotoxicity (ADCC). On a cellular level, ADCC dynamics are determined by the timing and level of Env expression on the surface of HIV-infected cells. This study aims to delineate the expression kinetics of HIV-1 Envelope and their relationship to virus production. We expect that it will provide new insights into the utility of bNAb-mediated ADCC in treating and possibly curing HIV-1 infection; therefore results might have substantial impact on future HIV treatment strategies.

Orthodontists, thanks to their keen appreciation of facial morphology and their skills in making functional examinations, are ideally placed to detect breathing problems in their patients that can require an ENT assessment, with special attention paid to the evaluation of sleep patterns. After a site of obstruction, resulting from a soft tissue anomaly and/or a narrowing of the airway passage through bone, has been established, orthodontists may be called upon to undertake corrective treatment sometimes in conjunction with a surgical procedure.

Obstructive Sleep Apnea Syndrome (OSAS) is a frequently occurring childhood malady, whose is incidence is largely underestimated, that can lead to serious complications. Because of their understanding of the anatomy and physiology of the masticatory apparatus and of the upper airways, orthodontist are in a strategic position to detect in their young patients developing disorders in these regions; accordingly when they suspect the existence of OSAS they should ask systematic and well targeted questions of parents who may not be aware of the relationship between certain symptoms their children display and sleep breathing problems. The goal of this article is to provide practitioners with a better understanding of the symptoms and the physio-pathological mechanics of OSAS so that they can spare their afflicted patients the complications it provokes by seeing that early treatment is undertaken.
Efficacy of Prevention by High-Efficiency Particulate Air Filtration or Laminar Airflow Against Aspergillus Airborne Contamination During Hospital Renovation

Objective:
To evaluate efficacy of laminar airflow facilities plus high-efficiency particulate air (HEPA) filtration and HEPA filtration alone in preventing environmental Aspergillus contamination during hospital renovation. To show the usefulness of environmental surveillance to facilitate protection of patients at risk for invasive pulmonary aspergillosis.
Design:Prospective sampling of air and surfaces for Aspergillus conidia during 2-year period.
Setting:A hematological department adjacent to building renovation at a university hospital.
Results:1,047 air samples and 1,178 surface samples were collected from January 1996 to December 1997. Significantly more air samples were positive for Aspergillus species during the period of building renovation than during the periods before and after renovation in a unit without a protected air supply adjacent to the building work area (51.5% vs 31.7%; odds ratio [OR], 2.3; 95% confidence interval [CI95, 1.4-3.7; P<.001). A major increase in the frequency of positive air samples was also found in another adjacent unit that was protected with HEPA filtration alone (from 1.8% to 47.5%; OR, 48.9; CI95,12-229; P<10-7). In addition, in this unit, the mean count of Aspergillus conidia in positive air samples increased significantly during construction (4 colony-forming units [CFU]/m3 to 24.7 CFU/m3; P=.04) and the proportion of positive surface samples showed a significant increase during renovation (from 0.4% to 9.7%; OR, 28.3; CI95, 3.4-623; P=104). However, none of 142 air samples collected during renovation in the area protected with laminar airflow plus HEPA filtration showed Aspergillus conidia. In a unit distant from the building renovation site, the results of air and surface samples were not affected by renovation.
Conclusion:This study showed a strong association between building renovation and an increase in environmental Aspergillus contamination. Results confirmed the high efficacy of laminar airflow plus HEPA filtration and a high air-change rate. Although filtration with HEPA was effective during normal conditions, it alone was unable to prevent the rise of Aspergillus contamination related to building renovation. This study emphasized the necessity of an environmental survey of airborne contamination related to construction, to facilitate prevention of nosocomial aspergillosis outbreaks. A standardized protocol for aerobiological surveillance is needed.

In the context of the electronic industry, thin films of aluminum are deposited on silica. However, the Al/SiO2 interface is unstable. A reaction leading to oxidation of Al and reduction of SiO2 occurs. We present an in situ study of this reaction, in thin films, at temperatures above 500°C. This experiment allows to understand the essential steps of the oxidation of Al by SiO2, and it shows that thermal cycles can be used to generate nucleation centers artificially. More generally, the morphological interpretation of out-of-equilibrium growth patterns gives important insight into the coupling between the thermodynamics and morphological aspects of a thin film chemical reaction.
