Intestinal brush-border-membrane vesicles were prepared from the porcine small bowel by magnesium precipitation and differential centrifugation, and were functionally intact. The influence of dietary ligands on 65Zn uptake was determined using a 65Zn concentration of 5 μm, an incubation time of 1 min and a reaction temperature of 27°, with a rapid filtration technique. At this low Zn concentration the addition of an excess of folate, histidine or glucose had no effect on Zn uptake. Addition of picolinate, citrate and phytate to the incubation medium significantly reduced Zn uptake at all concentrations of ligand examined. Any inhibitory effects of folic acid in vivo may thuss be due to a mucosal rather than lumen interaction. Those ligands inhibiting absorption may have done so through the formation of Zn-ligand complexes, which are either insoluble, or which reduce the binding of Zn to its mucosal receptor. This in vitro model of Zn absorption is useful for comparing the effects of potential Zn-binding ligands in the diet.

Thirty-two crossbred wether lambs (initial live-weight 31 kg) were fed on a diet (metabolizable energy (ME) 12.8 MJ/kg) ad lib. (ADLIB) or restricted to maintain body-weight (MAINT) for a 21 d period. On days 0, 7, 14 and 21, four lambs per treatment were slaughtered, visceral organs weighed and tissues sampled. During the 21 d period, ME intake in ADLIB lambs increased quadratically with an average rate of live-weight gain of 425 g/d. In MAINT lambs, live weight (30 kg) was maintained, and daily ME intake (kJ/kg empty body-weight (EBW)0.75) declined (P < 0.01) quadratically with time. Weights of liver, stomach and small intestines as a percentage of EBW were increased in ADLIB lambs and decreased by 10–33 % in MAINT lambs (treatment × day, P < 0.01). In vitro liver oxygen consumption was not affected by level of feed intake. Estimates of whole-liver O2 consumption (mmol O2/d per kg EBW) increased in ADLIB lambs and were relatively constant in MAINT lambs. These findings suggest that level of feed intake changes the relative proportion of visceral organs to body mass. In addition, the effect of level of feed intake on changes in the relative contribution of visceral organs to whole-body metabolic rate appears to be primarily a result of differences in organ size rather than tissue-specific metabolic activity.

Non-esterified fatty acid (NEFA) and glycerol kinetics were studied in lactating goats to gain insight into the mechanisms by which animals in early lactation can initially mobilize and later replenish body fat reserves. Kinetic measurements were made at days 10, 38 and 76 post-partum in ten multiparous lactating does. Plasma NEFA concentrations and NEFA entry rate decreased as lactation advanced, being significantly higher at day 10 than at either days 38 or 76 of lactation. Both plasma NEFA concentrations and NEFA entry rate were negatively correlated with calculated energy balance. In contrast, glycerol entry rate was significantly higher at day 76 than at day 10 of lactation and was positively related to both calculated energy intake and energy balance. Apparent intracellular fatty acid re-esterification was lower at day 10 than at later stages of lactation and was positively related to calculated energy balance. It is suggested that during early lactation, substantial shifts in adipose tissue fat reserves can occur via altering rates of fatty acid re-esterification and de novo lipogenesis, without major changes in the rate of lipolysis.

The effects of lectins in the diet have been mainly studied in rats. An important question is whether results obtained in rats can be extrapolated to larger animals like the pig. Phaseolus vulgaris beans are rich in toxic lectins. Therefore a study was carried out to compare the effects of diets containing 200 g Phaseolus vulgaris beans (raw or toasted)/kg in rats and piglets. Live-weight gain, nitrogen digestibility and N balance were much lower in piglets than in rats fed on diets containing raw beans. Live-weight gain and N balance were slightly negative in the piglets. When toasted beans were given, live-weight gain and N balance values were reduced in piglets but hardly at all in rats. Giving raw beans caused hypertrophy of the pancreas in the rats but in piglets the weight of the pancreas was reduced. Spleen weight was depressed in the piglets but not in the rats. Weight of liver was not affected in either animal species. When toasted beans were given no effects on the weights of pancreas, spleen or liver were found in piglets or rats. It was concluded that the piglet is much more sensitive to antinutritional factors in the Phaseolus vulgaris bean than the rat.

Maternal whole-body glucose entry rate and uterine and umbilical net uptakes of glucose and oxygen were measured in single-pregnant ewes which were either well-fed throughout, or fed at 0.3–0.4 predicted energy requirement for 7–21 d during late pregnancy. All ewes were studied while standing at rest and then while walking on a treadmill at 0.7 m/s on a 10° slope for 60 min. Underfed ewes suffered significant decreases in live weight and had lower fetal, but not placental, weights at 140–144 d gestation. Undernutrition also caused large decreases in maternal glycaemia and glucose entry rate, which were associated with equally large decreases in uterine and umbilical net uptakes and O2 quotients of glucose, and with a decrease in placental glucose transfer capacity. Exercise caused increases in maternal blood concentration, entry rate and uterine net uptake of glucose, the magnitudes of which were not significantly affected by plane of nutrition. Umbilical glucose uptake and placental glucose transfer capacity increased during exercise in underfed but not fed ewes. The fractional distribution of maternal glucose to the pregnant uterus, and of uterine glucose uptake to the fetus, were unaltered by undernutrition; during exercise, a disproportionately small fraction of the increased maternal glucose supply went to the uterus. The results confirm that the ovine conceptus responds to nutritional reduction in maternal glucose availability in a manner similar to non-uterine maternal tissues. Major reductions in glucose supply appear to override putative glucose-sparing mechanisms which may operate to favour the conceptus in better-nourished animals.

Fetal glucose kinetics were measured using a combination of isotope-dilution and Fick-principle methodology in single-pregnant ewes which were either well-fed throughout, or fed at 0.3–0.4 predicted energy requirement for 7–21 d during late pregnancy. All ewes were studied while standing at rest and then while walking on a treadmill at 0.7 m/s on a 10° slope for 60 min. Underfed ewes suffered major decreases in fetal total disposal rate, fetal-placental transfer and umbilical net uptake of glucose, each of which were significantly related to declines in maternal and fetal blood glucose concentrations respectively. In well-fed ewes, fetal endogenous glucose production was negligible, as indicated by the similarity between fetal utilization rate (total glucose disposal rate minus placental uptake of fetal glucose) and umbilical net uptake of glucose, and by nearly identical fetal and maternal arterial blood specific radioactivities of maternally infused D-[2-3H]glucose. By contrast, in underfed ewes, fetal utilization rate greatly exceeded umbilical net uptake of glucose, and the fetal:maternal [3H]glucose specific activity ratio declined significantly, suggesting induction of a substantial rate of fetal endogenous glucogenesis. Exercise caused increases in fetal total glucose disposal rate and glycaemia in fed and underfed ewes. In underfed ewes only, this was accompanied by increased placental uptake of fetal glucose and umbilical net glucose uptake, unchanged fetal glucose utilization and decreased fetal endogenous glucose production. It is concluded that fetal gluconeogenesis makes a major contribution to fetal glucose requirements in undernourished ewes. Increased maternal supply of fetal glucose during exercise substitutes for rather than adds to fetal endogenous glucogenesis.

A comparison was made of the effects of antinutritional factors present in Phaseolus vulgaris on piglets, rats and chickens. Also the hypothesis of whether the negative effect on weight gain due to the inclusion of raw Phaseolus vulgaris in the diet can be attributed to an insufficient supply of amino acids was tested. Test diets containing 200 g raw Phaseolus beans/kg were balanced for digestible protein and amino acids; in one diet extra casein was incorporated. The main response criteria were live-weight gain and the weight of various organs including the intestine. Live-weight gain in piglets was markedly reduced during feeding 200 g raw Phaseolus vulgaris/kg in the diet, but not in rats and chickens. Addition of casein did not improve the weight gain of the piglets, indicating that a toxic factor was responsible for the reduced weight gain and not an insufficient supply of amino acids. The weights of the spleen and thymus were markedly reduced in the piglets when the diets with raw Phaseolus beans were given, but not in the rats and chickens. Additional supply of casein did not change this effect. Indications were found that when the supply of dietary protein is adequate there is no reduction in pancreas weight with raw Phaseolus beans as was observed in previous experiments. The weight of the intestine was increased in all three species due to feeding raw Phaseolus vulgaris.

Two experiments are described in which changes in both nitrogen and mineral balance were monitored in ewes during lactation. In Expt 1, two groups (n 6) of ewes were fed on diets that were either high (15 g crade protein (N × 6.25; CP)/MJ metabolizable energy (ME)) or low (10 g CP/MJ ME) in protein content, and concurrent N and mineral balance studies were made at intervals during lactation with 45Ca and 32P being used to monitor faecal endogenous calcium and phosphorus losses. Ewes fed on the high-protein diet maintained a positive N balance during early lactation, whereas those fed on the low-protein diet were in negative N balance. Both groups, however, showed the same degree of negative Ca and P balance, with the amounts lost being directly related to milk yield. This indicates that loss of mineral from the skeleton at this time is not secondary to a loss of bone matrix due to a shortfall in dietary protein supply. In Expt 2, two groups of ewes (n 4) were fed on diets in late pregnancy that were either just adequate or generous in Ca and P supply relative to requirement (TCORN, 1990). After parturition both groups were fed on a diet which was formulated to meet their estimated Ca and P requirements for lactation. As in the previous experiment both groups were in negative Ca and P balance in early lactation and variation in dietary Ca and P supply during pregnancy had no effect on the extent of this loss. Alternative explanations for the cause of this loss of mineral from the skeleton are discussed.

Dietary polychlorinated biphenyls (PCBs) caused hypercholesterolaemia in rats. The concentration and output of biliary cholesterol was significantly lower than that of the control group. Biliary output of total bile acids was significantly decreased in rats given the PCB-supplemented diet. Faecal excretion of total steroids (sum of neutral steroids and acidic steroids) was not significantly changed in rats given the PCB-supplemented diet. The present results indicate that dietary PCBs cause hypercholesterolaemia without modifying the faecal total steroids excretion. These results suggest that PCBs produce hyper-cholesterolaemia accompanied by changes in biliary or faecal excretion of bile acids and neutral steroids in addition to an increase in hepatic cholesterol synthesis.

Using a defaunating method which preserved bacteria and fungi in the rumen, the effect of protozoa on protein and fibre digestion was studied in six adult wethers in relation to the nature of the diet. Sheep were given daily, 42 g dry matter (DM)/kg metabolic body-weight (W0.75), one of two isonitrogenous diets: one contained ammonia-treated wheat straw as the only energy source (diet S) and the other was supplemented with maize grain pellets (diet SM). Mean daily intakes (g/d) of nitrogen, neutral-detergent fibre and acid-detergent fibre were respectively 22, 573 and 373 for diet S and 23, 450 and 334 for diet SM. Elimination of protozoa increased duodenal non-ammonia-nitrogen flow. This result was mainly due to an increase in microbial protein flow and, to a lesser extent, to a higher dietary protein flow. Defaunation markedly increased the efficiency of microbial protein synthesis. Maize-grain supplementation had a net positive effect on this variable in defaunated sheep, but not in faunated sheep. Cell-wall carbohydrates were less well digested in the defaunated rumen, and the negative effect of defaunation was greatest with the diet SM. Intestinal fibre digestion increased in the defaunated sheep especially in those fed on diet SM, but not enough to compensate for the decrease in rumen digestion.

The effect in rabbits of giving isonitrogenous purified diets containing casein, ovalbumin, fish protein, milk-whey protein and soya-bean protein were compared. The diets were balanced for cholesterol and for the amount and type of fat. When incorporated into low-cholesterol diets (0.8 g cholesterol/kg), casein, ovalbumin and soya-bean protein produced similar levels of serum cholesterol. With a high background of dietary cholesterol (1.5 g/kg), serum cholesterol concentrations increased with soya-bean protein, whey protein, casein and fish protein, in that order. Thus, the hypercholesterolaemic effect of casein in carefully balanced diets was only seen against a high-cholesterol background. The development of hypercholesterolaemia produced by giving fish protein was different from that produced by casein. First, less cholesterol accumulated in the very-low-density-lipoprotein fractions and more in the lipoproteins of higher density with fish protein than with casein. Second, fish protein, unlike casein, did not increase liver cholesterol. Third, transfer of rabbits from a diet containing soya-bean protein to one containing casein resulted in an immediate marked depression in neutral steroid and bile acid excretion in faeces. However, when rabbits were fed on the diet with fish protein after the diet with soya-bean protein, there was no significant depression in neutral steroid output and the depression in bile acid output was delayed. The present study suggests that different animal proteins cause hypercholesterolaemia by different mechanisms.

The stool-bulking effect of dietary fibre (DF) is well-documented and believed to be important in the postulated beneficial effect of DF on human health. The aim of the present study was to investigate the digestibility of DF in relation to its stool-bulking properties and to study possible mechanisms for this effect. Four diets, based on conventional foods only, were studied in balance experiments on human subjects. Diet A contained DF mainly from whole-grain cereals while diets B1 and B2 contained DF mainly from pulses, vegetables and fruit. Diet C was a low-fibre diet. Faeces was fractionated into four fractions, each enriched in one of the following three components: undigested DF (fractions 1 + 2), faecal bacteria (fraction 3) and soluble components (fraction 4). The digestibility of DF in diets A, B1 and B2 was 0.62, 0.88 and 0.90 respectively. Subjects consuming diet A excreted slightly more fraction 3 than subjects consuming the other diets. Thus, the statement that DF of high digestibility stimulates microbial growth in the gut was not supported. The water-holding capacity of fraction 1 was studied in vitro and was found to be low. It is suggested that undigested soluble DF is important in the stool-bulking properties of DF.

Mongrel dogs were fed, from weaning to 9 months of age, on one of two diets that differed only in the type of fat content (virgin olive oil or sunflower oil) to compare the composition of exocrine pancreatic secretion in the basal period and in response to food. In resting pancreatic flow, electrolytes and the specific activities of amylase, lipase and chymotrypsin were similar in both experimental groups. However, lipase and amylase outputs, and amylase and protein concentrations were significantly higher in the group fed on the diet rich in sunflower oil. Food intake was not followed by any change in flow-rate or electrolyte or protein content in the group given the diet rich in olive oil. Amylase activity and output were also lower in this group, as was lipase output, whereas activity and specific activity of chymotrypsin were lower in dogs fed on the diet containing sunflower oil. The differences traceable to the composition of the two types of dietary fat supplied may be related to the balance between factors that stimulate and inhibit pancreatic secretion.

Little is known about the ability of carnitine to modulate cell functions. As carnitine plays an important role in lipid metabolism we investigated the acute effect of L-carnitine, L-acetyl carnitine and L-propionyl carnitine (300 mg/kg per d; 4 d) on the basal and calcium-ionophore (A23187)-stimulated release of arachidonic acid metabolites from rat carrageenan-elicited peritoneal macrophages. A decrease in the number of peritoneal carrageenan-elicited macrophages was observed after feeding all three compounds. The basal release of prostaglandin E2, 6 keto-prostaglandin F1α and leukotriene B4 was stimulated by all treatments. In contrast, thromboxane B2 production was diminished by feeding carnitine and acetyl carnitine. A23187-stimulated synthesis of 6 keto-prostaglandin F1α and leukotriene B4 was further enhanced by all three compounds. Acetyl carnitine and propionyl carnitine also enhanced thromboxane B2 synthesis. However, no effects on prostaglandin E2 formation were detected. The 6 keto-prostaglandin F1α: thromboxane B2 ratio, calculated from the basal and A23187-stimulated values, was increased by carnitine treatment. In the presence of A23187 there was also an increase in the 6 keto-prostaglandin F1α: leukotriene B4 ratio. We conclude that carnitine, and possibly some of its derivatives, could modify the macrophage component of an inflammation in vivo.

A sugar-beet fibre preparation (SBF) was incorporated into a semi-synthetic rat diet at a level of 100 g/kg. The material caused no feed aversion, and gain of live weight was unimpaired. SBF appeared to be slightly more fermentable than wheat bran and only marginally less effective as a faecal bulking agent when compared with equalized intakes of non-starch polysaccharide. SBF did not stimulate mucosal cell turnover in the small intestine. Some enlargement of the caecum was observed in animals given SBF, but it was no greater than that of animals given wheat bran. Animals given SBF had a lower serum cholesterol concentration than both the fibre-free controls and those given wheat bran. This hypocholesterolaemic effect was less than that of guar gum however. The partial digestibility of energy for SBF was 0.64, and its partial digestible energy value was 11.3 kJ (2.7 keal)/g. The partial digestibility of energy for non-starch polysaccharide in SBF was estimated to be 0.53 and its partial digestible energy value was 9.1 kJ (2.2 kcal)/g. This value was not significantly different from that expected for unavailable carbohydrate in mixed human diets.

Unlike severe zinc deficiency, marginal Zn deficiency is difficult to identify in rats because no reliable indicator of suboptimal Zn status is currently available. We have previously observed reduced pancreatic γ-glutamyl hydrolase (EC 3.4.22.12) activity and impaired pteroylpolyglutamate absorption in Zn-deficient rats. In the present study the effect of Zn depletion and repletion on the Zn concentration of various tissues and on the activity of this enzyme was investigated. The objective was to determine the sensitivity of these variables to Zn depletion and to evaluate their usefulness as indices of Zn status. Male Wistar rats (about 180 g), maintained from weanling on a purified Zn-adequate diet, were randomly allocated into twelve groups. A pretreatment control group was killed immediately. The remaining eleven groups were fed on a Zn-deficient diet and a group killed daily for 7 d (Zn-depleted groups). The remaining four groups were re-fed the Zn-adequate diet and a group killed daily (Zn-repleted groups). On analysis, pancreas and spleen Zn levels responded most rapidly to reduced Zn intake, followed by tibia, liver, kidney and plasma. Zn concentration was maintained in testes. Reduced plasma folate levels were also observed. A significant reduction in pancreatic γ-glutamyl hydrolase activity before the depletion of many tissue Zn stores confirms the Zn sensitivity of the enzyme. It was concluded that future investigation into the inter-relationship between Zn and folate metabolism may be useful in identifying a sensitive, biochemical index of Zn status.

The purpose of the 2 × 2 factorial study was to determine the effect of varying levels of dietary calcium (2.5 and 10 g/kg) and butterfat (50 and 200 g/kg) on lipid utilization and on development of colon tumours in animals initiated with 1,2-dimethylhydrazine dihydrochloride. Among rats fed on 200 g butterfat/kg, the fourfold increase in Ca intake induced more than a sevenfold increase in faecal excretion of total lipids and almost a fortyfold increase in faecal excretion of acid-extractable lipid. Among rats fed on 50 g butterfat/kg, the ingestion of supplemental Ca had a less dramatic effect and induced only a twofold increase in faecal excretion of total lipids and a threefold increase in acid-extractable lipid. The volume of intestinal adenocarcinomas was correlated with the excretion of acid-extractable lipid in faeces (R 0.369, P < 0.02). Caecal enzymic activity was not correlated with tumour incidence or size or faecal lipid excretion. Overall, the fourfold increase in Ca intakes decreased total lipid absorption significantly but by less than 6%.
