Associations between schizophrenia genetic risk, anxiety disorders and manic/hypomanic episode in a longitudinal population cohort study

Background
Studies involving clinically recruited samples show that genetic liability to schizophrenia overlaps with that for several psychiatric disorders including bipolar disorder, major depression and, in a population study, anxiety disorder and negative symptoms in adolescence.
AimsWe examined whether, at a population level, association between schizophrenia liability and anxiety disorders continues into adulthood, for specific anxiety disorders and as a group. We explored in an epidemiologically based cohort the nature of adult psychopathology sharing liability to schizophrenia.
MethodSchizophrenia polygenic risk scores (PRSs) were calculated for 590 European-descent individuals from the Christchurch Health and Development Study. Logistic regression was used to examine associations between schizophrenia PRS and four anxiety disorders (social phobia, specific phobia, panic disorder and generalised anxiety disorder), schizophrenia/schizophreniform disorder, manic/hypomanic episode, alcohol dependence, major depression, and – using linear regression – total number of anxiety disorders. A novel population-level association with hypomania was tested in a UK birth cohort (Avon Longitudinal Study of Parents and Children).
ResultsSchizophrenia PRS was associated with total number of anxiety disorders and with generalised anxiety disorder and panic disorder. We show a novel population-level association between schizophrenia PRS and manic/hypomanic episode.
ConclusionsThe relationship between schizophrenia liability and anxiety disorders is not restricted to psychopathology in adolescence but is present in adulthood and specifically linked to generalised anxiety disorder and panic disorder. We suggest that the association between schizophrenia liability and hypomanic/manic episodes found in clinical samples may not be due to bias.

Vortex-current filaments have been used to study phenomena such as coronal loops and solar flares as well as tokamaks, and recent experimental work has demonstrated dynamics akin to vortex-current filaments on a table-top plasma focus device. While MHD vortex dynamics and related applications to turbulence have attracted consideration in the literature due to a wide variety of applications, not much analytical progress has been made in this area, and the analysis of such vortex-current filament solutions under various geometries may motivate further experimental efforts. To this end, we consider the motion of open, isolated vortex-current filaments in the presence of magnetohydrodynamic (MHD) as well as the standard hydrodynamic effects. We begin with the vortex-current model of Yatsuyanagi, Hatori & Kato (J. Phys. Soc. Japan, vol. 65, 1996, pp. 745–759) giving the self-induced motion of a vortex-current filament. We give the ‘cutoff’ formulation of the Biot–Savart integrals used in this model, to avoid the singularity at the vortex core. We then study the motion of a variety of vortex-current filaments, including helical, planar and self-similar filament structures. In the case where MHD effects are weak relative to hydrodynamic effects, the filaments behave as expected from the pure hydrodynamic theory. However, when MHD effects are strong enough to dominate, then we observe structural changes to the filaments in all cases considered. The most common finding is reversal of vortex-current filament orientation for strong enough MHD effects. Kelvin waves along a vortex filament (as seen for helical and self-similar structures) will reverse their translational and rotational motion under strong MHD effects. Our findings support the view that vortex-current filaments can be studied in a manner similar to classical hydrodynamic vortex filaments, with the primary role of MHD effects being to change the filament motion, while preserving the overall geometric structure of such filaments.
Anhedonia and cognitive function in adults with MDD: results from the International Mood Disorders Collaborative Project

Background
Cognitive dysfunction is common in major depressive disorder (MDD) and a critical determinant of health outcome. Anhedonia is a criterion item toward the diagnosis of a major depressive episode (MDE) and a well-characterized domain in MDD. We sought to determine the extent to which variability in self-reported cognitive function correlates with anhedonia.
MethodA post hoc analysis was conducted using data from (N=369) participants with a Diagnostic and Statistical Manual of Mental Disorders, 4th Edition, Text Revision (DSM-IV-TR)-defined diagnosis of MDD who were enrolled in the International Mood Disorders Collaborative Project (IMDCP) between January 2008 and July 2013. The IMDCP is a collaborative research platform at the Mood Disorders Psychopharmacology Unit, University of Toronto, Toronto, Canada, and the Cleveland Clinic, Cleveland, Ohio. Measures of cognitive function, anhedonia, and depression severity were analyzed using linear regression equations.
ResultsA total of 369 adults with DSM-IV-TR–defined MDD were included in this analysis. Self-rated cognitive impairment [ie, as measured by the Adult ADHD Self-Report Scale (ASRS)] was significantly correlated with a proxy measure of anhedonia (r=0.131, p=0.012). Moreover, total depression symptom severity, as measured by the total Montgomery–Åsberg Depression Rating Scale (MADRS) score, was also significantly correlated with self-rated measures of cognitive dysfunction (r=0.147, p=0.005). The association between anhedonia and self-rated cognitive dysfunction remained significant after adjusting for illness severity (r=0.162, p=0.007).
ConclusionsThese preliminary results provide empirical data for the testable hypothesis that anhedonia and self-reported cognitive function in MDD are correlated yet dissociable domains. The foregoing observation supports the hypothesis of overlapping yet discrete neurobiological substrates for these domains.

Genetic diversity of Philippine Rubus moluccanus L. (Rosaceae) populations examined with VNTR DNA probes

Two synthetic DNA probes composed of tandemly repeated ‘core’ sequences (GACA and GATA) were used to examine genetic diversity at multiple variable-number-tandem-repeat (VNTR) loci within and among four spatially isolated Philippine populations of Rubus moluccanus L. (Rosaceae), an uncultivated bramble widely distributed throughout southeast Asia – Malesia. Central goals were to determine whether apomictic propagation was detectable in R. moluccanus and to examine whether populations isolated on separate islands, or on mountain tops on a single island, were genetically differentiated. Sampling 22 individuals per population, the findings include: (1) no two individuals shared identical VNTR band profiles and thus apomictic propagation was not detected; (2) the mean number of bands scored per individual was 24.3 (SD = 3.91) and the proportion of polymorphic loci within populations ranged from 0.69 to 1.00 (mean = 0.86 ± SD = 0.085); (3) the average proportion of bands shared between individuals within populations ranged from 0.39 to 0.67 (0.50 ± 0.067), while average interpopulation similarity ranged from 0.21 to 0.50 (0.32 ± 0.092); and (4) estimated heterozygosity within populations ranged from 0.42 to 0.79 (0.62 ± 0.083), while interpopulation heterozygosity ran from 0.62 to 0.81 (0.74 ± 0.062). Permutation tests were used to estimate the statistical significance of differences in similarity between populations. A Luzon population isolated by 1200 km was always significantly different in similarity tests when compared with each of the other three populations, all located on Mindanao (and separated by at least 100 km but less than 250 km). Of the latter three, only one population differed significantly from the other two in the degree of VNTR markers shared, possibly reflecting biogeographic partitioning suggested for the island. Across all populations, average estimated Fst was 0.154, although mean inter-island Fst (0.224) was significantly higher than mean intra-island Fst (0.085). These results suggest that, while intra-island gene flow is relatively high, significant differentiation of tropical species may occur even over short distances on individual islands. Preservation of only a limited number of populations may result in a significant loss of genetic diversity in such species.
Autism at the beginning: Microstructural and growth abnormalities underlying the cognitive and behavioral phenotype of autism

Autistic symptoms begin in the first years of life, and recent magnetic resonance imaging studies have discovered brain growth abnormalities that precede and overlap with the onset of these symptoms. Recent postmortem studies of the autistic brain provide evidence of cellular abnormalities and processes that may underlie the recently discovered early brain overgrowth and arrest of growth that marks the first years of life in autism. Alternative origins and time tables for these cellular defects and processes are discussed. These cellular and growth abnormalities are most pronounced in frontal, cerebellar, and temporal structures that normally mediate the development of those same higher order social, emotional, speech, language, speech, attention, and cognitive functions that characterize autism. Cellular and growth pathologies are milder and perhaps nonexistent in other structures (e.g., occipital cortex), which are known to mediate functions that are often either mildly affected or entirely unaffected in autistic patients. It is argued that in autism, higher order functions largely fail to develop normally in the first place because frontal, cerebellar, and temporal cellular and growth pathologies occur prior to and during the critical period when these higher order neural systems first begin to form their circuitry. It is hypothesized that microstructural maldevelopment results in local and short distance overconnectivity in frontal cortex that is largely ineffective and in a failure of long-distance cortical–cortical coupling, and thus a reduction in frontal–posterior reciprocal connectivity. This altered circuitry impairs the essential role of frontal cortex in integrating information from diverse functional systems (emotional, sensory, autonomic, memory, etc.) and providing context-based and goal-directed feedback to lower level systems.
