449 Progression of silica-induced pulmonary fibrosis is arrested after selective ablation of Col1a1+ fibroblasts

OBJECTIVES/GOALS: Silicosis is a highly fatal progressive fibrotic disease of the lungs characterized by accumulation and persistence of fibroblasts that excessively deposit Collagen1a1. We sought to eliminate Collagen1a1-expressing fibroblasts through a targeted genetic ablation strategy and hypothesized that this would arrest the progression of Silicosis. METHODS/STUDY POPULATION: Silicosis was induced with a single intratracheal (i.t.) instillation of silica particles ( RESULTS/ANTICIPATED RESULTS: Targeted ablation of Col1a1+ fibroblast in established Silicosis resulted in a decrease in: 1) Col1a1+ fibroblasts by flow cytometry and within fibrotic nodules by immunofluorescent staining, 2) total lung collagen content by histology and hydroxyproline assay, 3) tissue-associated disease by microCT and an increase in arterial oxygen saturation by pulse oximetry. Cessation of targeted Col1a1+ fibroblast ablation resulted in a rebound effect in Silicosis disease progression. Following ablation, Col1a1+ fibroblasts expanded by proliferation (Ki67+) and total lung collagen levels returned to pre-ablation levels. DISCUSSION/SIGNIFICANCE: Silicosis is a often fatal disease with no FDA approved therapies. These results suggest that targeted loss of Col1a1+ fibroblasts in Silicosis is sufficient to arrest disease progression. Thus, it is essential to understand how targeted loss of pro-fibrotic fibroblasts can alter disease progression as a tool to develop novel therapeutic strategies.

Lairage staff at 11 abattoirs were asked to rate which producers regularly provided pigs which were ‘easy’ (EH) or ‘difficult’ (DH) to handle, on a scale of one (very DH) to five (very EH). A postal questionnaire, dealing with various aspects of post-weaning farm management, was then given to the four or five producers sending the most EH and the four or five producers sending the most DH pigs to each abattoir. Of 105 questionnaires sent, information on 26 EH and 27 DH systems was returned. The median number of replies per abattoir was two for both EH and DH systems. In most systems (77%) pigs experienced three or four housing stages from weaning to slaughter. In each of the first five housing stages, more EH pigs had access to daylight (mean of 86% ± 11.5 (SD)) than DH pigs (mean of 64% ± 10.1 (SD), P < 0.05, two-sample t test). More EH systems provided straw in the first three housing stages, although over all stages the difference was not significant. During housing stage two, the difference in provision of straw between the systems was most marked, with 58 per cent of EH and 27 per cent of DH systems providing straw. Distance walked between housing stages three to four and four to five was significantly greater for EH compared to DH systems (EH mean of 64m ± 24.1 (SD), versus DH mean of 22m ± 14.0 (SD), and EH mean of 73m ±17.2 (SD), versus DH mean of 23m ± 8.5 (SD), P <0.001 and 0.01 respectively, two-sample t test). At loading for pre-slaughter transport, moving from daylight to daylight conditions occurred in 65 per cent of EH and 25 per cent of DH systems. Overall, the results provide circumstantial evidence that environmental factors can affect ease of handling, and hence pig welfare during pre-slaughter transport and lairage.

The role that vitamin D plays in pulmonary function remains uncertain. Epidemiological studies reported mixed findings for serum 25-hydroxyvitamin D (25(OH)D)–pulmonary function association. We conducted the largest cross-sectional meta-analysis of the 25(OH)D–pulmonary function association to date, based on nine European ancestry (EA) cohorts (n 22 838) and five African ancestry (AA) cohorts (n 4290) in the Cohorts for Heart and Aging Research in Genomic Epidemiology Consortium. Data were analysed using linear models by cohort and ancestry. Effect modification by smoking status (current/former/never) was tested. Results were combined using fixed-effects meta-analysis. Mean serum 25(OH)D was 68 (sd 29) nmol/l for EA and 49 (sd 21) nmol/l for AA. For each 1 nmol/l higher 25(OH)D, forced expiratory volume in the 1st second (FEV1) was higher by 1·1 ml in EA (95 % CI 0·9, 1·3; P<0·0001) and 1·8 ml (95 % CI 1·1, 2·5; P<0·0001) in AA (Prace difference=0·06), and forced vital capacity (FVC) was higher by 1·3 ml in EA (95 % CI 1·0, 1·6; P<0·0001) and 1·5 ml (95 % CI 0·8, 2·3; P=0·0001) in AA (Prace difference=0·56). Among EA, the 25(OH)D–FVC association was stronger in smokers: per 1 nmol/l higher 25(OH)D, FVC was higher by 1·7 ml (95 % CI 1·1, 2·3) for current smokers and 1·7 ml (95 % CI 1·2, 2·1) for former smokers, compared with 0·8 ml (95 % CI 0·4, 1·2) for never smokers. In summary, the 25(OH)D associations with FEV1 and FVC were positive in both ancestries. In EA, a stronger association was observed for smokers compared with never smokers, which supports the importance of vitamin D in vulnerable populations.

Preterm birth affects over 12% of all infants born in the United States; yet the biology of early delivery remains unclear, including whether epigenetic mechanisms are involved. We examined associations of maternal and umbilical cord blood long interspersed nuclear element-1 (LINE-1) DNA methylation with length of gestation and odds of preterm birth in singleton pregnancies in Project Viva. In white blood cells from maternal blood during first trimester (n = 914) and second trimester (n = 922), and from venous cord blood at delivery (n = 557), we measured LINE-1 by pyrosequencing [expressed as %5 methyl cytosines within the LINE-1 region analyzed (%5mC)]. We ran linear regression models to analyze differences in gestation length, and logistic models for odds of preterm birth (<37 v. ⩾37 weeks’ gestation), across quartiles of LINE-1. Mean (s.d.) LINE-1 levels were 84.3 (0.6), 84.5 (0.4) and 84.6 (0.7) %5mC for first trimester, second trimester and cord blood, respectively. Mean (s.d.) gestational age was 39.5 (1.8) weeks, and 6.5% of infants were born preterm. After adjustment for maternal age, race/ethnicity, body mass index, education, smoking status and fetal sex, women with the highest v. lowest quartile of first trimester LINE-1 had longer gestations [0.45 weeks (95% CI 0.12, 0.78)] and lower odds of preterm birth [OR 0.40 (0.17, 0.94)], whereas associations with cord blood LINE-1 were in the opposite direction (−0.45 weeks, −0.83, −0.08) and [OR 4.55 (1.18, 17.5)]. In conclusion, higher early pregnancy LINE-1 predicts lower risk of preterm birth. In contrast, preterm birth is associated with lower LINE-1 in cord blood.

Objective:
To evaluate the risk of phlebitis associated with chlorhexidine-coated polyurethane catheters in peripheral veins.
Design:A randomized, double-blinded trial comparing chlorhexidine-coated polyurethane catheters with uncoated polyurethane catheters.
Setting:A university hospital.
Patients:Adult medicine and surgery patients.
Interventions:Certified registered nurse anesthetists or an infusion team consisting of nurses and physicians inserted the catheters. Catheter insertion sites were scored twice daily for evidence of phlebitis. At the time catheters were removed, a quantitative blood culture was performed, and catheters were sonicated for quantitative culture.
Results:Of 221 evaluable catheters, phlebitis developed in 18 (17%) of 105 coated catheters, compared to 27 (23%) of 116 uncoated catheters (relative risk [RR], 0.74; 95% confidence interval [CI95], 0.43-1.26; P=.32). By survival analysis, chlorhexidine-coated catheters had a lower risk of phlebitis during the first 3 days (P=.06), but not when all catheters were considered in both patient groups (P=.31). In the absence of catheter colonization, the incidence of phlebitis was 21% (16/76) and 24% (20/86) for coated and uncoated catheters, respectively (P=.85), whereas in the presence of catheter colonization, the incidence of phlebitis was 14% (1/7) and 80% (4/5) for coated and uncoated catheters, respectively (RR, 0.18; CI95, 0.03-1.15; P=.07).
Conclusion:The risk of phlebitis in the presence of catheter colonization was 82% lower for chlorhexidinecoated polyurethane catheters compared to otherwise identical uncoated catheters.
Do 95-kg live weight pigs choose to stand, sit or lie during short journeys?

Although the need for pigs to lie down on long journeys is not in question, there is evidence that they may not choose to do so on journeys of less than 3h (Hunter et al., 1994). These observations were undertaken to determine how pigs in the 95 to 100 kg weight range behaved on short journeys. A three-tier floating-deck vehicle with weld-mesh flooring was used because the popularity of three-deck vehicles is increasing (Riches et al., 1996).

There is currently no published information on the conditions under which young pigs are transported. European Directive 95/29/EC concerns the protection of animals during transport but recommends no stocking density for pigs moved by road other than those of 100 kg live weight. A survey of 97 farms showed store pigs accounted for 64% of all inter-farm journeys. The most common weights were 26 to 30 kg. The mean stocking density for transport was 162kg/m2 (range 72-214 (s.d. 25) kg/m2). The mean journey time was 165 min and the distance 228 km. This experiment was designed within the range of stocking densities and distances travelled found in current commercial practice. Pigs were transported at high, medium and low densities. Posture and heart rate during transport were recorded.

The GALLEX collaboration aims at the detection of solar neutrinos in a radiochemical experiment employing 30 tons of Gallium in form of concentrated aqueous Gallium-chloride solution. The detector is primarily sensitive to the otherwise inaccessible pp-neutrinos. Details of the experiment have been repeatedly described before [1-7]. Here we report the present status of implementation in the Laboratori Nazionali del Gran Sasso (Italy). So far, 12.2 tons of Gallium are at hand. The present status of development allows to start the first full scale run at the time when 30 tons of Gallium become available. This date is expected to be January, 1990.
