Probing the consistency of cosmological contours for supernova cosmology

As the scale of cosmological surveys increases, so does the complexity in the analyses. This complexity can often make it difficult to derive the underlying principles, necessitating statistically rigorous testing to ensure the results of an analysis are consistent and reasonable. This is particularly important in multi-probe cosmological analyses like those used in the Dark Energy Survey (DES) and the upcoming Legacy Survey of Space and Time, where accurate uncertainties are vital. In this paper, we present a statistically rigorous method to test the consistency of contours produced in these analyses and apply this method to the Pippin cosmological pipeline used for type Ia supernova cosmology with the DES. We make use of the Neyman construction, a frequentist methodology that leverages extensive simulations to calculate confidence intervals, to perform this consistency check. A true Neyman construction is too computationally expensive for supernova cosmology, so we develop a method for approximating a Neyman construction with far fewer simulations. We find that for a simulated dataset, the 68% contour reported by the Pippin pipeline and the 68% confidence region produced by our approximate Neyman construction differ by less than a percent near the input cosmology; however, they show more significant differences far from the input cosmology, with a maximal difference of 0.05 in $\Omega_{M}$ and 0.07 in w. This divergence is most impactful for analyses of cosmological tensions, but its impact is mitigated when combining supernovae with other cross-cutting cosmological probes, such as the cosmic microwave background.

Laser–solid interactions are highly suited as a potential source of high energy X-rays for nondestructive imaging. A bright, energetic X-ray pulse can be driven from a small source, making it ideal for high resolution X-ray radiography. By limiting the lateral dimensions of the target we are able to confine the region over which X-rays are produced, enabling imaging with enhanced resolution and contrast. Using constrained targets we demonstrate experimentally a $(20\pm 3)~\unicode[STIX]{x03BC}\text{m}$ X-ray source, improving the image quality compared to unconstrained foil targets. Modelling demonstrates that a larger sheath field envelope around the perimeter of the constrained targets increases the proportion of electron current that recirculates through the target, driving a brighter source of X-rays.

Clostridium difficile, the most common cause of hospital-associated diarrhoea in developed countries, presents major public health challenges. The high clinical and economic burden from C. difficile infection (CDI) relates to the high frequency of recurrent infections caused by either the same or different strains of C. difficile. An interval of 8 weeks after index infection is commonly used to classify recurrent CDI episodes. We assessed strains of C. difficile in a sample of patients with recurrent CDI in Western Australia from October 2011 to July 2017. The performance of different intervals between initial and subsequent episodes of CDI was investigated. Of 4612 patients with CDI, 1471 (32%) were identified with recurrence. PCR ribotyping data were available for initial and recurrent episodes for 551 patients. Relapse (recurrence with same ribotype (RT) as index episode) was found in 350 (64%) patients and reinfection (recurrence with new RT) in 201 (36%) patients. Our analysis indicates that 8- and 20-week intervals failed to adequately distinguish reinfection from relapse. In addition, living in a non-metropolitan area modified the effect of age on the risk of relapse. Where molecular epidemiological data are not available, we suggest that applying an 8-week interval to define recurrent CDI requires more consideration.

A multichannel calorimeter system is designed and constructed which is capable of delivering single-shot and broad-band spectral measurement of terahertz (THz) radiation generated in intense laser–plasma interactions. The generation mechanism of backward THz radiation (BTR) is studied by using the multichannel calorimeter system in an intense picosecond laser–solid interaction experiment. The dependence of the BTR energy and spectrum on laser energy, target thickness and pre-plasma scale length is obtained. These results indicate that coherent transition radiation is responsible for the low-frequency component (${<}$1 THz) of BTR. It is also observed that a large-scale pre-plasma primarily enhances the high-frequency component (${>}$3 THz) of BTR.

Conodonts of the Williams Island and Long Rapids Formations (Upper Devonian, Frasnian-Famennian) of the Onakawana B Drillhole, Moose River Basin, Northern Ontario, with a revision of Lower Famennian species

The upper part of cores of the Onakawana B Drillhole in the Moose River Basin in northern Ontario includes the upper part of the upper member of the Williams Island Formation (22.5 m, 16 samples), and the entire overlying Long Rapids Formation (75.1 m, 49 samples). The sequence of conodonts from the drillhole was analyzed by graphic correlation as well as conventional zonation.
The upper carbonate member of the Williams Island Formation correlates with lower Frasnian zones 2 to 5. Below this, mixed Frasnian and Famennian conodont faunas occur partly in a brecciated interval within the member and represent stratigraphic leak below the Frasnian. The lower member of the Long Rapids Formation correlates in its lowest part with Zone 5, followed by a hiatus of zones 6 to 8. This is succeeded by zones 9 and 10. Zone 11 is missing, followed by an interval that correlates with upper Frasnian zones 12 and 13 to within the lower Famennian Middle triangularis Zone. The Frasnian-Famennian boundary occurs within a narrow interval in the lower member. The Upper triangularis Zone and perhaps part of the Middle triangularis Zone are missing.
The middle member of the Long Rapids Formation correlates with the Lower to Uppermost crepida zones. A sequence from high in the rhomboidea Zone to within the Lower marginifera to perhaps slightly into the Upper marginifera Zone occurs in the upper member of the formation.
Sixteen species are described, of which seven are new: Palmatolepis angularis, P. angusta, P. mystica, P. nodosa, P. parva, Palmatolepis n. sp. A, and Mehlina? unica. Two species that affect definition and identification of the Frasnian-Famennian boundary, P. triangularis and P. ultima (=P. praetriangularis), are revised.

Hay was fed to an in vitro continuous culture of the rumen microbial population and to sheep kept in metabolism cages, and the effects of potassium chlorate addition on the rumen fermentation were studied. The compound was given for 8 days in vitro and for either 3 or 8 days in vivo.
Potassium chlorate addition in vitro (13·7 mg/g hay D.M.) depressed the production of CH4 and acetate, had little effect on propionate production and caused a small increase in the production of n-butyrate and n-valerate. The treatment also depressed cellulose digestion and the concentration of DNA in fermentor liquor, but increased the CO2:CH4 ratio in fermentor gas.
When given in vivo for 8 days at 6·7 mg/g hay D.M., potassium chlorate progressively depressed total VFA concentration in rumen fluid, had no effect on VFA molar proportions but caused a small increase in the CO2:CH4 ratio in rumen gas. When administered in vivo for 3 days at 14·4–15·3 mg/g hay D.M. the treatment increased the molar proportions of propionate and depressed those of acetate in rumen fluid without affecting total VFA concentration. There were considerable differences between animals in propionate response, and the maximum responses were generally obtained 2–5 days after dosing had ceased. Potassium chlorate addition also caused a temporary reduction in appetite with some sheep.
It was concluded that potassium chlorate was toxic to the rumen microbial population when given for 8 days, but that large doses given over 3 days could be used to increase the ratio of propionic acid relative to acetic and butyric acids produced from the rumen fermentation.

Rumen fermentation characteristics were studied using sheep fitted with rumen cannulae. Diets of 100% hay and 20% hay:80% cooked flaked maize (concentrate diet) were fed at the maintenance level of energy intake as two equal portions per day. Both the gas and liquid phases of the rumen were continuously sampled over 33-h periods.
Concentrations of O2 and N2 in rumen gas inoreased during feeding, whilst concentrations of CO2 and CH4 decreased. Thereafter the concentrations of both CO2 and CH4 rapidly increased. The CO2: CH4 ratio increased rapidly following feeding; it declined to baseline levels 2–4 h after feeding the hay diet, but with the concentrate diet the decline took longer. O2 concentration declined rapidly following feeding and was stable within the range 1–3% for long periods. At no stage was O2 absent from the rumen gas phase. H2 comprised 100–1500 μ1/1 and 100–6000 μ1/1 of the gas phase in sheep fed the hay or concentrate diets respectively and its concentration increased very rapidly with the onset of eating. CO concentration varied between 2 and 16 μ1/1 in the rumen gas of hay-fed animals and was not related to time after feeding. In concentrate-fed animals CO comprised 0—130 μ1/1 of rumen gas and increased very slowly after feeding.
Eh and rH values ranged between —150 and —260 mV and 8·0 and 5·0 units respectively for rumen contents from animals fed the two diets. A diurnal cycle was evident, with the most oxidizing state being attained just before feeding, and the most reducing state just after feeding. The diurnal cycles were better denned by rH than by Eh. The magnitude of the decrease in rH (and pH) during feeding was greater for animals fed the concentrate than the hay diet. At no stage during feeding did Eh or rH change towards more oxidizing conditions. Changes in the concentration of H2 in rumen gas were related to changes in the rH of rumen contents.
VFA molar proportions showed no changes during the 24-h cycle with hay-fed animals but showed erratic variation with concentrate-fed animals. When two sheep fed the concentrate diet were sampled daily for 21 days, it was shown that VFA molar proportions were not constant.
It was concluded that the gas phase of the rumen was never completely O2-free, and that whilst a stable rumen fermentation existed in sheep fed the hay diet the fermentation appeared to be continuously changing in the concentrate-fed sheep, and on this last mentioned diet abnormal values for rumen gas composition and VFA proportions were produced from time to time.
Mesenteric venous blood flow and the net absorption of glucose in sheep fed dried grass or ground maize-based diets

A surgical procedure is described for placing a catheter in the mesenteric vein of sheep, such that blood draining the reticulo-rumen is not sampled. This catheter, together withothers in the mesenteric vein and carotid artery, was used to determine mesenteric blood flow and the net absorption of glucose in five sheep fed both dried grass and ground maize-based diets. The mean (± S.E.) mesenteric blood flow rates were 33 ±0·9 and 37 ± 5·7ml/min/kg live weight when fed the dried grass and maize-based diets respectively. These flow rates, together with the small venousarterial differences in plasma volatile fattyacid concentrations observed, indicated that the mesenteric catheter did not sample blood draining the reticulo-rumen in the conscious, fed animal. Further confirmation was provided by subsequent post-mortem examinations of the catheters. When fed the dried-grass diet, a significant net absorption of glucose could not be detected. However, when fed the maize-based diet a net glucose absorption of 19·3 ± 0·75 mmol/h was observed.

An impermeable continuous-culture artificial rumen was constructed and rumen microbial populations were cultured in vitro for periods of up to 23 days. Diets of hay and 20:80 hay:cooked flaked maize (concentrate diet) were fed to the in vitro cultures and also to sheep, and the performance of the in vitro fermentation compared with that of the in vivo fermentation.
With the hay diet, changing the initial inoculum from strained rumen liquor to unmodified rumen contents (containing solid ingesta) reduced the time required to attain a steady-state fermentation in vitro from 13 to 4 days. VFA molar proportions and the CO2: CH4 ratio indicated that the type of fermentation taking place in vitro was remarkably close to in vivo fermentation. However, rates of cellulose digestion, CH4 production and number of bacteria/unit volume (as judged by DNA concentration) were much less in vitro than in vivo, showing that the rate of fermentation was reduced in the artificial rumen. Total VFA concentration and the osmotic pressure of fermentor liquor were slightly higher than the maximum values found in vivo, and it was concluded that these were responsible for the reduced rate of fermentation.
Regardless of the duration of culture, a satisfactory steady-state fermentation was never attained when the concentrate diet was administered. DNA concentration in fermentor liquor declined to very low levels after 4 days; CH4 production rapidly declined and was replaced by H2 production after 5 days. VFA molar proportions bore little resemblance to those found in vivo. Both the osmotic pressure of fermentor contents and total VFA concentration rapidly increased with time, and by day 3 were approximately 2·5 times the values found in vivo. It was concluded that high VFA concentration, and the associated increase in osmotic pressure, were the main factors responsible for the unrealistic in vitro fermentation, and that this problem was much greater than encountered with the hay diet due to starch being digested at a faster rate than cellulose and hemicellulose.
Methods of regulating VFA concentration in continuous culture in vitro rumen systems are discussed and some improvements suggested.
Carbohydrase activity in the pancreatic tissue and small intestine mucosa of sheep fed dried-grass or ground maize-based diets

Two groups of six sheep were fed either dried-grass or ground maize-based diets for at least 4 weeks before slaughter. Samples of the small intestinal mucosa and spancreatic tissue were assayed for a-amylase, glucoamylase, maltase and oligo-l,6-glucosidase.
The pancreatic tissue contained high activities of α-amylase and much lower activities of glucoamylase, maltase and oligo-1,6-glucosidase. There was no effect of diet on the specific activities of any of these enzymes in the pancreatic tissue.
The activity of α-amylase adsorbed on to the mucosa of the small intestine was greatest in the proximal region of the small intestine, the activity generally declining with increasing distance away from the pylorus. There was no diet effect on the absorbed α-amylase activity.
Similar patterns of distribution along the small intestine were observed for maltase, glucoamylase and oligo-1,6-glucosidase with the highest activities in t he jejunum. There was no overall effect of diet on glucoamylase or maltase specific activities and glucoamylase total activity, although the total activities of maltase and oligo-1,6-glucosidase were significantly greater for the sheep fed the ground maize-based diet (P < 0·05).
It is suggested that ruminant animals may be capable of digesting large amounts of starch in the small intestine through an adaptation in the activity of the host carbohydrases.
Rotavirus infections and climate variability in Dhaka, Bangladesh: a time-series analysis

Attempts to explain the clear seasonality of rotavirus infections have been made by relating disease incidence to climate factors; however, few studies have disentangled the effects of weather from other factors that might cause seasonality. We investigated the relationships between hospital visits for rotavirus diarrhoea and temperature, humidity and river level, in Dhaka, Bangladesh, using time-series analysis adjusting for other confounding seasonal factors. There was strong evidence for an increase in rotavirus diarrhoea at high temperatures, by 40·2% for each 1°C increase above a threshold (29°C). Relative humidity had a linear inverse relationship with the number of cases of rotavirus diarrhoea. River level, above a threshold (4·8 m), was associated with an increase in cases of rotavirus diarrhoea, by 5·5% per 10-cm river-level rise. Our findings provide evidence that factors associated with high temperature, low humidity and high river-level increase the incidence of rotavirus diarrhoea in Dhaka.

Production responses of weaned pigs after chronic exposure to airborne dust and ammonia

Nine hundred and sixty weaned pigs were exposed for 5·5 weeks to controlled concentrations of airborne dust and ammonia in a single, multi-factorial experiment. Production and health responses were measured but only the former are reported here. The treatments were a dust concentration of either 1·2, 2·7, 5·1 or 9·9 mg/m3 (inhalable fraction) and an ammonia concentration of either 0·6, 10·0, 18·8 or 37·0 p. p. m., which are representative of commercial conditions. The experiment was carried out over 2·5 years and pigs were used in eight batches, each comprising five lots of 24 pigs. Each treatment combination was replicated once and an additional control lot (nominally ≈ 0 mg/m3 dust and ≈ 0 p. p. m. ammonia) was included in each batch to provide a baseline. The dust concentration was common across the other four lots in each batch in which all four ammonia concentrations were used; thus the split-plot design was more sensitive to the effects of ammonia than dust.
The pigs were kept separately in five rooms in a purpose-built facility. The pollutants were injected continuously into the air supply. Ammonia was supplied from a pressurized cylinder and its concentration was measured with an NOx chemiluminescent gas analyser after catalytic conversion. The endogenous dust in each room was supplemented by an artificial dust, which was manufactured from food, barley straw and faeces, mixed by weight in the proportions 0·5: 0·1: 0·4. The ingredients were oven-dried, milled and mixed and this artificial dust was then resuspended in the supply air. Dust concentration was monitored continuously with a tribo-electric sensor and measured continually with an aerodynamic particle sizer and gravimetric samplers.
Live weight per pig and cumulative food intake per pen of 12 pigs were measured after 5·5 weeks of exposure. Exposure to both aerial pollutants depressed live weight relative to the control (control v. pollutant, 25·7 v. 25·0 (s.e.d. = 0·33) kg, P = 0·043) and there was a trend for food intake to be lower for pollutant-exposed pigs (control v. pollutant 292 v. 280 (s.e.d. = 7·1) kg per pen, P = 0·124). The reduction in live weight and food intake was dependent upon the concentration of dust (mean across all ammonia concentrations for increasing dust concentration; live weight 25·3, 26·4, 24·0 and 24·5 (s.e.d. = 0·65) kg, P = 0·081; food intake 295, 316, 248 and 263 (s.e.d. = 14·3) kg per pen, P = 0·016) but not ammonia (mean across all dust concentrations for increasing ammonia concentration; live weight 24·4, 25·1, 25·3 and 25·3 (s.e.d. = 0·41) kg, P = 0·158; food intake 279, 275, 288 and 279 kg (s.e.d. = 9·0) kg per pen, P = 0·520). There was an interaction between dust and ammonia for live weight (P = 0·030) but the effects were complicated and may have been the result of a type I error. There was no interaction for food intake (P = 0·210). In general, both food intake and live-weight gain, but not food conversion efficiency, were lower for weaned pigs exposed to 5·1 and 9·9 mg/m3 dust concentrations compared with 1·2 and 2·7 mg/m3 treatments. Other measures of production were also analysed and supported the overall interpretation that dust concentrations of 5·1 mg/m3 and higher depress performance.
This study is the first to quantify the effects of chronic exposure to common aerial pollutants on the performance of weaned pigs. The results suggest that dust concentrations of 5·1 or 9·9 mg/m3 (inhalable fraction) across ammonia concentrations up to 37 p.p.m. adversely affect performance. The commercial significance of these findings depends on the financial benefits of the superior production at low dust concentrations relative to the cost of providing air of this quality.
