Prefrontal cortical thinning links to negative symptoms in schizophrenia via the ENIGMA consortium

Background
Our understanding of the complex relationship between schizophrenia symptomatology and etiological factors can be improved by studying brain-based correlates of schizophrenia. Research showed that impairments in value processing and executive functioning, which have been associated with prefrontal brain areas [particularly the medial orbitofrontal cortex (MOFC)], are linked to negative symptoms. Here we tested the hypothesis that MOFC thickness is associated with negative symptom severity.
MethodsThis study included 1985 individuals with schizophrenia from 17 research groups around the world contributing to the ENIGMA Schizophrenia Working Group. Cortical thickness values were obtained from T1-weighted structural brain scans using FreeSurfer. A meta-analysis across sites was conducted over effect sizes from a model predicting cortical thickness by negative symptom score (harmonized Scale for the Assessment of Negative Symptoms or Positive and Negative Syndrome Scale scores).
ResultsMeta-analytical results showed that left, but not right, MOFC thickness was significantly associated with negative symptom severity (βstd = −0.075; p = 0.019) after accounting for age, gender, and site. This effect remained significant (p = 0.036) in a model including overall illness severity. Covarying for duration of illness, age of onset, antipsychotic medication or handedness weakened the association of negative symptoms with left MOFC thickness. As part of a secondary analysis including 10 other prefrontal regions further associations in the left lateral orbitofrontal gyrus and pars opercularis emerged.
ConclusionsUsing an unusually large cohort and a meta-analytical approach, our findings point towards a link between prefrontal thinning and negative symptom severity in schizophrenia. This finding provides further insight into the relationship between structural brain abnormalities and negative symptoms in schizophrenia.

This study was motivated by the focal tendency of atherosclerotic plaque to appear near arterial junctions. A two-dimensional bifurcation was selected to provide preliminary information on branch flow dynamics. Superseding previous steady flow estimates in similar branches, digital solutions for two flux waves have been obtained to reveal temporal and spatial distributions of shear and separation in pulsatile flow. Marked differences from steady flow are illustrated as well as variation due to flux-wave shape.

Strong optical limiting and large two-photon absorptivities are reported for a class of bisdonor diphenylpolyene derivatives with varying polyene bridge lengths. These molecules exhibit strong optical limiting using nanosecond pulses over a broad spectral range. Bis(diphenylamino)stilbene exhibits a 90 nm red shift of its optical limiting band, and only a minimal shift of about 13 nm of its lowest one-photon electronic absorption band relative to bis(di-n-butylamino)stilbene. This suggests a potential for broadband optical limiting with high transparency using mixtures of such compounds. Pulse width dependent nonlinear transmission measurements suggest that two-photon pumped excited-state absorption contributes significantly to the limiting of nanosecond pulses.

Molecules exhibiting strong two-photon absorption hold great potential for a wide range of applications including two-photon fluorescence imaging, three-dimensional (3D) optical data storage, and 3D microfabrication. We have observed two-photon absorptivities as high as 1500×10− 50 cm4 s/photon in bis-donor diphenylpolyene derivatives that are correlated to simultaneous charge transfer from the end groups to the polyene bridge in the molecule. Many of these molecules are also excellent photoexcitable electron donors that can initiate charge-transfer reactions with acrylate monomers. Marcus theory is used to describe the efficiency of these charge-transfer reactions. Polymerization rates have also been measured and we show that these twophoton chromophores display increased sensitivity and recording speed over conventional UV photo-initiators. The fabrication of complex, three-dimensional structures by twophoton polymerization is demonstrated and discussed in the context of advanced photonic applications.

Cellular interactions at the interface between bone and a prosthetic device plays an important role in total joint reconstruction success or failure. Aseptic loosening is a typical long-term complication encountered due to a failure at this interface. A number of factors have been shown to influence cell mitogenic and morphological behavior including load and stability [1] surface topography and surface chemistry [2]. The selection of suitable surface coatings on a prosthetic device would be facilitated if the nature of the interaction of bone cells to surface could be predicted from appropriate behavior of cells in tissue culture [3]. Molecular biology techniques now enable investigators to study cell-substrate interactions and gene expression at the mRNA level.
