Mechanisms of glyphosate resistance in common ragweed (Ambrosia artemisiifolia): patterns of absorption, translocation, and metabolism

Glyphosate’s efficacy is influenced by the amount absorbed and translocated throughout the plant to inhibit 5-enolpyruvyl shikimate-3-phosphate synthase (EPSPS). Glyphosate resistance can be due to target-site (TS) or non–target site (NTS) resistance mechanisms. TS resistance includes an altered target site and gene overexpression, while NTS resistance includes reduced absorption, reduced translocation, enhanced metabolism, and exclusion/sequestration. The goal of this research was to elucidate the mechanism(s) of glyphosate resistance in common ragweed (Ambrosia artemisiifolia L.) from Ontario, Canada. The resistance factor for this glyphosate-resistant (GR) A. artemisiifolia biotype is 5.1. No amino acid substitutions were found at positions 102 or 106 of the EPSPS enzyme in this A. artemisiifolia biotype. Based on [14C]glyphosate studies, there was no difference in glyphosate absorption or translocation between glyphosate-susceptible (GS) and GR A. artemisiifolia biotypes. Radio-labeled glyphosate metabolites were similar for GS and GR A. artemisiifolia 96 h after application. Glyphosate resistance in this A. artemisiifolia biotype is not due to an altered target site due to amino acid substitutions at positions 102 and 106 in the EPSPS and is not due to the NTS mechanisms of reduced absorption, reduced translocation, or enhanced metabolism.

Data from rock shelters in southern Belize show evidence of tool making, hunting, and aquatic resource exploitation by 10,500 cal b.c.; the shelters functioned as mortuary sites between 7600 and 2000 cal b.c. Early Holocene contexts contain stemmed and barbed bifaces as part of a tradition found broadly throughout the neotropics. After around 6000 cal b.c., bifacial tools largely disappear from the record, likely reflecting a shift to increasing reliance on plant foods, around the same time that the earliest domesticates appear in the archaeological record in the neotropics. We suggest that people living in southern Belize maintained close ties with neighbors to the south during the Early Holocene, but lagged behind in innovating new crops and farming technologies during the Middle Holocene. Maize farming in Belize intensified between 2750–2050 cal b.c. as maize became a dietary staple, 1000–1300 years later than in South America. Overall, we argue from multiple lines of data that the Neotropics of Central and South America were an area of shared information and technologies that heavily influenced cultural developments in southeastern Mesoamerica during the Early and Middle Holocene.
Real-world evidence use in assessments of cancer drugs by NICE

Objective
To establish how real-world evidence (RWE) has been used to inform single technology appraisals (STAs) of cancer drugs conducted by the National Institute for Health and Care Excellence (NICE).
MethodsSTAs published by NICE from April 2011 to October 2018 that evaluated cancer treatments were reviewed. Information regarding the use of RWE to directly inform the company-submitted cost-effectiveness analysis was extracted and categorized by topic. Summary statistics were used to describe emergent themes, and a narrative summary was provided for key case studies.
ResultsMaterials for a total of 113 relevant STAs were identified and analyzed, of which nearly all (96 percent) included some form of RWE within the company-submitted cost-effectiveness analysis. The most common categories of RWE use concerned the health-related quality of life of patients (71 percent), costs (46 percent), and medical resource utilization (40 percent). While sources of RWE were routinely criticized as part of the appraisal process, we identified only two cases where the use of RWE was overtly rejected; hence, in the majority of cases, RWE was accepted in cancer drug submissions to NICE.
DiscussionRWE has been used extensively in cancer submissions to NICE. Key criticisms of RWE in submissions to NICE are seldom regarding the use of RWE in general; instead, these are typically concerned with specific data sources and the applicability of these to the decision problem. Within an appropriate context, RWE constitutes an extremely valuable source of information to inform decision making; yet the development of best practice guidelines may improve current reporting standards.

Background
We examined demographic, clinical, and psychological characteristics of a large cohort (n = 368) of adults with dissociative seizures (DS) recruited to the CODES randomised controlled trial (RCT) and explored differences associated with age at onset of DS, gender, and DS semiology.
MethodsPrior to randomisation within the CODES RCT, we collected demographic and clinical data on 368 participants. We assessed psychiatric comorbidity using the Mini-International Neuropsychiatric Interview (M.I.N.I.) and a screening measure of personality disorder and measured anxiety, depression, psychological distress, somatic symptom burden, emotional expression, functional impact of DS, avoidance behaviour, and quality of life. We undertook comparisons based on reported age at DS onset (<40 v. ⩾40), gender (male v. female), and DS semiology (predominantly hyperkinetic v. hypokinetic).
ResultsOur cohort was predominantly female (72%) and characterised by high levels of socio-economic deprivation. Two-thirds had predominantly hyperkinetic DS. Of the total, 69% had ⩾1 comorbid M.I.N.I. diagnosis (median number = 2), with agoraphobia being the most common concurrent diagnosis. Clinical levels of distress were reported by 86% and characteristics associated with maladaptive personality traits by 60%. Moderate-to-severe functional impairment, high levels of somatic symptoms, and impaired quality of life were also reported. Women had a younger age at DS onset than men.
ConclusionsOur study highlights the burden of psychopathology and socio-economic deprivation in a large, heterogeneous cohort of patients with DS. The lack of clear differences based on gender, DS semiology and age at onset suggests these factors do not add substantially to the heterogeneity of the cohort.
The development of a consensus-based nutritional pathway for infants with CHD before surgery using a modified Delphi process

Introduction
Despite improvements in the medical and surgical management of infants with CHD, growth failure before surgery in many infants continues to be a significant concern. A nutritional pathway was developed, the aim of which was to provide a structured approach to nutritional care for infants with CHD awaiting surgery.
Materials and methodsThe modified Delphi process was development of a nutritional pathway; initial stakeholder meeting to finalise draft guidelines and develop questions; round 1 anonymous online survey; round 2 online survey; regional cardiac conference and pathway revision; and final expert meeting and pathway finalisation.
ResultsPaediatric Dietitians from all 11 of the paediatric cardiology surgical centres in the United Kingdom contributed to the guideline development. In all, 33% of participants had 9 or more years of experience working with infants with CHD. By the end of rounds 1 and 2, 76 and 96% of participants, respectively, were in agreement with the statements. Three statements where consensus was not achieved by the end of round 2 were discussed and agreed at the final expert group meeting.
ConclusionsNutrition guidelines were developed for infants with CHD awaiting surgery, using a modified Delphi process, incorporating the best available evidence and expert opinion with regard to nutritional support in this group.
Glyphosate-Resistant Giant Ragweed (Ambrosia trifida) Control in Dicamba-Tolerant Soybean

Glyphosate-resistant (GR) giant ragweed has been confirmed in Ontario, Canada. Giant ragweed is an extremely competitive weed and lack of control in soybean will lead to significant yield losses. Seed companies have developed new herbicide-resistant (HR) crop cultivars and hybrids that stack multiple HR traits. The objective of this research was to evaluate the efficacy of glyphosate and glyphosate plus dicamba tank mixes for the control of GR giant ragweed under Ontario environmental conditions in dicamba-tolerant (DT) soybean. Three field trials were established over a 2-yr period (2010 and 2011) on farms near Windsor and Belle River, ON. Treatments included glyphosate (900 g ae ha−1), dicamba (300 g ae ha−1), and dicamba (600 g ha−1) applied preplant (PP), POST, or sequentially in various combinations. Glyphosate applied PP, POST, or sequentially provided 22 to 68%, 40 to 47%, and 59 to 95% control of GR giant ragweed and reduced shoot dry weight 26 to 80%, 16 to 50%, and 72 to 98%, respectively. Glyphosate plus dicamba applied PP followed by glyphosate plus dicamba applied POST consistently provided 100% control of GR giant ragweed. DT soybean yield correlated with GR giant ragweed control. This is the first report in Canada of weed control in DT soybean, specifically for the control of GR giant ragweed. Results indicate that the use of dicamba in DT soybean will provide an effective option for the control of GR giant ragweed in Ontario.
Control of Glyphosate-Resistant Horseweed (Conyza canadensis) with Dicamba Applied Preplant and Postemergence in Dicamba-Resistant Soybean

Herbicide-resistant crops, such as glyphosate-resistant (GR) soybean, allow for broad-spectrum, flexible weed control with minimal crop injury; however, the development of GR weeds, such as horseweed, has forced reliance on alternative herbicides for control of these weeds. While preplant (PP) herbicides provide excellent control of GR-horseweed, there are currently no POST herbicide control options within soybean. The objective of this study was to evaluate the efficacy of dicamba for the control of GR-horseweed when applied PP, POST, and sequentially in dicamba-resistant soybean. Dicamba applied PP at 600 g a.e. ha−1 provided 90 to 100% control of GR-horseweed 8 wk after application (WAA) across three field trials conducted in Ontario in 2011 and 2012. Similarly, sequential applications provided 91 to 100% control. This technology provides a much-needed POST option of dicamba to be applied as a rescue treatment to control weed escapes caused by late emergence or poor initial control following a PP herbicide application.
Response of Corn to Simulated Glyphosate Drift Followed by In-Crop Herbicides

Thirteen field experiments were conducted in Illinois, Indiana, Ohio, and Ontario from 2005 to 2007 to determine the effects of simulated glyphosate drift followed by in-crop applications of nicosulfuron/rimsulfuron plus dicamba/diflufenzopyr or foramsulfuron plus bromoxynil plus atrazine on nontransgenic corn injury, height, stand count, shoot dry weight, and yield. Simulated glyphosate drift at 100 and 200 g/ha, resulted in 11 to 61% visual crop injury and a 19 to 45% decrease in corn height. Simulated glyphosate drift at 200 g/ha caused a reduction in shoot dry weight by 46%, stand count by 28% and yield by 49 to 56%. Generally, simulated glyphosate drift followed by the in-crop herbicides resulted in an additive response with respect to visual crop injury, height, stand count, shoot dry weight, and yield.

We present an overview of the survey for radio emission from active stars that has been in progress for the last six years using the observatories at Fleurs, Molonglo, Parkes and Tidbinbilla. The role of complementary optical observations at the Anglo-Australian Observatory, Mount Burnett, Mount Stromlo and Siding Spring Observatories and Mount Tamborine are also outlined. We describe the different types of star that have been included in our survey and discuss some of the problems in making the radio observations.

Summary
Like many of the social sciences, sociology is a multi-paradigm science (Ritzer, 1975). Different approaches explain social phenomena at different scales (from the individual to the world system) and focus on different aspects of social reality (e.g. the distinction between the objective world and the subjective world). Another distinction is between descriptive and normative analysis. Descriptive analyses focus on explanation and understanding cause and effect relationships. Normative analyses focus on moral dimensions of issues and what we ‘ought to do’. While sociological approaches often entail elements of both of these approaches, most work tends to emphasise one or the other. Also, there is a distinction between sociological work that has further theoretical explanation as a primary goal and work that is more applied – that is, work that applies past theory and research to practical empirical problems.
In sociology, there are a variety of views about conflict, and the orientation of any given analysis depends upon the theoretical framework and objectives of the researcher. Thus, the approach that a sociologist might take regarding conflict depends on where her work is situated with regard to these different considerations. Some sociologists might focus primarily on explaining social conflict, such as someone studying the causes of a revolution, while others might focus on trying to resolve it, such as those supporting a land management planning process. In some instances sociologists might actually be interested in facilitating conflict, such as those who work to mobilise collective action among members of an oppressed group. Some sociologists might focus on the mechanisms that generate conflict (descriptive analysis) – such as the factors that might underlie a conflict over clear-cut logging, while others might focus primarily on the moral dimensions of conflict (e.g. how can gender inequality be reduced in forest-dependent communities). Some sociologists might focus primarily on ‘objective’ indicators of conflict (e.g. the size of a social protest, and its political outcomes), while others might focus on subjective dimensions, such as how conflict is socially constructed (e.g. such as perceptions about the social values that underlie the conflict).
