Methods for Performing Atomic Force Microscopy Imaging of Clay Minerals in Aqueous Solutions

Three methods were developed that allow for the imaging of any clay mineral in aqueous solutions with atomic force microscopy (AFM). The methods involve fixing the particles onto special substrates that do not complicate the imaging process, but hold the particles sufficiently so that they do not move laterally or float away during imaging. Two techniques depend on electrostatic attraction under circumneutral pH conditions, between the negatively charged clay particles and the high point of zero charge substrate (either aluminum oxide or polyethyleneimine-coated mica) whereas the third technique depends on adhesion to a thermoplastic film. The first electrostatic technique involves a polished single crystal α-Al2O3 (sapphire) substrate. This was used successfully as a substrate for clay-sized minerals with high permanent layer charge localized on the basal planes (phlogopite and vermiculite) and when the AFM was operated in TappingMode to limit the lateral forces between the probe tip and the particles. However, electrostatic attraction between the sapphire surface and clay minerals such as smectite and kaolinite (low or no permanent layer charge) is not sufficiently strong to adequately fix the particles for imaging. The second electrostatic technique involves a polyethyleneimine-coated mica surface designed to immobilize a larger variety of clay minerals (phlogopite, vermiculite, montmorillonite, and kaolinite), and in this technique weak bonding between the clay and the organic film is also a factor. The third technique, which does not depend on electrostatic attraction, fixes clay particles into the surface of a thermoplastic adhesive called Tempfix. This has proven useful for fixing and imaging relatively large clay particles with well-defined morphology. The Tempfix mount also requires imaging in TappingMode because the Tempfix is relatively soft.
Emotions and behaviours of child and adolescent psychiatric patients during the COVID-19 pandemic

Background
Previous pandemics have had negative effects on mental health, but there are few data on children and adolescents who were receiving ongoing psychiatric treatment.
AimsTo study changes in emotions and clinical state, and their predictors, during the COVID-19 pandemic in France.
MethodWe administered (by interview) the baseline Youth Self-Report version of the CoRonavIruS Health Impact Survey v0.3 (CRISIS, French translation) to 123 adolescent patients and the Parent/Caregiver version to evaluate 99 child patients before and during the first ‘lockdown’. For 139 of these patients who received ongoing treatment in our centre, treating physicians retrospectively completed longitudinal global ratings for five time periods, masked to CRISIS ratings.
ResultsThe main outcome measure was the sum of eight mood state items, which formed a single factor in each age group. Overall, this score improved for each age group during the first lockdown. Clinician ratings modestly supported this result in patients without intellectual disability or autism spectrum disorder. Improvement of mood states was significantly associated with perceived improvement in family relationships in both age groups.
ConclusionsConsistent with previous studies of clinical cohorts, our patients had diverse responses during the pandemic. Several factors may have contributed to the finding of improvement in some individuals during the first lockdown, including the degree of family support or conflict, stress reduction owing to isolation, limitations of the outcome measures and/or possible selection bias. Ongoing treatment may have had a protective effect. Clinically, during crises additional support may be needed by families who experience increased conflict or who care for children with intellectual disability.

Depression is highly prevalent in people living with HIV (PLWH) and has negative consequences for daily life and care. We evaluated for the first time the acceptability, feasibility and benefits of group interpersonal therapy (IPT), combined with a task-shifting approach, to treat depression in PLWH in Senegal. PLWH with depression received group IPT following the World Health Organization protocol. Acceptability and feasibility criteria were defined from the literature data. The PHQ-9, the WHODAS, and the 12-item-stigma scale were used, pre- and post-treatment, including a 3-month follow-up, to assess depressive symptom severity, functioning and stigma, respectively. General linear mixed models were used to describe changes in outcomes over time. Of 69 participants, 60 completed group IPT. Refusal to enroll and dropout rates were 6.6 and 12.7%, respectively. Ninety-seven percent of participants attended at least seven out of eight sessions. Patients and facilitators endorsed group IPT, with willingness to recommend it. Depressive symptoms and disability improved drastically and sustainably. We showed that group IPT is well accepted and feasible in Senegal as treatment for depression in PLWH. Combined with a task-shifting approach, it can narrow the gap in mental health treatment. Implementation may be enhanced by refining patient identification procedures and increasing treatment accessibility.

The understanding of the genetic architecture of psychiatric disorders has made significant advances in the last decade and some scientific findings can now be translated into clinical practice. The rise of genetic testing and the awareness of patients and their families motivate psychiatrists to examine this approach. The COST Action EnGagE (CA17130) is promoting these developments in Europe. Whereas the findings of common variants are the domain of research, screening for rare variants at the genome-wide level is already applicable in clinical practice. It is now possible to return meaningful results to the individual to help him/her understanding the disease and the comorbidities, to guide treatment and to perform genetic counseling. In this presentation, we will give meaningful examples for psychiatric practice. For instance, around one-third of the patients diagnosed with autism spectrum disorder can benefit from a molecular diagnosis (fragile X syndrome, SHANK3 deletion…). Microdeletion or microduplication may explain a fraction of schizophrenic cases (e.g. del22q11). Identification of rare variants causing the disease may decrease the stigma and feeling of guilt often reported by patients and families. This could also help to detect and manage other comorbidities. It is expected that treatment guidelines and clinical trials would be developed in the near future for patients carrying a rare variant, opening the way to personalized psychiatry. Finally, this effort has a huge impact on the family, by enhancing genetic counseling in psychiatry. The rise of psychiatric genetics might align our field more closely with the other medical specialties.

The $\Omega $ numbers—the halting probabilities of universal prefix-free machines—are known to be exactly the Martin-Löf random left-c.e. reals. We show that one cannot uniformly produce, from a Martin-Löf random left-c.e. real $\alpha $ , a universal prefix-free machine U whose halting probability is $\alpha $ . We also answer a question of Barmpalias and Lewis-Pye by showing that given a left-c.e. real $\alpha $ , one cannot uniformly produce a left-c.e. real $\beta $ such that $\alpha - \beta $ is neither left-c.e. nor right-c.e.

Distributed models and a good knowledge of the catchment studied are required to assess mitigation measures for nitrogen (N) pollution. A set of alternative scenarios (change of crop management practices and different strategies of landscape management, especially different sizes and distribution of set-aside areas) were simulated with a fully distributed model in a small agricultural catchment. The results show that current practices are close to complying with current regulations, which results in a limited effect of the implementation of best crop management practices. The location of set-aside zones is more important than their size in decreasing nitrate fluxes in stream water. The most efficient location is the lower parts of hillslopes, combining the dilution effect due to the decrease of N input per unit of land and the interception of nitrate transferred by sub-surface flows. The main process responsible for the interception effect is probably uptake by grassland and retention in soils since the denitrification load tends to decrease proportionally to N input and, for the scenarios considered, is lower in the interception scenarios than in the corresponding dilution zones.

Summary
The rotation rate of a planet is a fundamental parameter, no less than its mass or composition, and planetary investigators require this rate to assess various other phenomena such as planetary wind speeds, internal and atmospheric models, ring dynamics and so forth. Saturn presents a conundrum, however, because none of its various planetary periods indicates the “true” rotation of the planet. Thus, although the planet displays an abundance of periodicities near 10.7 hours, the exact rotation period of Saturn is unknown. In the magnetosphere, “planetary-period oscillations” (PPOs) appear in charged particles, magnetic fields, energetic neutral atoms, radio emissions and motions of the plasma sheet and magnetopause. In Saturn’s rings, the spoke phenomenon can exhibit periodicities near 10.7 hours, and ring phenomena themselves may be related to the interior rotation of the planet. In the high-latitude ionosphere, modulations near this period appear in auroral motions and intensities. In the upper atmosphere, some cloud features rotate near this period, although wind speeds are generally faster, and the well-known polar hexagon rotates with a period close to 10.7 hours. Some of the magnetospheric/ionospheric oscillations differ in the northern and southern hemispheres and their periods do not remain constant, sometimes varying on long time scales of a year or longer and sometimes on much shorter time scales. These variations in the period argue against a cause related to changes interior to Saturn, and because the magnetic and spin axes of Saturn are reported to be axisymmetric (unlike those of any other known planet), Saturn’s periodicities cannot be explained as “wobble” caused by a geometric tilt or by a nondipolar magnetic anomaly. Several models have been proposed to account for the observed periodicities, including rotating atmospheric vortices, periodic plasma releases and a flapping magnetodisk, but none can satisfactorily explain all of Saturn’s periodicities nor their common origin, and none can determine the exact rotation rate of the planet. This chapter reviews Saturn’s periodicities, theories thereof, and how they might be used to determine the elusive rotation rate of the planet.

Summary
Introduction
MicroRNAs (miRNAs) are small, single-stranded RNAs of about 22 nucleotides involved in gene regulation by binding to 3′ untranslated regions (UTRs) of messenger RNAs (mRNAs) (Bartel, 2004). By bringing the RNA-induced silencing complex (RISC) to the mRNAs, they enable gene expression inhibition (gene silencing), either by affecting protein translation or by destabilizing mRNAs through deadenylation or decapping (Fabian et al., 2010). Target mRNAs are recognized by miRNAs through Watson–Crick matching between the nucleotides two to seven of the 5′ end of miRNAs (seed sequences), and complementary sequences called seed sites in the 3′ UTR of mRNAs (Bartel, 2009). Gene silencing by miRNAs is an important mechanism in physiological processes, and its deregulation can lead to complex diseases such as cancer (Garzon et al., 2006).
Complex diseases that are heritable are commonly analysed by studying genomic DNA variants, such as single nucleotide polymorphisms (SNPs), which are a change of one nucleotide that occurs in more than 1% in a population (Frazer et al., 2009). A SNP can take several forms called alleles. Because recombination events between closely located SNPs are less likely than recombination between SNPs that are located far apart on a chromosome, alleles at close SNPs often co-occur, or correlate, in which case the SNPs are said to be in linkage disequilibrium (LD; Reich et al., 2001; Clague et al., 2010).
SNPs in the coding sequence of mRNAs have been well studied for their role in changing the amino-acid chain, as it may result in protein isoforms with affected function, leading to phenotypic differences and also disease. Nevertheless, SNPs can also occur in non-coding regions of the genome such as the 3′ UTR of mRNAs, which harbors many functional sequence elements involved in gene regulation. One type of functional element that can be disrupted by SNPs is the miRNA target site. SNPs in miRNA target sites (miRSNPs) can change the affinity between the miRNA seed sequence and its target mRNA, resulting in deregulation of gene expression (Figure 10.1), and possibly in phenotype differences and diseases (Sethupathy and Collins, 2008).
One classic example that a miRSNP determines phenotype is the single nucleotide change in the myostatin gene (GDF8) of Texel sheep (Clop et al., 2006).

We analysed Mycobacterium tuberculosis strains from children, hospitalized from January 2004 to July 2008 in the largest paediatric hospital complex in Cambodia. Specimens were tested for drug susceptibility and genotypes. From the 260 children, 161 strains were available. The East African-Indian genotype family was the most common (59·0%), increasing in frequency with distance from the Phnom Penh area, while the frequency of the Beijing genotype family strains decreased. The drug resistance pattern showed a similar geographical gradient: lowest in the northwest (4·6%), intermediate in the central (17·1%), and highest in the southeastern (30·8%) parts of the country. Three children (1·9%) had multidrug-resistant tuberculosis. The Beijing genotype and streptomycin resistance were significantly associated (P < 0·001). As tuberculosis in children reflects recent transmission patterns in the community, multidrug resistance levels inform about the current quality of the tuberculosis programme.
Detection of methicillin-resistant Staphylococcus aureus carrying the mecC gene in human samples in Slovenia

Following the recognition of a mecC MRSA isolate from a patient hospitalized in the northeastern region of Slovenia, a national collection of 395 community-associated methicillin-resistant Staphylococcus aureus (CA-MRSA) isolates from 2006 to 2013 was screened. An additional six mecC MRSA strains were found and characterized as spa types t843, t9397 and t10009, and multilocus sequence type ST130. The low oxacillin minimum inhibitory concentrations and absence of the mecA gene make recognition of these MRSA strains problematical for diagnostic laboratories. In such strains the presence of mecC should be determined.

We have written a theoretical description of one-way and two-way satellite time and frequency transfer and developed a model of the micro-wave link in the frame of the ACES/PHARAO mission. This is used to write a data analysis software and a simulation to test it. A very short description of the mission and of the micro-wave link is given here. Summary
Neutron stars generate powerful winds of relativistic particles that form bright synchrotron nebulae around them. Polarimetry provides a unique insight into the geometry and magnetic configuration of the wind, but high-energy measurements have failed until recently. The INTEGRAL-IBIS telescope has been used in its Compton mode to search for linearly polarized emission for energies above 200 keV from the Crab nebula. The asymmetries in the instrument response are small and we obtain evidence for a strongly polarized signal at an angle parallel to the pulsar rotation axis. This result confirms the detection recently reported by, and extends the polarization measure for all the pulsar's phases. We also report the recent observation of a variable polarization signal from the long GRB 041219A. The achieved sensitivity opens a new window for polarimetric studies at energies above 200 keV.
Introduction
Gamma-ray polarimetry has been possible with Compton telescopes since the 1970s. Photons that are Compton scattered between two detectors follow an azimuthal distribution around the source direction that allows quantification of the degree and direction of linear polarization because the photon is preferentially scattered in a plane at right angles to its incident electric vector. It was unsuccessful until recently because of intrinsic asymmetries in the detector response and nonuniformities in the large background signals. They induce pseudo-polarimetric signals, even from an unpolarized source, that limit the sensitivity to any detection.

The presented study deals with the influence of the contacts materials on the evolution of the arc in low voltage circuit breakers. The commutation from the mobile contact onto the electrode is especially investigated in order to improve the breaking device performances. Hence, to preserve the qualities of the contact, the arc must leave this area as soon as possible. To undertake this study, several diagnostics are implemented. A measuring device is composed of magnetic induction sensors located outside the chamber. It allows the determination of the position and the volume of the arc during the breaking. Optical (ultra-speed camera) and electrical measurements are also performed with the technical support of Schneider Electric. Different current values (assumed peak current of 3 kA, 8 kA and 12 kA) and different natures of materials composing the electrical contacts have been tested. Copper contacts, different contacts on silver base obtained with different processes have been used. Experimental results show the influence of these various parameters on the arc volume and on the transfer of the arc towards the quenching chamber. Indeed, different dynamic behaviors are observed during the current transfer phase.

Chemically Peculiar (CP) stars have been the subject of systematic research for more than 50 years. With the discovery of pulsation of some of the cool CP stars, and the availability of advanced spectropolarimetric instrumentation and high signal-to-noise, high resolution spectroscopy, a new era of CP star research emerged about 20 years ago. Together with the success in ground-based observations, new space projects are developed that will greatly benefit future investigations of these unique objects. In this contribution we will give an overview of some interesting results obtained recently from ground-based observations and discuss the future outstanding Gaia space mission and its impact on CP star research.

Objective.
To describe an outbreak of extended-spectrum β-lactamase (ESBL)–producing Klebsiella pneumoniae in the intensive care units (ICUs) of a hospital and the impact of routine and reinforced infection control measures on interrupting nosocomial transmission.
Design.Outbreak report.
Setting.A 31-bed intensive care department (composed of 4 ICUs) in a university hospital in Belgium.
Intervention.After routine infection control measures (based on biweekly surveillance cultures and contact precautions) failed to interrupt a 2-month outbreak of ESBL-producing K. pneumoniae, reinforced infection control measures were implemented. The frequency of surveillance cultures was increased to daily sampling. Colonized patients were moved to a dedicated 6-bed ICU, where they received cohorted care with the support of additional nurses. Two beds were closed to new admissions in the intensive care department. Meetings between the ICU and infection control teams were held every day. Postdischarge disinfection of rooms was enforced. Broad-spectrum antibiotic use was discouraged.
Results.Compared with a baseline rate of 0.44 cases per 1,000 patient-days for nosocomial transmission, the incidence peaked at 11.57 cases per 1,000 patient-days (October and November 2005; rate ratio for peak vs baseline, 25.46). The outbreak involved 30 patients, of whom 9 developed an infection. Bacterial genotyping disclosed that the outbreak was polyclonal, with 1 predominant genotype. Reinforced infection control measures lasted for 50 days. After the implementation of these measures, the incidence fell to 0.08 cases per 1,000 patient-days (rate ratio for after the outbreak vs during the outbreak, 0.11).
Conclusion.These data indicate that, in an intensive care department in which routine screening and contact precautions failed to prevent and interrupt an outbreak of ESBL-producing K. pneumoniae, reinforced infection control measures controlled the outbreak without major disruption of medical care.
